Objective To study the therapeutic effect of arbutin combined with glycyrrhizin on immune-inflammation. Methods The effect of arbutin combined with glycyrrhizin on adjuvant arthritis and on oxzolone induced mice ear swelling were investigated. PGE2 level in inflammatory tissue and LTB4 content from peritoneal macrophage of rat treated with arbutin and glycyrrhizin were determined. Results Arbutin combined with glycyrrhizin could inhibit Freund's complete adjuvant induced rat primary and secondary foot swelling. The combined group could also inhibit oxazolone induced mice ear swelling and decrease CD4+ T cell in blood. Compared with control group,PGE2 level in combined group was remarkable lower, and there was no difference between combined group and GL 400 mg/kg group. Arbutin combined with glycyrrhizin could inhibit LTB4 release from rat peritoneal macrophage. Conclusion Arbutin and glycyrrhizin has synergetic therapeutic effect on immune-inflammation.

Protamines are a group of highly basic proteins first discovered in spermatozoon that allow for denser packaging of DNA than histones and will result in down-regulation of gene transcription[1].It is well recognized that the Autographa californica multicapsid nucleopolyhedrovirus (AcMNPV) encodes P6.9,a protamine-like protein that forms the viral subnucleosome through binding to the viral genome[29].Previous research demonstrates that P6.9 is essential for viral nucleocapsid assembly, while it has no influence on viral genome replication[31].In the present study, the role of P6.9 in viral gene transcription regulation is characterized. In contrast to protamines or other protamine-like proteins that usually down-regulate gene transcription,P6.9 appears to up-regulate viral gene transcription at 12-24 hours post infection (hpi),whereas it is non-essential for the basal level of viral gene transcription.Fluorescence microscopy reveals the P6.9's co-localization with DNA is temporally and spatially synchronized with P6.9's impact on viral gene transcription, indicating the P6.9-DNA association contributes to transcription regulation.Chromatin fractionation assay further reveals an unexpected co-existence of P6.9 and host RNA polymerase Ⅱ in the same transcriptionally active chromatin fraction at 24 hpi, which may probably contribute to viral gene transcription up-regulation in the late infection phase.

Objective To investigate the influence of factors related to metabolic syndrome(MS)on the outcome in subjects without diabetes mellitus in a community. Methods A two-year follow-up study was conducted in 885 subjects who were enrolled in the epidemiologic survey carried out in Pingliang Community, Shanghai in 2002.Oral glucose tolerance test, lipid prefde, blood pressure(BP),body mass index(BMI),waist and hip circumferences were measured. Results (1)The baseline of BMI,fasting plasma glucose(FPG),2h plasma glucose after glucose loading(2hPG),BP,triglyceride(TG)in the subjects with impaired glucose regulation(IGR)increased significantly as compared to those with normal glucose regulation(NGR)(all P

Objective To observe the dynamic changes of NO,NOS in serum and CSF in patients with CNS infection and lay an experimental basis for discrimination of CNS infection due to different agents. Methods The method of nitrate redutase and the method of chromometry were employed to measure NO,NOS in serum and CSF at different time. The dynamic changes of NO,NOS in serum were observed on admission,on 3,5,9,14 day after admission. The dynamic changes of NO,NOS in CSF before treatment and two weeks after treatment were ob- served, too. Results There were no difference between the the concentration of NO and the vigor of total NOS in serum and in CSF of viral meningitis, bacterial meningitis and tubercular meningitis patients due to different agents. Conclusion The changes of the concentration of NO in serum and CSF,the vigor of total NOS in serum and CSF could not he seen as laboratory basis for discrimination of CNS infection due to different agents.

Objective To explore the rule of the Nrf2-ARE in renal interstitial fibrosis and the mechanism of butylphthalide on renal protective effect. Methods Seventy-two male CD-1 mice were divided into 4 groups,Sham group,UUO group,NBP group,ACEI group. The Sham group and UUO group were gavaged with physiological saline. The NBP group was gavaged with butylphthalide. The ACEI group was gavaged with benazepril. After 3,7,14 days,6 mice were executed and collection of kidney tissue. The expression of Nrf2,γ-GCS and type Ⅰ collagen were detected by immunohistochemisty,RT-PCR and Western blot. The correlation of Nrf2 and γ-GCS was assessmented by linear regression. Results The expression of type Ⅰ collagen in UUO group was increased compared with the Sham group,However, the expression of Type Ⅰ collagen in NBP group or ACEI group were reduced compared with the UUO group.Compared with Sham group, the expression of Nrf2mRNA,γ-GCSmRNA and type Ⅰ collagen mRNA in uuogroup were increased in 3,7,14 days after surgery.Compared with UUO group at 7th,14u day,the lelve of Nrf2 mRNA were apparently increased in NBP group (P<0.05).It was a positive correlation between the Nrf2 and γ-GCS and negative correlation between the γ-GCS and Type Ⅰ collagen. Conclusion The renal protective effect of butylphthalide in the renal interstitial fibrosis was more predominant than benazepril. The roles maybe occurred through increased the expression of Nrf2 and γ-GCS and alleviated the expression of Type Ⅰ collagen in nephridial tissue.

Objective:To investigate the molecular interaction between non-structural protein 3 serine protease of hepatitis C virus(HCV)and wild type P53,and to lay the basis for elucidating the mechanism of oncogenesis of hepatocellular carcinoma(HCC)after infection of HCV.Methods:The recombinant plasmids,pGAD424-NS3,pGAD424-NS315aa- and pGAD424-NS330aa-,were constructed and the interaction between NS3 serine protease and its cofactor NS4A,the interaction between wild type P53 and NS3 serine protease and its N-truncated mutants were dectected qualitatively and quantitatively in yeast two-hybrid system. Results:The results indicated that interaction existed not only between full-length NS3 serine protease and P53,but also between N-truncated mutants of NS3 serine protease and P53.Furthermore,the difference between enzyme activity unit(IU)of β-gal induced by these interactions was not significant(P>0.05).Conclusions:NS3 serine protease of hepatitis C virus and its N-truncated mutants can interact with wild type P53,and the region of NS3 serine protease involved in the interaction may be located in its C-terminal,but not in its N-terminal.

Objective To investigate the supply of iodized salt in non-excessive iodine counties and iodine-free salt in excessive iodine counties at household level in Jiangsu Province so as to provide a basis for the prevention and control of iodine deficiency disorders(IDD).Methods According to the National Iodine Deficiency Disorders Monitoring Program(Trial),county(city, district) was taken as a elementary sampling unit in Jiangsu Province.Townships(towns) and administrative villages were selected by systematic sampling and random sampling in every county and households were chosen by random sampling to collect their edible salt samples. The salt iodine content in non-and excessive iodine regions was detected by direct titration method and semi-quantitative method, respectively. Results All 30 840 salt samples were collected from 106 non-excessive iodine counties, and qualified iodized salt was 30 303 copies,iodine-free salt 199 copies.Weighted by the population of counties, the rate of iodine-free salt was 0.71%,the coverage rate of iodized salt accounted for 99.29%,out of which,98.93% was qualified and the consuming rate of qualified iodized salt was 98.23%.All 1296 salt samples were collected in 5 counties with excessive water iodine content and the coverage rate of iodine-free salt was 98.99% (1283/1295).Conclusions The national targets for preliminary elimination of IDD have been achieved in regions of nonexcessive and excessive iodine of Jiangsu Province. But it still should be strengthen the management work of iodinefree salt in excessive iodine counties and iodine saft in non-excessive iodine counties.
