Objective To research the occurrence of liver cirrhosis and portal hypertension splenectomy treatment,and analyze the risk factors for postoperative complications.Methods Retrospectivly analyzed the clinical data of 390 liver cirrhosis cases from March 2009 to May 2013 who were admitted to department of digestion and vascular surgery,First Affiliated Hospital of Xinjiang Medical University,for splenic resection.Child classification,diabetes,amount of fluid drainage,drainage time,blood loss,emergency surgery,combined with other surgery were alalysised by Logistic regression.Results Among three hundred and ninty cases of splenectomy patients,liver cirrhosis complications occurred in 53 cases (53/390,13.58％).In these 53 cases,45 cases combined diabetes,Child grade A 12 cases,B 22 cases,C 19 cases,and theirs 24 hours drainage volume was (422.45 ±297.09) mL,drainage time was (4.96 ± 1.48) d,blood loss was (874.72 ± 106.19) mL,K value was 0.10 ±0.06,R15 value was 0.27 ±0.016,albumin concentration was (30.09 ±6.87) g/L,Among the rest 337 cases,228 cases were combined diabetes,Child grade A 134 cases,B 129 cases,C 74 cases,and theirs 24 hours drainage volume was (234.06 ± 171.15) mL,drainage time was (3.18± 1.72) d,blood loss was (261.90±98.27) mL,K value was 0.14 ± 0.09,R15 value was 0.19 ± 0.015,albumin concentration was (32.51 ±5.58) g/L.Multivariate regression analysis had indicated that Child grade,drainage volume of 24 hours,drainagetime,and K value were independent factors affecting the occurrence of complications.Conclusion The major risk factors of incidence happen on postoperative complications in cirrhosis and portal hypertension splenectomy are high Chill grading,draining a long time,and 24 hours blood loss,and k value.

Objective To investigate the expression level of miR-133b in cancer tissues of patients with prostate cancer and its effect on the proliferation of prostate cancer cells.Methods The total RNAs in resected prostate cancer tissues and adjacent tissues from 30 patients with prostate cancer were extracted and reversely transcripted into cDNA,and then the expression levels of miR-133b were detected by real-time quantitative PCR.The correlations between the expression levels of miR-133b and the patients' clinicopathological features were analyzed.The expression of positive regulatory domain I-binding factor 16 (PRDM16) and proliferation of PC-3 cells transfected with miR-133b mimics by LipofectamineTM 3000 were determined by real-time quantitative PCR and the CCK8 method,respectively.Results The expression levels of miR-133b in prostate cancer tissues [(16.85 ± 0.94) × 10-4] was significantly lower than that in adjacent tissues [(22.95 ± 1.567) × 10-4,t =3.335,P ＜ 0.01].The expression levels of PRDM16 in PC-3 cells transfected with miR-133b mimics were significantly lower than that in the control group (0.371 ±0.031 vs 1.000 ±0.022,t =12.53,P ＜ 0.01).The proliferation ability of PC3 cells transfected with miR-133b mimics for 72 hours was significantly lower than that in the control group (t =6.811,P ＜ 0.01).Similarly,the proliferation ability of PC-3 cells transfected with PRDM16 inhibitor for 72 hours was also significantly lower than that in the control group (t =9.048,P ＜0.01).Conclusion The expression levels of miR-133b in prostate cancer tissues are significantly down-regulated,which regulate the proliferation of prostate cancer cells possibly through PRDM16.

AIM: To research the protective effects of crocetin on cerebral ischemia-reperfusion(CIR) injury in mice.METHODS: The effects of crocetin on brain edema,brain capillary permeability,the activities of lactate dehydrogenase(LDH),superoxide dismutase(SOD),glutathione peroxidase(GSH-Px) in brains and the contents of malondialdehyde(MDA) and nitric oxide(NO) in CIR model mice were observed.RESULTS: Compared with the CIR group,crocetin(50,100 mg/kg i.g.for 5 d) decreased the content of water,Evans blue,MDA and NO in mice brain tissue,and increased LDH、SOD and GSH-Px activity in brains of CIR mice,with statistical significance.CONCLUSION: Crocetin has protective effects on cerebral ischemia-reperfusion injury in mice,the mechanism may be related to its anti-oxidative activity and inhibition of NO overproduction.

Objective To study factors associated with necrosis and calcification of cyst echinococcosis. Methods This study retrospectively analyzed 191 cases of hepatic cycstic echinococcosis admitted from 2005 to 2008 in the First Affiliated Hospital of Xinjiang Medical University Department of General Surgery, possible relevant factors screened were the patients' gender, age, operation frequency, hepatitis, gall stones, calcium of blood serum, multiple hepatic hydatid cyst, biliary fistulae and hepatic cycstic echinococcosis degeneration. Single-factor analysis and multi-factors analysis were conducted. Results The single-factor independent samples t-test showed that age is significantly related to hepatic cycstic echinococcosis degeneration (t = - 2. 455, P = 0. 008). Single factor Chi-square test showed that factors impacting on hepatic cycstic echinococcosis degeneration were multiple hepatic hydatid cyst and the biliary fistulae (x2 = 6. 848, P = 0. 009; x2 = 7. 149, P = 0. 008). The multiple-factor Logistic regression analysis showed that the age, the multiple hepatic hydatid cyst, and biliary fistulae are the related risk factors(x2 =5. 306,P = 0. 021,OR = 1. 665;x2 = 10. 377,P =0. 001,OR =3. 300;x2 = 4.455,P =0. 035, OR =2. 164) of hepatic cycstic echinococcosis patients with cyst echinococcosis degeneration. Conclusions Age, multiple hepatic hydatid cyst and biliary fistulae are the risk factors associated with degenerative necrosis and calcification of hepatic echinococcosis.

Objective To establish a HPLC method for the determination of strychnine content in Cidan Capsules. Methods Using Hypersil- C18 as the column and 1 % Acetic acid-acetonitrile (88 ∶ 12) as the mobile phase, HPLC was performed. The detection wavelength was 254 nm. Results The linear range was 0.112～ 0.560 ? g(r=0.999 9). The average recovery rate was 97.36 % , RSD was 1.00 % ( n=5) . Conclusion This method is accurate and specific.

Objective To study on the relationship of PTEN with clinicopathological parameters and prognosis of esophageal squamous cell carcinoma patients.Methods The expression of PTEN was detected in 62 cases of esophageal squamous cell carcinomas by immunohistochemistry.Results(1)PTEN expres- sion is negatively correlated with the depth of invasion,lymph node metastasis,distant metastasis,pTNM stage and degree of differentiation.(2)The difference of survival is significant between high and low expres- sion groups.Conclusion PTEN is correlated negatively with the clinicopathological parameters reflecting the malignant biological behavior,and is one of the significant prognostic predictors by univariate analysis.

The host protein SAMHD1 has been identified as the first mammalian deoxynucleoside triphosphate triphosphohydrolase (dNTPase), which blocks the infection of HIV-1 in non-cycling immune cells.SAMHD1 protein is highly expressed in human myeloid-lineage cells and resting CD4+T lymphocytes, which restricts HIV-1 replication by hydrolyzing the cellular dNTPs, thus inhibiting reverse transcription and viral complementary DNA ( cDNA) synthesis. Recent studies have revealed that SAMHD 1 plays an important role in virus whole life by promoting HIV -1 genome recombination, degenerating viral genome RNA and restricting virus transmission between cells .In this review, these progress on SAMHD1 research are summarized and the mechanisms by which SAMHD 1 mediates retroviral restriction are analyzed .

Objective To explore the role of ppk1 gene in E.coli CFT073 strain during urinary tract infection (UTI). Methods C57BL/6 mouse models of acute UTI with the wild-type(CFT073) and ppk1 mutant (△pk1) infected, were used to compare the bacteria colonization and inflammation induction abilities of bladder tissues. Results In the mouse models, the △pk1 strain showed a significantly lower infection rate (73.3% vs 93.3%) and lower adhesion frequency of bladder (0.01%vs 0.5%) than those of the CFT073 strain. The expression of IL-6 and TNF-αwere reduced in the bladder of △pk1 infected group (P<0.05). Hematoxylin-Eosin tissue staining showed that the damage degree of bladders in △pk1 infected mice were less serious than the CFT073 infected mice. Conclusion ppk1 gene plays an important role in E.coli colonization to bladder and the inflammation induction ability.

Objective To investigate the effect on differentiation of denervated skeletal muscle-derived stem cells (MDSCs) induced by TGF-β1 in vitro.Methods MDSCs were obtained from the rat denervated skeletal muscle by preplate technique,with TGF-β1 adding on medium.Cultured cells were divided into two groups.A,control group; B,10 ng/ml TGF-β1 group.Cell growth was observed with phase contrast microscope.lmmunocytochemistry,quantitative RT-PCR and Western blot was used to detect the expression of Sca-1,COL-Ⅰ,COL-Ⅲ,α-SMA and vimentin in denervated MDSCs.Results The synthesis of COL-Ⅰ,COL-Ⅲ,α-SMA and vimentin by denervated MDSCs was extremely low at protein level in vitro,while Sca-1 level was really high.Belong to the treatment with TGF-β1,COL-Ⅰ,COL-Ⅲ,oα-SMA and vimentin in the denervated MDSCs had strong expression,but Sca-1 in which had a weak expression.Under the stimulation of TGF-β1,COL-Ⅰ expression reached peak at the 2nd day (12.5591 ± 0.3389),which was about 3 times as control group.COL-Ⅲ reached highest value at the 5th day (0.8956 ± 0.0438),which was about 23 times as control group.α-SMA topped out to 18 times at the 5th day (1.1090 ± 0.0018).Vimentin expression rose by 8.5 times and peaked at the 5th day (0.1794 ± 0.0019).The expression of Sca-1 began to decline at the 2nd day,with a remarkable reduction at the 5th day (0.0636 ± 0.0015).Conclusion TGF-β1 could induce differentiation of the denervated MDSCs to myofibroblasts in vitro,and promote the synthesis and excretion of extracellular matrix.

Objective:To investigate ulinastatin (UTI) combined with Xingnaojing injection in the treatment of acute cerebral hemorrhage (ACH) and its effect on the serum high sensitivity C reactive protein (hs-CRP),D-dimer (D-D) and neuron specific enolase(NSE) levels.Methods:110 cases of ACH patients admitted in our hospital from January 2015 to December 2016 were selected and divided into two groups according to the random number table method.The control group was given UTI treatment,while the observation group was given UTI combined with Xingnaojing treatment.Then the brain edema absorption effect,NIHSS score,serum hs-CRP,D-D and NSE levels before and after the treatment of the two groups were recorded and compared;the safety ofmedicidstion of the two groups was evaluated.Results:At the 14th day after treatment,the total effective rate of cerebral hematoma absorption in the observation group was 89.1％,which was significantly higher than 67.3％ of the control group (P＜0.01).At the 14th day after treatment,the NIHSS scores of both groups were significantly lower than those before the treatment (P＜0.01);compared with that of the control group of the same time period,at the 14th day after treatment,the improvement effect of NIHSS score in the observation group was more significant (P＜0.01).Compared with those before the treatment,the serum hs-CRP,D-D and NSE levels of both groups at the 14th day after treatment were significantly decreased (P＜0.01);at the 14th day after treatment,the serum indicators of the observation group improved more significantly than those of the control group (P＜0.01).The incidence rate of adverse reaction in the observation group was 3.6％ compared with 5.5％ of the control group (P＞0.05).Conclusion:Ulinastatin combined with Xingnaojing Injection could rapidly relieve or eliminate hematoma in the treatment of acute cerebral hemorrhage,control the inflammatory response,improve the blood coagulation system and fibrinolytic system,protect the nerve cells and reduce the neurological damagee.
