1.Clinical research of psychological intervention combined with celecoxib and physiotherapy on vertebral artery type of cervical spondylosis

Objective To analyse effects of psychological intervention combined with celecoxib and physiotherapy on vertebral artery type of cervical spondylosis.Methods 134 patients with vertebral artery type of cervical spondylosis were grouped into two groups.Two groups were treated with physiotherapy and celecoxib,observation group was treated another with psychological intervention.Results Total efficiency of observation group was higher than that of control group(P<0.05).After treatment,symptom score,CASCS score,mean blood flow velocity in left vertebral artery and right vertebral artery,TNF-α and IL-6 level of observation group were better than that of control group(all P<0.05).During treatment, there was no obvious adverse reaction in the two groups.Conclusion Sychological intervention can improve the therapeutic effect of patients with cervical spondylosis of vertebral artery type.
2.Luteolin ameliorates endothelial dysfunction induced by oxidative stress

AIM: To evaluate the protective effect of luteolin on endothelial dysfunction induced by tert-butyl hydro-peroxide (t-BOOH). METHODS: We observed the effect of luteolin on t-BOOH-induced contractions in the aorta rings with or without endot helium, which were incubated with luteolin (10-6 to 10-4 mol/L) for 30 min before determining the concentration-response to t- BOOH. Cultured endothelial cell line (ECV304) was pretreated with different concentrations of luteolin (10-6 to 10-4 mol/L) for 30 min and then exposed to 10 -5 mol/L t-BOOH for 24 hours. Cell morphology was observed , and cell viability was determined by MTT assay. Meanwhile, RT-PCR was used to measure the expression of eNOS and COX-1. RESULTS: Increasing concentrations of t-BOOH produced concentration-dependent cont ractions in aorta rings isolated from rats, luteolin effectively attenuated the contraction in a concentration-dependent manner, and the relaxation response was greater in intact endothelium segments. In MTT and RT-PCR assays, luteolin effectively reduced the cytotoxicity of t-BOOH to endothelium cell and increased the expression of nitric oxide synthase (eNOS) mRNA, which was greatly down-regulated by t-BOOH. CONCLUSION: Luteolin effectively protects the endothelium from the impairment of oxidative stress, and the protection could be related to its negative modulation towards t-BOOH-induced contractions in the aorta.
3.Effect of laparoscopic cholecystectomy on systemic stress response

Objective To investigate the systemic stress responses of laparoscopic cholecystectomy(LC). Methods Seventy patients with cholecystolithiasis were randomly divided into two groups: LC and open cholecystectomy(OC). Peripheral blood samples were obtained during the preoperative day and 1, 3, 5, 7 day after operation. Serum cytokines levels (TNF-α, IL-1β and IL-6) and C-reactive protein(CRP) were measured. Results Changes of TNF-α and IL-1β before or after surgery in both groups showed no significant difference(P>0.05). Serum concentrations of IL-6 and CRP after surgery in both groups were significantly higher than those before surgery(P<0.05); but there were no significant difference between the two groups(P>0.05). Conclusions The systemic stress responses of LC have no significant difference comparing with OC.
4.Effect of TRPC6 knockdown on puromycin aminonucleoside-induced podocyte injury.

This study was aimed to construct eukaryotic expression vectors carrying the small hairpin RNA (shRNA) targeting TRPC6 gene and investigate the effect of TRPC6 knockdown on puromucin aminonucleoside (PAN)-induced podocyte injury. Two DNA sequences containing the small hairpin structure targeting TRPC6 were designed, synthesized and then inserted into the green fluorescence protein (GFP)-contained plasmids (pGC) to establish the plasmids pGCsi-TRPC6A and pGCsi-TRPC6B. Plasmids expressing scrambled shRNA were used as negative control and named pGCsi-NC. These plasmids were transfected into a conditionally immortalized murine podocyte cell line by using liposome. Flow cytometry was used to examine the transfection efficiency. TRPC6 mRNA and protein expression levels were detected by RT-PCR and Western blotting. Cultured podocytes were divided into four groups: control group, PAN treatment group, PAN+TRPC6 shRNA transfected group and PAN+scrambled shRNA transfected group. The paracelluar permeability to BSA was evaluated by Millicell-PCF Inserts and cell viability was measured by the trypan blue assay. Immunofluorescent assay was used to observe the distribution of α-actinin-4 and α-tubulin. The results showed that the transfection efficiency of the shRNA expression vector was about 45%. Expression levels of TRPC6 mRNA and protein were downregulated after transfection with pGCsi-TRPC6A and pGCsi-TRPC6B. Knocking down TRPC6 gene could effectively reverse the PAN-induced increase in the paracelluar permeability to BSA. The distribution of α-actinin-4 and α-tubulin was disrupted after treatment with PAN, which was reversed by knocking down TRPC6 gene. It was concluded that knocking down TRPC6 gene could effectively prevent podocytes from the permeability increase induced by PAN, which may be related to the regulation of podocyte cytoskeleton.
5.Value of ultrasonography in the diagnosis of hermaphroditism

Objective To evaluate the clinical value of ultrasonography in the diagnosis of hermaphroditism.Methods Ultrasonographic appearances of 45 cases with hermaphroditism were reviewed retrospectively,and the results were compared with clinical data.Results All cases were confirmed pathologically.5 cases showed true hermaphroditism (11.1%),14 cases showed male pseudohermaphroditism (31.1 %),24 cases showed female pseudohermaphroditism (53.3 %),2 cases showed gonadal dysgenesis(4.4%).Compared with pathological results in the ultrasound examination,4 cases showed true hermaphroditism,the coincidence rate was 80%.11 cases showed male pseudohermaphroditism,the coincidence rate was 78.6%.20 cases showed female pseudohermaphroditism,the coincidence rate was 83.3%.1 cases showed gonadal dysgenesis,the coincidence rate was 50.0%.Conclusions Ultrasonography can not only evaluate internal reproductive organs in the patients,but also estimate the site,size,morphology and structure,which provides important imaging evidence for the clinical diagnosis and treatment.
6.Comparation and considerations for general notice between the Chinese Pharmacopoeia and the Japanese Pharmacopoeia

Objective: The characteristics and differences of the general notice between the Chinese Pharmacopoeia and the Japanese Pharmacopoeia were investigated to provide references and suggestions for the compilation of the Chinese Pharmacopoeia.
Methods: From the perspective of frame structure and main contents, the general notice between the Chinese Pharmacopoeia and the Japanese Pharmacopoeia was compared.
Results: Each volume of the Chinese Pharmacopoeia had its general notice, including 34 to 48 items and 10 to 12 chapters based on different varieties collected in each volume. The Japanese Pharmacopoeia had 49 items not arranged by chapters. There are many differences on the general notice between the Chinese Pharmacopoeia and the Japanese Pharmacopoeia, such as the definitions and expressions of names, determination of appearance, revision rules, risk assessment and quality control conception. The framework of the general notice in the Chinese Pharmacopoeia was clear, the content was specific and the operation was friendly. The term description of the general notice in the Japanese Pharmacopoeia was concise, and some terms need to be implemented under the guidance of professional knowledge.
Conclusion: In light of comparative study, every volume’s general notice of the Chinese Pharmacopoeia has its own characteristics. By integrating advanced analytical technique, combining the requirements with laws and regulations, and optimizing content and terms, all volume’s general notice could be explored to be coordinated and unified.
7.Correlation of serum ZO-1 and aβ2-GP levels with infarct lesion extent and degree of carotid atherosclerosis in patients with cerebral infarction

Objective To investigate the correlation of serum atresia zonule protein 1(ZO-1)and anti-β2-glycoprotein 1 antibody(aβ2-GP1)levels with infarct lesion extent and degree of carotid atherosclerosis in pa-tients with cerebral infarction.Methods A total of 103 patients with cerebral infarction treated in Baoding No.1 Central Hospital from January 2020 to January 2023 were selected as the study group,and 51 healthy subjects in the same hospital during the same period were selected as the control group.Patients with cerebral infarction were divided into large infarction group(infarct lesion extent>50 cm2),middle infarction group(infarct lesion extent as 10-50 cm2),and small infarction group(infarct lesion extent<10 cm2)according to the infarct lesion extent.According to the results of carotid ultrasound,the patients with cerebral infarction were divided into plaque stage group,thickening stage group and normal vessel wall group.Enzyme-linked im-munosorbent assay was used to detect serum ZO-1,aβ2-GP1 levels,and the levels of ZO-1 and aβ2-GP1 in each group were compared.Spearman correlation analysis was used to analyze the correlation between serum ZO-1,aβ2-GP1 levels and the infarct lesion extent and the degree of carotid atherosclerosis in patients with cerebral infarction.Results The study group had a significantly lower serum level of ZO-1 and a significantly higher serum level of aβ2-GP1 than the control group(P<0.05).Among the 103 patients with cerebral in-farction,there were 32 cases in the large infarction group,34 cases in the middle infarction group,and 37 cases in the small infarction group.The small infarction group had a significantly higher level of ZO-1 and a signifi-cantly lower level of aβ2-GP1 than the medium infarction group and large infarction group(P<0.05).The middle infarction group had a significantly higher level of ZO-1 and a significantly lower level of aβ2-GP1 than the large infarction group(P<0.05).Among 103 patients with cerebral infarction,there were 29 cases in the plaque stage group,38 cases in the thickening stage group,6 cases in the normal vessel wall group.The plaque stage group had a significantly lower serum level of serum ZO-1 and a significantly higher level of aβ2-GP1 than the thickening stage group and normal vessel wall group(P<0.05).The thickening stage group had a significantly lower serum level of ZO-1 and a significantly higher serum level of aβ2-GP1 than the normal ves-sel wall group(P<0.05).Spearman correlation analysis showed that the serum level of ZO-1 was negatively correlated with the degree of carotid atherosclerosis and the extent of infarction(P<0.05),and the serum level of aβ2-GP1 was positively correlated with the degree of carotid atherosclerosis and the extent of infarc-tion(P<0.05).Conclusion The levels of serum ZO-1 and aβ2-GP1 in patients with cerebral infarction are significantly correlated with the degree of carotid atherosclerosis and the extent of infarction,which have po-tential value in the diagnosis,treatment and prognosis of patients with cerebral infarction.
8.Effect of lateral position ventilation combined with vibration sputum drainage on patients with acute respiratory distress syndrome: a prospective randomized controlled trial

Objective To investigate the curative effect of lateral position ventilation combined with vibration sputum drainage on the patients with acute respiratory distress syndrome (ARDS). Methods A prospective randomized controlled trial was conducted. The patients with ARDS undergoing mechanical ventilation (MV) admitted to intensive care unit (ICU) of Linyi Central Hospital from January 2013 to June 2017 were enrolled, and they were divided into simple ventilation group and combined treatment group according to random number table. The patients in both groups received etiological treatment, protective ventilation strategy, sensitive antibiotics for anti-infection, and calefacient and humidifying treatment. The patients in the simple ventilation group received bilateral discontinuous alternative lateral position ventilation [pressure controlled ventilation (PCV), tidal volume (VT) ≤ 6 mL/kg, the inhaled oxygen concentration (FiO2) and positive end expiratory pressure (PEEP) was adjusted to maintain the airway plateau pressure (Pplat) ≤ 30 cmH2O (1 cmH2O = 0.098 kPa)], and those in the combined treatment group received lateral position ventilation combined with vibration sputum drainage, twice a day, 15 minutes each time. The parameters of respiratory function and inflammation as well as excretion of sputum before and after treatment were compared between the two groups, and the complication and prognosis were recorded. Results A total of 200 patients with ARDS were included, 4 patients were excluded because of severe pneumothorax, massive hemorrhage of the digestive tract, or elevated intracranial pressure, so 196 patients were finally enrolled in the analysis. There were 98 patients in the simple ventilation group and the combined treatment group, respectively. There were no significant differences in parameters of respiratory function and inflammation as well as excretion of sputum before treatment between the two groups. After treatment, the above parameters in both groups were improved, and the effect of combined treatment was more significant. Compared with the simple ventilation group, white blood cell count (WBC), procalcitonin (PCT), C-reactive protein (CRP) at 24 hours of treatment in the combined treatment group were significantly decreased [WBC (×109/L):9.1±1.6 vs. 11.8±3.6, PCT (μg/L): 14.5±2.4 vs. 22.7±3.2, CRP (mg/L): 32.2±6.3 vs. 67.2±7.2, all P < 0.01], the quantity of sputum excretion was significantly increased (mL: 49.3±12.5 vs. 36.8±11.8, P < 0.01); 72 hours after treatment, the oxygenation index (PaO2/FiO2) in the combined treatment group was significantly increased [mmHg (1 mmHg = 0.133 kPa): 278±28 vs. 238±39, P < 0.01], and PEEP, FiO2, airway resistance (Raw) were significantly lowered [PEEP (cmH2O): 5±2 vs. 7±3, FiO2: 0.40±0.11 vs. 0.49±0.12, Raw (cmH2O): 12.8±0.7 vs. 16.2±0.8, all P < 0.01]. There was no significant difference in static lung compliance (Cst) or Pplat between the two groups. Compared with the simple ventilation group, the duration of MV (days: 5.5±3.0 vs. 8.6±2.1), the length of ICU stay (days: 7.5±5.4 vs. 11.3±4.6) and the extraction time of artificial airway (days: 6.6±2.8 vs. 9.4±3.3) in the combined treatment group were significantly shortened (all P < 0.01). However, there was no significant difference in the 28-day mortality rate [4.1% (4/98) vs. 3.1% (3/98)], the rate of tracheotomy [2.0% (2/98) vs. 3.1% (3/98)] or the incidence ofⅠ-Ⅱ skin pressure sore [1.0% (1/98) vs. 2.0% (2/98)] between simple ventilation group and the combined treatment group (all P > 0.05). No artificial airway disappearance, pneumothorax, grade Ⅲ or Ⅳ pressure sore was found in the two groups. Conclusions Compared with lateral position ventilation alone, the treatment of combined with vibration sputum drainage is more effective for improving the respiratory function of patients with ARDS, reducing infection action, shortening the duration of MV and the length of ICU stay, and improve the recovery, promote the rehabitation of patients.

10.Antiviral effects of Bovine antimicrobial peptide against TGEV in vivo and in vitro

Background:
In suckling piglets, transmissible gastroenteritis virus (TGEV) causes lethal diarrhea accompanied by high infection and mortality rates, leading to considerable economic losses. This study explored methods of preventing or inhibiting their production.Bovine antimicrobial peptide-13 (APB-13) has antibacterial, antiviral, and immune functions.
Objectives:
This study analyzed the efficacy of APB-13 against TGEV through in vivo and in vitro experiments.
Methods:
The effects of APB-13 toxicity and virus inhibition rate on swine testicular (ST) cells were detected using 3-(4,5-dimethylthiazol-2-yl)-2, 5-diphenyl tetrazolium bromide (MTT). The impact of APB-13 on virus replication was examined through the 50% tissue culture infective dose (TCID50 ). The mRNA and protein levels were investigated by real-time quantitative polymerase chain reaction and western blot (WB). Tissue sections were used to detect intestinal morphological development.
Results:
The safe and effective concentration range of APB-13 on ST cells ranged from 0 to 62.5 µg/mL, and the highest viral inhibitory rate of APB-13 was 74.1%. The log10 TCID50 of 62.5 µg/mL APB-13 was 3.63 lower than that of the virus control. The mRNA and protein expression at 62.5 µg/mL APB-13 was significantly lower than that of the virus control at 24 hpi. Piglets in the APB-13 group showed significantly lower viral shedding than that in the virus control group, and the pathological tissue sections of the jejunum morphology revealed significant differences between the groups.
Conclusions
APB-13 exhibited good antiviral effects on TGEV invivo and in vitro.
