1.Expression of human oncostatin M in the methylotrophic yeast Pichia pastoris and its activity identification

Objective: To establish the yeast expression system for oncostatin M (OSM). Methods:The cDNA encoding human OSM was inserted into pPIC9K vector and transformed into Pichia pastoris. The correct phenotype of recombinants was screened through MD and MM auxotrophic culture medium. The OSM mRNA transcription in the P.pastoris and molecular weight were identified with Northern blot and Western blot. MTT method was used to test the activity of OSM in inhibiting the growth of human melanoma cells. Results: Established P.pastoris system could accomplish human OSM secreting expression. The recombinant OSM with a 30 000 molecular weight amounted to 70% of the total protein in medium supernatant. The OSM showed an inhibitory effect on the growth of A375 cells in vitro. Conclusions:The P.pastoris system of OSM was proved effective with a high expressing level. The secreted recombinant OSM present in supernatant of culture medium would be easy to be purified. It may lay the foundation for the large-scale preparation of OSM in future.

3.Clinical experience of antegrade radical retropublic prostatectomy in 30 patients

Objective To discuss the experience of antegrade radical retropublic prostatectomy. Methods Thirty cases of prostate cancer were treated by antegrade radical retropublie prostateeto-my. The mean age of patients was 64 years. There were 8 cases of T_1, 15 cases of T_2, and 7 cases of T_3. The surgery included following procedures: Firstly the vesical neck was divided. Then the seminal vesicle and the prostate were freed from above downward. After the urethra was separated from the prostatic apex, the proximal end of urethra was sutured to the new vesical neck. Results All the 30 laparoscopic surgeries were accomplished successfully. The mean operative time was 2.5 hours. The mean blood loss was 350 ml in the operation. No ureteral injury, rectal injury and other severe compli-cation was observed. Histopathologic study showed prostate cancer for all the cases. Four cases had positive surgical margins. All cases were followed up from 6 to 48 months (average 25 months) with-out dysuria and permanent incontinence. No death occurred. Sexual function recovered in 7/13(54%) cases after the operation. Conclusions Antegrade radical retropublic prostatectomy provides low complication, tittle bleeding and positive surgical margins. It is an effective and sale procedure to treat prostate cancer.
4.Cardiac tissue engineering scaffold material: How far is it from research to application

OBJECTIVE: To seek the bionic material myocardial extracellular matrix in the myocardial tissue engineering for the cultured myocardial cells and to provide a reliable scaffold.METHODS: A computer-based online search of CNKI and PubMed database to search articles published between January 1996 and August 2007 on scaffold material of tissue engineering.RESULTS: Natural biological scaffold materials with good biocompatibility and mechanical properties are ideal choice in the cardiac tissue engineering rapid prototyping technology. Natural biological materials such as collagen, chitosan have poor mechanical properties and plasticity, but good biocompatibility, which can be used for rapid prototyping of cardiac stents. The scaffolds which are prepared by the technology such as three-dimensional printing, laser sintering, three-dimensional pressing,selective laser sintering rapid prototyping technology on have the advantages such as high porosity, high surface area volume ratio,completely transparent among the holes, controllable macro-shape, porosity, and independent control of aperture.CONCLUSION: Through the use of dispersion/accumulation forming principle, surface modification of traditional biological materials in cardiac tissue engineering using computers and other high-tech nano-polymer technology is expected to provide ideal myocardial scaffolds.

7.The relationship of serum placental growth factor and carotid atherosclerosis in patients with type 2 diabetes mellitus

Objective To explore the relationship between serum placental growth factor (PLGF) and carotid atherosclerosis (CAS) in patients with type 2 diabetes mellitus (T2DM ). Methods Serum PLGF were determined by ELISA in 53 patients with T2DM ,who were divided into T2DM without carotid plague (T2DM group ,n= 27) and T2DM with carotid plague (T2DM + CAS group ,n= 26). Fasting plasma glucose (FPG) ,glycosylated hemoglobin A1c (HbA1c) and some other metabolic variables were also measured. The results were compared to those of 27 healthy controls (NC group). Results Serum PLGF levels were significantly higher in T2DM group and T2DM + CAS group than in NC group (P<0.01). T2DM+CAS group showed significantly higher serum levels of PLGF compared to that of T 2DM group(P<0.01).Correlation analysis showed that serum PLGF was positively correlated with TC ,LDL‐C , hsC‐RP and IL‐6 in T2DM patients (P<0.01). Similarly ,multiple regression analysis showed that LDL‐C and IL‐6 were the independent factors of serum PLGF level (P<0.05). Furthermore ,Serum PLGF was positively associated with carotid plague. Conclusion PLGF is positively related with chronic inflammation factors in T2DM patients. Serum PLGF may play an important role in the occurrence and development of carotid atherosclerosis and other macroangiopathy in T 2DM.
8.Determination of the Concentration of Isorhynchophylline in Tissue of Rats by RP-HPLC

Objective To establish a pretreatment mthod of the tissue sample of rat and a determination method of the concentration of isorhynchophylline in tissue of rats by RP-HPLC. Method The tissue sample were treated by adding methanol, and the supernatant was dried by nitrogen gas, the residue was redissolved in methanol, then detected by RP-HPLC directly. Waters? RP18 (4.6 mm?250 mm, 5 ?m) and pre-column Wondasil C18 (4.0 mm?10 mm, 5 ?m) were used as the stationary phase. Methanol-water (70∶30) added 0.03% triethylamine was taken as mobile phase with flow rate of 0.8 mL/min. The wavelength of detection was 254 nm. Results The standard curve of Isorhynchophylline concentraton in rat tissues:Y=48.558X+0.207 6 (n=6), r=0.999 6, it appears a good linear relationship within the range of 0.16~16.0 ?g/mL. The average recovery of was 99.66%, RSD was 1.56%. Conclusion This mehod is sensitive, rapid, simple, specific and reproducible.
9.Lymphocyte infusion with lymphoma and leukemia immunotherapy

Remarkable achievements have been made for lymphocyte infusion of lymphoma and leukemia, especially of lymphoproliferative disease within twenty-first century. The donor lymphocyte infusion or mobilized donor lymphocyte infusion (DLI/DSI) offer an opportunity of second remission for relapse patients post-transplantation. Cytokine-induced killer/DC cytokine-induced killer play an anti-tumor activity beyond non-MHC restricted. Cytotoxic lymphocyte infusion activated by synthetic tumor antigen produces targeted effects of anti-tumor. Transgenic CTL of anti-tumor TCR has brought the dawn in lymphoma and leukemia patients with defective lymphocytes. Maternal lymphocyte infusion play an anti-tumor/viral activity by avoiding the immune barrier of HLA mismatch.
10.Analysis the complications after trabeculectomy in 405 cases in department of ophthalmology

AIM: To analyze and summarize the effect of glaucoma trabeculectomy in the department of ophthalmology in basic hospital.
METHODS: Postoperative intraocular pressure ( IOP ) , filtering bleb and complications of 316 cases (405 eyes) of patients with glaucoma after trabeculectomy were analyzed.
RESULTS: After follow- up 12mo, 76. 5% IOP was controlled in normal level. 42. 5% was filtration bleb typeⅠ, 33. 1% typeII, 14. 6% in typeⅢand 9. 9% in typeⅣ. Intraoperative complication rate was 2. 5%, that was 31.4% at postoperative early stage ( before discharge ) , and 6. 7% at postoperative long - term ( 6mo after discharge) .
CONCLUSION:Trabeculectomy for glaucoma can better solve the problem of high IOP. It is more mature in primary hospitals, but there are still a variety of intraoperative and postoperative complications.
