1.Effect of sevoflurane preconditioning on autophagy during ischemia-reperfusion injury to isolated rat hearts and the role of PI3K/Akt signaling pathway

Objective To evaluate the effect of sevoflurane preconditioning on autophagy during ischemiareperfusion (I/R) injury to isolated rat hearts and the role of PI3K/Akt signaling pathway.Methods Healthy adult male Wistar rats,aged 6-8 weeks,weighing 250-280 g,were anesthetized.Their hearts were excised and perfused in a Langendorff apparatus.Eighty-four isolated rat hearts with I/R injury were randomly divided into 7 groups (n =12 each):normal control group (NC group),I/R group,sevoflurane preconditioning group (S + I/R group),normal control plus wortmannin group (NC + W group),I/R plus wortmannin group (I/R + W group),sevoflurane preconditioning plus wortmannin group (S + I/R + W group),and solvent group (S + I/R + D group).In NC group,the hearts were continuously perfused with K-H solution for 180 min.In I/R group,the hearts were perfused with K-H solution for 30 min,and then subjected to ischemia for 30 min followed by 120 min of reperfusion.In S+ I/R group,the hearts were perfused with K-H solution for 10 min,and then with K-H solution saturated with 2.5% sevoflurane for 15 min,followed by 5 min washout with K-H solution before ischemia.In NC + W group,wortmannin (PI3K inhibitor) 15 μg/kg was injected intraperitoneally at 30 min before chest opening,and the other procedures were similar to those previously described in group NC.In I/R + W group,wortmarnin 15 μg/kg was injected intraperitoneally at 30 min before chest opening,and the other procedures were similar to those previously described in group I/R.In S + I/R + W group,wortmannin 15 μg/kg was injected intraperitoneally at 30 min before chest opening,and the other procedures were similar to those previously described in S + I/R group.In S + I/R + D group,dimethyl sulfoxide 1.5 ml/kg was injected intraperitoneally at 30 min before chest opening,and the other procedures were similar to those previously described in group S + I/R.The HR,± dp/dtmax,left ventricular developed pressure (LVDP) and left ventricular end-diastolic pressure (LVEDP) were recorded at the end of equilibration and reperfusion.At the end of reperfusion,coronary effluent was collected to detect lactate dehydrogenase (LDH) activity,and myocardial specimens were obtained to calculate the percentage of myocardial infract size.The expression of autophagy marker LC3-Ⅱ and Akt,phosphor-Akt (p-Akt),mammalian target of rapamycin (mTOR),and phosphor-mTOR (p-mTOR) was determined by Western blot.Results Compared with NC group,no significant change was found in the parameters of hemodynamics in NC + W group,and HR,± dp/dtmax and LVDP were significantly decreased,LVEDP,myocardial infract size,and LDH activity were increased,LC3-Ⅱ expression was up-regulated,and the expression of p-Akt and p-mTOR was down-regulated in the other groups.Compared with group I/R,HR,± dp/dtmax,and LVDP were significantly increased,LVEDP,myocardial infract size,and LDH activity were decreased,LC3-Ⅱ expression was downregulated,and the expression of p-Akt and p-mTOR was up-regulated in S + I/R and S + I/R + D groups,and no significant change was found in each parameter in S+ I/R+ W group.Compared with S + I/R group,HR,± dp/ dtmax and LVDP were significantly decreased,LVEDP,myocardial infract size and LDH activity were increased,LC3-Ⅱ expression was up-regulated,and the expression of p-Akt and p-mTOR was down-regulated in S + I/R + W group,and no significant change was found in each parameter in S + I/R + D group.Conclusion Sevoflurane preconditioning can decrease autophagy of myocardial cells during I/R through activating PI3K/Akt signaling pathway and enhancing mTOR activity in the downstream,thus protecting isolated rat hearts against I/R injury.
2.Effect of hypoxia-inducible factor-1α on proliferation and invasion of prostate cancer PC-3 cell in hypoxic situation

ObjectiveWE transfected the recombinant expression plasmid of pcDNA3. 1-HIF-1α into the prostate cancer cells, to research the effect of HIF-1α on proliferation of prostate cancer cell PC-3.MethodsWe selected a stable expression cell line with G418 were selected by transfection of the recombinant expression plasmid of pcDNA3. 1-HIF-1α into the prostate cancer. The protein and mRNA expression of HIF-1α was assayed by western - blot and RT-PCR. The cells growth curves were described by MTT and the ability of invasion was assayed by Transwell.ResultsThe expression of HIF-1α mRNA was not obviously increased compared to the untransfected prostate cancer cell by RT-PCR, but the expression of HIF-1α protein was up-regulated by western-blot after the recombinant expression plasmid transfected into PC-3. The ability of cell proliferation and invasion was significantly enhanced by MTT and Transwell assays.ConclusionThe stable expression cell model of HIF-1α was successfully constructed, which enhanced the proliferation and invasion of prostate cancer cell PC-3.
3.Designing and applying study of a new iono-chamber

Objective To design a new X-ray sensor of iono-chamber type to achieve automatic exposure control(AEC) of the X-ray machine. Methods We designed the chamber acording to the ionization theory and the requirements of automatic exposure control. Results (1) The device had double-negative poles, so the distance between positive and negative poles was 1/2 shorter and its test-volume was equal to odd-negative pole. (2)It had good property of electromagnetism-resistance because of the double shielded design by double-negative poles and screen-rings.(3) Iono-chamber was made of low-density material, so it had low absorb-ratio of X-ray and could not form image on film. Conclusion The property of the device was steady and reliable. It had applying and spreading value.
4.Testing and analysing the characteristics of new double three-field iono-chamber

Objective To test and analyse the characteristics of double three field iono chamber developed by ourselves. Methods We tested the time characteristic, dose rate characteristic and X ray quality characteristic by using direct test method, and current characteristic, voltage characteristic and thickness characteristic of double three field iono chamber by using photograph test method with NERO 6000B test meter, RF 500 mA X ray equipment and so on. The results were analysed. Results The time characteristic and dose rate characteristic had satisfied linear scales(the return coefficients of linety scales were R =0.999 61 and R = 0.998 88 respectively). The X ray quality characteristic was steady (the maximum output variation was ? max ≤?5%). The good characteristics of current, voltage and thickness were found(the maximum variations of film density were ?D max ≤?0.01, ?D max ≤?0.15 and ?D max ≤?0.06 respectively). The excellent repeat was showed (repeating error of exposure time and dose were ?T max ≤?0.004 s and ?X max ≤0.01?10 -5 C/kg). Conclusion The performace of the new Iono chamber is excellent and steady, so it has practical value.

group was also higher than that in the control group, Z=2.39, P<0.05,all which had significant difference, P<0.05.While no significant difference was found in the scores of the Geriatric Depression Scale or the Cornell Scale of Depression Disease between two groups, Z=1.21, P>0.05. Conclusions Reminiscence therapy improves cognitive function, the ability of self-care and social communicate of patients with mild to moderate dementia patients,so that it can improve further patients′quality of life.
6.One-stage total knee arthroplasty for knee osteoarthritis combined with ipsilateral femoral extra-articular deformity:individual design

BACKGROUND:Total knee arthroplasty for knee osteoarthritis accompanied with extra-articular deformity is stil chal enging. An individual replacement plan should be designed to obtain good mechanical axis and to recover suitable soft-tissue balance. OBJECTIVE:To observe the safety and feasibility of individual y designed one-stage total knee arthroplasty for knee osteoarthritis combined with ipsilateral femoral extra-articular deformity. METHODS:A total of eight patients with knee osteoarthritis combined with ipsilateral femoral extra-articular deformity were enrol ed in this study. According to preoperative plan, five patients underwent intra-articular compensatory osteotomy, and the remaining three received extra-articular corrective osteotomy. Multi-planar deformities as wel as soft-tissue conditions were recorded. Main outcome measures included the hospital for special surgery score, Western Ontario and McMaster Universities Arthritis Index score and alignments deviation. RESULTS AND CONCLUSION:The patients were averagely fol owed up for 29 months. The hospital for special surgery score changed from preoperative 32.5 points to postoperative 87.5 points. The Western Ontario and McMaster Universities Arthritis Index score was increased from preoperative 37.1 points to postoperative 88.8 points (P<0.05). Mean mechanical axis deviation (either varus or valgus) changed from preoperative 17.1° to postoperative 1.4° (P<0.05). Al cases with extra-articular corrective osteotomy healed within 3 months, and none had postoperative infection, loosening or venous thromboembolic events. One patient was with 5° residual knee anti-sheets. Results verified that one-stage total knee arthroplasty procedures are effective in the treatment of knee osteoarthritis accompanied with ipsilateral femoral extra-articular deformity in accordance with individual preoperative plan.
7.Combined interventional therapy for malignant vena cava obstruction

Objective To evaluate the effectiveness of combined treatment with thrombolysis,PTA,and endovascular stent placement for malignant obstruction of vena cava.Methods The study included 29 patients with malignant obstruction of vena cava(superior,8 patients;inferior,21 patients).The patients underwent treatment of PTA and endovascular stent placement(for patients with complete vena cava obstruction or patients with incompletely expanding stent),or endovascular stent placement(for patients with incomplete vena cava obstruction),or endovascular stenting combined with thrombolysis(for patients with thrombosis in vena cava),respectively.Results A total of 29 stents was implanted,with a success rate of stent placement of 100%.Symptoms of SVC obstruction completely disappeared in the 8 patients.Scores of patients' IVC syndrome were declined from 4~5(median,4.2) preoperatively to 0~2(median,0.3) postoperatively,with statistically significant difference(Z=-55.245,P=0.000).Follow-up checkups for 1~20 months(mean,6.2 months) in the 29 patients found patent vena cava.There were no serious complications except for vena cava laceration in 1 patient and stent displacing to the right atrium on the third day after therapy in 1 patient.Conclusions The combined treatment of thrombolysis,PTA,and endovascular stenting is safe and effective in the treatment of vena cava obstruction.
8.MR IMAGING OF THE PANCREAS

Objective To evaluate the ability of showing the pancreatic diseases on various MRI sequences. Methods Eighty-four subjects included 50 normal individuals and 34 patients(22 patients investigated for pancreatic neoplasia and 12 pancreatitis) were presented. The MR protocol included conventional SE T1WI, FSE T2WI, Pre-and post-contrast T1-weighted fat-suppressed and GRE imaging. Results The best diagnostic information was provided by T1-weighted fatsuppressed imaging before and after gadolinium enhancement on 27 of 34 cases with abnormal pan creas, followed by immediately postcontrast GRE images. Precontrast GRE imaging better showed the features of a cute pancreatitis. FSE T2WI obviously exhibited islet cell tumor and metastases of liver from pancreatic adenocarcino ma. Conclusion The standard MR protocol included T1-weighted fat-suppressed image and dynamic GRE imaging.
9.Experimental study of renal damage assessment after injection of iodine contrast medium: the role of blood oxygen level-dependent functional MRI with a 3.0 T system

Objective To evaluate blood oxygen level-dependent (BOLD) MRI on assessing renal damage after injection of iodine contrast medium with a 3.0 T system. Methods Routine MRI examination,including T1WI and T2WI, and BOLD MRI were performed in 29 SD rats with a 3.0 T system before the injection of iodine contrast agent and 20 min, 24 h, 48 h, 72 h after the injection, respectively. T2 * and R2 * ( = 1/T2 * ) measurements were obtained in the cortex, inner and outer medulla of kidney,respectively. The results obtained before contrast agent administration were considered as the self-controls.AVONA test were used for the comparison of R2 * values in different parts of both kidneys before contrast agent administration. Two-sample t test was used to compare R2 * values before and at each time point after contrast agent administration, and R2 * values in different parts of the kidneys. Results Before contrast agentadministration, R2 * values in outer medulla in both sides of kidney [R2 *left oM = (31.76 ± 2.73 ) / s,R2 * right OM = (32.77±3.07) /s] were higher than those in cortex [R2 * left c=(30.20±3.48) /s,R2 * right c = (28.84 ±3.11 )/s] and in inner medulla [ R2 * letf IM = (29.54 ± 2.42) /s, R2 * right IM =(28.37±2.80) /s ] ( F = 3.357 and 14.961, P< 0.05 ). There was no statistical significance in R2 *values in the three parts between left and right kidney, including cortex, outer and inner medulla (P>0.05). After contrast agent administration, R2* values in outer medulla changed obviously, which reached to the peak values at 20 minutes after contrast agent administration [R2* left OM = ( 43.57 ± 3.84 )/s,R2*right OM= (44.58±3.13) /s] and dropped from 24 hours [R2*left OM = (42.07 ±4.82) /s,R2* right OM = (42.89±3.40) /s]. R2* values in inner medulla and cortex only presented slight changes.Conclusion R2* values reflected the changes of oxygen content in renal cortex and medulla quantitatively,which helped for detecting medullar ischemia and hypoxia. BOLD MR imaging could offer a feasible method for evaluating oxygen metabolism and renal injury in cortex and medulla.
10.Role of phosphatidyl-inositol 3-kinase-Akt signal pathway in attenuation of ischemia-reperfusion injury by sevoflurane preconditioning in isolated rat hearts

Objective To investigate the role of phosphatidyl-inositol 3-kinase-Akt (PI3k-Akt) signal pathway in the attenuation of ischemia-reperfusion (I/R) injury by sevoflurane preconditioning in isolated rat hearts. Methods Ninety-six adult male SD rats weighing 220-280 g were randomly divided into 6 groups ( n = 16 each): sham operation group (group S); I/R group; sevoflurane preconditioning group (group SP); wortmannin group (group W); dimethyl sulfoxide (DMSO) group (group D) and sevoflurane preconditioning + wortmannin group (group SW) . Their hearts were excised and perfused in a Langendorff apparatus with K-H solution saturated with 95%O2-5%C02 at 37 ℃ . The hearts were continuously perfused for 180 min in group S. After 15 min of equilibration, the isolated hearts were subjected to 30 min of ischemia followed by 120 min of reperfusion in SP, W, D and SW groups. Croups SP, W, D and SW received 10 min of perfusion with K-H solution containing 2. 4% sevoflurane, 100 nmol/L wortmannin, 20 μmol/L DMSO, and 2.4% sevoflurane + 100 nmol/L wortmannin, respectively, followed by 5 min washout before I/R. Eight hearts in each group were selected and HR, left ventricular end-diabetic pressure (LVEDP), left ventricular developed pressure (LVDP), and ± dp/dtmax were recorded at the end of equilibration and at 15 min of reperfusion, Myocardial tissues were obtained at 15 min of reperfusion for determination of apoptosis (by TUNEL) and phosphorylated Akt (p-Akt) expression (by Western blot) . Another 8 hearts were selected at 120 min of reperfusion for determination of myocardial infarct size by TTC staining. Result Compared with group S, LVDP and ± dp/dt,^ were significantly decreased and LVEDP was significantly increased in groups I/R, SP, W, D and SW, and myocardial p-Akt expression was up-regulated in groups I/R, SP and D ( P < 0.05). Compared with group I/R, LVDP and ± dp/dtmax were significantly increased, LVEDP and apoptosis index were significantly decreased, myocardial p-Akt expression was up-regulated, and myocardial infarct size was significantly reduced in group SP (P <0.05) . Conclusion Activation of PI3K-Akt signal pathway is involved in the attenuation of I/R injury by sevoflurane reconditioning in isolated rat hearts.
