1.Effects of Maca on kidney mitochondrial respiratory function and anti-aging capabilities in aged rats after exhausted exercise

BACKGROUND: Studies have shown that Maca can enhance immune cel function, improve mitochondrial respiratory chain enzymes, and play a role in anti-oxidation and anti-aging.
OBJECTIVE: To observe Maca effects on kidney mitochondrial respiratory function and anti-aging capabilities in elderly rats after exhausted exercise.
METHODS: 10-month-old elderly rats were intragastrical y administrated Maca powder 5 g/kg, once a day, and did treadmil exercise, 5 days in a week. Rats were randomly divided into control group, Maca group and training group and Maca+training group. At 6 weeks after training, rats in each group did exhausted exercise (35 m/min), and immediately received intraperitoneal injection of 2% sodium pentobarbital. The kidney was obtained, and mitochondria were extracted by differential centrifugation. A spectrophotometer was used to measure mitochondrial respiratory chain activity.
RESULTS AND CONCLUSION: (1) Mitochondrial respiratory chain enzyme and antioxidant enzyme activity: mitochondrial respiratory chain enzyme I-III, superoxide dismutase and glutathione peroxidase activities were higher in the Maca+training group than in the control group, Maca group and training group (P < 0.05 or P < 0.01), but malondialdehyde content was lower (P < 0.05 or P < 0.01). (2) Results suggested that the combination of supplement Maca and incremental exercise can improve mitochondrial respiratory function and delay aging in the kidney of aged rats after exhausted exercise. Moreover, supplemented Maca and incremental exercise have a synergistic effect.

4.Effects of vasopressin and adrenaline on cerebral resuscitation in rats undergone cardiopulmonary resuscitation

Objective To compare the effects of vasopressin and adrenaline on cerebral resuscitation in rats. Method Sixty male SD rats were used to set the animal model of cardiac arrest and were randomly (random number) divided into 4 groups (n = 15/group): sham operation group (sham-gro), negative control group (neg-gro), vasopressin group (vas-gro) and adrenaline group (adr-gro). Blood pressure was recorded, and brain tissue samples were obtained. Results There was no significant difference in the recovery rate between vas-gro and adr-gro 30 min after resuscitation (P <0.05). The mean blood pressure in vasgro was higher than that in adr-gro within 30 min (1,2, 5, 10, 20, 30 min) after restoration of spontaneous circulation. The measured MDA, SOD and GSH were significantly different (P < 0.05) between each two groups among vas-gro, sham-gro and adr-gro. Conclusions The recovery rate was similar between vasopressin group and adrenaline group during cardiopulmonary resuscitation in rats. However, vasopressin can to maintain the mean arterial pressure at a higher level after resuscitation, which can the increase the cerebral perfusion and reduce brain cell damage.
5.Effects of Hormone Replacement Therapy and Sport Walk with Weight on Serum Lipid and Bone Mineral Density in Postmenopausal Women

Objective To explore the effects of estrogen,sport walking with weight and estrogen plus sport walk on serum lipid,bone metabolism and bone mineral density(BMD) in postmenopausal women.Methods 52 postmenopausal women were divided into the hormone replacement therapy(HRT) group(n=12),sport walking with weight group(Sp group,n=14),HRT + Sp group(n=12) and control group(n=14).HRT group took compound nylestriol,Sp group took sport walking with 5 kg weight and exercise intensity was from 60% to 80% VO2max,HRT+Sp group received two treatments as HRT group and Sp group.The experimental duration consisted of six months.Results The total cholesterol(TC),TC/high density lipoprotein cholesterol(HDL-C),alkaline phosphatase(APL) and urine calcium(Ca)/creatinine(Cr) were markedly lower in three experimental groups than the control group and before experiment(P<0.05 or P<0.01),HDL-C in Sp and HRT+Sp groups were markedly higher than the control group and before experiment(P<0.05),BMD of spine(L2~L4) and left trochanter in three experimental groups were markedly higher than the control group(P<0.01) and before experiment(P<0.0).Conclusion Both estrogen replacement therapy and sport walking with weight effectively improve serum lipid,prevent and reverse osteoporosis induced by menopause in women.
6.Analysis of multi-resistant organisms in the hospital during the past 4 consecutive years

Objective To investigate the change trend and the drug resistance rate of the multi-resistant organisms in the hospi-tal during the past four years,so as to guide the clinical medication,and strengthen the management of antibiotics application.Meth-ods Kirby-Bauer test which was recommended by the clinical and laboratory standards institute was used to conduct the suscepti-bility test,with the results evaluated based on US Clinical and Laboratory Standards Institute.Results Strains of bacteria were de-tected from January 201 1 to December 2014 in each year for the basic trend.The gram-negative bacterium was in the dominant posi-tion.The bacteria in the top five were:Escherichia coli,Staphylococcus epidermidis,Staphylococcus aureus,Pseudomonas aeruginosa and Pneumonia crayresearch coli.In each year,the overall situation of bacteria distribution had little changed.From the antimicrobial susceptibility test results,425 strains of multi-resistant bacteria were detected during the past four years.The detection rate and constituent ratio of multi-resistant bacteria then fell slightly in the first three years,but rebound in 2014.There was no statistically significant difference between the four years.Conclusion In the past four years,our hospital actively participates in monitoring and controlling the multiple drug-resistant bacteria,but nosocomial infection of the multiple drug-resistant bacteria has not been effec-tively controlled because of a variety of factors.We should continue to strengthen monitoring the clinical multiple drug-resistant bacteria,and understand the change tendency.In order to strengthen the management of the use of antibiotics and to guide clinical rational drug use,the drug sensitive test and the resistance situation of the hospital should be considered for the reasonable selection of antimicrobial agents.Strengthening the biological monitoring of clinical departments,enhancing the consciousness of sterile oper-ation of clinicians can help prevent the spread of drug-resistant strains.
7.Expression and clinical significance of muti-drug resistance factors:P-gp,GST-?,TOPO? in lung cancer

Objective: To examine the expression of P-gp,GST-?,TOPO? and Ki67 in lung cancer and study the clinical relationship of P-gp,GST-?,TOPO?and Ki67 Co-expression in lung cancer.Methods: The expression of P-gp,GST-?,TOPO? in 52 cases of lung cancer without chemotherapy was detected before operation by immunohistochemical staning.Results: The positive rates of P-gp,GST-?,TOPO?,Ki67 were respectively 73%,80.5%,62.5%,23.7%.The P-gp,GST-? expression was correlated with the histology and differentiation,The expression of TOPO? was obviously lower in adenocarcinoma than in other typers of lung cancers;The coexpression rate of more than one MDR gene production appareatly exceeded individual expression rate.The expression of P-gp,GST-?,TOPO? was not correlated with the expression of Ki67.Conclusion: P-gp,GST-?,TOPO? can be expressed in different types of lung cancers and with different expression levels,They are all correlated with multidrug resistance of lung cancer,Thus,they can be regarded as a valuable indicators for clinical chemotherapy and for prognosis in lung cancer patients.
8.High lever soluble expression and purification of recombinant human IL-18 in E.coli

Objective: Objective To study high-level expression and purification of recombinant mature human IL-18(mhIL-18)in E.coli with soluble form.Methods: The gene of mhIL-18 was amplified with PCR.Then,the recombinant expression plasmid pTYB1IL-18 was constructed by cloning mhIL-18 into pTYB1 vector and transformed to E.coli ER2566.IPTG was added into the E.coli ER2566 culture to express recombinant fusion protein.After the harvested bacteria dispersed with ultrasound,SDS-PAGE and Western blot were used to analyze the expression of recombinant fusion protein.The activity of mhIL-18 which had been purified by affinity chromatography was tested by MTT.Results:The cloned sequence of mhIL-18 was identical with the sequence in GenBank.After the induction by IPTG inducing,the fusion protein of mhIL-18 covered over 26% total bacterial proteins,which included 80% active fusion protein.Purification of recombinant expressed mhIL-18 purity was 95%,which had the activity of the mhIL-18.Conclusion: High level expression and purification of recombinant human IL-18 is succeeded which has the activity.
9.Study on unscheduled DNA synthesis of Alternaria alternata 261-B2-3 in human amnionic cells in vitro

Tne Unscheduled DNA syntheris (UDS) was induced in cultured human amnionic cell line FL in vitro by exposure to 261-B2-3, an extract of Alternaria alternata separated from crops in Linxian county which is a high risk area of esophageal cancer in China. In the absence of S-9M, 261-B2-3 was capable of inducing UDS in target cells and the ratio of UDS elicited by the extract (the range of concentration being 10~(-6) to 10~(-3) mg/ml) was significatly different from the ratio by solvent control (P0.05) with S-9M. The results demonstrated that 261-B2-3 has stronger mutagenicity without S-9M and it's mutagenicity is rising with increasing concentrations in a certain range. On the contrary, 261-B2-3 has no or only a weak mutagenicity with S-9M. It is suggested that there arc direct mutagens in this extract. It is possible that Alternaria alternata contaminated the crops play an important role in the high incidence of esophageal cancer in Linxian county.
10.Dynamic changes of biventricular and pulmonary circulatory function and relations among them after left coronary artery ligation in dogs

In anesthetized and thoracotomized dogs the dynamic changes of biventricular contractility, relaxation, pump function, pulmonary circulatory pressure and resistance were observed and the relations among them were analysed in 2 hours after left anterior descending coronary artery (LAD) ligation. The myocardial ischemic size made by LAD ligation amounted to 35.2?1.4% of left ventricular (LV) mass. After LAD ligation, the decrease of LV relaxation and increase of pulmonary artery pressure happened at first. Then left and right ventricular pump function, LV contractility and right ventricular (RV) relaxation decreased in turn. And pulmonapy vascular resistance increased by a big margin at last. Biventricular pump functions decreased more obviously while pulmonary vascular resistance increased. RV contractility did not change significantly.The results show that the decrease of LV function results in the decrease of RV relaxation directly and RV pump function directly and indirectly by means of increased pulmonary circulatory pressure and resistance but do not effect on RV contractility during early stage of LV myocardial infarction (MI). Factor analysis indicates that the alterations of LV contractility and biventricular pump function are very consistent and play a leading role in the changes of cardiac function after LVMI, the alterations of biventricular relaxation and pulmonary circulatory function take second place in due order.
