1.Determination of Baicalin in Oral Liquid for Gastritis by HPLC

OBJECTIVE: To establish an HPLC method to determine the content of Baicalin in Oral liquid for gastritis. METHODS: Zorbax SB-C18(150 mm?4.6 mm,5 ?m) was used as chromatographic column, and the mobile phrase consisted of methanol-water-phosphoric acid(50∶50∶0.2) with detection wavelength set at 276 nm.RESULTS: The calibration curve of baicalin was in good linearity over the range of 0.065~0.520 ?g (r=0.999 8). The average recovery of Baicalin was 99.79% with RSD at 1.28% (n=6).CONCLUSION: The method is simple and accurate and applicable for the quality control of Oral liquid for gastritis.
2.Protective effects of desflurane, sevoflurane and isoflurane preconditioning on myocardium against ischemia-reperfusion injury in rabbits

0. 5mv. LAD occlusion was maintained for 50min and then released for reperfusion (120min) . Myocardial infarct size was measured by nitroblue tetrazolam at the end of experiment. Results There was no statistically significant difference in HR, MAP, cardiac output (CO), left ventricular systolic pressure (LVSP), left ventricular end-diastolic pressure ( LVEDP) and rate-pressure products ( RPP) among all groups during ischemia and early reperfusion period except for LVEDP in control group which was much higher than that in all preconditioning groups during the late reperfusion period. Compared with the control group, desflurane, sevoflurane, isoflurane and G+ desflurane reduced myocardial infarct size by 41%, 47%,31.7% and 17.8% respectively without significant homodynamic effects. Myocardial infarct size in G + desflurane groups was significantly larger than that in desflurane group. Conclusion Preconditioning with desflurane, sevoflurane and isoflurane reduce myocardial infarct size in rabbits to some extent. The protective effects may be partly medialed via activation of KATP channel.

Objective To evaluate the MRI findings and clinical features of methotrexate-induced acute encephalopathy in children.Methods The clinical data and brain MRI obtained in 13 children with methotrexate-induced acute encephalopathy were retrospectively reviewed.The MRI features were analyzed , including information on the location , the signal intensity and follow-up MRI study was performed.Results Of the 13 patients , 2 patients suffered from seizure.Five patients had dysphasia , of which 4 patients had evidence of hemiparesis , 1 patient had right facial palsy.Five patients had unilateral weakness.And left hemiparesis was observed in 1 patient.DWI revealed well demarcated asymmetrical hyperintensity lesions within the centrum semiovale and/or periventricular white matter in 10 patients, corresponding to areas of hypointensity on ADC maps.One case showed hyperintensity areas in the bilateral supratentorial cortex and subcortical white matter on T 2-weighted images with subtle high-intensity on DWI.In all 10 cases there were resolution of the diffusion abnormality , 8 cases displayed residual FLAIR signal abnormalities involving areas of previously seen diffusion restriction , 5 cases showed decreased range of the lesion , 1 case was progressive, and 2 cases were stable.One case with hyperintensity areas in the supratentorial cortex and subcortical white matter showed small residual hyperintensity on T 2-weighted images and resolution of the diffusion abnormality.Conclusions MTX-induced acute encephalopathy often manifests as stoke-like symptoms.DWI is the imaging modality of choice for the detection of acute MTX neurotoxicity , and asymmetrical restricted diffusion in the deep white matter is the characteristic sign.Cytotoxic edema induced by MTX is transient and reversible .

8.Study on the role of MAT1 in estradiol regulated MMP-9,TIMP-1 expression in ER positive breast cancer

Obej ctive To investigate the role of metastasis associated protein 1(MTA1)in estrogen reg-ulated expression of matrix metalloproteinase -9(MMP-9)and tissue inhibitor of metalprotease -1(TIMP-1) in estrogen receptor( ER ) positive breast cancer cells .Methods MTA1 knockdown cell model was generated based on MCF-7breast cancer cell line by transfected with MTA 1-shRNA.The mRNA and protein level of MMP-9 and TIMP-1 in wild type MCF-7(MCF-7WT)and MCF-7MTA1-shRNA before and after 17β-estradiol ( E2) treatment were examined by Real -time PCR and Western blot respectively .Results The MTA1-shRNA showed maximally 84.9%suppression of MTA1 expression in MCF-7,suggesting a satisfied MTA 1 knockdown cell model was established for subsequent experiments .After treated with E2 for 48 h,MCF-7WT showed an incre-ment of 46%(P<0.05)and 37%(P<0.05)of the mRNA and protein level of MMP -9 and a decrement of 32.3%( P<0.05)and 18.2%(P<0.05)of TIMP-1;MCF-7MTA1-shRNA showed a decrement of 32.3%(P<0.05)and 18.2%(P<0.05)of mRNA and protein expression of MMP -9 respectively but no significant differ-ence in TIMP-1 comparing with MCF-7WT before treated with Estradiol.After E2 treatment,MCF-7MTA1-shRNA didn′t show significant change of MMP -9 except decrements of 32.3%(P<0.05)and 18.2%(P<0.05)in the mRNA and protein levels of TIMP -1.Conclusion MTA1 may be involved in the pathway by which estrogen regulated the expression of MMP -9 but not TIMP-1 in ER positive breast cancer cells .

Objective To characterize the manifestations of non-Hodgkin's lymphoma in nervous system with in-flammation-like presentation. Methods We reviewed clinical and laboratory data obtained from 3 cases of non-Hodgkin's lymphoma in nervous system with inflammation-like presentation.Those data include clinical manifestations,CSF examina-tions neuroimaging,pathology of biopsies,treatment and prognosis.Results The clinical manifestations of NHL in nervous system were variable and the findings of cerebrospinal fluid and imaging were not characteristic.Parital relief of symptoms by steroid cortisone could be achieved in some cases which maght further increased the difficulty in differentiating NHL from CNS inflammation.Several signs including no evidence of CNS inflammation,multiple organ involyements,especially the organ involvements outside CNS,and deterioration after a transient relief of symptoms by steroid cortisone,strongly suggest the possibility of NHL.Condusions We should increase physicians'awareness to NHL to reduce the misdiagnosis even though the final diagnosis relies on pathological examination.
10.Assessment of the error of measurement technique on DSA

Objective To explore the creation and control of measurement technique error on digital subtraction angiography (DSA). Methods The data was obtained from Advantx LCV Plus DSA system made by GE Corporation. We divided the screen into three areas, per area account for 1/3, ie, central area, middle area and outlying area. The enlargement rate or reduction rate of the target object was respectively calculated according to the different calibration, different height of the bed and different X-ray source to image distance (SID). Results The target object was enlarged gradually from the central area to the outlying area, and the lengthwise enlargement rate was more obvious than transverse. The different of target object measured by coin (diameter was 20.4 mm) with steel ball (diameter was 7.7 mm) was not significance, but the target object was underestimated significantly used the calibration by 4F catheter. When the target object was measured by the calibration in same area and same axis, the error of measurement technique was controlled rang from 1.0% to-2.5%. Conclusion This systematic investigation suggest that the screen was divided into the central area, middle area and outlying area will be beneficial to control DSA measurement error for the interventional physician. The target object was close to real size when it measured by transverse of the calibration, and the error was better controlled when the calibration was in the same area and same axis as the target object.
