1.Relationship between the rs3179060C/A polymorphism in TNF- gene and the pathogenesis of hyperandrogenism of polycystic ovary syndrome in Han Chinese racial origin

Objective To explore the association between rs3179060C/A polymorphism in the ex-on 1 of TNF-a gene and hyperandrogenism and polycystic ovary syndrome (PCOS). Methods One hundred thirty PCOS women and one hundred seventy five normal women as controls were enrolled in this study. The genotypes were screened by polymerase chain reaction-On/off switch and the product was isolated by e-lectrophoresis on a 2. 5% agarose gel containing ethidium bromide and visualized using an ultraviolet transil-luminator. The relationship of TNF-a alleles to serum testosterone level was analyzed in PCOS patients. Results Three genotypes were identified, corresponding to CC, CA, AA, and two alleles were screened, corresponding to C and A. The frequencies of the CC, CA, AA genotypes were 58. 4% ,23.1% ,18.5% vs. 72. 0% , 17.7% , 10. 3% in PCOS group and control group, showing statistically significant difference between two groups( P < 0.05 ). The allelic frequency was 70.0% for the C allele and 30.0% for the A in P-COS group, and 80. 9% for the C allele and 19. 1% for the A in control group, respectively, showing statistically significant between two groups ( P <0.05). The relationship was not observed between rs3179060C/ A polymorphism and serum testosterone level in PCOS patients in Han Chinese racial origin ( P >0.05). Conclusions The TNF-a gene rs3179060C/A polymorphism may be a risk factor for the pathogenesis of P-COS in Chinese women, but it was not effect on hyperandrogenism in PCOS women.
2.Analysis of Vancomycin serum concentration and adverse reaction in 213 neonates

Objective To investigate the relationship of Vaneomycin serum concentration and drug adverse reactions (ADRs) in neonates.Methods Two hundred and thirteen neonates who were treated by Vancomycin and had their serum concentration monitored were recruited.The number of cases which had liver,kidney and hearing damage after treatment was calculated.The correlation between 3 different serum concentrations (<5 mg/L,5-10 mg/L,> 10 mg/L) and ADRs were analyzed.Results The mean level of Vancomycin in serum was (4.36 ± 4.99) mg/L The total incidence of ADRs was 17.8% (38/213 cases).The main ADRs were liver damage and hearing impairment,whose incidences were 9.9% (21/213 cases) and 7.5% (16/213 cases),respectively,and they were were significantly higher than kidney danage (0.5%,1/213 cases) (x2 =19.172,P =0.000;x2 =13.785,P =0.000).There was no significant difference be-tween the inidence ratio of liver damage and that of the hearing impairment (x2 =0.330,P =0.566).The ADRs ratio among 3 different serum concentrations was 17.0% (24/141 cases),17.6% (9/51 cases) and 23.8% (5/21 cases).There was no significant difference between the ADRs ratios and distribution of different serum concentrations (x2 =0.576,P =0.750).Moreover,the ADRs rates didn't increase along with the serum concentrations (Z =0.648,P =0.517).Except for aspartate transaminase,the indicators of liver and kidney function varied significantly after vancomycin treatment (P <0.05).But there was no clinical significance because the mean value was in normal range.By Bivariate Correlation analysis,Vancomyein serum concentration had no significant influence on liver and kidney function (P > 0.05).Conclusions Neonates treated by Vancomycin had reatively high ratio of the liver damage and the hearing impairment.There is no significant correlation between Vancomyein serum concentration and ADRs in neonates.

Aim To study the effect of Macrophage colony-stimulating factor(M-CSF) on MMP-9 in RAW 264.7 cell and explore the relationship between atherosclerosis caused by M-CSF and the activity of MMP-9. Methods Gelatin zymography analysis was used to investigate the effect of M-CSF and PD98059 on the activity of MMP-9 in cultured RAW 264.7 cell.Western blot was used to study the effect of M-CSF and PD98059 on the express of p-ERK1/2 in cultured RAW 264.7 cell. Results The enzyme activity of MMP-9 was significantly increased after 24-hour M-CSF treatment.Meanwhile, M-CSF upregulated the expression of p-ERK1/2. Pre-treatment with PD98059 blocked partly the increased expression of p-ERK1/2 and the activity of MMP-9 induced by M-CSF. Conclusion M-CSF can induce the secretion of MMP9 in RAW 264.7 cell, which may be mediated by the phosphorylation of ERK1/2.

OBJECTIVETo investigate the effect of the fibronectin extra domain A on the aggressiveness of salivary adenoid cystic carcinoma (SACC) cells, via the clustered regularly interspaced short palindromic repeats (CRISPR)/ associated proteins (Cas) system.
METHODSOne sgRNA was designed to target the upstream of the genome sequences of extra domain A(EDA) exon and the downstream. Then the sgRNA was linked into plasmid PX-330 and transfected into SACC-83 cells. PCR and DNA sequence were used to testify the knockout cells, and the monoclones of EDA absent SACC cells were selected (A+C-2, A+C-6, B+C-10). CCK-8 cell proliferation and invasion was then tested in control group and the experimental group.
RESULTSThe sgRNA was successfully linked into PX-330 plasmid. Part of adenoid cystic carcinoma cells' SACC-83 genomic EDA exon was knocked out, and the knockdown efficiency was above 70%, but the total amount of fibronectin did not change significantly. Three monoclones of EDA absent SACC- 83 cells were successfully selected with diminished migration and proliferation.
CONCLUSIONSThe CRISPR/Cas9 system was a simplified system with relatively high knockout efficiency and EDA knockout could inhibiting SACC cell's mobility and invasiveness.

5.CT initial diagnosis value of nasopharyngeal fibroangioma

Objective To explore the diagnostic value of multi-slice spiral CT in clinical stage of nasopharyngeal fi-broangioma. Methods The clinical data and imaging findings of CT in 40 patients with nasopharyngeal fibroangioma confirmed by operation and pathologic examination were retrospectively analyzed. The size, shape, density, boundary, strengthen and the changes of the adjacent structure of the mass were observed and analyzed. Results According to Radkowski classification, 3 cases were evaluated as stageⅠa, 9 cases were evaluated as stage Ⅰb, 8 cases were eval-uated as stage IIa, 10 cases were evaluated as stage IIb, 5 cases were evaluated as stage IIc, 3 cases were evaluated as stageⅢa and 2 cases were evaluated as stageⅢb. Most corrosive location were in nasopharynx, pterygopalatine fos-sa, sphenoid sinus, maxillary sinus, eye socket and followed occurred in fratemporal fossa, and few corrosive location were in sclerotin of pars buccalis, cavernous sinus and basis cranii. The tumor showed uniform and slightly lower den-sity but obviously strengthening after administration of Gd-DTPA. Conclusion Multi-slice spiral CT has important clinical value for the clinical stage diagnosis, preoperative localization, evaluating the postoperative extent and the prognosis of the nasopharyngeal fibroangioma.
6.Effect of core symptoms of attention deficit hyperactivity disorder on behavioral problems in children with ADHD propensity

ObjectiveTo explore the influence of the core symptoms of attention deficit hyperactivity disorder (ADHD) on behavioral problems of children with ADHD propensity, so as to provide references for early identification and targeted intervention for children with ADHD propensity. MethodsFrom July to August 2021, 25 children with ADHD propensity were screened as the ADHD propensity group, and 25 children matched for age, gender and grade were included as the normal group in an elementary school in Guangzhou. ADHD core symptoms were assessed by the Chinese version of the Swanson Nolan and Pelham, version IV-parent form for ADHD (SNAP-IV), and behavioral problems were assessed by Questionnaire-Children with Difficulties (QCD) and Conners Parental Symptom Questionnaire (PSQ). Spearman correlation analysis was used to examine the correlation between ADHD core symptoms and QCD and PSQ scores, and hierarchical linear regression analysis was used to explore the effect of ADHD core symptoms on behavioral problems. Results① The differences between the groups showed that both attention deficit and hyperactivity-impulsivity factor scores were higher in the ADHD propensity group than those in the normal group (t=7.771, 6.726, P<0.01). ② Correlation analysis showed that the attention deficit factor score was negatively correlated with QCD total score (r=-0.440, P<0.05), and positively correlated with the learning problem factor score of PSQ (r=0.457, P<0.05). The score of hyperactivity-impulsivity was negatively correlated with score of anxiety factor in PSQ (r=-0.457, P<0.05), and positively correlated with impulse-hyperactivity factor score (r=0.552, P<0.01). ③ Hierarchical linear regression analysis showed that the attention deficit factor score negatively predicted the total score of QCD (B=-0.682, P<0.05, R2=0.468). The hyperactivity-impulsivity factor score had a negative predictive effect on the anxiety factor score of PSQ (B=-0.048, P<0.05, R2=0.367), and had a positive predictive effect on the impulsivity-hyperactivity factor score (B=0.077, P<0.01, R2=0.424). ConclusionChildren with ADHD propensity have significant attention deficit symptoms, hyperactivity-impulsivity symptoms and behavioral problems, and the attention deficit may be the main cause of their daily behavioral problems, while hyperactivity-impulsivity may be the main cause of their impulsive-hyperactivity problems.

The successful retrieval of ancient mitochondrial DNA (mtDNA) from Neanderthals provides powerful experimental evidence that clarifies the arguments between the out-of-Africa and multiregional models of evolution. However, the lack of nuclear DNA from Neanderthal fossils and mtDNA of early modern human fossils dating back to approximately the same time in the Pleistocene constitutes a limitation that may compromise the significance of mtDNA phylogenetic analysis. In this report, we introduce a mitochromic analysis using Neanderthal mtDNA as a foreign transgene and humans as a naturally occurring transgenic species. Forty Neanderthal mtDNA retrievable nuclear fragments were identified by blasting human genome data with Neanderthal mtDNA. Five of the 40 fragments exhibited higher correlation with Neanderthal mtDNA than those with modern human mtDNA. Furthermore, these five nuclear fragments harbor Neanderthal mtDNA-unique haplotypes. Based on the 98%+ identity between Neanderthal and modern human mtDNA when compared by groups, we suggest that some of the modern human nuclear fragments retrieved using Neanderthal mtDNA may aid in decoding Neanderthal genetic information, and also may simultaneously demonstrate a close genetic evolutionary relationship between modern humans and Neanderthals.
