1.In vitro cultivation and identification of epithelial cells of keratocystic odontogenic tumor

Objective:To establish a method of in vitro cultivation of epithelial cells of keratocystic odontogenic tumors(KCOT).Methods: Tissue block and enzyme digestion techniques were used for primary cultivation of KCOT cells.The cells were grown in keratinocyte-serum free medium(K-SFM).The biological characteristics of the cultivated keratinocytes were identified by phase microscopic observation and by immunohistochemistry of cytokeratin,vimentin,cytokeratin10 and 14.Results: KCOT keratinocytes could survive for 30-50 days in K-SFM after passing 2-4 generations.Cells were polygon and showed typical slabstone-like appearance.Immunostaining showed positive staining for cytokeratin antibody,and negative for vimentin.Conclusion: KCOT epithelial cells could be serially cultured in vitro in K-SFM by techniques suggested in this study.
2.Clinical observation of via -nasal ileus tube interventional treatment on 27 patients with early postoperative inflammatory intestinal obstruction

Objective To explore the method and clinical effect of via -nasal ileus tube combined urografin to treat early postoperative inflammatory intestinal obstruction.Methods Retrospective analyzed the data of 27 patients with early postoperative inflammatory intestinal obstruction who treated through via -nasal ileus tube intervention. Results The 27 patients were cured through conservative treatment and the cure rate was 100%.The symptom of 10 cases got complete remission 4 to 7 days after put the ileus tube,17 cases got complete remission 8 to 13 days. The passage of gas by anus recovery time was average (12.3 ±2.6)days,time of therapy was average (16.1 ± 3.7)days.The patients were followed up for 6 months to 1 year,none of the patients relapsed.Conclusion The early postoperative inflammatory intestinal obstruction should be treated through conservative treatment positively,decom-pression by via -nasal ileus tube combined urografin has important role to diagnose and treat the disease,the therapy can enhance the therapeutic effect validly and shorten the treatment cycle,it is safe to use in clinic and the effect is satisfactory.
3.Study on Inhibitory Effects of Triptolide on the Proliferation of Fibroblast-like Synovial Cells from Pa-tients with Rheumatoid Arthritis in vitro

OBJECTIVE:To investigate the effects of triptolide(TP)on the proliferation of fibroblast-like synoviocytes(FLS) from patients with rheumatoid arthritis(RA)in vitro. METHODS:5 RA patients received knee arthroplasty or synovectomy to ob-tain synovial tissue. FLS was isolated,cultured and identified,and then incubated in the presence of TP [0 (blank control),50, 100 and 200 nmol/ml] for 24,48 and 72 h. The effects of TP on FLS was evaluated by MTT,and then proliferation inhibitory rate was calculated;flow cytometry was used to detect the apoptosis and cell cycle of FLS. RESULTS:The inhibitory rates of TP(50, 100 and 200 nmol/ml)on the proliferation of FLS were 17.46%-52.56%,which was positively correlated with drug concentration. Compared with blank control group,100 and 200 nmol/ml TP could increase the percentage of cells in G0/G1 phase and decrease the percentage of cells in S phase,with statistical significance(P<0.05);200 nmol/ml TP could induce cell apoptosis. CONCLU-SIONS:TP could inhibit the proliferation and also could induce the apoptosis of FLS in RA patients in vitro,which may be one of its mechanism for treating RA.
4.A new phenolic amide glycoside from Cimicifuga dahurica.

A new phenolic amide glycoside, cimicifugamide A (1) along with four known compounds, trans-feruloyl tyramine 4-O-beta-D-glucopyranoside (2), (+)-isolariciresinol 3-O-beta-D-glucopyranoside (3), cimidahurine (4), and 24-epi-7, 8-didehydrocimigenol-3-O-beta-D-xylopyranoside (5) were isolated from the rhizomes of Cimicifuga dahurica. Compound 3 was identified as a lignan and has been obtained from Cimicifuga genus for the first time. The structure of compound 1 was elucidated by IR, UV, HR-MS and NMR spectroscopic methods.
5.Comparative Study on the Therapeutic Efficacy between Medicinal Cupping and Bloodletting for Chronic Low Back Pain

Objective To compare the therapeutic efficacy between medicinal cupping and collateral-pricking bloodletting therapy in treating lumbar intervertebral disc herniation (LIDH) and non-specific low back pain.Method Sixty LIDH patients were randomized into treatment group 1 and control group 1, 30 cases in each group. Sixty non-specific low back pain patients were randomized into treatment group 2 and control group 2, 30 cases in each group. Treatment group 1 and 2 were intervened by medicinal cupping, while control group 1 and 2 were by collateral-pricking bloodletting. The Visual Analogue Scale (VAS) and Oswestry Disability Index (ODI) were observed before and after intervention.Result The VAS and ODI scores were significantly changed after intervention in treatment group 1, treatment group 2, and control group 2 (P<0.05). The VAS score was significantly changed after intervention in control group 1 (P<0.05); the ODI score didn’t show significant change after intervention in control group 1 (P>0.05). After treatment, there was a significant difference in comparing the ODI score between control group 1 and control group 2 (P<0.05).Conclusion Medicinal cupping and collateral-pricking bloodletting are both effective in treating chronic low back pain, and medicinal cupping has its advantage in improving ODI compared to the bloodletting therapy.

7.Effects of long-term low-dose γ-rays exposure on radiosensitivity of human B lymphoblast cells

Objective To investigate the effects of long-term low-dose radiation (LDR) of γ-rays on the proliferation and radiosensitivity of human lymphoblast cells HMy2.CIR (HMy) and to elucidate the underlying mechanism.Methods HMy cells were divided into control group and long-term LDR group.For the long-term LDR treatment,HMy cells were fractionally exposed to a low dose of γ-rays,which could enhance cell proliferation,3 times per week for 4 weeks.After the long-term LDR exposure,part of the control and long-term LDR exposed cells were further irradiated with a challenging dose (2 Gy) of γ-rays.Then cell proliferation and radiosensitivity were assayed by CCK-8 kit,cell apoptosis,and γ-H2AX formation was measured by flow cytometry.Gene expressions of cyclinD1,PCNA,bcl-2 and bax were detected by RT-PCR.Results The long-term LDR significantly increased cell proliferation (t =9.607,P < 0.01) accompanied with up-regulation of cell cycle regulation gene cyclinD1 (t =6.869,P < 0.01),proliferation regulation gene PCNA (proliferating cell nuclear antigen) (t =9.229,P < 0.01) and bcl-2 gene (t =2.662,P < 0.05),but decreased the expression of pro-apoptotic gene bax (t =19.908,P <0.01) in HMy cells.Compared to untreated cells,the long-term LDR decreased cell radiosensitivity (t =8.896,P < 0.01),including apoptosis induction (t =4.762,P < 0.01) and γ-H2AX formation (t =10.264,P<0.01).Conclusions The long-term LDR promoted cell proliferation by up-regulating cell cycle related genes,while it reduced the radiosensitivity of HMy cells with acquisition of apoptotic resistance.
8.Anti-endothelin receptor type A autoantibody in lupus associated pulmonary arterial hypertension

Objective To investigate autoantibody against endothelin receptor type A (ENRA-Ab) in patients with systemic lupus erythematosus associated pulmonary arterial hypertension (SLE-PAH).The possibility of autoantibody-mediated pathogenesis in the development of SLE-PAH has also been explored.Methods ENRA-Ab in the serum of SLE-PAH and controls were detected by using a human ETRA epitope peptide-based ELISA.The clinical relevance of ENRA-Ab in SLE-PAH was analyzed.Proliferation of vascular smooth muscle cells (SMCs) and permeability of endothelial cells in vitro under the stimulation of polyclonal ENRA-Ab IgG were assessed.The expressions of PAH-related markers, i.e., 5-HTT, PDGFR-b, VEGF-A and PDGF-B were measured by qPCR.The effect of ENRA-Ab in vivo was also determined in a suboptimaldose monocrotaline-induced model with the assessment of right ventricle hypertrophy index and pathology parameters.Independent t-test, Tukey-Kramer test of variance analysis and Pearson' s correlation analysis were used for statistical analysis.Results ENRA-Abs was presented in a higher occurrence in SLE-PAH (35/85,41%) compared with controls (0/60;0, 13/80, 16%).There was a significant correlation between ENRA-Ab and echocardiograph estimated pulmonary arterial systolic pressure (r=0.392, P=0.002) in SLE-PAH.ENRA-Ab could promote SMCs proliferation, disrupt endothelial barrier and up-regulate PAH-related markers expression,which could be blocked in the presence of ETR antagonist.ENRA-Ab aggravated right ventricle hypertrophy and vascular remodeling in vivo.Conclusion ENRA-Ab is a new biomarker, in SLE-PAH, which may mediate PAH development in SLE.
9.Effect of Alkanes on the Production of a Solvent-stable Extracellular Protease by the Organic Solvent Tolerant Bacterium Bacillus licheniformis YP1

In this paper, the effect of 5% (V/V) n-alkanes (e.g, n-Heptane, n-Octane, n-Decane, n-Dodecanen-Tetradecane and n-Hexadecane) on the growth and protease production of organic-solvent-tolerant- bacte-rium Bacillus licheniformis YP1 was studied. 5%(V/V) n-alkanes had no effect on the stability of YP1 prote-ase. 5% (V/V) n-alkanes had no notable influence on the yield of strain YP1 but dramatically affected theprotease production. The presence of n-Heptane, n-Octane and n-Decane deeply repressed the protease pro-duction; however n-Dodecane, n-Tetradecane and n-Hexadecane enhanced the protease production promi-nencely. The concentration of n-Tetradecane (1%-8%, V/V) had a direct ration with the protease production.The detailed experiments showed that the notable increase of protease activity appeared at the late logarithmof cultivation compared with the blank. The cell shape of YP1 strain remarkably decreased when grown inthe presence of n-Tetradecane. This is the first report about the effect of n-alkanes on the protease productionby the solvent tolerant bacterium.
