2.Detection of intracranial aneurysms with spiral CT angiography and MPVR

Objectives:To explore the optimal method of spiral CT angiography and MPVR in the detection of intracranial aneurysms. Methods:Eleven patients with clinical suspicion of intracranial aneurysms underwent spiral CT angiography, then postprocessed with MPVR. Results:,Five anterior communicating aneurysms,1 posterior communicating aneurysm and 1 base aneurysm were found with MPVR. Conclusions: Lesions at the circle of Willis, the main trunk of the carotid and the vertebrobasilar arteries, including their branches could be visualized perfectly with MPVR by optimizing the thickness, center, angle of rotation and window level, by which more intracranial aneurysms could be found.
3.Expressions of MMP-2 and MMP-9 in esophageal cancer

Objective To analyze the expression levels of matrix metallo proteinase-2 (MMP-2) and matrix metallo proteinase-9 (MMP-9) in serum of patients with esophageal cancer,and to analyze the interrelations among MMP-2,MMP-9 and clinicopathologic characteristics of esophageal cancer.Methods The levels of serum MMP-2,MMP-9 from 60 patients with esophageal cancer were detected by enzyme linked immunosorbent assay.The interrelations among MMP-2,MMP-9 and clinicopathologic characteristics of esophageal cancer including age,gender,histological type,whether or not lymph nodes metastasis and TNM staging were analyzed.Results The levels of MMP-2 and MMP-9 in control group were 61.31 ng/ml and 87.67 ng/ml,the levels of MMP-2 and MMP-9 in patients with esophageal cancer were 121.66 ng/ml and 169.73 ng/ml.The levels of MMP-2 and MMP-9 were significantly higher in patients with esophageal cancer than that of general population (t =3.015,P =0.007; t =2.037,P =0.04).The level of MMP-2 was significantly associated with whether or not lymph node metastasis (t =2.150,P =0.04) and clinical staging (t =2.186,P=0.03).The level of MMP-9 was significantly associated with whether or not lymph node metastasis (t =2.390,P =0.02) and clinical staging (t =2.149,P =0.03).The levels of MMP-2 and MMP-9 have no reference to age,gender and histological type (all P>0.05).Conclusion MMP-2 and MMP-9 may correlate with tumor invasion and metastasis in patients with esophageal cancer.
4.The lethal activity of lymphokine activated killer cells from umbilical blood on some human cancer cells

Objective To investigate the feasibility of lymphokine activated killer(LAK) cells induced from cord blood used as adoptive cellular immunotherapy for human cancer.Methods Mononuclear cells were separated from umbilical blood(UBMC) by Ficoll,and stimulated by IL-2.The phenotypes(CD3/ CD4/ CD8) of the mononuclear cells were assayed by Flow cytometry,and their lethal activity on K562 or SKOV6 assayed by MTT colometric.The peripheral blood mononuclear cells were used as the control.Results The in vitro anti-tumor effect of LAK from cord blood was significant.Conclusion LAK from cord blood can be a source of adoptive cellular immunotherapy in the treatment of human cancer.

6.Effect of capacity for liquor on vomiting induced by chemotherapy in male patients with nasopharyngeal carcinoma

Objective To explore the effect of the single maximum of alcohol consumption on vomiting induced by chemotherapy in male patients with nasopharyngeal carcinoma.Methods According to the single maximum of alcohol consumption,48 cases of male patients with nasopharyngeal carcinoma were divided into two groups:group A (small capacity for liquor group 10-50 g),group B (large capacity for liquor group >50 g).The amount of alcohol intake was assessed by a questionnaire.The responses of vomiting and the effects of antiemetic therapy between the two groups during the first cycle after induction chemotherapy were observed.Results The incidence rates of vomiting induced by chemotherapy between the two groups were 52.0% and 17.4%,respectively.There were significant differences between the two groups (x2 =6.273,P <0.05).The incidence rates of grade 0,1,2 and 3 of vomiting in group A were 48.0%,20.0%,20.0%,12.0%,in group B were 82.6%,13.0%,4.4%,O,respectively.There were significant differences between the two groups (x2 =6.013,P =0.024).In the two groups,the complete control rate for acute vomit were 48.0% and 82.6% (x2 =6.273,P =0.012),for delayed vomit were 36.0% and 65.2% (x2 =4.090,P =0.043).There were also significant differences between the two groups.Conclusion With the increasing of capacity for liquor,the incidence of vomiting is significantly reduced,the degree of vomiting is distinctly alleviated,and the antiemetic efficacy is distinctly increased.
7.Absolute quantification of calf muscle metabolites by proton 1H-MR spectroscopy

Objectiye To measure longitudinal (T1) and transverse (T2 ) relaxation time of metabolites in m. soleus (SOL) and m. tibialis anterior TA of healthy volunteers at 3.0 T through 1H-MRS and optimize measurement protocols. Methods Altogether 24 healthy volunteers were recruited in the study. All subjects signed a letter of informed consent. After divided into 2 groups randomly by the table of random number, 1H-MRS measurements with stimulated echo acquisition mode (STEAM) sequence were undertaken in SOL and TA separately. Progressive saturation method was used for T1 measurement. Spectra with 8 different TRs (770,900,1000, 1100,1200,1500,2000 and 3000ms ) were acquired with TE=20 ms.T2 time was measured by changing TE. Altogether 8 TEs (20,30,45,60,90,135,200 and 270 ms) were used with TR = 3000 ms. Metabolites' concentration was calculated through T1 and T2 correction using water as internal reference. The t test was used for statisties. Results Altogether 22 groups of data were gained ( 12 for SOL, 10 for TA ) . T1 value of water, Creatine-CH3 ( Cr3 ), Trimethyl amonium ( TMA ),extramyocellular lipid (EMCL) and intramyocellular lipid (IMCL) in SOL were ( 1384. 0 ± 36. 9 ),( 1064. 0 ± 167.0), (964. 2 ± 144. 0 ), ( 373.0 ± 46. 8 ), ( 374. 7 ± 20. 6) ms respectively and T2 value were (26.5 ±1.2), (100.2±19.3), (149. 1 ±32.7), (81.4±5.2), (84.7±4.2) ms. InTA T1 value of water, Cr3, TMA, EMCL, and IMCL were ( 1307. 0 ± 24.4), (945.7 ± 132. 0), (968.3 ± 127. 0),(372. 7 ± 39. 2), (412. 8 ±80. 2) ms respectively and T2 value were (27. 1 ± 0. 9), (135.3 ± 18. 2 ),(62.1 ± 6. 0), ( 84. 3 ± 4. 0 ), ( 90. 7 ± 3.2 ) ms. After corrected by the calculated relaxation times, the concentrations of Cr3 in SOL and TA were (33. 1 ± 3.7) and (31.7 ± 3. 1 ) mmol/kg respectively, TMA (35.2±3.2) and (32.9 ±5.2) mmol/kg, EMCL (12.2 ±5.0) and (8.9 ±4.9) mmol/kg, IMCL (9. 0 ± 2. 4) and (3.0 ± 0. 8 ) mmoL/kg. IMCL in TA was much lower than SOL with statistical significant ( t = 8. 044, P < 0. 01 ), the difference between other metabolites were not statistically significant( t = 0. 926,1. 264, 1. 542, P > 0. 05 ) . Conclusions Accurate relaxation time was measured at 3.0 T of the metabolites in skeletal muscles of healthy adult human. After corrected by the relaxation times, the absolute concentrations calculated were consistent with the reported results. Quantitative knowledge of muscle NMR relaxation time was a prerequisite for absolute quantification of metabolites using the 1H-MRS and also was useful for optimizing measurement protocols.
8.The effects of vardenafil on high altitude pulmonary hypertension in rats

Objective To investigate the effect of vardenafil on high altitude pulmonary hypertension in rats, and the possible mechanism thereof. Methods Thirty rats were randomly divided into three groups:control group with normal-pres?sure and normal-oxygen (group C), pulmonary hypertension group with low-pressure and low-oxygen (group P), and the group treated by vardenafil in low-pressure and low-oxygen condition (group V). The rats of group P and group V were ex?posed to low-pressure and low-oxygen condition in an auto-modulating hypobaric and hypoxic cabin to simulate 5 000 m high altitude environment (air pressure 50 kPa, oxygen concentration 10%) for 8 hours daily. Vardenafil (1 mg/kg) was given by gastrogavage to rats in group V once daily for 4 weeks, while the isodose distilled water was given by gastrogavage to rats in group C and group P. The mean pulmonary arterial pressure and right ventricular mass index were measured respectively after 4-week treatment. Morphologic changes of peripheral pulmonary artery were detected by light microscope. The serum levels of nitric oxide (NO) and endothelin-1 (ET-1) were detected as well. Results The pulmonary arterial pressure and right ventricular mass index were significantly higher in group P than those of group C and group V (P<0.05). The ratio of vascular medial wall thickness to external diameter (WT%) and the ratio of pulmonary artery wall area to tube area (WA%) were significantly increased in group P than those of group C and group V (P<0.05). Furthermore, the serum level of NO was significantly lower in group P than that of group C and group V, but the serum level of ET-1 was significantly increased compared with that of group C and group V (P<0.05). Conclusion Vardenafil can effectively reduce the pulmonary arteri?al pressure, and attenuate pulmonary vessels and right ventricle remodeling induced by high altitude pulmonary hypertension.
9.Homology of carbapenem-resistant Klebsiella pneumoniae in an intensive care unit

Objective To understand antimicrobial resistance and homology of carbapenem-resistant Klebsiella pneumoniae (CRKP)from an intensive care unit (ICU).Methods 11 CRKP isolates from patients and environment of an ICU in a hospital were performed antimicrobial susceptibility testing,the homology of CRKP was analyzed by randomly amplified polymorphic DNA (RAPD)method.Results Of 11 CRKP isolates,8 were from patients,and 3 from environment,the main specimen was sputum(n =6,54.55%).Antimicrobial susceptibility testing results re-vealed that 10 (90.91%)CRKP isolates were resistant to ciprofloxacin;11 isolates were susceptible to compound sulfame-thoxazole,intermediate to imipenem,and resistant to other antimicrobial agents(100%).All 11 CRKP isolates had 3 bands,and were divided into two types:(typeⅠ,n=10;type Ⅱ,n=1).Conclusion Antimicrobial resistance of Kleb-siella pneumoniae in ICU is serious,CRKP isolated from ICU patients and environment are of the same clone. Cleaning,disinfection,and monitoring of ICU environment should be strengthened,which is helpful for reducing, timely warning of multidrug-resistant organisms,and reducing healthcare-associated infection.
10.Determination of Copper Ion in Rabbit with Implantation of Copper Intrauterine Device

Objective To develop a method for determination of serumal copper ion in rabbit with implantation of copper intrauterine device.Methods At different time points after implantation,the serumal copper ion concentrations were determined by microwave digestion-inductively coupled plasma mass spectrometry(ICP-MS) using scandium(Sc) and indium(In) as internal standards to compensate sample matrix effects,and the pharmacokinetics parameters were calculated in order to reflect copper ion release and metabolic rule.Results The serumal copper ion concentrations were kept at a low and stable level.The recoveries were in the range of 98.7%-113.3%,with the relative standard deviations of less than 5.0%.Conclusion The analytical method is simple,fast,and can be used for the determination of serumal copper ion in rabbit.
