1.Effect of Vaspin in vitro expressed on insulin signal transduction pathway of GDM

Objective To investigate the relationship between Vaspin and GDM insulin resistance. Methods The GDM preadipocyte cells were recoveried, extended and differentiated.Vaspin overexpression carriers were made to transformate, cultivate and extract the plasmid. The fat cells were transfected and extended using 4 overexpression levels (0.0 μg, 1.0 μg, 2.5 μg, 5.0 μg). Q-PCR was used to detect mRNA expression of Vaspin, insulin receptor substrate-1/2 (IRS-1/2), phosphatidy inositol 3 kinase (PI3K (P85a) Western Blot was used to detect protein expression of Vaspin, IRS-1/2, PI3K (P85a) and IRS-1/2 phosphorylation levels, and [3H]-2-deoxidation-D-glucose uptake assay was used to detecte glucose uptake rates. Results (1) According to the Q-PCR and WB results, the constructed Vaspin overexpression carrier was effective; (2) With the Vaspin expression increased, the mRNA and protein expression of IRS-1/2, PI3K (P85a) and IRS-1/2 tyrosine phosphorylation levels had no significant changes;(3) Glucose uptake rate of fat cells had no obvious correlation with Vaspin. Conclusion High expression of Vaspin in GDM serum and omental adipose tissue has no obvious link with the insulin resistance of GDM.
2.Correlation betw een lipoprotein-associated phospholipase A2 gene R92H polymorphism and ischemic stroke and its subtype in a Chinese Han population of Shandong province

Objective To investigate the correlation between lipoprotein-associated phospholipase A2 (Lp-PLA2) gene R92H polymorphism and ischemic stroke and its subtype in a Chinese Han population of Shandong province. Methods A total of 386 patients w ith first-ever ischemic stroke and 368 healthy controls in China Shandong region w ere enrol ed. According to the TOAST criteria, the patients w ere further divided into large artery atherosclerosis (LAA) and smal artery occlusion (SAO). Enzyme linked immunosorbent assay w as used to detect the serum Lp-PLA2 level. Polymerase chain reaction and directly sequencing w ere used to detect R92H gene polymorphism. Results The serum Lp-PLA2 levels in the ischemic stroke group, LAA group and SAO group w ere higher than that in the control group, and there w ere significant differences (al P<0.01). The distribution frequencies of GA (P=0.006), AA (P=0.020), AA+AG (P=0.009), and A al ele (P=0.001) in the ischemic stroke group w ere significant higher than those in the control group. There w ere also significant differences in distribution frequencies of GA+AA genotype (P=0.007) and A al ele (P<0.001) betw een the LAA group and the control group, w hile the SAO group w as not the case. Multivariate logistic regression analysis showed that GA+ AA genotype (odds ratio [OR] 1.43, 95%confidence interval [CI] 1.02-2.00; P=0.029), GA genotype (OR 1.42, 95%CI 1.01-2.00; P=0.037), and A alele (OR 1.44, 95%CI 1.11-2.18; P=0.028) were the independent risk factors for ischemic stroke. GA+AA genotype (OR 1.73, 95%CI 1.18-2.55; P<0.001) and GA genotype (OR 1.67, 95%CI 1.13-2.48; P<0.001) w ere the independent risk factors for LAA, and they w ere not significantly independent correlated w ith SAO. Conclusions The serum Lp-PLA2 levels increased in patients w ith ischemic stroke, and they increased most significantly in the LAA group. The R92H gene polymorphism might be associated w ith the susceptibility of ischemic stroke in a Chinese Han population of Shandong province.

OBJECTIVE To evaluate the value of blood lactic acid in elder pneumonia patients and indicate its relation with the classification of peverity of patients.METHODS Seventy-five elder patients with pneumonia and 50 younger patients were divided into sepsis group and no sepsis group.The sepsis group was divided into MODS and non-MODS sub groups.The level of blood lactic acid,CRP and PaO2 in them were measured.RESULTS The incidence of sepsis was remarkably higher in the elder pneumonia patients group than in the younger one.There was a significent difference of the blood lactic acid between them((1.74?0.33)mmol/L vs(1.35?0.66)mmol/L,P

7. Preparation process and in vitro evaluation of Shufei in situ gel optimized by central composite design-response surface methodology

objective To optimize the preparation technology of sulfuric in situ gel and study the infiltration experiment of different dosage forms. Methods Shufei in situ gel was prepared by cold cut method with poloxamer 188 (P188) and poloxamer 407 (P407) as gel base. Using gelling temperature index, the dosage range of gel matrix P407 and P188 in Shufei in situ gel was determinated by the single factor and star design-response surface methods to get the best prescription of Shufei in situ gel. The transdermal diffusion process of Shufei in situ gel was carried out in Franz diffusion cells to explore the permeation mechanism. Results The optimized prescription of Shufei in situ gel was as follow: The ratio of gel matrix to drug at 1:3, P188 dosage of 4%, P407 dosage of 22.5%, and the phase transition temperature within 32-36 ℃ to form gel. The releases of sinapine thiocyanate and genkwanin in Shufei in situ gel were all in line with the Higuchi release model. Conclusion The preparation process of Shufei in situ gel is stable and feasible with reliable product quality and good application prospect, which is suitable for industrial production and clinical application.
8.Influence of visfatin in expressions of insulin receptor substrate and PI3K in fat cells in vitro

Objective:To investigate the relationship between visfatin and insulin resistance (IR)of type 2 diabetes mellitus(T2DM) through the classic insulin signaling pathway phosphatidy inositol-3 kinase (PI3K).Methods:The human T2DM preadipocyte cells were recoveried, extended and differentiated.The visfatin overexpression vectors were built, transformated, cultivated and extracted.The fat cells were transfected with different overexpression levels (0.0, 1.0, 2.5 and 5.0 μg);0.0 μg group was used as control group,and the remaining three groups as observation groups.The mRNA expression levels of visfatin, insulin receptor substrate-1(IRS-1), insulin receptor substrate-2(IRS-2) and PI3K (P85α) were detected by Q-PCR.The protein expression levels of visfatin, IRS-1, IRS-2, PI3K (P85α), IRS-1 and IRS-2 phosphorylation levels were measured by Western blotting method.The glucose uptake rates of fat cells were determined by [3H]-2-deoxidation-D-glucose uptake assay.Results:The expression levels of visfatin mRNA and protein in various groups were increased with the increase of transfection concentration gradient (P<0.01);and the constructed overexpression vector of visfatin was effective.With the increase of visfatin expression, the mRNA and protein expression levels of IRS-1 and PI3K (P85α) and IRS-1 phosphorylation degrees in various groups were increased (P<0.01), but the IRS-2 mRNA and protein expression levels had no obvious changes(P>0.05).The glucose uptake rates of fat cells were elevated with the increasing of visfatin expression (P<0.05).Conclusion:The visfatin overexpression of fat cells in vitro can increase the expression levels of IRS-1 and PI3K.
9. Study on preparation of Compound Xiongqi Maichong Tablets

Objective: To prepare Compound Xiongqi Maichong Tablets (CXMT) by selecting ligustrazine and Panax notoginseng saponins (PNS) in the treatment of coronary heart disease as a model drug, using hypromellose (HPMC) as coating material, and control of drug release delay as 4 h. Methods: The core system of CXMT was prepared by direct powder compression method; The best coating prescription and preparation of CXMT by press coated was optimized by central composite design and response surface method. The release model was fitted by DDsolver software, and the synchronous release of drug was studied. Results: The in vitro cumulative release fitting order of CXMT preparation was Logistic > Higuchi; Logistic model fitting degree was 0.950 1, and AIC was 29.432 0. So the drug release mechanism of CXMT was type S dissolution process, and the in vitro cumulative release model was the Logistic model. Conclusion: Through the study of CXMT preparation technology and the drug release mechanism, the quality of the prepared CXMT is stable, and the lag time is 4 h; Different components of the cumulative dissolution rate are over 90%, which reach the pulse drug release standard.
10.Association of diet and C-reactive protein with atherosclerosis in type 2 diabetes mellitus patients

Objective To compare the characteristics of food and nutrition intake in type 2 diabetes mellitus (T2DM) patients with or without carotid atherosclerosis and analyze the relationship between diets/C-reactive protein (CRP) and carotid intima-media thickness (C-IMT). Methods Sixty patients with T2DM were enrolled and divided into two groups based on C-IMT: group A (C-IMT < 1 mm, n=30) and group B (C-IMT≥1 mm, n=30). All subjects were investigated with questionnaires including 3-day food recall They all took somatometric measurement. Blood and urine samples were collected in all subjects to examine the levels of high sensitive-CRP,C-peptide, blood glucose, glycosylated hemoglobin, blood lipid, renal function, urine microalbumin, and other indicators. Results The intakes of vegetables, fruits, and aquatic products were significantly higher in group A than in group B ( P < 0. 05 ). The intake of vitamin C in group A was significantly higher than that in group B ( P <0. 05 ). The levels of CRP in group B was significant higher than that in group A (P = 0. 000). Positive correlation was found between CRP level and C-IMT in T2DM patients ( r = 0. 36, P = 0. 004). Furthermore, CRP was negatively correlated with the intakes of vegetables and fruits ( r = - 0. 334, P = 0. 01 ), aquatic products ( r = -0. 315, P = 0. 016), and vitamin C ( r = - 0. 2786, P = 0. 038 ), respectively. The intake of fruits was negatively correlated with C-IMT (r, = -0. 33, P = 0. 01 ). Conclusions T2DM patients without carotid atherosclerosis intake more vegetables, fruits, aquatic products and vitamin C than those with atherosclerosis. Vegetables, fruits,sea foods and vitamin C may be the protective factors against atherosclerosis in T2DM patients. CRP is associated with the development of atherosclerosis in T2DM patients.
