Journal of Applied Clinical Pediatrics 2006;0(16):-
Asthma is one kind of chronic respiratory tract inflammatory disease.Recently,it has been discovered that the small airway also participates in asthmatic pathogenesis. The small airway which diameter is less than 2 mm is one of the smallest regions in the lung. Its function may be affected by the small airway disease and related with the airway hyperresponsiveness. Some structural changes in the small airway is the latent reason which aggravates asthma. The change of small airway function in the mild asthma can cause the change of airway hyperresponsiveness. So it is considered that the small airway function is one of the methods to monitor the asthmatic situation.

Objective To observe the electrophysiological characteristics of the inward current of cardiomyocyte-like cells derived from rat bone marrow mesenchymal stem cells (MSCs). Methods Bone marrow specimens was extracted from SD rats aged 1 month. MSCs were isolated by gradient centrifugation and obtained by adherence culture. The second-passage MSCs were treated with 10 ?mol/L 5-azacytidine and incubated for 24 h. After 4 weeks, cardiomyocyte-like cells were identified by immunocytochemical staining technique, then used to detect the inward current with the whole-cell patch-clamp technique, which results were compared with the normal cardiomyocytes and the MSCs without induction. Results Some cardiomyocyte-like cells were stained positive for troponin T, and the quickly activated and inactivated inward currents were elicited 4 weeks after 5-aza cytidine treatment. The inward currents of cardiomyocyte-like cells recorded in the same square pulse were weaker than that of the normal cardiomyocytes, and the I-V curve of cardiomyocyte-like cells was right-shifted. Conclusion After treated with 5-azacytidine, MSCs differentiate into cardiomyocyte-like cells which express the excitable inward current.

Objective To detect the expression of SSX and to correlate it with clinical indicators of primary hepatocellular carcinoma (HCC).Methods The expression of SSX1-5 mRNA and SSX1 protein were respectively detected by RT-PCR and Western blot and immunohistochemistry staining. The relation between the expression of SSX mRNA and SSX1 protein with clinical indicators were analysed. Results SSX1,SSX2,and SSX3 mRNA were expressed in hepatocellular carcinoma cell lines BEL-7404,Hep G2,and SMMC-7721.In 26 HCC samples,SSX1-SSX5 mRNA was detectable in 53.8%,42.3%,50.0%,46.2% and 26.9%.The expression of SSX1 mRNA was not related to serum AFP levels (P >0.05).Specific expression was both found in the normal group and the high value group. The expression rate of SSX1 mRNA was 85.7% in the older group, which was higher than in the younger group (16.7%,P < 0.05).The expression rate of SSX1 protein was 50% in HCC tissues, which was not seen in the caner-adjacent or cirrhosis tissues. In 49 HCC paraffin tissue section samples, the expression rate of SSX1 protein was higher than that in caner-adjacent tissues (46.9% vs 18.4%,P < 0.05).The expression rate of SSX1 protein was 68.3% in the large hepatocellular carcinoma group, which was higher than in the small hepatocellular carcinoma group (29.6%),(P < 0.05).Conclusions SSX1 mRNA is expressed with a high percentage and specificity in HCC and their products are new potential promising targets for antigen-specific immunotherapy of HCC.The detection of SSX1 expression has the potential value for auxiliary diagnosis of HCC.

Objective To explore the repair method for refractory diabetic wound. Methods A total of 206 patients with refractory diabetic foot ulcers were treated with proper surgical treatments. Results Of all, 106 patients were treated by skin flap (51.5 % ), with one stage wound healing rate of 85.8%; 122 patients were repaired with split-thickness skin graft ( 59.2% ), with survival rate of the graft for 79.5%. Simple toe amputation was made in 34 patients (46 toes). The high level amputation was performed in 56 patients (27.2%). Of all, 132 patients were followed up for 6-18 months, which showed that ulcer recurred in 12 patients (9.1%). Conclusion Timely and effective treatment as well as flap and skin graft repair could reduce high level amputation rate of diabetic foot ulcer and promote the quality of life.

Objective To explore the therapeutic effects of 660 nm red light on sciatic nerve injury in adult rats. Methods Forty-five adult, male rats were divided into a control group and treatment groups 1,2,3 and 4.Sciatic nerve injury was modeled by crushing the nerve. The treatment groups received irradiation with red light once daily for 21 consecutive days. The power density of red light and irradiation time varied among the groups. The latency and amplitude of compound muscle action potentials (CMAPs) and nerve conductive velocity were examined at different time points. The Sciatic Function Index (SFI) was used to evaluate walking function. Results After 21 days of red light therapy no statistically significant differences were observed between the control group and treatment groups 1 to 4 with regard to the latency or the amplitude of the CMAPs.There was a significant difference between the control group and treatment group 3 in terms of sciatic nerve conduction velocity. The average Sciatic Function Indexes of treatment groups 2,3 were significantly different from that of the control group. Conclusion Red light irradiation can promote recovery after sciatic nerve injury,at least in rats, thereby improving walking function.

The means of OD value measurement and plate counting were used to choose the bacterial growth stimulants. Among the 120 substances sorted to 13 kinds (the trace elements, REE, carbohydrate, amino acids and amino acids derivatives, vitamins, nucleosides, plant hormones, animal hormones, plant and animal extracts, Echlonia Kurome Okum water extract and yeast extract), Echlonia Kurome Okum water extract and yeast extract were found to stimulate the growth of Escherichia coli significantly.

Many researchers employed mammalian expression system to artificially express cannabinoid receptors, but immunoblot data that directly prove efficient protein expression can hardly be seen in related research reports. In present study, we demonstrated cannabinoid receptor protein was not able to be properly expressed with routine mammalian expression system. This inefficient expression was rescued by endowing an exogenous signal peptide ahead of cannabinoid receptor peptide. In addition, the artificially synthesized cannabinoid receptor was found to aggregate under routine sample denaturing temperatures (i.e., [Symbol: see text]95°C), forming a large molecular weight band when analyzed by immuno-blotting. Only denaturing temperatures [Symbol: see text]75°C yielded a clear band at the predicted molecular weight. Collectively, we showed that efficient mammalian expression of cannabinoid receptors need a signal peptide sequence, and described the requirement for a low sample denaturing temperature in immuno-blot analysis. These findings provide very useful information for efficient mammalian expression and immuno-blotting of membrane receptors.
