BACKGROUND: Bone defect repair is a difficulty in orthopedic field all the time, researches on the tissue engineered bone tissue have provided completely new thoughts and methods for bone defect repair, and it is an important link to detect the biocompatibility of the biomaterials.OBJECTIVE: To investigate the biocompatibility of PCL with bone marrow stromal cells (BMSCs).DESIGN: A controlled observation.SETTING: Department of Orthopaedics, the First Affiliated Hospital of Xinjiang Medical University.MATERIALS: The experiments were carried out in the orthopedic laboratory of the First Affiliated Hospital of Xinjiang Medical University. Healthy 4 to 8-week-old New Zealand rabbits of about 2 kg were used.METHODS: ① Bone marrow was extracted from bilateral femurs of the rabbits, then mixed into 10 mL RPMI1640 complete medium, and then entered the passage culture. ② The cells were inoculated and then divided into PCL group and control group, the cells were only inoculated in the control group, and the BMSCs were co-cultured with PCL in vitro in the PCL group. The morphological observed, cell proliferation, protein content and enzymological determinations were conducted.MAIN OUTCOME MEASURES: The growth and the adhesion of BMSCs on PCL biomaterials were mainly observed.RESULTS: In the control group, most of the BMSCs changed to the shape of fusiform or multi-angles. In the PCL group, the BMSCs could adhere and proliferate on PCL, and the growth and function were not affected, PCL also played a certain role in promoting the cell proliferation.CONCLUSION: PCL possess satisfactory biocompatibility, and it is possible to be used as the carrier of BMSCs in tissue engineering.
6.Breeding Selenium-Enriched Yeast by Protoplast Mutagenesis.
Microbiology 1992;0(02):-
This paper studied on breeding selenium-enriched yeast by protoplast mutagenesis. A strain which the content of selenium is the highest is selected from thirteen strains yeast. The optimum conditions to form protoplast are lysed by 1 g/100 mL lywallzyme for 120 min, the formation and regeneration being 95.2% and 21.8% respectively. By mutating breed a strain of A1 which the content of selenium is 821 mg/kg and the amount of dry cell of 0.84 g/100 mL is obtained.

Objective:To observe the clinical efficacy of acupuncture with medication for post-stroke depression. Methods:A total of 96 patients with post-stroke depression were randomized into an observation group and a control group, with 48 cases in each group. The control group was treated with oral fluoxetine hydrochloride capsules, and the observation group was treated with acupuncture on the basis of the treatment of the control group. After 6 months of treatment, the traditional Chinese medicine (TCM) symptom scores, and Hamilton depression scale (HAMD) component scores and total scores of the two groups before and after treatment were observed, and the clinical efficacy and adverse reaction rate were compared between the two groups. Results:The total effective rate was 95.8% in the observation group and 83.3% in the control group, and the difference between the two groups was statistically significant (P<0.05).After treatment, the intra-group differences in TCM symptom scores and HAMD score of both groups were statistically significant (all P<0.01).In the observation group, the scores of indifferent expression, emotional restlessness, and sentimentality in the TCM symptoms were statistically different from those in the control group (all P<0.01).The scores of desperate factor and sleep disorder factor and total score in HAMD in the observation group were significantly different from those in the control group (all P<0.01).The incidence of adverse reactions was 6.3% in the observation group and 4.2% in the control group, and the difference was not significant between the two groups (P>0.05).Conclusion:Acupuncture plus medication is an effective method for post-stroke depression, and it can further improve the patient's bad mood.

Objective To explore the application of particle-gun technique in transfecting dendritic cells(DCs),and investigate the expression of DNA vaccine particle IgHV1-GM-CSF in DCs induced by particle-gun.Methods Monocytes were isolated from donor peripheral blood collected by Ficoll density gradient method and adherent culture, and then differentiated into immature DCs(imDCs) by rhGM-CSF and rhlL-4 induction. The plasmids for transfection were IgHV1-GM-CSF/pcDNA3.0 and pEGFP,which were prepared with endofree plasmid purification kit.The DCs were transfected with plasmid pEGFP by particle-gun at different transfection conditions, the green fluorescence positive cell and the total cell numbers were counted respectively under fluorescence microscope, and the transfection efficiency was calculated;the viable cell count was performed after trypan blue staining;the transfection efficiencies of particle-gun,liposome-mediated transfection and electroporation method were compared. The plasmids IgHV1-GM-CSF/pcDNA3.0 and pEGFP were cotransfected into imDCs by particle-gun under the optimum conditions, and then DNA was extracted, the expression of IgHV1-GM-CSF was determined by RT-PCR,and of GM-CSF by ELISA.Results The optimum conditions of particle-gun transfection in imDCs were: gas pressure at 120psi,PVP concentration in 0.02mg/ml,microcarrier loading in 0.5mg gold/shot,and DNA loading ratio in 1.5?g plasmid/mg gold. The transfection efficiencies of particle-gun and electroporation were 10.5%?2.4%(n=3) and 45.2%?5.6%(n=3) respectively, while that of liposome-mediated transfection was very low,and significant difference existed among the three methods(P
