1.Preliminary study effect of artificial liver support on HBV cccDNA in patients with fulminant hepatitis B

Objective:To establish a new fluorescence-based quantitative PCR assay for detecting hepatitis B virus covalently closed circular DNA(HBV cccDNA).To explore the effect of artificial liver support system(ALSS)on total HBV DNA and HBV cccDNA in sera of patients with fulminant hepatitis B.Methods:The serum specimens from 50 patients of fulminant hepatitis B prior to and after treatment with ALSS were collected.Then HBV cccDNA was quantitatively detected by fluorescent PCR with specific primer set and Taqman probe.And the results were analyzed by SAS8.0 statistics software.Results:We successfully established a new fluorescence-based quantitative PCR method for HBV cccDNA.HBV cccDNA was detectable in sera of the patients with fulminant hepatitis B.The level of total HBV DNA and HBV cccDNA decreased significantly following plasma exchange in ALSS.The levels of total HBV DNA and HBV cccDNA had positive correlation with degree of PT,ALT,andAST.Total HBV DNA also had positive correlation with HBV cccDNA.Conclusion:The level of total HBV DNA and HBV cccDNA has positive correlation with degree of PT,ALT,and AST,which might be indication of hepatocyte damage.The finding that significant decrease in total HBV DNA and HBV cccDNA after plasma exchange in ALSS may provide a clue of further research about the fulminant hepatitis B treated with ALSS.

4.Optimal proportion of VEGF 165, Ang-1 and bFGF concentration on angiogenesis of transplanted fat

Objective To explore the optimal formula of vascular endothelial growth factor,angiopoietin and basic fibroblast growth factor on autologous fat angiogenesis and tissue proliferation.Methods Wistar rat mesenteric fat in 9 groups trimmed into particles were added a 6μl complex solution of the three factors in the formula of 1 ∶ 0 ∶ 0,0 ∶ 1 ∶ 0,0 ∶ 0 ∶ 1,1/2 ∶ 1/2 ∶ 0,0 ∶ 1/2 ∶ 1/2,1/2 ∶ 0 ∶ 1/2,1/3 ∶ 1/3 ∶ 1/3,2/3 ∶ 1/6 ∶ 1/6,1/6 ∶ 2/3 ∶ 1/6,then injected in each rat subcutaneously.15 weeks later the transplantated fat was removed,to observe the fat cell appearance and counting the number of capillaries in the graft.Statistical genetic algorithm was used to set up a Scheffer egression model to optimize fat angiogenesis number.Results In one growth factor group fat vacuoles was uneven size;in two growth factors group,fat cell size tended to be homogeneous and in the three growth factor group,fat cell size was uniformity;the highest angiogenesis rate was 12.092,when the formula of VEGF165,Ang-1 and b-FGF was 0.380∶0.280:0.340.Conclusions Formula of VEGF165,Ang-1,bFGF at 0.380 ∶ 0.280 ∶ 0.340 could improve blood vessels density and increase implanted fat survival.
5. Clinical comparision of the treatment of cervical prolapse of intervertebral disc by using cage or titanium plate

Objective: To compare the clinical effects of treating cervical prolapse of intervertebral disc by using Solis cage, titanium cage and autogenous iliac crest graft (AICG) combined with titanium plate. Methods: 64 cases of cervical prolapse of intervertebral disc were analyzed retrospectively. All the patients were followed up for about one year. Group A is composed of 20 patients (30 intervertebral spaces) treated with microdiscectomy and Solis cage fusion; group B is composed of 21 patients(22 intervertebral spaces) treated with titanium cage and group C is involved of 23 patients(28 intervertebral spaces) which were dealt with AICG combined with titanium plate. The differences in the total X ray exposure time, time working on iliac bone, incidence of complications, fusion rate, incidence of JOA score recovery rate more than 50% and rehabilitation time of the three groups are analyzed. Results: All indexes from group A and B were more satisfactory than those from group C (P<0.05). The statistic analysis results of the incidence of complications of cervical part, rehabilitation time in group A are 5.0±1.8, 5.1±1.2; and that is 14.3±2.6, 7.5±1.6 and 26.1±6.2, 8.6±2.3 in group B and group C respectively. There is significant difference between group A and group B on the incidence of complications of cervical part and rehabilitation time (P<0.05). Results of the incidence of JOA score recovery rate >50% in group A(95.8±3.2) was more satisfactory than that in group B(93.6±2.9), while there was no statistical difference between group A and group B (P>0.05). Conclusion: The Solis cage is better in treating the cervical prolapse of intervertebral disc than other ways. Also, it makes the operation easier, rehabilitation time shorter, and it causes fewer complications.
7.The quality of basic research in ophthalmology needs to be improved in China

Basic research program is essential to the investigation of the pathogenesis of ocular diseases and the development of novel strategies for the prevention and treatment for these diseases. With increasing support of research grants at various levels, basic research in ophthalmology has gained great achievement in China in recent years. A number of studies have recently been published in well known peer-review international journals and won the State Scientific and Technological Progress Awards. However, we have to keep it in mind that basic research in ophthalmology should be improved qualitatively meanwhile, the imbalance in basic study among different areas needs to be resolved in the near future.
8.Advances in treatment of Vogt-Koyanagi-Harada syndrome

Vogt-Koyanagi-Harada (VKH)syndrome is an autoimmune disease attacking against pigmented cells, resulting in blindness and usually affecting multiple organs including ears, meninges, hair and skin.Correct diagnosis and immediate treatment in the early stage is vital to visual prognosis.Currently, corticosteroids is first-line drug.In addition, VKH patients refractory to corticosteroids can choose other treatment such as immunosuppressive agents and biological agents.
10.The Expression and clinical Significance of RhoA protein in Colorectal Carcinoma

Objective To study the expression of RhoA protein in colorectal carcinoma. Methods The expression of RhoA protein in 92 cases of colorectal ccarcinoma and 30 cases of normal rectal tissue was detected by SP immunohistochemical technique. Results RhoA protein were negative in 40 cases of normal colorectal tissue. In 92 cases of colorectal carcinoma, the positive rate of RhoA protein was 70.7%. It was significantly higher than that in the normal colorectal tissue. The positive rate of RhoA protein in high-differentiated 、moderate-differentiated and lowdifferentiated rectal carcinoma were 43.5% 、74.4% and 86.7% ,respectively(P <0.05). The positive rates of RhoA in phase Ⅰ、phase Ⅱ、phase Ⅲ and phase Ⅳ were 48.6% 、77.3% 、83.3% and 94.1%, respectively (P < 0.05).The positive rate of RhoA protein in patients with lymphnode mestastasis and non-lymphnode mestastasis were 82% and 57.1% (P < 0.05). Conclusion RhoA protein could be related with the differentiation、invasion、metastatsis and prognosis of colorectal carcinoma.
