Nuclear receptors(NRs)superfamily signaling plays an important role in regulating the expression of gene involved inenergy homeostasis. The nuclear receptors regulate target gene expression through the recruitment of coregulatory proteins, which act as scaffolds or plateforms for linking NRs with enzyme complexes that modify DNA and histones. As a transcriptional co-repressor for NRs,RIP140 negatively regulates the transcription of target gene in metabolic tissue, such as adipose tissue, muscle and liver. In the absence of RIP140,genes expressions involved in metabolic pathways were upregulated including glycolysis, triglyceride synthesis,TCA cycle, fatty acid ? oxidation, mitochondrial electron transport and oxidative phosphorylation.RIP140 may be a candidate therapeutic target for metabolism syndromes.

Aim The limited transfection efficiency for plasmid in primary neonatal rat cardiomyocytes, which are terminal differentiated cells, and long foreign DNA (the RIP140 gene sequence are as long as 3.5 kb) cause us to choose a better system to study RIP140 gene expression in primary non-replicative cells. Methods Full-length of RIP140 was cloned into pAdTracker-CMV shuttle vector, and then recombined with virus backbone pAdEasy-1 vector in BJ5183 bac-teria.Positive recombinant plasmid was confirmed by sequence analysis and restriction enzyme determina-tion,and then transfected into AD293 cells for amplifi-cation.Titers of virus particles were determined by Tis-sue Culture Infectious Dose 50 (TCID50 )method and cell vitality was analyzed by CCK-8 kit in cardiomyo-cytes.RIP140 gene was identified by Western blot. Results Sequence analysis suggested that full-length RIP140 gene was cloned correctly into AdEasyTM sys-tem.Virus titers of Ad-RIP140 and Ad-GFP were 1011.3 and 1011.7 PFU·mL-1 ,respectively.Cell vitali-ty was not affected when the Multiplicity of Infection (MOI)was lower than 200.Green fluorescent protein (GFP)and Western blot analysis showed RIP140 gene was remarkably increased in cardiomyocytes for 12h in-fection by Ad-RIP140 (P<0.05 ).Conclusion Re-combinant adenovirus containing RIP140 gene was suc-cessfully constructed and effectively expressed in car-diomyocytes.These will be helpful for further research on the function of RIP140 in cardiomyocytes.

BACKGROUND: Without any external stimuli in natural environment, it is applicable to observe the infrared radiant track over human body surface that is in conformity basically with the running courses of ancient fourteen meridians.OBJECTIVE: To probe into the influence of electroacupuncture on infrared radiant track along meridian courses over human body surface.DESIGN: Self-controlled opening experiment.SETTING: Department of Meridian Research of Academy of Traditional Chinese Medicine in Fujian Province.PARTICIPANTS: The experiment was performed in Key Laboratory of Meridian Research of Fujian Province from January 2000 to December 2002. Totally 53 students from Department of Acupuncture and Massage of Fujian College of Chinese Medicine were observation, of which, 38 students were males, 15 students were females, aged varied from 19 to 23years.METHODS: The receptors exposed completely the body areas required and sat quietly for 20 to 30 minutes to adapt to the environmental temperature. In natural state, 6T67 infrared thermo tracer produced by Japanese San-Ei Company was used to observe 1 to 2 infrared radiant tracks in advance as the control before acupuncture (scanned once per second). Afterwards, acupuncture was done on Guangming (GB 37), Neiguan (PC 6) and Dazhui (GV 14). The stimulation was given by a pair of stainless filiform needles, 6.6 cm long. The frequency of effectric pulse was 2 Hz, and 0.2 ms in duration. The video record was taken once every 5 to 10 minutes during stimulation. After acupuncture, 1 to 2 infrared radiant tracks were scanned.MAIN OUTCOME MEASURES: Tracks of isothermal display and whole thermal display on infrared thermography.infrared radiant track along meridian courses over the human body surface high basic temperature, acupuncture decreased skin temperature on infrared radiant track along meridian courses over the human body surface in a part of subjects. The skin temperature of a part of subjects was decreased acupuncture increased skin temperature on infrared radiant track along though it did not great affect the skin temperature on infrared radiant track along meridian courses over the human body surface in a part of subjects, acupuncture induced infrared radiant track to be more continuous and human body surface, acupuncture stimulates the infrared radiant track displayed along the meridians over human body surface, too.CONCLUSION: Infrared radiant track along meridian courses over the human body surface is a kind of normal life phenomenon existed commonly among peoples. Acupuncture not only induces the infrared radiant track of the related meridian, but also alters the skin temperature of the displayed infrared radiant track along meridian courses and makes it become more continuous and regular. Changes in tissue temperature are relevant necessarily with altered energetic metabolism. It is suggested that infrared radiant track along meridian courses is closely contributed to functional regulation of the human body.

BACKGROUND: It is reported that ceramide signal pathway may play an important role in the apoptosis of vascular endothelial cell and then lead to the progress of atherosclerosis, such as the formation of foam cells and the proliferation of smooth muscular cells.OBJECTIVE: To study the changes of the sphingomylinase activity and ceramide content in aorta of rabbits with experimental atherosclerosis and investigate the regulative effects and mechanism of emodin on them as compared with positive fenofibrate.DESIGN: Completely randomized controlled design.SETTING: Laboratory of Pharmacology & Toxicology, School of Pharmaceutical Science of Sun Yat-sen University.MATERIALS: The experiment was completed at the Laboratory of Pharmacology & Toxicology, School of Pharmaceutical Science of Sun Yat-sen University from July to December 2003. Totally 48 New Zealand male rabbits were selected. Forty animal models of atherosclerosis were made with high cholesterol feed, and the other 8 rabbits were selected as the normal controls. Model animals were divided randomly into model group, 5 mg/kg emodin group, 10 mg/kg emodin group, 20 mg/kg emodin group and 25mg/kg fenofibrate group with 8 in each group.METHODS: At the seventh weeks of model duplication, 5 mg/kg, 10 mg/kg and 20 mg/kg emodin were perfused in rabbits of emodin groups respectively, and 25 mg/kg fenofibrate was perfused in rabbits of fenofibrate group. Emodin and fenofibrate were diluted or suspensed with 2 mL saline once per day respectively. Rabbits in normal control group and model group were administrated with the same volume of saline for 4 weeks. The rabbits were raised separately and were fed with 135-150 g food per day.MAIN OUTCOME MEASURES: [1] The area of the lipid plaque in aortal intima; [2] the content of serum TC and TG; [3] SOD activity and MDA content; [4] SMase activity and CER content in aorta.RESULTS: Totally 48 rabbits entered the final analysis. [1] Area of the lipid plaque: Area of the lipid plaque was (48.87±15.5) % in the model group, which was larger than that in each emodin group (P < 0.05 or 0.01),especially larger than that in the 10 mg/kg emodin group (22.19±12.9)%while that in the fenofibrate group was similar to that in the model group (P > 0.05). [2] Content of serum TC and TG: The anrtal intima of control was smooth. Content of serum TC and TG in each emodin group were similar to those in the model group (P > 0.05), but those in the 25 mg/kg fenofibrate group were lower than those in the model group (P < 0.05). [3]Content of SOD and MDA in plasma: SOD activity of rabbits in each emodin group was higher than that in the model group (P < 0.05, P < 0.01),but the MDA activity in the 10 mg/kg and 20 mg/kg emodin group was lower than that in the model group (P < 0.05). The MDA activity in the25 mg/kg fenofibrate group was similar to that in the model group (P > 0.05).[4] Content of SMase and CER: Those in the model group were higher than those in the normal control group, but those in the 5, 10 and 20 mg/kg emodin groups were lower than those in the model group; those in the 25 mg/kgfenofibrate group were similar to those in the model group (P > 0.05). [5]Analysis of correlation: Content of SMase was in positive relation with blood cholesterol (r=0.542, P < 0.01), in positive relation with blood MDA (r=0.789, P > 0.01), and in negative relation with blood SOD(r=-0.936, P > 0.01); content of CER was in positive relation with blood cholesterol (r=0.433, P > 0.05), in positive relation with blood MDA (r=0.673, P > 0.01), and in negative relation with blood SOD (r=-0.876, P > 0.01).CONCLUSION: The study finds that emodin, despite its insignificant effects on decreasing TG or TC, can protect vascular endothelial cells and reduce the area of lipid-laden plague of aortal intima by antioxidation, inhibition of the sphingomyelinase activity and reduction of the content of ceramide. It is suggested that moderate dosage of emodin employed in the study may be most appropriate to atherosclerosis treatment.

AIM: To study the roles and mechanisms of ERKs and intracellular free calcium in cardiomyocyte hypertrophic response induced by endothelin-1(ET-1). METHODS: (1) Neonatal rat cardiomyocyte hypertrophic response was assayed by measuring cell surface area and protein content; (2) ERKs activity was determined by Whatman Paper Filter method; (3) Intracellular free calcium concentration ([Ca2+]i) was measured using Fura-2/AM as a fluorescent indicator. RESULTS: (1) ET-1 could increase total protein production, surface area, ERKs activity and [Ca2+]i in cultured cardiomyocyte in dose-dependent manner at concentrations ranging from 10-9 to 10-7 mol/L. And this effect could be abolished by BQ123, an antagonist of ETA receptor, partly inhibited by PTX, but not by BQ788, an antagonist of ETB receptor.(2)The activation of ERKs and the increase of [Ca2+]i induced by ET-1 were obviously inhibited by PD98059, a selective ERKs kinase inhibitor, and nifedipine, a calcium channel blocker, respectively. Both antagonists partially inhibited ET-1-stimulated cardiomyocyte hypertrophic response. (3) Staurosporine, a selective PKC inhibitor, could inhibit ET-1-stimulated cardiomyocyte hypertrophic response and increase of [Ca2+]i, but not affect the activation of ERKs. CONCLUSION: Cardiomyocyte hypertrophic response induced by ET-1 is mediated by ETA receptor coupled to PTX-sensitive G-protein, which involves at least two signalling pathways: PKC-mediated increase of [Ca2+]i , and PKC-independent activation of ERKs.
