Objective Hes5 is one of the critical effectors of the Notch pathway and its expression Call inhibit neuronal differentiation.Which protein expression is influenced in cell differentiation by HeS5 remains to be determined.Methods Experiments were designed to study changes in protein expression in the subventricular zone(SVZ)of the HesS knockout mice at the postnatal day 8 using the new method of proteomic isobaric tags for relative and absolute quantification(ITRAQ),and confirmed expression of the up-gnlated and down-regulated proteins by Western blotting.Results Nineteen proteins showed differences in expression levels in the SVZ of Hes5 knockout mice.When compared tO the wild type mice,expressions of 11 proteins in the knockout mice were up-regulated,and the differentially expressed proteins showed ratios of knockout/widtype between 1.20 and 1.32.Expressions of 9 proteins were down-regulated,with their ratios were between 0.77 and 0.83.The upregulated proteins have been shown to be involved in neurogenesis,cell adhesion,migration and signal transduction.The downregulated proteins have been mainly shown to be involved in growth inhibition and cytoskeleton.Westem bloning analysis further confirmed the expression of proteins from iTRAQ.Conclusion Our findings suggest that Hes5 may influence cell differentiation by regulating expression of some proteins involved in cell growth and migration,and its cytoskeleton.

3.EFFECTS OF DIFFERENT DIETARY FATTY ACID COMPOSITION ON T LYMPHOCYTE FUNCTIONS AND LIPID PEROXIDATION IN MICE

Objective: To study the effects of dietary n-6/n-3 PUFA ratio on T lymphocyte functions in mice. Methods: 96 male BALb/c mice were randomly divided into 8 groups with dietary S:M:P 1:1.5:3.7 or 1:1.5:1, respectively. The n-6/n-3 PUFA ratio was 1, 7.5, 15, 30 respectively in each S:M:P diet. Each diet contained 70.0g/kg lipid,and different n-6/n-3 PUFA ratios by mixing different oils. At the end of 12w, all mice were killed and blood was collected. Objective To assess whether echocardiographic parameters of M-mode,pulsed-wave Doppler and tissue Doppler imaging which were usually used for selecting cardiac resynchronization therapy (CRT) candidates could predict CRT responders.Methods Twenty-nine patients with NYHA class Ⅲ or Ⅳ heart failure,left ventricular ejection fraction(LVEF)≤35%,QRS≥130 ms were enrolled.All of them underwent biventricular pacemaker implantation.The septal-to-posterior wall motion delay(SPWMD) from M-mode recording,and the standard deviation of time to peak systolic velocity(Ts-SD) were used to evaluate intraventricular dyssynchrony.The interventricular mechanical delay (IVMD) by pulsed Doppler echocardiography was calculated by subtracting the right ventricular preejection intervals (RV-PEI:the onset of the QRS on the electrocardiogram to the onset of pulmonary outflow) from the left ventricular preejection intervals (LV-PEI:the onset of the QRS on the electrocardiogram to the onset of aortic outflow).At least 15 % reduction in left ventricular end-systolic volume(LVESV) at the 6-month after CRT was defined as responder.Results In the 29 patients,there were 20 (68.97%) responders.The area under the receiver operating curve(ROC) of SPWMD,Ts-SD,LV-PEI,RV-PEI and IVMD to predict CRT responders was 0.448 (95% CI 0.184-0.712,P>0.05),0.557 (95% CI 0.317-0.798,P>0.05),0.479 (95% CI 0.186-0.773,P>0.05),0.568 (95 % CI 0.272-0.863,P>0.05) and 0.411 (95% CI0.136-0.687,P>0.05),respectively.Conclusions M-mode echocardiography,pulsed Doppler and tissue Doppler imaging failed to predict CRT responders.New echocardiographic techniques including threedimensional echocardiography and speckle tracking image will be investigated,and may improve the predictive power.

7.An anatomical and biomechanical study of the forearm interosseous membrane

Objective To observe the anatomical and biomechanieal properties of the forearm in terosseous membrane (IOM), providing the scientific theoretical basis for the diagnosis and treatment of the IOM injury. Methods Ten radius-IOM-ulna structures (left for 5 and right for 5) were harvested from 5 fresh-frozen adult cadavers to measure the length, width and thickness of the tendinous part. Then the tendi-nous part with its radial and ulnar insertions were isolated, embedding the proximal part of the radius and the distal part of the ttina in commercially available dental base acrylic resin powder. The embedded speci-mens were mounted on MTS 858 testing machine using a 10 000 N load cell for all tensile tests. The speci-mens were tested at a displacement rate of 50 mn/min until failure. The load-displacement curve was de-scribed by the computer while the maximum load and stiffness were recorded. Results The IOM consisted of three components: the tendinous part, the membranous part, and the dorsal oblique accessory cord. The IOM was fiat in the neutral position, while it flexed during the pronation and supination. Six specimens ob-tained the tear of tendinous portion at a maximum load of (1021.50±250.13) N. The stiffness was (138.24±24.29) N/mm with displacement to failure of (9.77±1.77) mm. Other 4 specimens failed by fracture through the ulnar fixed site before there was failure of the IOM at a maximum load of (744.40±109.85) N. The stiff-ness was (151.17±30.68) N/mm with displacement to failure of (6.51±0.51) ram. Conclusion The IOM has stiffness comparable to the ligament between the radius and the ulna and play an important role for the maintenance of longitudinal stability of the forearm. These results can provide objective criteria for the eval-uation of reconstructive methods.

Objective To discuss the pathogenesis of cerebral hemorrhage in maintenance hemodialysis patients.Methods From Jan.2002 to Dec.2004all the 261 hemodialysis patients in changai Hospital were divided into two groups on the basis of with or without cerebral hemorrhage :(1)the control group,(2)the group of cerebral hemorrhage.Clinical data of 261 hemodialysis patients were retrospectively analyzed.Results In the group of cerebral hemorrhage,the incidence of high blood pressure was 81.1%:the blood pressure after medical therapy not up to standard was 83.8%;the usage of ordinary heparin was 78.4%;the average RRF was(3.8?1.9)and those of the control group were 62.5%,52.7%,52.7% and(7.1?3.3).There were significant differences between the two groups.Conclusion The maintenance hemodialysis patients with cerebral hemorrhage result from multiple factors.Positive control of the blood pressure,selection of appropriate anticoagulant and protection of RRF have important clinical significance.
