Objective:To probe into the mechanism of Shenshuai Capsule in delaying advance of chronic renal failure(CRF)and against interstitial renal fibrosis.Methods:74 cases of CRF at decompensation phase were randomly divided into a treatment group and a control group.Changes of their serum urea nitrogen(BUN),creatinie(SCr),endogenous creatinine clearance rate(Ccr),collegen Ⅳ (Col Ⅳ),procollagen Ⅲ(PC Ⅲ),laminin(LN),and improvement of urinary protein(uPro),urinary ?_2-microglobulin(?_2-MG)and clinical symptoms and signs were observed.Results:After treatment of Shenshuai Capsule,BUN,SCr,Col Ⅳ,PC Ⅲ,LN,urinary protein and ?_2-MG decreased and Ccr increased in the treatment group,with a significant differences between the two group in the above mentioned indexes except LN(P
2.Role of adenovirus-mediated cardiac-specificRIP140 overexpression in cardiac function and inflammation pathway

Aim To explore the role of cardiac-specific overexpression of RIP140 in cardiac function and inflammation signaling pathway.Methods Direct intra-myocardial injection of adenovirus vector expressing RIP140 drove transgene expression in heart tissue.RIP140 overexpression was confirmed using immunofluorescence technique in heart.Cardiac function was assessed by echocardiographic and hemodynamic assessment.TNF-α,IL-2 and IL-1β inflammatory cytokines were detected by ELISA,and the protein levels of p65 and IκB-α were measured by Western blot.Results Adenovirus-mediated foreign gene of RIP140 was successfully transferred in cardiac tissue.RIP140 overexpression in heart induced ventricular dilation,decreased left ventricular ejection fraction and cardiac malfunction,as well as increased releases of TNF-α,IL-2 and IL-1β inflammatory cytokines,p65 protein translocation into the nucleus and IκB-α protein degradation in cytoplasm.Conclusion Adenovirus-mediated RIP140 overexpression in cardiac tissue impairs cardiac function,activates NF-κB/p65 inflammatory signaling pathway and induces the release of inflammatory cytokines.
3.Influence of Invigorating Kidney and Activating Blood Formulae on Expression of IL-6 and MMP-13 in Synovial Fluid of Rabbits with Knee Osteoarthritis

Objective To investigate the expression levels of Interleukin(IL)?6 and matrix metalloproteinases(MMP)?13 in synovial fluid by in?vigorating kidney and activating blood formulae in treating rabbit knee osteoarthritis model. Methods A total of 30 New Zealand rabbit were ran?domly divided into blank group,model group,invigorating kidney group,activating blood group and invigorating kidney and activating blood group. Rabbits model with knee osteoarthritis were established by improved Hulth method. To give corresponding respectively the medicinal broth,model group was given saline,knee joint synovial fluid was collected after 4,8 and 12 weeks. Enzyme linked immunosorbent assay(ELISA)was used to measure the levels of IL?6 and MMP?13. Results The levels of IL?6 in rabbit knee osteoarthritis were obviously higher than that of normal control group at both 4 weeks and 8 weeks(P<0.001). But there was no statistical difference on the levels of IL?6 compared with controls in 12 weeks. In addition,the level of MMP?13 at 4 weeks,8 weeks and 12 weeks were significantly higher than the blank control group(P<0.001). After 8 weeks of Chinese medicine administration,the levels of IL?6 in synovial fluid were significantly decreased in invigorating kidney group,activating blood group and invigorating kidney and activating blood group(P<0.001),but there was no statistical difference among groups in 12 weeks. The MMP?13 levels of synovial fluid was significantly lower than the model group(P<0.001). Conclusion Our results indicate that IL?6 and MMP?13 par?ticipate in the pathological development of the rabbit knee osteoarthritis. Invigorating kidney and activating blood formulae could reduce the expres?sion of IL?6 and MMP?13 and alleviate osteoarthritis progression,and which is superior to the pure invigorating kidney formulae and activating blood formulae.
4.Effectiveness and safety of high frequency electronic associating stents insertion and subsequent radiotherapy guided by fiberoptic bronchoscope in treating malignant tracheobronchial stenosis

ObjectiveTo evaluate the effectiveness and safety of high frequency electronic associating stents insertion and subsequent radiotherapy guided by fiberoptic bronchoscope in treating tracheobronchial stenosis caused by lung and esophagus carcinoma.MethodsFifty-two patients which were ascertained with serious stenosis in trachea or bronchi by fiberoptic bronchoscope were subjected to the study.Fiberoptic bronchoscope was used to investigate the location,extent,blood supply and the degree of tracheobronchial stenosis before the study.After the focus in the airways was cleared up with high frequency electronic,electrocoagulation and snare under the guide of the euthyphoria of fiberoptic bronchoscope,Ni-Ti memory alloy stents were placed into the stenosis airways.The 52 patients were divided into two groups with 26 cases each:the stent group and the stent associating radiotherapy group(radiotherapy group).The later group accepted subsequent radiotherapy after the therapy.The following indexes of the two groups were investigated:short-time clinical effect,dyspnea index class,rate and average time of airway restenosis,life span and survival rate.ResultsAfter therapy,the 52 patients had obvious improvements in dyspnea and the diameter of the stenosis airways.The short-time total effective rate was 100.0% (52/52),but there was not significant difference between the two groups(P> 0.05 ).After therapy,there was significant difference in two groups in dyspnea index class(P < 0.01 ).Although there was not significant difference in the rate of airway restenosis between the two groups(P>0.05),the average time of airway restenosis in the radiotherapy group was obviously longer than that in the stent group(P< 0.01 ).The survival rate of the radiotherapy group in the 6th and 9th month after therapy was higher than that in the stent group (P < 0.05),but there was no significantdifferenee in the 3rd and 12th month (P >0.05).ConclusionHigh frequency electronic associating stents insertion and subsequent radiotherapy guided by fiberoptic bronchoscope is an effective and safe treatment for the patients suffering tracheobronchial stenosis caused by lung and esophagus carcinoma.
5.Morphology and proliferation of bone marrow mesenchymal stem cells and expressions of CD34 and CD44 under stomach cancer microenvironment

BACKGROUND:Gastric cancer mesenchyal stem cel s from clinical stomach cancer specimens and tumorigenic tissues in nude mice are similar to the bone marrow mesenchymal stem cel s in biological characteristics, which have been proved to be an important component of tumor microenvironment to promote tumor growth. It is speculated that biological characteristic of bone marrow mesenchymal stem cel s may change in stomach cancer microenvironment. OBJECTIVE:To observe the effect of stomach cancer microenvironment on morphology and proliferation of bone marrow mesenchymal stem cel s and expressions of CD34 and CD44. METHODS:Rat bone marrow mesenchymal stem cel s were cultured alone as control group. In the test group, rat bone marrow mesenchymal stem cel s were co-cultured with human stomach cancer BGC-823 cel s using Transwel chamber assay to establish the stomach cancer microenvironment. Then, cel morphology, proliferation, cel cycle and CD34, CD44 expressions were observed and detected using inverted phase contrast microscope, MTT assay, and flow cytometry, respectively. RESULTS AND CONCLUSION:In the test group, bone marrow mesenchymal stem cel s were similar to human stomach cancer cel s BGC-823 that arranged disorderly and irregularly, were interconnected loosely, became thinner and longer, and grew in clusters with smal er nuclei. The cel proportion in G 1 phase significantly decreased, but that in S and G 2/M phases significantly increased (P<0.01, P<0.05). The positive rate of CD44 significantly declined, and the CD34 expression significantly raised (P<0.01). In conclusion, stomach cancer microenvironment by non-contact co-culture with BCG-823 cel s has an obvious effect on the morphology, proliferation and surface antigens expressions of bone marrow mesenchymal stem cel s that wil tend to be malignant gastric cancer cel s.
6.In vitro study of inhibitory effect of Ulinastatin against serine protease activity

Objective To study inhibitory effect of serine protease activity by Ulinastatin in vitro .Methods Different chromogenic peptides were designed and synthesized.Highly sensitive fluorescence detection was performed to optimize the concentration of each serine proteases and their chromogenic substrates.Multi-point method was used for the calculation of half maximal inhibitory concentration of Ulinastatin .ResuIts Ulinastain could inhibit Polymorphonuclear leukocyte elastase ( PMNE ) and plasmin with IC50 lower than 100 U/mL.For factor Xa, and Kallikrein, the IC50 of Ulinastatin was higher than 1000U/mL.No thrombin IC50 could be calculated at the present experiments.ConcIusion Similar to Ulinastatin injection from Japan, domestic Ulinastatin shows the strongest inhibitory effects on PMNE among those serine proteases.As important references, this study gives reliable data for dose range of domestic Ulinastatin in anti-inflammation, coagulation/anti-coagulation and anti-shock therapy.
7.A retrospective study of the treatment for epithelial-myoepithelial carcinoma

Objective To evaluate the risk of locoregional recurrence (LRR) and the influencing factors for long-term survival in patients with epithelial-myoepithelial carcinoma (EMCa).Methods A retrospective analysis was performed for 18 EMCa patients, who received initial therapy or initial adjuvant therapy in our hospital from 1999 to 2015, to investigate their survival.Among these patients, 8(44%) underwent surgery alone, 9(50%) received adjuvant radiotherapy, and 1(6%) received radical concurrent chemoradiotherapy.Locoregional recurrence-free survival (LRFS) and overall survival (OS) rates were compared between these groups.The Kaplan-Meier mtthod was used to calculated survival rates and log-rank test was used to compare the LRFS.Results With a median follow-up time of 46 months, 5 patients developed LRR, and the 5-year LRFS and OS rates were 69% and 93%, respectively.The patients treated with radiotherapy had a significantly higher 5-year LRRFS rate than those not treated with radiotherapy (71% vs.57%, P=0.569).Conclusions LRR is the main failure mode of EMCa treatment, and further improving local control is the key to improved survival.
8.Rhubarb extract protects ischemia-reperfusion injury in mice through the antioxidative mechanism

Objective To investigate the neuroproteetive effect of Rhubarb extract (RE) on cerebral ischemia-reperfusion injury in mice and its mechanism.Methods Twenty-eight male ICR mice were randomly divided into sham operation,ischemia-reperfusion,low-dose RE (100 mg/kg),and high-dose RE (100 mg/kg) groups.A model of middle cerebral artery occlusion and reperfusion in mice was induced by the suture method.The drug intervention groups were given intragastric RE administration (once a day) on the third day before model preparation,and the same volume of normal saline was injected into mice of the cerebral ischemia-reperfusion group.The volume of cerebral infarction was detected by triphenyltetrazolium chloride staining.The neuron-specific nuclear protein (NeuN),glial fibrillary acidic protein (GFAP),and ionized calcium binding adapter 1 (IBA-1) were used as markers of the ischemic cortical neurons,astrocytes,and microglial cells,respectively,and detected by inmunohistochemistry.The expression levels ofsuperoxide dismutase (SOD)-1,SOD-2,and catalase (CAT) in ischemic cortex were detected by Western blot analysis.Results Compared with the cerebral ischemia-reperfusion group,the neurological function score of the high-dose RE group was significantly reduced,the infarct volume was significantly reduced,and the number of neurons in the ischemic cortex was increased significantly,and the activation degree of astrocytes and microglia was decreased significantly (all P <0.05),the expression levels of SOD-1,SOD-2,and CAT were increased significantly (all P< 0.05);in contrast,there was no significant difference between the low-dose RE group and the cerebral ischemia-reperfusion group.Conclusions High-dose RE may play a protective role for cerebral ischemia-reperfusion mice through anti-oxidative mechanism.
9.JNK and Akt genes in pancreatic tissue of diabetic rats and the effects of protein expression and phosphorylation modification

Objective To observe the effects of Yutang pill on pancreatic tissue, JNK, Akt gene, protein expression and phosphorylation of diabetic rats. Methods A total of 150 male SD rats were divided into blank group, model group, western medicine control group, Chinese medicine control group, Yutang Pill low dose group and Yutang pill high dose group according to random number table, 20 rats in each group. In addition to the blank group, the other groups of rats were prepared for the diabetes model. The western medicine control group was intragastrically administered with metformin hydrochloride suspension 0.09 g/kg, the Chinese medicine control group was administered with the suspension of glycoside tablets 2.43 g/kg, and the low-dose and high-dose groups of Yutang pills were respectively administered with Yutang pills 1.35 and 2.70 g/kg. The blank group and the model group were intragastrically administered with equal volume of saline. 1 time/d, 8 weeks. After continuous administration, the material was taken. The expression levels of JNK and Akt in pancreatic tissue were detected by Real-time PCR. The expression of JNK and Akt protein and phosphorylation in pancreatic tissue were detected by Western blot. Results Compared with the model group, the expression of JNK mRNA (0.57 ± 0.07, 0.95 ± 0.13 vs. 1.14 ± 0.19) and P-JNK/JNK (0.222 ± 0.038, 0.817 ± 0.104 vs. 1.140 ± 0.136) in the low- and high dose group significantly decreased (P<0.01). The expression of Akt mRNA (2.09 ± 0.13 vs. 1.63 ± 0.12) and P-Akt(Ser473)/Akt (0.385 ± 0.072 vs. 0.130 ± 0.027) significantly increased (P<0.01). Conclusions The Yutang pill can reduce the expression of JNKmRNA in pancreatic tissue of T2DM rats and inhibit the phosphorylation of JNK protein. The expression of AktmRNA increased and the phosphorylation level of Akt protein increased.
10.Clinical Feature Analysis of 525 Cases of Patients with Ulcerative Colitis

Objective: To conduct retrospective analysis of clinical features of patients with ulcerative colitis in JiangsuProvince Hospital of Traditional Chinese Medicine (TCM) . Methods: Ulcerative colitis medical records in jiangsuprovince hospital of TCM were selected and used to retrospectively analyze the feature such as gender, age, course of thedisease, clinical type, stage, severity, and pathological parts. Results: The study include 290 cases of male and 235 casesof female with the ratio of 1.2:1. Peak incidence was between 20 to 60 years old. Dietary factor was the most commoncause, followed by fatigue, environment, medicine and emotional factors. There were 28 (5.33%) patients at remissionstage, and 497 (94.67%) patients at active stage. In patients at active stage, there were 78 (15.69%) patients newlydiagnosed, and 419 (84.31%) patients with chronic recurrent type. There 92 (18.51%) rectum type patients, 156 (31.39%) left half colon type patients and 249 (50.10%) extensive colon type patients. There were 275 (55.33%) mild patients, 149 (29.98%) moderate patients and 73 (14.69%) severe patients. The main symptoms were bloody purulent stool, diarrheaand abdominal pain. There were 375 (71.43%) patients with bloody purulent stool, and symptom disappearance rate was89.07% after treatment. There were 335 (63.81%) patients with diarrhea, and symptom disappearance rate was 71.64%after treatment. There were 319 (60.76%) patients with abdominal pain, and symptom disappearance rate was 85.89%after treatment. There were 3 cases of complications. Drug therapy was mainly based on integrated Chinese and westernmedicine (86.29%) . For Chinese medicine treatment, there were 453 patients (87.45%) with oral Chinese medicine, 401 patients (77.41%) with Chinese medicine enema, and 10 patients (1.93%) with foot bath. For western medicine treatment, there were 392 patients (75.68%) treated with oral amino salicylate drugs, 100 patients (19.31%) with anal used aminosalicylate drugs, 88 patients (16.99%) with glucocorticoid, 18 patients (3.47%) with immunosuppressive agent and 441 patients (85.14%) with microecological preparation. Conclusion: The types of ulcerative colitis patients in our hospitalwere mainly chronic recurrent type at active stage with mild to moderate and extensive colon type. The treatment wasmainly based on integrated Chinese and western medicine, with significant improvement in symptoms of bloody purulentstool, diarrhea and abdominal pain. 99.61% patients were discharged with a better condition
