Objective To detect the expressive levels of hepatocyte growth factor and D2-40 and its correlation with clinicopathological factors and microlymphatic density in gastric carcinoma. Methods Immunohistochemistry method was used to detect the expression of HGF and D2-40 in gastric cancer tissue, with microlymphatic density(MLD) counted. Results The expression of HGF had the positive correlation with histologic types, infiltration depth, clinical stage and lymph node metastasis (P <0.05). No correlation was found in age, tumor diameter(P >0.05). In gastric cancer, the MLD in the HGF-positive group (22.8±10.9) was higher than that in the HGF-negative group(14.0±4.2, P <0.05). Conclusion The expressive levels of HGF have the positive correlation with histologic types, infiltration depth, clinical stage, lymph node metastasis and MLD in gastric carcinoma. HGF can be used to predict prognosis of gastric carcinoma.

Multidrug resistance (MDR) plays a major obstacle to successful gastric cancer chemotherapy. The purpose of this study was to investigate the MDR reversal effect and mechanisms of hyperthermia in combination with neferine (Nef) in adriamycin (ADM) resistant human SGC7901/ADM gastric cancer cells. The MDR cells were heated at 42°C and 45°C for 30 min alone or combined with 10 μg/mL Nef. The cytotoxic effect of ADM was evaluated by MTT assay. Cellular plasma membrane lipid fluidity was detected by fluorescence polarization technique. Intracellular accumulation of ADM was monitored with high performance liquid chromatography. Mdr-1 mRNA, P-glycoprotein (P-gp), γH2AX expression and γH2AX foci formation were determined by real-time PCR, Western blot and immunocytochemical staining respectively. It was found that different heating methods induced different cytotoxic effects. Water submerged hyperthermia had the strongest cytotoxicity of ADM and Nef combined with hyperthermia had a synergistic cytotoxicity of ADM in the MDR cells. The water submerged hyperthermia increased the cell membrane fluidity. Both water submerged hyperthermia and Nef increased the intracellular accumulation of ADM. The water submerged hyperthermia and Nef down-regulated the expression of mdr-1 mRNA and P-gp. The water submerged hyperthermia could damage DNA and increase the γH2AX expression of SGC7901/ADM cells. The higher temperature was, the worse effect was. Our results show that combined treatment of hyperthermia with Nef can synergistically reverse MDR in human SGC7901/ADM gastric cancer cells.

To investigate the role of nitric oxide (NO) in steroid-induced femoral head necrosis, NO production was measured indirectly as nitrite in serum and synovial fluid samples from patients with steroid-induced femoral head necrosis together with serum samples from healthy volunteers matched for age and sex. The results showed that:① serum nitrite concentration in patients with femoral head necrosis were significantly lower than that in controls (P<0.001); ② nitrite level of synovial fluid was markedly higher than that of serum. In addition, there was a positive correlation between them (r=0.378,P<0.05). We reach the conclusion:① NO is synthesized by synovium and chondrocytes; ② the decreased NO concentration in serum suggests a protective role in steroid-induced femoral head necrosis.

Objective To explore the changes of gene and protein expressions of nuclear factor-κB(NF-κB) and tumor necrosis factor-α (TNF-α) in immature rats with pilocarpine-induced epilepsy treated with ghrelin. Methods The pllocarpine-induced epilepsy model in immatured rats were built, then the rats were divided into three groups: Ghrelin-treated group, saline-treated group and model group, meanwhile the normal control group was set. The NF-κB and TNF-α levels of gene and protein in the cerebral cortex of immature rats were detected. Results The expression levels of gene and protein of NF-κB and TNF-α were increased in model group, but decreased in the normal control group;NF-κB and TNF-α levels in Ghrelin treated group were obviously lower than those of saline-treated group and model group(P < 0. 05). Conclusion The protective mechanism of Ghrelin for nerve cell is cutting down the expressions of NF-κB and TNF-α in the cerebral cortex of immature rats with epilepsy and lessening inflammatory reaction in neurocytes.

Objective:To investigate the best preservation conditions of N-terminal pro-brain natriuretic peptide( NT-proBNP) and provide detection references with stable performance for detection kits. Methods: ELISA was used to quantitatively detect the changes in NT-proBNP contents in various preservation solutions. The effects of basic buffer system, preservative Proclin300 and antibiotics on the preservation of NT-proBNP were analyzed using univariate analysis. The combination of various factors was then optimized using orthogonal experiments, to identify the best preservation system for NT-proBNP.Results: The univariate analysis determined that the basic buffer system for NT-proBNP was 0.02 mol/L phosphate buffered saline(PBS) at pH7.2,the addition of pre-servative Proclin300 could extend the preservation time of NT-proBNP at 37℃ by one day, the combined addition of penicillin and streptomycin prolonged the preservation time of NT-proBNP by one day compared with individually adding penicillin or streptomycin. The orthogonal experiments identified a preservation solution for NT-proBNP as 20%calf serum,1/1 000 Proclin300,120 U/ml penicillin and 80 U/ml streptomycin in a basic buffer system of 0.02 mol/L PBS at pH7.2.This solution was used to preserve an NT-proBNP reference sample at 37℃.Seven days later, the calibrated fixed-value of the sample at 37℃was only 1.3%lower than that at 4℃.Conclusion:Optimized NT-proBNP serum preservation solution could preserve NT-proBNP standard sample at 37℃ for seven days.

Cytoplasmic processing bodies, termed P bodies, are involved in diverse post-transcriptional processes including mRNA decay, nonsense-mediated RNA decay (NMD), RNAi, miRNA-mediated translational repression and storage of translationally silenced mRNAs. Regulation of the formation of P bodies in the context of multicellular organisms is poorly understood. Here we describe a systematic RNAi screen in C. elegans that identified 224 genes with diverse cellular functions whose inactivations result in a dramatic increase in the number of P bodies. 83 of these genes form a complex functional interaction network regulating NMD. We demonstrate that NMD interfaces with many cellular processes including translation, ubiquitin-mediated protein degradation, intracellular trafficking and cytoskeleton structure. We also uncover an extensive link between translation and RNAi, with different steps in protein synthesis appearing to have distinct effects on RNAi efficiency. Moreover, the intracellular vesicular trafficking network plays an important role in the regulation of RNAi. A subset of genes enhancing P body formation also regulate the formation of stress granules in C. elegans. Our study offers insights into the cellular mechanisms that regulate the formation of P bodies and also provides a framework for system-level understanding of NMD and RNAi in the context of the development of multicellular organisms.

Journal of Biomedical Engineering 2018;35(5):774-778
In order to realize brain-computer interface (BCI), optimal features of single trail motor imagery electroencephalogram (EEG) were extracted and classified. Mu rhythm of EEG was obtained by preprocessing, and the features were optimized by spatial filtering, which are estimated from a set of data by method of common spatial pattern. Classification decision can be made by Fisher criterion, and classification performance can be evaluated by cross validation and receiver operating characteristic (ROC) curve. Optimal feature dimension determination projected by spatial filter was discussed deeply in cross-validation way. The experimental results show that the high discriminate accuracy can be guaranteed, meanwhile the program running speed is improved. Motor imagery intention classification based on optimized EEG feature provides difference of states and simplifies the recognition processing, which offers a new method for the research of intention recognition.
