There are some common analysis challenges in the hormone detection in agriculture science, including matrix interference, complicated sample preparation, poor reproducibility, trace analyte content. An automated on-line SPE and innovative fast polarity switch analysis method employing dual-gradient liquid chromatography ( DGLC ) coupled with tandem mass spectrometry ( DGLC-MS/MS ) was established and validated for the simultaneous determination of gibberellic acid ( GA3 ) , indole acetic acid ( IAA ) , zeatin ( ZT) and abscisic acid ( ABA) in the soybean plant ( leaf, grain and pod) . The method was applied in the actual sample detection successfully. In order to acquire higher sensitivity, recovery, stability and precision, some conditions including SPE column, analytical column, mobile phase, additive etc were optimized according to the selection and retain of hormone. Beans were cryogenically grinded by liquid nitrogen, extracted by 80% methanol, certrifugatel and dilluted with water, and then injected directly. Samples were transported and gradient eluted on the analytical column Acclaim PA2 by 0 . 1% formic acid in water and methanol, after retaining and separation on the SPE column Hypersep Retain AX. All analytes were detected in selection reaction monitoring ( SRM) mode in both positive and negative channels. The quantification was based on linear regression. The linear ranges of GA3, IAA and ZT were 0. 1-50 μg/L with the LOQ of 0. 0002 μg/g, and the linear of ABA was 0. 5-50 μg/L with the LOQ of 0. 0010μg/g. The recoveries of four kinds of plants hormones were 76 . 1%-93 . 5%, and RSDs were 0 . 82%-6 . 02% at low ( 0 . 8 μg/L ) , medium (4. 0μg/L) and high (40μg/L). The results noted that the content of ABA in seeds was apparently higher than others. This method could be used for the rapid and accurate detection of hormone in different parts of soya beans.

Objective:To detect the expression of Shank1 protein in renal cell carcinoma (RCC), to investigate its difference between the tumor and carcinoma adjacent tissue, and to analyze its correlation with RCC clinicopathological characteristics and prognosis. Methods:The renal carcinoma and carcinoma adjacent tissues of 120 patients were selected from Cangzhou Central Hospital and Ji'nan Central Hospital from May 2008 to December 2014. The expression level of Shank1 was determined by immunohistochemistry and Western blot. Statistical analysis was performed to determine the relationship between the expression of Shank1 and the clinicopathological features of RCC patients. Results:Results of immunohistochemistry showed that the expression level of Shank1 in renal cancer tissue was significantly higher than that in carcinoma adjacent tissue, and the difference was statistically significant (P<0.05). Western blot results showed that the expression level of Shank1 protein in renal cancer tissue was also significantly higher than in carcinoma adjacent tissue. Correlation analysis found that the high expression level of Shank1 in renal cancer tissue was not significantly related to gender, age, tumor size, and TNM stage, but was significantly associated with the histological differentiation of RCC (P<0.05). Conclusion:Shank1 is abnormal y expressed in RCC renal cancer tissues and is correlated with the histological differentiation of RCC.

Objective To investigate the expression of a proliferation-inducing ligand (APRIL) in urothelial carcinoma tissue and its clinical significance. Methods We investigated the expression of APRIL mR-NA in urothelial carcinoma tissue of 33 patients in urothelial carcinoma in Cangzhou Central Hospital from July 2013 to March 2015, Among of them, 23 cases of adjacent pericancerous tissues were detected by qRT-PCR. Immunohistochemistry and western blot were used to detect the APRIL protein expression level in urothelial carci-noma tissue and adjacent pericancerous tissues. We analyzed the relationship between APRIL expression and clinical pathology in patients with urothelial carcinoma by statistical methods. Results The qRT-PCR revealed that the expression of APRIL in urothelial carcinoma tissue was significantly higher than that in the adjacent peri-cancerous tissues (P < 0.05). Immunohistochemistry and western blot showed that the expression of APRIL in urothelial carcinoma tissue was higher than that in the adjacent pericancerous tissues. Correlation analysis found that the high expression of APRIL in urothelial carcinoma tissue was not significantly related to gender , age , size of tumor, lymph node metastasizing or not, and clinical stages (P > 0.05). Conclusion The APRIL is a high abnormal expression in urothelial carcinoma tissues , and maybe related to the occurrence and development of urothelial carcinoma.

The occurrence and development of malignant tumors is closely related to the tumor microenvironment. Tumor-associated neutrophils (TAN) are important inflammatory cells in the tumor microenvironment. Studies have confirmed that TAN plays an important role in tumor growth, invasion, metastasis, pathological angiogenesis, and immunity escapes. TAN can be activated as an anti-tumor N1 type and a tumor-promoting N2 type. This article reviews the biological characteristics of TAN and the role of TAN in the malignant tumors and its mechanism.
9.Investigation on family caregiver preparation of cognitive impairment patients after leaving ICU and its influence factors
Chinese Journal of Practical Nursing 2020;36(24):1892-1897
Objective:To investigate family caregiver preparation of cognitive impairment patients after leaving ICU and analyze the influencing factors. Methods:A total of 169 family caregivers of cognitive impairment patients after leaving ICU who met the inclusion criteria were selected as subjects. The General Information Questionnaire, Care Preparedness Scale and Health Promoting Lifestyle Profile Ⅱwere used to investigate the family caregivers. Multiple linear regression was used to explore the influencing factors of patient caregiver preparation. Results:The score of family caregiver preparation for cognitive impairment patients after leaving ICU was 12.58±4.57, with the highest physiological demand preparation score (2.18±0.63) and the lowest nursing care preparation score (1.07±0.51). The results of multiple linear regression analysis showed that the patient's gender, Mini-Mental State Examination score, caregiver gender, education level, experience of care and the presence or absence of a co-career were the influencing factors of family caregiver preparation, which explained 41.6% of the variation of family caregiver preparation. Conclusions:The family caregiver preparation of cognitive impairment patients after leaving ICU is at a low level. The caregiver preparation is affected by many factors. Improving the caregiver health promotion lifestyle will help improve caregiver preparation.
