1.Selective Inhibitory Effect of Sinomenine on Activity of Cyclooxygenase 2

[Objective] To explore the anti - inflammatory mechanism of sinomenine, a monomer alkaloid from Caulis Sinomenii. [ Methods] In - vitro enzyme was applied to observe the effect of sinomenine on cyclooxygenase (COX) 1 and COX 2. [Results] Sinomenine at the concentration of 1 - 250 ?mol/L could inhibit the formation of PGE2 induced by COX 2 in a good dose - effect manner. [Conclusion] Sinomenine selectively inhibiting the activity of COX 2 may be the anti - inflammatory and anagesic mechanism of sinomenine with less gastrointestinal adverse effect.
2.Establishment and Identification for Macrophage-derived Foam Cell Model of THP-1 Cell Line

Objective To establish a macrophage-derived foam cell model of human monocytic THP-1 cell line and to identify the foam cell.Methods THP-1 cells were differentiated into macrophages after induction with 160 nmol/L phorbol-1-myristate-13-acetate for 24 hours and then incubation with 80 mg/L oxidized low density lipoprotein(oxLDL) for 48 hours.The differentiated cells were observed after oil red O staining under light microscope.High performance liquid chromatography/atmospheric pressure chemical ionization tandem mass spectrometry(HPLC-MS) was used for quantitative analysis of cellular cholesterol and cholesteryl esters contents.Results There was a large number of red lipid droplets in phorbol-1-myristate-13-acetate and oxLDL treated THP-1 cells after oil red O staining under light microscope.Cellular contents of cholesterol and cholesteryl esters in the differentiated cells increased markedly compared with the normal THP-1 monocytes by HPLC-MS assay.Conclusion A human monocyte-derived foam cell model has been established by incubating THP-1 cells with 120nmol/L phorbol-1-myristate-13-acetate for 24 and then with 80 mg/L oxidized low density lipoprotein for 48 hours.
3.Studies on In-Vitro Biological Activity of Albiflorin R1 in Counteracting Cell Proliferation

[Objective] To study the in-vitro biological activity of albiflorin R1 (AR1), a new monoterpene glycoside from Paeonia lactiflora,in counteracting cell proliferation. [Methods] Two kinds of human cancer cell strains (HeLa cells and K562 cells) and J774 macrophage cell strain were cultured in vitro to observe the effects of albiflorin Rl on cell proliferation. [Results] Albiflorin Rl could markedly inhibit the proliferation of J774 macrophages and also inhibit the proliferation of HeLa and K562 cell stains in vitro. [Conclusion] Albiflorin Rl may have an anti-neoplastic, anti-inflammatory and immunosuppressive activity.
4.The clinical efficacy of olecranon osteotomy approach and bilateral approach through triceps brachii for the treatment of intercondylar humeral fractures

ObjectiveSystematically evaluate the clinical efficacy and complications of olecranon osteotomy and bilateral approach through triceps brachii for the treatment of intercondylar humeral fractures. MethodsTo search the domestic and foreign literatures published from January 1980 to March 2008 about the treatment of intercondylar humeral fractures, which use olecranon osteotomy approach and bilateral ap-proach through triceps brachii. Systematically evaluate these literatures using the methods of systematic re-view provided by the Cochrane collaboration. ResultsAfter the screening and evaluation to the literatures, 33 published literatures that met the inclusive criteria of this study were collected. There were a total of 876 cases of patients whose follow-up data was integrity (olecranon osteotomy approach group for 509 cases;bilat-eral approach through triceps brachii group for 367 cases) and these cases had provided the postoperative scores of elbow joint function. The excellent and good rates of postoperative score of elbow joint function for the type C2 and C3 fracture patients in the olecranon osteotomy approach group were 82.32% and 79.38% respectively; while those in the bilateral approach through triceps brachii group were 70.59% and 64.71% respectively. The former was significantly higher than the latter. The excellent and good rate of the type C1 fracture patients between two approaches groups showed no statistical significant. Fourteen literatures provid-ed the data about postoperative complications, the incidence of complications of patients that received ole-cranon osteotomy approach was 4.72%(95%CI 4.70%-4.74%), and the results also showed that compared with bilateral approach through triceps brachii (8.17%, 95%CI 8.13%-8.21%). ConclusionTo treat inter-condylar humeral fractures, postoperative efficacy of olecranon osteotomy approach is better than that of the bilateral approach through triceps brachii. Using olecranon osteotomy approach can get adequate exposure, easy open reduction and internal fixation, quick recover and less complication.

Objective: To establish a new method for the identification of Schisandra chinensis(Turcz.) Baill. (Beiwuwei) and Schisandra sphenanthera Rehd. Et Wils. (Nanwuwei). Methods: Random amplified polymorphic DNA method was applied to screen random primers. Results:Screening from 80 primers,only S429, which can be used to identify Schisandra chinensis(Turcz.) Baill.(Beiwuwei) and Schisandra sphenanthera Rehd. Et Wils. (Nanwuwei) accurately and is of good reproducibility. Conclusion: S429 can be used to identify Schisandra chinensis(Turcz.) Baill. (Beiwuwei) and Schisandra sphenanthera Rehd. Et Wils. (Nanwuwei) accurately.
6.Effects of Dachengqi Decoction on Systemic Inflammatory Response Syndrome in Mice

[ Methods ] Mouse models induced by zymosan A were administered with DCD orally. Serum endotoxin level was detected by limulus assay and TNF-? and IL-6 levels by radioimmunoassay 6, 12 and 24 hours after medication. 【Objective】To establish a simple,rapid and practical method for the in-vitro isolation,purification and proliferation of rat marrow mesenchymal stem cells(MSCs).【Methods】Rat MSCs were isolated and purified by differential adhesion method.The effects of conditioned medium on the morphologic characteristics and growth of rat MSCs were observed.【Results】Rat MSCs were effectively purified by differential adhesion method,and grew well with good proliferation and normal morphology in conditioned medium.【Conclusion】Differential adhesion method and conditioned medium are effective for the purification and culture of rat MSCs.
9.Effects of sinomenine on IL- 1?, LL-8 gene expression of human peripheral blood

Objective:To study the anti-inflammatory and anti-rheumatic mechanisms of sinomenine, a pure alkaloid extracted from the Chinese medical plant sinomeniuim acutum, on cytokines-related genes modulation. Methods: Cultured normal human peripheral blood mononuclear cytes(PBMIN) with sinomenine in vitro, isolated total RNA from different groups of treated and controlled cells to investigate the effects on mRNA expression of IL-1?, LL-8 with LPS stimulation by one-step and two-step RT-PCR techniques. Results: The results from the one-step RTPCR showed that the inhibition ratio on IL-l?mRNA expression in the group of mmol/L sinomenine was about( 15.7?5.52) % ; with two-step RT-PCR techneque demonstrated that the inhibition ratio of sinomenine in the group of sinomenine with the concentration of 1 mmol/L, 100 umol/L on IL-1?mRNA expression were ahout( 17.07 ? 7 .69)% ,(25 .99 ? .84)% respectively and the inhitition ratio in the group with sinomenine concentration of 1 mmol/L,100umol/L on IL-8mRNA expression were(2l.01 ?7.79)%,(16.04?7.55)% respectively .Conclusion: The therapeutic effects of sinomenine on rheumatoid arthritis(RA) may partially related to downregulating gene expression of IL-1?, IL-8 cytokines of the peripheral blood mononuclear cells.
10.Inhibition of clotrimazole on Paneth cell defensins

Aim To investigate the Clotrimazoles (CLT) inhibitive effect on the secretion of ?-defensins of the Paneth cells.Methods The ?-defensin mRNA expression is determined by semi-quantitative RT-PCR. The secretion of defensins is determined by dot-blot and ELISA. Results CLT has no influence on the defensin mRNA expression. All of 125 nmol?L -1, 250 nmol?L -1 and 500 nmol?L -1 CLT can inhibit the secretion of ?-defensins. Conclusion CLT could inhibit the secretion of defensins, which suggests that CLT maybe influence the function of small intestine
