1. Chemical Constituents from Polygonum cuspidatum

OBJECTIVE: To study the chemical constituents from Polygonum cuspidatum. METHODS: The petroleum ether-ethyl acetate solvents at different ratios of 9:1, 7:1, 5:1, 3:1 and 1:1 were used as gradient eluents for silica gel column chromatography, and samples collected were further isolated and purified by preparative high performance liquid chromatography. Their structures were elucidated by physico-chemical constants and spectroscopic methods. RESULTS: Eight compounds were obtained and identified as 22(Z)-ergosterol-4,6,8,22-tetraene-3-one(1), (22E,24R)-stigma-1,4-diene-3-one(2), (E)-ethyl octadecanoic-16-enoic acid ethyl ester(3), oleanolic acid(4), emodin(5), resveratrol(6), trans-resveratrol glycoside(7) and 6'-gallic acid-4-O-D-resveratrol ester(8).CONCLUSION: Compound 1 is a new natural compound (patent No. ZL 2013 10205435.5), compound 2 and 3 are isolated from this plant for the first time.
2.Three-dimensional conformal radiotherapy for 56 patients with liver neoplasms

Purpose:To evaluate the curative effects and side effects of 3-dimensional conformal radiotherapy (3DCRT) in the treatment of liver carcinoma.Methods:56 patients with liver carcinoma (Forty three patients were primary hepatic carcinoma and 13 liver metastatic tumors) with 62 lesions were treated with 3DCRT for 30~45 Gy/5~10 f/1~2 weeks with fractionated doses ranging from 4Gy to 8Gy.Results:In the 1~3 months after 3DCRT the results examined by CT or MRI showed that the 62 lesions(56 patients) had CR 24% (15/62),PR 50%(31/62),NC 16% (10/62) and PD 10% (6/62).The overall response rate (CR+PR) was 74%.Conclusions:3DCRT is an effective and safe treatment for hepatic malignant tumor and it can be tolerated by most of the patients.The immediate response of tumor is encouraging. The survival rate and late complications remain to be observed.
3.Effect of Ningshen Ling and dehydroepiandrosterone on the cognitive function in mice subjected to chronic mild stress

ObjectiveTo investigate the alteration of behavior,such as spontaneous movement,spatial memory ability and cholinergic M receptors in the brain of mice subjected to chronic mild stress (CMS). And to determine whether Ningshen Ling (NSL)and dehydroepiandrosterone(DHEA)could reverse the cognitive impairment in this model.MethodsForty mice were divided into four groups: control group,CMS group,CMS+NSL(p.o.5g·kg-1·d-1) group and CMS+DHEA (i.p.5 mg·kg-1·d-1) group. Morris water maze procedure was used to determine the spatial memory ability. M receptor binding activity was measured with radioactivity assay by3 H-QNB.ResultsAfter 3 weeks CMS,the mice exerted a decrease in spontaneous movement test,and the latency in Morris water maze was obviously prolonged. Treating CMS mice with NSL and DHEA for 1 weeks could improve the spontaneous movement and latency declined. Whereas the3 H-QNB binding ability to M receptor showed a significantly decrease in cerebral cortex and hippocampus of CMS mice (P<0.05),the decreased ability of M receptor binding was reversed by NSL treatment in hippocampus(P<0.05).ConclusionNSL and DHEA can alleviate the stress response and reversed cognitive impairment induced by CMS,and it may be concerned with the central cholinergic M receptors activity.

Based on their multiple years of personal experiences in hospital administration,authors of this article point out that medical service,humanistic service,and emotional service should be the core aspects in the current hospital competition.Medical professionalism embodied in the three levels of consciousness,behaviors,and regulations should be the main subject in the research on the role of medical ethics in hospital administration and medical treatment behaviors.It is an urgent ethical requirement and the chief task for hospital administrators to achieve humanistic medical service through humanistic administration of hospitals.
5.CD151 promotes proliferation and migration of PC3 cells via the formation of CD151-integrin α3/α6 complex.

Over-expression of CD151 was found to be associated with metastasis and poor prognosis of prostatic carcinoma. This study was designed to examine the mechanism by which CD151 promotes the proliferation and migration of prostatic cancer cells. The pAAV-CD151, pAAV-GFP and pAAV-CD151-AAA mutant plasmids were constructed and used to transiently transfect PC3 cells (a prostatic carcinoma 3 cell line) by the mediation of Fugene HD. Then, the cells were assigned to control group, pAAV-GFP group, pAAV-CD151 group, and pAAV-CD151-AAA group respectively. Cell proliferation was evaluated by using the 3-[4,5-dimet-hylthiazol-2-yl]-2,5, diphenyltetrazolium bromide (MTT) method. Cell migration assay was performed by using Boyden chambers. The formation of CD151-integrin α3/α6 complex was determined by the method of co-immunoprecipitation. The protein expression levels of CD151 and extracellular signal-regulated kinase (ERK) were measured by Western blotting. The results showed that transfection of pAAV-CD151 or pAAV-CD151-AAA mutant increased the expression of CD151 protein in PC3 cells. Co-immunoprecipitation showed that more CD151-integrin α3/α6 complex was formed in the pAAV-CD151 group than in the control group, the pAAV-GFP group and the pAAV-CD151-AAA mutant group. Furthermore, the proliferative and migrating capacity of PC3 cells was substantially increased in the pAAV-CD151 group but inhibited in the pAAV-CD151-AAA mutant group. CD151 transfection increased the expression of phospho-ERK. Taken together, it was concluded that CD151 promotes the proliferation and migration of PC3 cells through the formation of CD151-integrin complex and the activation of phosphorylated ERK.

7.Role of prostaglandin E2 receptors in H9c2 cardiomyocyte hypertrophy induced by prostaglandin E2

Objective To evaluate the role of prostaglandin E2 (EP) receptors in H9c2 cardiomyocyte hypertrophy induced by prostaglandin E2 (PGE2).Methods Primary cultured H9c2 cardiomyocytes were seeded in culture flasks (3 ml/flask) or in 24-well plate (1 ml/hole) or 6-well plate (2 ml/hole) with density of 4 × 104/ml.The cells were randomly divided into 4 groups (n=24 each): control group (group C),PGE2 group,AH6809 (EP1 and EP2 receptor antagonist) group (group A) and GW627368X (EP4 receptor antagonist) group (group G).The cells were continuously cultured for 48 h.PGE2 (final concentration 1 μmol/L) was added to the culture medium in PGE2 group.PGE2 (final concentration 1 μmol/L) and A H6809 (final concentration 10 μmol/L) were added to the culture medium in group A.PGE2 (final concentration 1 μmol/L) and GW627368X (final concentration 10 μmol/L) were added to the culture medium.The cells were then cultured for 48 h in groups PGE2,A and G.Then the cell morphology was observed by using fluorescent microscope.The cell diameter was measured by using the Image J medical image analysis system.Total protein content in the cells was measured with BCA method.The expression of atrial natriuretic peptide (ANP) mRNA and brain natriuretic peptide (BNP) mRNA in the cytoplasm was determined using RT-PCR.Results Compared with group C,the total protein in the cells and cell diameter were significantly increased,and the expression of ANP mRNA and BNP mRNA in the cytoplasm was up-regulated in groups PGE2,A and G (P < 0.05).Compared with group PGE2,the total protein in the cells and cell diameter were significantly decreased,and the expression of ANP mRNA and BNP mRNA in the cytoplasm was downregulated in group G (P < 0.05),and no significant change was found in the parameters mentioned above in group A (P > 0.05).Conclusion EP4 receptor mediates H9c2 cardiomyocyte hypertrophy induced by PGE2 and the effect is not related to EP1 and EP2.
8.Clinical research of rhGH on pregnancy rate of polycysticovary syndrome

Objective To investigate the effect of recombinant human growth hormone (rhGH) action on pregnancy rate of polycystic ovarian syndrome(PCOS).Methods The PCOS who were client in our hospital in February-August,2012 were investigated.The rhGH group (41 cases) were that all patients took rhGH at the same time ovulation.The control group (41 cases) were no medication group.The two groups had sex action after ovulation.Ovulation rate,pregnancy rate and endometrial score of the two groups were compared.Results Ovulation rate of rhGH group was 90.2% (37/41),the control group was 68.2% (28/41),the difference was statistically significant (x2 =3.459,P =0.036).Clinical pregnancy rate of RhGH group was 32.7% (12/37),the control group was 25.0% (7/28).The difference was statistics significance (x2 =4.319,P =0.032).Endometrial score of rhGH group was (14.96 ± 2.18),the control group was (12.77 ± 2.60) points,the difference between groups was statistically significant (t =-3.356,P =0.032).Conclusion Giving rhGH ovulation cycle specification can promote ovulation PCOS patients and improve the adverse factors affecting pregnancy,improving the pregnancy rate.

Objective: To establish a mini invasive way to evaluate endothelium function. Methods: Group 1 consisted of 10 subjects with no risk factors for coronary artery disease, group 2 consisted of 11 subjects with risk factors for coronary artery disease and angiographically normal coronary arteries.
