1.Effect of stem cell transplantation on expression of SMA, bFGF and VEGF in myocardium of dogs with ischemic mitral regurgitation

Objective To analyze effect of stem cell transplantation on the expression of smooth muscle actin (SMA), basic fibroblast growth factor (bFGF) and vascular endothelial growth factor (VEGF) in the myocardium of dogs with ischemic mitral regurgitation.Methods 24 healthy male experiment dogs were selected, 6 were collected randondy as normal group, the others were used to construct mitral regurgitation model.After that, the modelized dogs were divided into three groups, 5 dogs in model group, 6 dogs in experiment group and 6 in control group.The experimental group were given 2 mL stem cells suspension to ischemic myocardial, control group was infused without stem cells in DMEM medium, and then the SMA, bFGF and VEGF expression levels of myocardial tissues in each groups were detected and analyzed.ResuIts Compared with control group and model group, the heart function of experiment group were improved more obviously, such as cardiac output (CO) and stroke volume (SV) increased significantly (P<0.05).Myocardialα-SMA were significantly lower (P<0.05).bFGF contents in myocardial tissue increased significantly (P<0.05).The content of VEGF in myocardial tissue significantly increased (P<0.05).ConcIusions Stem cell transplantation can inhibit the expression of SMA in myocardium of ischemic mitral regurgitation, and promote the expression of bFGF and VEGF, has a better effect on heart function.
2.Study in hospice home care method of dying children

Objective To explore hospice home care method of dying children.Methods In 58 cases of dying children at home were given personalized hospice care plan,then the questionnaire were adopted to investigate the related indexes and the results were analyzed.Results In home hospice care,parents had a high recognition and satisfaction to children's pain management and symptom care.The parents with different education background had significant difference in recognition extent of psychological support,games support and death education.The parents anxiety and depression mood got effective mitigation through hospice home care of dying children.Conclusions Implementation of personalized home hospice care can increase life quality of dying children and improve adverse mood of their parents.

4.The X-ray Findings and Clinical-pathological Study of Primary Non-Hodgkin's Lymphoma of Bone

Objective To improve the knowledge of X-ray findings and clinic-pathology of primary non-Hodgkin's lymphoma of bone(PNHLB).Methods X-ray findings and clinical-pathological manifestatioms of PNHLB proved by operation and pathology in 5 cases were reported and review of literature.Results The X-ray findings included infiltrative,osteolytic and cystic destruction of bone, among them,4 cases were B-lymphocytes,while 1 case was T- lymphocytes.Conclusion PNHLB is rare,its clinical symptoms are not in accord with X-ray findings,the finial diagnosis depends on pathology.
5.Qualitative study of three cell culture methods.

Primary rat hepatocytes were cultured using different in vitro models and the enzyme leakage, albumin secretion, and cytochrome P450 1A (CYP 1A) activity were observed. The results showed that the level of LDH was decreased over time in culture. However, on day 5, LDH showed a significant increase in monolayer culture (MC) while after day 8 no LDH was detectable in sandwich culture (SC). The levels of AST and ALT did not change significantly over the investigated time. The CYP 1A activity was gradually decreased in a time-dependent manner in MC and SC. The decline of CYP 1A was faster in MC than in SC. This effect was partially reversed by using cytochrome P450 (CYP450) inducer such as Omeprazol and 3-methylcholanthrene (3-MC) and the CYP 1A induction was always higher in MC than in SC. In bioreactor basic CYP 1A activity was preserved over 2 weeks and the highest albumin production was observed in bioreactor followed by SC and MC. Taken together, it was indicated each investigated model had its advantages and disadvantages. It was also underlined that various in vitro models may address different questions.

6.Analysis of Sodium Intake via Drugs Used by Hypertensive Inpatients

OBJECTIVE:To investigate sodium intake via drug used by hypertensive inpatients during hospitalization. METH-ODS:290 inpatients with hypertension were collected from vasculocardiology department of our hospital between Jan. and Oct. 2013. The types of sodium-containing drugs and sodium-intake during hospitalization were analyzed statistically. RESULTS:275 pa-tients with hypertension were prescribed 38 sodium-containing drugs(94.8%);82.7% were administered by iv gtt with a average drug treatment course of 6.7 days(average hospitalization days of 6.8 days). Solvent containing sodium such as 0.9% Sodium chlo-ride injection and Creatine phosphate sodium for injection were the most sodium donors. 43.3% patients’sodium intake exceeded upper limit of 2 g/day recommended by WHO. CONCLUSIONS:Hypertensive inpatients are prescribed too much sodium-contain-ing drugs,and less intravenous solutions and less adjuvant medicines should be took to decrease sodium intake. Only when prospec-tive benefit are greater than risk,the patients can be prescribed sodium-containing drug prescription.At the same time,manufacturers should annotate the content of sodium in package inserts,which is convenient for physicians to choose drugs.
7.Effect of rhGH on JAK2-STAT3 signal pathway after GHR was down-regulated by siRNA in gastric cancer cell.

To investigate the effect of recombinant human growth hormone (rhGH) on JAK2-STAT3 pathway and the growth of gastric cancer cell lines at different GHR expression status, the eukaryotic expression vector targeting human GHR (pGPU6/GFP/Neo-shGHR and pGPU6/GFP/Neo-scramble) was constructed and transfected into MGC803 cells by Lipofectamine 2000. Stable expressive cell lines were obtained by G418 screening. The expression of GHR was analyzed by Western blotting. After being stimulated with rhGH, cell growth was detected by MTT assay. Cell cycle and apoptosis were examined by flow cytometry. The components of JAK2/STAT3 signaling pathway were detected by Western blotting. There is no significant difference of GHR expression between MGC803 and pGPU6/GFP/Neo-scramble-transfected cells (named as MGC803-NC) (P > 0.05). Compared with MGC803, the GHR expression in pGPU6/GFP/Neo-shGHR-transfected cells (named as MGC803-shGHR) decreased significantly (protein decreased 50%). The cells were treated with rhGH at 0, 150 and 300 ng x mL(-1), the growth rate of MGC803 and MGC803-NC increased significantly, PI and the number of G2/M phase cells all increased significantly, and apoptosis decreased significantly. Western blotting revealed that the expression of pJAK2 and pSTAT3 was up-regulated after being treated with rhGH in MGC803 and MGC803-NC cells. In contrast, similar change was not observed in MGC803-shGHR cells. Knockdown of GHR gene may decrease the sensitivity of gastric cancer cells to rhGH, and down-regulating of components of the expression of JAK2/STAT3 signaling pathway may be the potential mechanisms.
8.Effects of Bio-feedback and Pelvic Electric Stimulation on Stress Urinary Incontinence

Objective To investigate clinical effects of bio-feedback and pelvic electric stimulation on stress urinary incontinence (SUI).Methods Fifty-six patients with SUI were selected randomly.All the patients received one or two courses of treatment and the effects were observed.Results Forty-three patients finished the treatment and the symptoms were improved.Scores of ICI-Q-SF and rotational angle of bladder were found to be significantly different from those before the treatment.Conclusions Stress urinary incontinence could be treated by bio-feedback and pelvic electric stimulation.Both the combination of treatment and individualized treatment should be taken into account.
9.Association of chronic kidney disease and cerebral microbleeds in patients with ischemic stroke

Objective To investigate the association between the presence of cerebral microbleeds and chronic kidney disease in patients with ischemic stroke.Methods Patients with ischemic stroke within 1-6 months were consecutively recruited.Cranial MRI was taken within two weeks after recruitment.Cerebral microbleeds were assessed using Microbleed Anatomical Rating Scale on gradient echo MRI.Demographics including sex,age and risk factors were obtained.Chronic kidney disease was defined and classified according to National Kidney Fundation-Kidney Disease Outcome Quality Initiative Guideline.Glomerular filtration rate (GFR) was estimated by using the abbreviated Modification of Diet in Renal Disease equation.Results Of the 636 patients included,mean age was (59.8 ± 10.1) years,435 (68.4%) were male.Sixty-six had decreased estimated GFR (eGFR; < 60 ml · min-1 · 1.73 m-2).Two hundred and one (31.6%) patients had cerebral microbleeds,which were most commonly located in deep or infratentorial location (133/201,66.2%).The presence of cerebral microbleeds was much higher in patients with decreased eGFR than the others (48.5% (32/66) vs 29.6% (169/570),x2 =9.709,P =0.002).Age,history of hypertension and decreased eGFR were associated with the presence of cerebral microbleeds in univariate analysis.In multivariate analysis,decreased eGFR was independently associated with the presence of cerebral microbleeds in deep or infratentorial location (OR =1.457,95% CI 1.044-2.034,P =0.027),but not associated with the presence of cerebral microbleeds in pure lobe.Conclusion Impaired kidney function is associated with the presence of cerebral microbleeds in deep or infratentorial regions in patients with ischemic stroke.
10.Stress ulcer survey of anti-ulcerative drug use in patients undergoing arthroscopic operation

Objective To investigate anti-ulcerative drug use in patients undergoing arthroscopic operation against stress ulcer. Methosds A total of 297 patients undergoing arthroscopic operation during Jan. to Aug. 2013 in our hospital were recruited and retrospectively analyzed with Excellspreadsheet. Results Among 297 patients in the study, ninety-four patients (31.6%) used AUD before operation, most (94.9%) administered by intravenous, and drug utilization index was 1.35. Ninety-eight point nine patients used proton pump inhibitors (PPIs) in which Lansoprazole was prescribed most. Conclusion This study highlights the overuse of PPIs in patients undergoing arthroscopic operation.
