We determined the nucleotide sequences of enzymatically amplified second exons of DRB1and DRB3 genes derived from three Chinese DRw12 homozygous cells and found two distinctDRw12 haplotypes.The combinations between DRB1 and DRB3 genes of the two haplotypeswere different from that in Caucasoid DRw12 HTCa.Thise result might explain the consquenceof generating“new”Dw specificities by cellular assay.

Colon cancer possesses hypoxia microenvironment. The essence of hypoxia is a kind of reaction and adaptive change. Hypoxia reaction plays an important role in the genesis and development of colon cancer,including the effect produced by hypoxia inducible factor-let and reactive oxygen species under hypoxia reaction and influence on cell apoptosis and energy metabolism directly generated by hypoxia reaction in colon cancer. The alteration of hypoxia reaction in colon carcinoma as tumor adjunctive therapy becomes focus research in the field of colon cancer.

Objective: To understand the situation of mosquito larva breeding and adult mosquito density among schools in Yangpu District, Shanghai Municipality before the start of the fall semester, so as to provide insights into mosquito prevention and control in schools. Methods: A stratified sampling method was used to select four schools each from universities, primary and secondary schools, and preschool institutions as survey sites. A survey of mosquito larva breeding sites and adult mosquito density was carried out in August each year from 2021 to 2023. The mosquito larva breeding sites in small and large water bodies were investigated by visual inspection and spoon trapping methods, and adult mosquito density was investigated by human-induced stinging method. The positive rate of mosquito larva breeding sites and adult mosquito landing index were analyzed. Results: Totally 1 253 mosquito larva breeding sites were investigated from 2021 to 2023, and the positive rate was 4.07% (51 positive sites). The positive rates of mosquito larva breeding sites in universities, primary and secondary schools and preschool institutions were 2.36%, 4.05% and 8.30%, respectively. The top three positive breeding sites in small water bodies were tyres (6.67%), small stagnant water in green belts (6.04%) and unused containers (5.92%), and Aedes albopictus was the dominant species (97.21%). The top three positive breeding sites in large water bodies were rainwater wells (3.90%), landscape water bodies (3.45%) and sewage wells (3.23%), and Culex pipiens pallens and Aedes albopictus each accounted for 50.00%. A total of 200 adult mosquitoes were captured from 2021 to 2023, with Aedes albopictus being dominant (93.00%). The total landing index decreased and then rebounded slightly from 2021 to 2023, with the highest seen in 2021 as 1.78 mosquitoes/0.5 hours per person. The highest mean adult mosquito landing index was 1.17 mosquitoes/0.5 hours per person in preschool institutions. Conclusions The positive rates of mosquito larva breeding sites and landing index of adult mosquitoes among schools in Yangpu District before the start of the fall semester from 2021 to 2023 were higher than the relevant provisions of the Shanghai municipal local standards. Aedes albopictus was the dominant species. Mosquito surveillance in schools should be carried out on a regular basis and hygiene management should be strengthened.

Objective To express human thyroid peroxidase (hTPO) epitopes gene and apply its products in clinical assay. Methods hTPO epitopes gene was cloned into expression vector pGEX 4T 3 then transformed into E. coli BL21. Expression of hTPO gene was induced by isopropyl ? D thiogalactoside, expressed product (GST hTPO) was purified by affinity chromatography and their immunoactivity was demonstrated. ELISA technique using GST hTPO as antigen was established for determining TPOAb. Serum TPOAb level was determined, and HLA DR antigen, dendritic cells and lymphocytes in the thyroid gland tissue were observed in these same AITD patients.Results GST hTPO acquired from procaryotic expression had high purity and good immunoactivity. The CVs of the ELISA technique established with GST hTPO were between 5.93%～7.59%. A significantly positive correlation was found between the TPOAb levels determined respectively by ELISA and RIA method. Serum TPOAb level and distribution of HLA DR antigen and dendritic cells showed the same ascending tendency following the aggravation of lymphocyte infiltration. Conclusion Product of genetic engineering, GSH hTPO, can be used to establish a clinical assay for TPOAb. The correlation between the level of serum TPOAb and the detriment of thyroid tissue is demonstrated.

Objective This work aimed at study the rel ationship between the pathologicomorphological change of gastrocneminus muscle cells and deep venous insufficiency (DVI).Methods Patients with DVI in three different stage of developing process are grouped into early t eam?middle team and late team group ,each group consists of 9 cases.HE stain , special ATPase histoenzymic methds was quantitatively employed to study histo chemical cytomorphology of gastrocneminus and musculus sartorius specimens in th ose subjects.Results There is no significant difference of the pathologicomorphological change,either between early-stage group and mid-sta ge group or between early-stage group & mid-stage and control group.But there is significant difference comparing advanced stage group and early-stage group & m id-stage group or between advanced-stage and control group.Conclusion As the process of DVI develops,marked morphological degeneration can be found in gastrocneminus muscle cells.It could be regarded as the pathogenic fundament of impairment of calf muscle pump and development of DVI.

Objective:To investigate if DQB1 promotor allele polymorphism is associated with the susceptibility to unexplained habitual abortion (UHA).Methods:Polymerase chain reaction-sequence specific oligonucleotide (PCR-SSO) genotyping method was performed to detect HLA-DQB1 promotor (QBP) alleles in 32 cases of unexplained habitual abortion and 53 women with normal pregnancy history.Results:The frequency of QBP alleles was not different between two groups. The frequency of QBP6 2-DQB1*0604/0605 haplotype was 12 5% in patients group and 2 83% in control group, which got statistical significance. Meanwhile, six new haplotypes were first observed in Chinese population.Conclusion:No direct association was found between QBP polymorphism and UHA, but QBP-DQB1 haplotype might play a role in the susceptibility to UHA.

Liver is the main target organ of the metastasis of colorectal cancer.More and more researches demonstrate that including micro-environment of cancer,signal transduction pathway,abnormality of chromosomes and the gene which control cell proliferation and apoptosis are the most important mechanisms in liver metastasis of colorectal cancer.The metastasis of colorectal cancer is the result of all these factors.To study the mechanism can shed some light on prevention and therapy in liver metastasis of colorectal cancer.

Objective To investigate T cell receptor(TCR)variable β(BV)chain usage at the maternal-fetal interface and explore the relationship between the skewed TCR BV usage and unexplained recurrent spontaneous abortion(BSA).Methods Eighteen cases with unexplained RSA,together with matched 41 women with normal pregnancies in first trimester from Renji Hospital,Shanghai Jiao Tong University were studied.A high-resolution spectrum typing analysis of complementarity-determining region 3 (CDR3)was used to detect and compare the degree and frequency of TCR BV family expression in deciduas between RSA patients and normal controls.Results(1)The expression degree of BV19(0.029±0.031 vs.0.013±0.010,P=0.038)in RSA group showed a higher usage,while BV5.2(0.040±0.035 vs.0.067±0.052,P=0.046)showed a significantly lower usage when compared with normal controls.No significant difference in the expression of the other TCR BV families between RSA and controls were observed(P>0.05).(2),TCR BV2,3,6,and 7 were the four most common BV families in deciduas of patients with RSA and normal controls,whose frequencies were all mors than 50%.In RSA group,higher frequencies of BV15 (33.3%vs.7.3%,P=0.018),BV19(38.9%vs.14.6%,P=0.049)and BV20(33.3%vs.7.3%,P=0.018)were observed;meanuhile lower frequencies of BV4(33.3%vs.65.9%,P=0.026)and BV7 (66.7%vs.92.7%.P=0.018)distributions were observed.The other TCR BV families did not display significantly different freqencies of distribution(P>0.05).Conclusions It is suggested that a significant skewed TCR BV family occurs at the maternal-fetal interface in patients who undergo abortion.The specific skewed usages of TCR BV might be associated with the susceptibility to unexplained pregnancy loss.

Objective To prepare polyclonal antibodies of anti Nogo-66, the extracellular region of one central nervous system neurite regeneration inhibitor Nogo, which could be used to further identification and functional study of Nogo molecule.Methods Preparing rabbit anti rat Nogo-66 polyclonal antibodies with a purified Nogo-66 fusion protein expressed in E.coli system. Studying its specificity by Western-blot and immuno-histochemical techniques and identifying its biological activity in PC12 cells.Results The high titer (1∶[KG-*2]10 000) anti rat Nogo-66 polyclonal antibodies were obtained.This antibody could specifically recognize the Nogo protein expressed in E.coli system.Immuno-histochemical staining indicated that the Nogo was widely expressed in rat spinal cord neurons and oligodendrocytes.It could effectively block the neurite extensioninhibition of Nogo protein in PC12.Conclusion Successful preparation of anti rat Nogo polyclonal antibodies provides a useful tool in identification or further functional study of Nogo molecule.

Objective To explore and observe the clinical characteristics and laboratory testing results of children with infectious mononuclear cells syndrome (IM),in order to improve diagnostic level.Methods 152 children with IM were selected,and the clinical manifestations,laboratory testing were retrospectively analyzed.Results There were a variety of clinical symptoms,the main clinical manifestations included fever 138 cases(90.8％),swollen lymph nodes 145 cases(95.4％),angina 112 cases (73.7％),hepatosplenomegaly 28 cases(18.4％),double eyelid edema 25 cases(16.4％),rash 7 cases(4.6％).Laboratory-testing of white blood cell count ＞ 10 × 109/L in 125 cases (82.2 ％),atypicallymphocyte proportion were more than or equal to 10％ in 48 cases (31.5 ％),liver function damage in 58 cases (38.2％),myocardial damage in 38 cases (25％),abnormal urine analysis in 30 cases (19.7％),thrombocytopenia 2 cases (3.4％).Conclusion Clinical symptoms of IM children are diversity,in some cases the clinical sympotoms are not typical,to improve the understanding of this disease can reduce the rates of missed diagnosis and misdiagnosis.
