Chinese Journal of Biologicals 2024;37(8):952-956
ObjectiveTo obtain the soluble expression of P protein of norovirus(NV)G Ⅱ. 2[P16]and analyze its antigenic characteristics.MethodsThe recombinant plasmid GⅡ.2-NV-pET-28a was constructed by optimizing the NV GⅡ.2[P16]P protein gene, synthesizing and cloning it into pET-28a(+)vector. The recombinant plasmid was transformed into DH5α competent cells, and the expression strain was optimized to make the P protein express in soluble form. The expressed P protein was purified by Ni-NTA affinity chromatography,of which the antigenicity was detected by Western blot, the purity was detected by size exclusion high performance liquid chromatography(SEC-HPLC),and the antigenantibody binding ability was identified by ELISA.ResultsThe target protein of recombinant BL21 Star(DE3)pLySs was highly expressed in soluble form. After purification by affinity chromatography, the purity of GⅡ.2 P protein was 95. 53%,and the antigenicity was good.ConclusionSoluble NV P protein was successfully prepared by prokaryotic expression strain, and the antigenicity of P protein was confirmed, which laid a foundation of the development of NV G Ⅱ.2[P16]detection kits and the recombinant subunit vaccines.

Chinese Journal of Orthopaedic Trauma 2012;14(3):228-231
Objective To study the effect of articular step-off on knee joint contact pressures in fractures of the medial tibial plateau. Methods Eight specimens of embalmed adult knee joint were used to establish models of articular step-off of the medial tibial plateau by 0,1,2,3 mm respectively (Schatzker Ⅳ fracture) .The technique of pressure sensitive film was used to measure the contact pressures on the weight-bearing area of the knee joint in the 3 models. Results When the step-offs of medial tibial plateau were 0,1,2,3 mm,the average contact pressures on the lateral compartment were respectively 3.34 ± 0.06 MPa,3.48 ± 0.06 MPa,4.19 ± 0.05 MPa,4.19 ± 0.04 MPa,while the average contact pressures on the medial coMPartment were 4.04±0.17 MPa,4.57±0.20 MPa,5.64±0.20 MPa,5.64±0.19 MPa.When the step-off of medial tibial plateau was 2 mm,the average contact pressures on the medial and lateral plateaus increased by 39.6% and 25.4% compared with the 0 mm step-off,with statistically significant differences (P <0.05). Conclusions If the step-off of the medial tibial plateau reaches ≥ 2mm,open reduction and internal fixation should be indicated to prevent secondary traumatic arthritis. When the step-off of the medial tibial plateau is < 2 mm,conservative treatment can be a proper choice.

Journal of Traditional Chinese Medicine 1993;0(04):-
Conventional therapy was given to 260 cases of advanced stage of cancer of colon during the period of 1975 - 1988. Radical operation, palliative resection or changing bowel passage were selected according to the condition of the lesions. Post operative chemo -therapy with integrated Chinese and western medicine were given, or with radiotherapy in some of the cases. Results showed that the 5- year survival rates of II, III, IV stages were 80. 5%, 56. 12%, 21. 73% respectively, the mean 5 -year survival rate being 52. 78%, 45 cases with liver metastasis were also treated with integrated medicine with a 5 - year surivial rate of 22. 2%. The above figures of therapeutic effect and survival rates were all superior to that in cases treated by western medicine alone as reported by domestic exponent units.
9.Study on correlation between serum Omentin-1 and gestational diabetes mellitus
International Journal of Laboratory Medicine 2017;38(3):291-294
Objective To observe the change of serum Omentin-1 in gestational diabetes mellitus(GDM)and to investigate its clinical significance. Methods Serum Omentin-1 level was detected in 85 cases of GDM and 85 matched cases of normal glucose tol-erance(NGT)by ELISA,the glycolipids biochemical indexes, inflammation indexes and fasting insulin (FINS)level were simultane-ous detected, then the insulin resistance index (HOMA-IR)was calculated in both groups. Results The levels of prenatal prepreg-nant BMI,FPG,hs-CRP,blood lipids, blood glucose,FINS and HOMA-IR in the GDM group were significantly higher than those in the NGT group, while serum Omentin-1 level was obviously lower than that in the NGT group, difference was statistically signifi-cant (P<0.05);when prenatal obesity and/or HOMA-IR≥2,the serum Omentin-1 levels was significantly decreased.Serum Omentin-1 level had significant positive correlation with HDL,and negative correlation with prepregnant BMI,prenatal BMI,FPG, FINS and HOMA-IR.The multiple stepwise regression analysis showed that prepregnant BMI,TG,FPG and FINS were independ-ent influencing factors of serum Omentin-1 in GDM.Conclusion Serum Omentin-1 is associated with GDM closely, which can re-flect the degrees of sugar, lipid metabolic disorder and insulin resistance in pregnant women, and may be involved in the occurrence and development of GDM.
10.Predictive value of epicardial adipose tissue thickness for the recurrence of atrial fibrillation after concomitant valve replacement and bipolar radiofrequency ablation
Clinical Medicine of China 2016;32(5):447-450
Objective To investigate the predictive value of epicardial adipose tissue(EAT) thickness for the recurrence of atrial fibrillation (RAF) after concomitant valve replacement and bipolar radiofrequency ablation. Methods From January 2012 to May 2014,156 patients with permanent atrial fibrillation and organic valvular diseases underwent concomitant valve replacement and successful bipolar radiofrequency ablation in the First People's Hospital of Shangqiu. After 1 year of follow-up,the recurrence of AF was analyzed and the correlation factors were analyzed statistically. Results All patients completed a regular follow-up.During a 12-month follow-up period,RAF was observed in 39 patients (25.0%).Echocardiography-derived regional EAT thickness was significantly greater in RAF patients than that in non-RAF patients((6.9±0.6) mm vs.(5.5±0.7) mm,t=4.649,P<0.05).The EAT thickness strongly correlated with patients age, body mass index,Ratio of left atrial diameter, mitral diastolic blood flow velocity and early diastolic velocity of mitral annulus, low density lipoprotein-cholesterol and thyroglobulin (r =0.62,0.57,0.68,0.59,0.73,0.76;P < 0.05).Cox multivariable analysis showed that, duration of AF history (95%CI:1.01-1.15,P =0.040),left atrial diameter (95%CI:1.07-1.24,P =0.021) and EAT thickness (95%CI:1.05-1.79,P =0.009) were independent predictors of RAF.Conclusion Echocardiography-derived regional EAT thickness combined with left atrial diameter may serve as effective parameters in identifying patients at the highest risk for RAF.
