1.Expression of metastasis associated antigen 1 in endometrial carcinoma and its clinical significance

Objective To investigate the relationship between the expression of metastasis associated antigen 1 (MTA1) and the biological behavior of endometrial carcinoma. Methods From May 2005 to May 2007, 30 patients with endometrial carcinoma and 20 patients with normal endometrial tissues, were admitted to Changzheng Hospital of the Second Military Medical University for surgical treatment were involved in the present study. The expression of MAT1 mRNA in two different groups was detected by real-time RT-PCR, and of MAT1 protein was detected by immunohistochemical streptavidin-peroxidase (SP) method and Western blotting. The relationship between their expression and pathological features were analyzed. Results The positive expression rate of MTA1 protein was 80% in endometrial carcinoma group, which was significantly higher than that in control group (30%, P0.05). Conclusion The expression of MTA1 might play a potential role in the pathogenesis and development of endometrial carcinoma, and it might be a novel target for the therapy and prognostic indicator of endometrial carcinoma.
2.Exploring the Anatomical Relationship between the Umbilicus and Major Retroperitoneal Vessels during Laparoscopy

Objective To study the anatomical relationship between the umbilicus and the retroperitoneal major vessels. Methods A total of 80 female patients undergoing laparoscopy in our department were enrolled in this study. The patients were divided into 4 groups according to their body mass index (BMI):low-weight group,normal-weight group,overweight group,and obese group. During the operation,the surgeons palpated the abdominal aorta,right common iliac artery,and left common iliac artery with hand to determine their relationship to the umbilicus. The abdominal aortic bifurcation was used as a reference point to determine its position in relation to the umbilicus. Results (1) The bifurcation was above the level of the umbilicus in 17 (21.2%) cases,at the umbilicus level in 29 (36.3%) cases,and below that in 34 (42.5%) cases. Among the four groups,the position of the bifurcation was significantly different (?2=16.769,P=0.010). In the overweight and obese groups,most of the patients had the umbilicus located under the level of the aortic bifurcation [65.0% (13/20) and 75.0% (6/8) respectively],while in the normal-weight group,47.6% (20/42) of the cases had the umbilicus above the aortic bifurcation(P

4. Real-time fluorescent quantitative PCR detection of key enzyme genes expression of alkaloid biosynthesis promoted by endophytic fungal elicitor in Sophora alopecuroides

Objective To establish a method of detecting the expression of Lysine decarboxylase (LDC) -a key enzyme for the synthesis of alkaloid in the host promoted by the endophytic fungal elicitor of Sophora alopecuroides by using real-time fluorescence quantitative PCR (qRT-PCR). Methods Target gene primers QLDC-F/QLDC-R and reference gene primers Lectin-F/Lectin-R were designed according to LDC and Lectin gene sequences of S. alopecuroids; Five-fold gradient dilution of cDNA was used as the standard sample for the construction of the standard curve of target gene and the reference gene. Reaction system and reaction conditions of qRT-PCR were optimized, and the sensitivity of semi-quantitative PCR and qRT-PCR were analyzed and compared. Under different eliciting time of endophytic fungal elicitors NDZKDF13 of S. alopecuroides, the content of oxymatrine in the host was determined by HPLC, the expression of LDC gene was detected by qRT-PCR, and the relationship between LDC gene expression and the accumulation of OMA was analyzed. Results The results of qRT-PCR were better when the cDNA content in the system was 200 ng/μL and the annealing temperature was 61 ℃. The standard curve of the target gene and the reference gene was constructed, in which the cycle threshold and template concentration showed a good linear relationship, the amplification efficiency was above 99%, and the sensitivity was 25 times that of semi-quantitative PCR. Under the induction effect of endophytic fungal elicitor NDZKDF13, expression of host LDC gene reached the peak on the 6th day, which was 25.58 times that of the control. The increase of OMA content lagged the change of the LDC gene expression and reached the highest amount on the 9th day after the induction. Conclusion The qRT-PCR technique was successfully applied to the functional gene research of S. alopecuroides. Through the optimization of various conditions, a platform for accurate and simple detection of functional gene expression in S. alopecuroides was established.
5.Recent advances in the application of nanobody technology: a review.

As a malleable and novel tool for antigen recognition and modulation, nanobodies have the advantages of small size, easiness of expression, screening and modification, as well as high affinity and stability. Nanobodies are capable of recognizing more cryptic antigenic epitopes that are difficult to be recognized by traditional antibodies, making them increasingly used in the diagnosis and treatment of various diseases and assays. Nanobodies are also playing an irreplaceable role in the basic research. This review summarized the recent development of nanobodies and their derivatives in the detection of small molecules, pathogenic microorganisms and diagnosis of diseases, as well as in the fields of targeted therapies, cellular and molecular imaging. Broad prospects of nanobodies in the field of protein conformation studies were also reviewed.

6.Relationship between fetal distress and endogenous opioid peptides levels of the venous blood and the umbilical blood in pregnant women

BACKGROUND: Endogenous opioid peptide is an important medium and regulator that participate in many physical and pathologic processes of the body. Its relationship with fetal encephalopathy has attracted much attraction.OBJECTIVE: To evaluate the role of endogenous opioid peptides (EOP) in fetal distress.DESIGN: A case-control observatory study based on healthy pregnant women.SETTING: Wards of the Department of Obstetrics and Gynecology of Changzheng Hospital Affiliated to Second Military Medical University of Chinese PLA.PARTICIPANTS: Eighty-three healthy women who were hospitalised in Changzheng Hospital and Changhai Hospital affiliated to the Second Military Medical University of Chinese PLA and met the inclusion criteria. Among them, 40 were normal healthy pregnant women(the control group) and 43 were healthy pregnant women with fetal distress(the fetal distress group).METHODS: Radioimmunoassay was used to measure the levels of blood EOP(β-endorphin, dynorphin A1- 13 and leu-enkephalin) of the venous blood of the pregnant women in fetal distress group and the control group and the EOP level in the umbilical blood of the newborns. Also, blood gas analysis of the blood from the umbilical artery was conducted.MAIN OUTCOME MEASURES: The levels of EOP in the venous blood of two groups of pregnant women and the umbilical blood of newborns and the correlation of EOP level with fetal distress.RESULTS: The levels of the umbilical artery blood EOP(β-endorphin,dynorphin A1-13 and leu-enkephalin) in the fetal distress group[(453± 68 ) ng/L, (242 ± 33)ng/L, and(498 ± 68)ng/L respectively] were significantly higher than those in the control group[ (251 ± 39) ng/L, (103± 22 )ng / L and(322 ± 40 )ng / L respectively ( t = 2. 713,2. 762, P< 0.01; t = 2. 132, P < 0.05 ) ]. The umbilical artery blood gas analysis;pH was (7.0 ± 0. 1 ) , PO2 was ( 1.7 ± 0.6) kPa, PCO2 was (8.9 ± 0. 7) kPa.The levels of β-endorphin were negatively correlated with pH and PO2 of the umbilical artery blood(r= -0.418 and -0.437, P < 0.01), but they were positively correlated with PCO2( r = 0. 442, P < 0. 05) . The level of dynorphin A1-13 was negatively correlated with pH and PO2( r = -0. 337,-0.383, P < 0.05), but it was positively correlated with PCO2(r= 0. 346, P < 0. 05). There was no significant difference among the three kinds of blood EOP of the two groups( P > 0.05).CONCLUSION: EOP participates in the pathological progress of the fetal distress and was closely correlated with the occurrence and development of the fetal distress. This finding has a reference value for early rehabilitation and intervention after the fetal was born that can be tested quantitatively.
7.Efficacy of epithelial-off accelerated corneal cross-linking in the treatment of advanced keratoconus

AIM: To investigate the efficacy of epithelial-off accelerated corneal cross-linking(CXL)in the treatment of advanced keratoconus.METHODS: A retrospective study was performed on data collected from 32 patients(43 eyes)with advanced keratoconus who underwent epithelial-off accelerated CXL at Ningxia Eye Hospital from April 2020 to December 2021. Slit-lamp, intraocular pressure, uncorrected visual acuity(UCVA), corrected visual acuity, specular microscope, Pentacam and Corvis ST were tested before and at 1, 3 and 6mo after surgery. Preoperative and postoperative corneal condition, UCVA, best corrected visual acuity(BCVA)and the values of corneal endothelial, maximum keratometry(Kmax), thinnest corneal thickness(TCT), anterior and posterior surfaces of the cornea K1, K2, biomechanically corrected intraocular pressure(bIOP), applanation time 1(A1T), applanation length 1(A1L), applanation velocity 1(A1V), applanation time 2(A2T), applanation length 2(A2L), applanation velocity 2(A2V), highest concavity deformation amplitude(HCDA), radius at highest curvature(HCR), highest concavity peak distance(HCPD)and stiffness parameter at first applanation(SP-A1)were recorded.RESULTS: There were differences between UCVA(LogMAR; 1.06±0.49, 0.78±0.39)and BCVA(LogMAR; 0.48±0.34, 0.38±0.29)before and at 6mo after surgery(P<0.05), but there were no differences in corneal endothelial cells(2917.39±288.38 vs. 2959.19±336.27 cells/mm2, P=0.477). There were differences among Kmax, TCT, anterior surface K1 and K2 and posterior surface K1 before and after surgery(P<0.05), and all increased at 1mo after surgery then returned to preoperative level at 3mo after surgery, while there was no difference in the posterior K2. Furthermore, there were statistical significance in A1T, HCPD and SP-A1 before and after surgery(P<0.05), while there were no statistical significance in A1L, A1V,A2T, A2L, A2V, HCDA, HCR and bIOP(P>0.05).CONCLUSION: Epithelial-off accelerated CXL can prevent the progression of keratoconus within half year after surgery, and it has certain safety.
8. The transcriptional study of tetramethylpyrazine against cerebral ischemia/reperfusion injury in diabetic rats

Aim To investigate the protective effect of tetramethylpyrazine ( TMP ) on cerebral ischemia/reper¬fusion ( I/R) injury in diabetes mellitus ( DM ) rats based on full-length transcriptome sequencing tech¬nique and the underlying mechanism. Methods The rat model of DM was induced by intraperitoneal injec- tion of streptozotocin (STZ,50 mg • kg" ) . The global cerebral I/R model of rats was established by double vessel occlusion (2-VO) for 10 min and reperfusion for 16 h. TMP (40 mg • kg ) was injected intraperitone-ally every day. Brain water content detection, HE stai¬ning, Nissl staining and TUNEL staining were used to evaluate the neuroprotective effect of TMP on cerebral I/R injury in DM rats. The differentially expressed genes were screened by full-length transcriptome se¬quencing. The possible candidate genes and metabolic pathways of TMP for cerebral ischemia protection in DM rats were screened by functional annotation and metabolic pathway enrichment analysis, and verified by RT-qPCR and Western blot. Results TMP treatment could reduce the degree of brain edema, increase the number of Nissl bodies and neurons, and significantly reduce cell necrosis and apoptosis after cerebral I/R in DM rats. GO enrichment analysis showed that the dif¬ferentially expressed genes after TMP treatment were mainly enriched in biological functions such as inflam¬matory response, transcription factor complex, growth factor activity and so on. KEGG pathway analysis showed that it was mainly concentrated in apoptosis, Toll-like receptor and other signaling pathways. Among them TMP down-regulated genes included TLR2, MyD88, IL-lp, Caspase-3, etc. The results of RT-qPCR and Western blot confirmed that TMP could down-regulate TLR2, MyD88, NF-KB, IL-10, IL-6 and Caspase-3 , and up-regulate the mRNA and protein expression of Bcl-2. Conclusions TMP may amelio¬rate neuronal apoptosis by inhibiting inflammatory re¬sponse, which improves cerebral I/R injury in DM rats, and the pathway may be TLR2/MyD88/NF-KB signaling pathway.
9.Preventing laryngeal anterior commissure adhesion after operation: a study in canines.

OBJECTIVE:
To explore the better operational methods by the laser surgery to treat diseases involving the laryngeal anterior commissure. It can excise the diseases as well as avoid anterior commissure adhesion, laryngostenosis and dyspnea after operations.
METHOD:
Twelve dogs were divided into 4 groups at random. There were three dogs in one group. A: excising experimental dog's anterior commissure by twice operations, the interval time was two weeks; B: excising experimental dog's anterior commissure in one time, at the same time suturing the silica gel sheet on the anterior commissure; C: excising experimental dog's anterior commissure in one time and then applying MMC on the wound of the anterior commissure; D: excising experimental dog's anterior commissure in one time, without any treatment.
RESULT:
All of the dog's surgery were completed successfully by laser. Four weeks later, we observed the raw surfaces. A: the neonatal membrane covered the wound, inflammatory reaction slight, we could not see obvious adhesion in the anterior commissure. B: the membrane covered the wound, appearing the dark chronic inflammation, we could see the adhesion in the anterior commissure slight. C: the membrane covered the wound, edematization, we could see the moderate adhesion in the anterior commissure. D: edematization, we could see the adhesion in the anterior commissure obviously. Four groups were all appeared hoarsenesses, the most slightly in group A, secondly in B and C, the worst in D. The analysis of vocal cord length of 4 groups, we used matched-pairs t-test, A, B, C groups' P>0.05, the vocal cord length didn't become shorter than before obviously. Group D's P<0.01, that meant the vocal cord length became shorter obviously. Between each group,we used reiterature-measurement analysis of variance (P<0.05), the change of vocal cord length had disparity in different groups. The comparison in two groups suggested that group A is the best. B and C are inferior ,but no disparity between them. The analysis of glottis area of 4 groups, as the same method above, A,B groups' P>0.05, the glottis area didn't shrink than before obviously. C and D groups' P<0.05, that meant having statistical significance and glottis area shrinked obviously. Between each group, we used reiterature-measurement analysis of variance (P>0.05), we could not think that the changes of four groups have disparity.
CONCLUSION
Excising the experimental dog's anterior commissure by laser, compared the 4 different operation methods, group A is the best method. B and C are inferior. The results are valuable in clinic when we perform operations by laser to treat the disease involving the anterior commissure.

10.Intra-nucleus accumbens shell injection of baclofen blocks the reconsolidation of conditioned place preference in morphine-addicted mice.

Preclinical studies suggest that the GABA receptor is a potential target for treatment of substance use disorders. Baclofen (BLF), a prototypical GABA receptor agonist, is the only specific GABA receptor agonist available for application in clinical addiction treatment. The nucleus accumbens shell (AcbSh) is a key node in the circuit that controls reward-directed behavior. However, the relationship between GABA receptors in the AcbSh and memory reconsolidation was unclear. The aim of this study was to investigate the effect of intra-AcbSh injection of BLF on the reconsolidation of morphine reward memory. Male C57BL/6J mice were used to establish morphine conditioned place preference (CPP) model and carry out morphine reward memory retrieval and activation experiment. The effects of intra-AcbSh injection of BLF on morphine-induced CPP, reinstatement of CPP and locomotor activity were observed after environmental cues activating morphine reward memory. The results showed that intra-AcbSh injection of BLF (0.06 nmol/0.2 μL/side or 0.12 nmol/0.2 μL/side), rather than vehicle or BLF (0.01 nmol/0.2 μL/side), following morphine reward memory retrieval abolished morphine-induced CPP by disrupting its reconsolidation in mice. Moreover, this effect persisted for more than 14 days, which was not reversed by a morphine priming injection. Furthermore, intra-AcbSh injection of BLF without morphine reward memory retrieval had no effect on morphine-associated reward memory. Interestingly, administration of BLF into the AcbSh had no effect on the locomotor activity of mice during testing phase. Based on these results, we concluded that intra-AcbSh injection of BLF following morphine reward memory could erase morphine-induced CPP by disrupting its reconsolidation. Activating GABA receptor in AcbSh during drug memory reconsolidation may be a potential approach to prevent drug relapse.
