Professional development of faculty is an essential component of strategy for medical schools to construct teaching team.Development of education is dependent upon the professional quality of teaching staff.Starting from connotation of faculty's professionalism,this paper compared present status of composition of teaching staff in China with that in foreign countries,explored the potential approaches for medical schools to strengthen the construction and sustainable development of teaching staff.

3.Thioredoxin-interacting protein: a new potential target for diabetes and related vascular complications therapy.

Thioredoxin-interacting protein (TXNIP), also known as vitamin D3-up-regulated protein (VDUP1), is an endogenous inhibitor of thioredoxin (Trx), which regulates the cellular reduction-oxidation (redox) state. TXNIP regulates cellular survival, apoptosis and inflammation induced by glucotoxicity, heat shock and mechanical pressure. The above functions of TXNIP are regulated by carbohydrate response element binding protein (ChREBP) and AMP-dependent protein kinase (AMPK). In recent years, numerous studies showed that TXNIP is involved in diabetes and diabetic complications. On the one hand, TXNIP functions in diabetes by increasing insulin resistance and hepatic gluconeogenesis. TXNIP expression is induced by high glucose, which is implicated in pancreatic beta cell glucotoxicity and endothelial cells dysfunction. TXNIP may contribute to the development and progression of diabetes and its vascular complications. TXNIP may be a new target for diabetes and its vascular complications therapy.

4.Research on some sero-enzymes measurement by applying three different vacuum tubes for venous blood samples

Objective To discuss the comparability of ALT, AST, CK and LDH results by ap- plying three different vacuum tubes (no additive tubes, separation gel coagulation promoting tubes and other coagulation promoting tubes) for venous blood samples, and to provide evidence for clinical tests. Methods Fast venous blood from 26 healthy blood donors were collected in test tubes without additives, separation gel coagulation promoting tubes and other coagulation promoting tubes. Olympus AU5400 Biochemical Analyzer was applied to examining serum ALT, AST, CK and LDH for all pre- pared serum samples. Results There was no statistical difference of serum ALT, AST, CK and LDH level collected in test tubes without additives, separation gel coagulation promoting tubes and other co- agulation promoting tubes. With the no additive tubes as the standard tubes, the above result bias be- tween separation gel coagulation promoting tubes and other coagulation promoting tubes were clinically acceptable. Conclusion There is credible comparability of serum ALT, AST, CK and LDH level a- mong different kinds of venous blood samples collected in test tubes without additives, separation gel coagulation promoting tubes and other coagulation promoting tubes. The separation gel coagulation promoting tubes and other coagulation promoting tubes can substitute test tubes without additives for ALT, AsT, CK and LDH determination.
6.Determination of serum prealbumin and C-reactive protein in 68 burn patients and its clinical significance

Objective To investigate the change of serum prealbumin (PA) and C-reactive protein (CRP) in burn patients and its clinical significance.Methods Sixty eight cases of burn patients were divided into two groups,group Ⅰ [n=38,with total burn surface area less than 15% of total body surface area (TBSA)] and groupⅡ(n=30,with total burn surface area more than 50% of TBSA). Thirty healthy people were set as healthy control group. Particle enhanced immunoturbidimetic assay was applied to detecting the serum levels of PA and CRP for all subjects on the day 1 after burns.Results The CRP level was much higher and serum level of PA was much lower in group Ⅱ than that of group Ⅰ,respectively. There existed significant negative correlation between PA and CRP level.Conclusion The level of serum PA decreases significantly along with total burn surface area expansion. PA can be used as a sensitive marker for estimation of inflammation degree after burns.
7. Effect of phosphatase activity of PTEN on migration ability of human breast cancer cell line ZR-75-1

Objective: This study was to explore the effects of phosphatase activity of PTEN (phosphatase and tensin homolog deleted on chromosome 10) on migration ability of human breast cancer ZR-75-1 cells. Methods: Three kinds of plasmids, wt-PTEN plasmid, G129R-PTEN plasmid, and G129E-PTEN plasmid, were constructed and transfected into human breast cancer cell line ZR-75-1, with a deletion of PTEN gene, via liposome mediation. The expression of PTEN protein as well as the Tyr397 in phospho-focal adhesion kinase (p397.FAK) was measured by Western blotting. The effect of phosphatase activity of PTEN was observed in a cell scratch wound model in vitro. The inhibitory effects of phosphatase activity of PTEN were measured by using cell-matrix adhesion test and reconstituted basement membrane invasion technique. The expression level of MMP-2 was determined by immunohistochemical staining. Results: Three kinds of plasmids (wt-PTEN, G129R-PTEN, and G129E-PTEN) were successfully transfected into ZR-75-1 cells, respectively. All the three stably transfected cells had PTEN protein expression. Transfection of wt-PTEN and G129R-PTEN inhibited the migration of ZR-75-1 cells. There was significant difference in the inhibitory degree of cell adhesion and invasion between wt-PTEN or G129E-PTEN groups and G129R-PTEN transfected or non transfected groups (P < 0.01). The level of p397.FAK was significantly lower in wt-PTEN and G129E-PTEN groups compared with G129R-PTEN transfected groups. There was significant difference in the expression level of MMP-2 between G129R-PTEN-transfected and non transfected groups(P < 0.01). Conclusion: Both wild-type PTEN with dual specific phosphatase activities and G129E-PTEN with only protein phosphatase activity have inhibitory effects on migration of human breast cancer cell line ZR-75-1. However G129R-PTEN without phosphatase activity have no effects.

10.Association of general level of total anti-oxidation capacity, superoxide dismutase, nitric oxide synthase activity in serum with the lifestyle of residents of Bin County of Heilongjiang Province

BACKGROUND: When the dynamic equilibrium between oxidation system and anti-oxidation system in the body is upset, the overly produced active oxygen-derived free radicals will attack the target organs, thus resulting in the damage to organs and big molecules, and then diseases. Total anti-oxidation capacity (T-AOC), superoxide dismutase (SOD), and nitric oxide synthase (NOS) activity are the majorindexes for the defense system of the body, and their status is closely related to diseases. OBJECTIVE:To explore the level of T-AOC, SOD and NOS in serum,and find out its relationship with lifestyle. DESIGN:Single sample investigation. SETTING:Department of Health Laboratory Technology, College of Public Health of Harbin Medical University. PARTICIPANTS:Totally 531 residents of Bin County aged 20 to 70years were recruited between September 2000 and May2001. MATERIALS:The reagents kits of T-AOC, SOD and NOS were manufactured by Nanjing Jiancheng Bio-engineering Institute. METHODS:The investigators surveyed the residents with the same questionnaire. Questionnaire was made and the investigators were trained in advance. The questionnaire was filled in item by item as required. Questionnaire items consisted of the general situation, financial status, dietary habit, hobby, status of health care, and mental factor. Finally 95 investigation indexes were completed. The activity of serum T-AOC, SOD and NOS of the 531 residents in Bin County was determined, respectively, with reagent kits produced by Nanjing Jiancheng Bio-engineering Institute. The lifestyle factors were analyzed by multivariate stepwise regression analysis,and informed consent was obtained from the participants. MAIN OUTCOME MEASURFS:① Serum T-AOC, SOD and NOS in the participants; ② lifestyle factors affecting T-AOC, SOD and NOS.RFSULTS:According to actual analysis, the 531 participants entered the results analysis. T-AOC was measured in 489 participants, SOD in 525participants, and NOS in 531 participants. ① Indexes of the participants:T-AOC of the males was obviously higher than that of the females [(20.01±7.21), (15.25±6.22) kU/L, P < 0.05]. SOD of the males was slightly lower than that of the females while NOS of the males was slightly higher than that of the females, but there was no significant difference. ②Multivariate stepwise regression analysis showed that risk factors related to T-AOC were gender difference (OR=2.188), educational level (OR=1.859),and the presence of rheumatism, respectively (OR =1.142). SOD-related risk factors were educational level (OR=1.584), years of spirit drinking (OR =1.048), presence of nephritis (OR=1.093), and irradiation (OR=1.770);frequency of tea drinking was a beneficial factor (OR=0.800). NOS-related risk factors were the average amount of cigarette smoking (OR=1.194) and the times of weekly spirit drinking (OR=1.368). However, the beneficial factor that affected serum NOS was the frequency of weekly mutton eating (OR=0.458) CONCLUSION:This experiment revealed that the ability to clean free radical in the 531 subjects was good and that it was better in males than in females. The increased educational level and presence of rheumatism can decrease T-AOC. Drinking, smoking and irradiation decrease SOD activity while the frequency of tea drinking and mutton eating are beneficial factors.
