3.Effect of Hyperbaric Oxygen on p38 MAPK Signal Transduction Pathway in Neuropathic Rats

Objective To investigate the effect of hyperbaric oxygen(HBO)on p38 MAPK signal pathway in neuropathic pain,and explore its mechanism. Methods The experiment was divided into two parts with thirty SD rats in each part. Each part was then randomly divided into 3 groups:sham operation group(S group),chronic constriction group(CCI group)and hyperbaric oxygen group(HBO group),with 10 rats in each group. Part one:Pain?related behavior were detected on 3 day,7 day,14 day,28 day after operation. Phosphorylative p38 was detected by Western blot method and the P2X4 receptor was detected by immunohistochemistry method on the 28th day. Part two:All the rats were treated with SB203580. Pain?related behavior were detected on 3 day,7 day,14 day,28 day after operation. P2X4 receptor was detected by immunohistochem?istry method on the 28th day. Results CCI and HBO groups were significantly lower than S group in pain?related behaviors(P<0.05). CCI group was significantly lower than that in HBO group(P<0.05). The content of phosphorylative p38 and P2X4 receptor in CCI and HBO group were increased(P<0.05),and the expression of phosphorylative p38 and P2X4 in CCI group was significantly increased than that in HBO group (P<0.05). When p38 MAPK was inhibited,pain?related behavior in HBO and CCI group increased significantly than the pain?related behavior of HBO and CCI group in Part 1(P<0.05). But the difference between group CCI and HBO was not significant(P>0.05). Though the expression of P2X4 receptor in CCI group and HBO group was significantly higher than that in S group(P<0.05),the difference between group CCI and HBO was not significant(P>0.05). Conclusion Hyperbaric oxygen therapy can affect the expression of p38 MAPK through P2X4 receptor in rats of neuropathic pain.
4.Allele frequencies of 5 short tandem repeat loci of Kashin-Beck disease patients on chromosome 12

Objective To analyze the allele frequencies of 5 short tandem repeat(STR)loci(D12S313,D12S304,D12S1640,D12S1708 and D12S1583)on chromosome 12 among Kashin-Beck disease(KBD)patients and the control population living in the area suffered from KBD.Methods Fifty KBD patient8 and 50 non-KBD patients were chosen in endemic afea of Shaanxi Province,5 STR loci on chromosome 12 were genotyped by the technology of polymerase chain reacfion(PCR)and capillary electmphoresis.The pelymorphisms of STR in these popIllations were analyzed.The allele and genotype frequencies of each STR in the corresponding groups were caleulated and compared. Results In KBD group,the 5 STR loci had 8,6,7,5 and 11 types ofalleles and 17,11,15,8 and 28 genotypes, respectively;while in the control group,the number of aUele types of 5 STR loci were 6,8,6,4 and 10,the number of genotype of those loci were 13,21,14,8 and 23,respectively The allele frequence of D12S304 locus was statiBtically significant between KBD patients and controls(P<0.05),especially for the 319 bp allele(P<0.006 25). Conclusion There is an association between D12S304 locus and KBD.The 319 bp allele might play the key role.

6.Hyperbaric Oxygen Induced Autophagy in Neuropathic Rats

Objective To investigate whether hyperbaric oxygen(HBO)affected neuropathic pain through autophagy and verify the HBO effect. Methods A total of 60 male SD rats were selected for the study and randomly divided into 4 groups:control group(C group),sham operation group (S group),chronic constriction group(CCI group)and hyperbaric oxygen group(HBO group). LC3Ⅱand P62 were determined by Western blot method and the autophagy was detected by electron microscopy on the 7th day. Results CCI and HBO groups were significantly higher than C and S group in Western blot(P<0.05). There was no difference between group C and group S(P>0.05). The expression of LC3Ⅱin HBO group was significantly higher than that in CCI group(P<0.05). And the expression of P62 in CCI group was significantly higher than that in HBO group(P<0.05). The autophagy in CCI and HBO group were significantly increased comparing to the C and S groups(P<0.05). However,the difference be?tween group C and S was not significant(P>0.05). Conclusion Hyperbaric oxygen therapy can induce autophagy for anti?neuropathic pain.
7.Sparc~(TM) sling system for the treatment of female stress urinary incontinence under local anesthesia (report of 42 cases)

Objective To introduce and evaluate the procedure of suprapubic tensionless Sparc TM sling system in treating female stress urinary incontinence (SUI). Methods A total of 42 patients (mean age,54 years) diagnosed with SUI (a mean course of 12 years) were treated by Sparc TM sling method.Of them 4 cases had detrusor instability.Under local anesthesia,this procedure took about a mean of 20 min.The urinary catheters were removed the next morning after the procedure.The patients were counseled on abstaining from heavy lifting,exercises and intercourse for 4 weeks. Results After removal of catheters,all the 42 patients successfully voided without incontinence.Detrusor instability to various degrees developed in 7 patients and was relieved by expectant treatment.All the patients were followed up for 3 to 12 months (mean,6 months).Neither recurrence of SUI nor lower urinary tract obstruction was found.No implant related complications such as local irritation,extrusion,erosion through the urethra,migration from the desired location,fistula formation and inflammation occurred. Conclusions Sparc TM sling system is a simple,safe and efficacious method of treating SUI under local anesthesia.
9. In vivo tissue distribution of hydroxyapatite nanoparticles modified with aminopropyltriethoxysilane

OBJECTIVE: To investigate the in vivo tissue distribution of hydroxyapatite nanoparticles (HANPs) and provide important information for the in vivo biosafety evaluation of HANPs. METHODS: The HANPs were modified with aminopropyltriethoxysilane (APTS) to introduce amino groups on the surface. The modified HANPs were labeled with 125I and injected into mice. A γ-counter was used to quantitatively assess the radioactivity of the tissues. The accumulation of HANPs in the tissues was expressed as the percentage injected dose per gram tissue (%ID · g-1). RESULTS: The HANPs mainly accumulated in the lung, liver, and spleen. One day post-injection, the accumulation in these tissues was over 5% ID · g-1. During 30 d after the injection, the accumulation of HANPs in the lung decreased quickly, while the accumulation in the liver and spleen reduced moderately and maintained at more than 2% ID · g-1. CONCLUSION: This study indicates that lung, liver and spleen are the main tissues for the in vivo biosafety evaluation of HANPs. KEY
