1.Legal issues of human subjects protection and ethics review pertaining to vulnerable groups

The ethics review for clinical trials pertaining to human body highly emphasizes human subjects protection.This study discussed how to balance the relationship between the rights,safety and health of the subjects and interests of the science and the community,how to review the damage compensation provisions and insurance provisions in the informed consent,how to scientifically define the human subject biomedical trials involving vulnerable groups,and how to conduct trials on vulnerable groups,as well as other sensitive issues in ethics review.
2.Effect of intrathecal gastrodin on skin cancer pain in mice

Objective To investigate the effect of intrathecal gastrodin on skin cancer pain in mice.Methods Thirty-two female Balb/c mice,aged 8 weeks,weighing 20-25 g,were randomly divided into 4 groups (n=8 each) using a random number table:sham operation group (group S),skin cancer pain group (group SCP),gastrodin group (group G),and artificial cerebrospinal fluid (ACSF) control group (group ASCF).Skin cancer pain was produced by injecting phosphate buffer solution 20 μl containing about 2 ×105 4T1 breast cancer cells into the plantar surface of the left hindpaw.At 14th day after inoculation of cancer cells,ASCF 5 μl was injected intrathecally in S and ACSF groups,and gastrodin 150 μg/kg (5 μl) was injected intrathecally in group G.Before inoculation,at 30 min before intrathecal injection,and at 15,30,60,90,120 and 150 min after intrathecal injection,the thermal paw withdrawal latency (TWL) was measured.The expression of acid-sensing ion channel (ASIC)-3 mRNA in the spinal dorsal horn was detected using the real-time reverse transcriptase polymerase chain reaction after the last measurement of the pain threshold.Results Compared with group S,the TWL was significantly decreased at each time point before and after intrathecal injection in SCP,ACSF and G groups,and the expression of ASIC-3 mRNA in the spinal dorsal horn was significantly down-regulated in group G (P<0.05).Compared with group SCP,the TWL was significantly increased at each time point after intrathecal injection,and the expression of ASIC-3 mRNA in the spinal dorsal horn was significantly down-regulated in group G (P<0.05),and no significant change was found in the parameters mentioned above in group ACSF (P>0.05).Conclusion Intrathecal gastrodin can reduce skin cancer pain and down-regulate ASIC-3 expression in the spinal dorsal horn which is helpful in maintaining the analgesic effect in mice.

Objective To study the injury of rat cardiomyocytes exposed to nonionizing radiation and its mechanism. Methods Primarily cultured cardiomyocytes were irradiated by 9 GHz 950 mW/cm2 microwave pulse (MWP)and 0~100 MHz 6?104 V/m electromagnetic pulse (EMP)for 72 h respectively ,then a series of apparatus including atom force microscope,laser scanning confocal microscope and flow cytometer were used to detect the changes of cell membrane conformation,structure and function. 5.Quality Analysis of Different Specifications of Frutus Aurantii Immaturus

Objective To study the quality standard of different specifications of Frutus Aurantii Immaturus. Methods The contents of synephrine, volatile oil, total ash, acid- insoluble ash and water in different specifications of Frutus Aurantii Immaturus were determined by HPLC or other methods according the china pharmacopoeia. Results The contents of volatile oil and synephrine in different specifications of Frutus Aurantii Immaturus were obviously different, and in accordance with empirically traditional classification. The contents of the total ash, acid- insoluble ash and water were similar in the different specifications and the total ash content was lower than 7 % which was in accordance with that recorded in China Pharmacopoeia.Conclusion Volatile oil, acid- insoluble ash, water and synephrine with a definite content limit can be chosen as the parameters for the research of quality standard of Frutus Aurantii Immaturus, which will supply evidence for the medicinal trade and research of Frutus Aurantii Immaturus.

Background The impact of statins on inflammation are independent of cholesterol-lowering effect.Recent studies showed that Toll-like receptor 4(TLR4),a mediator of innate immune responses,is involved in the initiation and progression of atherosclerosis.Objective To investigate the effects of atorvastatin on LPS-induced TLR4 expression and downstream signals and to explore the molecular mechanisms of anti-inflammation by statins.Methods Human umbilical vein endothelial cells(HUVEC)were pretreated with atorvastatin(1 or 10 ?mol/L)or NF-?B inhibitor CAPE for 30 min,then incubated by purified LPS for 24 hours.TLR4,ICAM-1 and E-selectin mRNA were measured by RT-PCR;the percentage of TLR4 positive cells were detected by flow cytometry.The activation of NF-?B(p65)were detected by Western blot.Results Atorvastatin(1-10 ?mol/L)prevented LPS-induced increases in TLR4,ICAM-1 and E-selectin expression [TLR4 mRNA(1.24?0.21)vs LPS(1.82?0.27),P
7.Analysis on results of neonatal ABO blood group reciprocal stereotypy and crossmatching test

Objective To analyze the coincidence situation of the results of neonatal ABO blood group reciprocal stereotypy with crossmatching test of allotype blood and to investigating the limitations of cross-matching test in infant blood transfusion and effec-tive measures for ensuring the neonatal safe blood transfusion.Methods The micro-column gel test was adopted to identify the ABO blood group and conduct the crosshatching test of allotype blood in 1 095 cases of neonatal blood samples.Results Among the 1 095 samples,the detected rates of weak A and weak B antigen were 3.99% and 17.93% respectively,and the weak B antigen was predominant.The negative rates of anti-A and anti-B antibody were 53.72% and 60.70% respectively;in the cross-matching test with allotype blood,the main side without appearing agglutination accounted for 52.87% and weak agglutination accounted for 33.27%,and the secondary side appearing weak agglutination accounted for 9.49%.Conclusion The maturity of antibody and an-tigen and the coincidence rates of group typing and reciprocal stereotypy in the newborns are less than those in the adults;so blood transfusion according to the cross-matching test results has certain limitation;high attention should be paid to the accuracy of neo-natal ABO blood type,the individual blood transfusion strategy in the newborn should be determined in order to avoid hemolytic blood transfusion reaction caused by ABO allotype blood transfusion and ensure the blood transfusion safety in newborns.
8.The correlations between the different pathological types gastritis and the expressions of AQP3 and AQP4

Objective To explore the correlations of expressions of gastric mucosa water channel aquaporin AQP3 and AQP4 and different gastritis types. Methods The gastric mucosa was mounted under gastroscope. The types of gastric mucosa pathology and activity were tested by the common pathohistology. The expressions of AQP3 and AQP4 were determined by immunohistology. Results The expressions of AQP3 and AQP4 of chronic superficial gastritis were significantly higher than those in the non-gastritis group and chronic atrophic gastritis group (P < 0.01), especially in the activity period of chronic superficial gastritis. The expressions of AQP3 and AQP4 of chronic atrophic gastritis group were reduced when compared to those in the non-gastritis group, in spite of no statistical differences between them. While compared to the non-gastritis group , the expression of AQP3 of chronic atrophic gastritis group during the active stage was remarkably decreased (P < 0.05). Conclusion The expressions of AQP3 and AQP4 of gastric mucosa in chronic gastritis in various pathological types are different. AQP3 and AQP4 may be the targeted point , which could be used for the differential diagnosis and treatment of chronic gastritis of different pathological types.
