International Journal of Cerebrovascular Diseases 2006;0(01):-
Bone marrow mesenchymal stem cells(BMSCs) are a population of multipotent cells. Many researchers have studied whether BMSCs could differentiate into neurons in vivo and ex vivo. Although some studies didn't agree with about some chemical induction medium inducing neurons, the application of cell transplantion and gene therapy of BMSCs has achieved certain progress showing good future for BMSCs in therapy of nervous system diseases.

OBJECTIVE:To investigate advantages and disadvantages of preexisting metal post, casting metal post, ceramic post, and fiber post for clinical application, and to evaluate mechanical function, esthetical performance, and biocompatibility of fiber post.METHODS:Articles were retrieved with the key words of "fiber post,post-core restoration, dental materials, cement material, binder" in both English and Chinese between January 1990 and October 2008.Inclusion criteria:Articles addressing clinical application of fiber post for repairing residual root and crown were included.Exclusion criteria:Duplicated studies or Meta analysis was excluded.A total of 24 articles were included to investigate mechanical function, biocompatibility, and stickiness and retention of fiber post.RESULTS:Traditionally, metal post was easily folded and corrosive; in addition, esthetical performance and biocompatibility were still dissolved.Intension, hardness, esthetical performance, corrosion proof, fatigue resistance, and biocompatibility of fiber post were superior to those of traditional metal post.Furthermore, fiber post was able to highly adhere to root canal dentin via resin splicing agent, which could prolong useful life of dental prosthetic restoration, reduce root folded onset, and benefit for dental storage and repairing.Light mass of fiber post was another advantage compared with metal post.If the repair was failure, fiber post was easily taken out and reused. The combination of fiber post and resin nucleus was beneficial for dental prosthetic restoration which was highly closed to original structure of natural teeth.CONCLUSION:Fiber post was characterized by suitable elastic modulus, great esthetical repairing effect, reliable stickiness, easy identification and removal, high achievement ratio of repairing, and convenient operation; therefore, fiber post was a new and ideal material for clinical application.

BACKGROUND:Previous in vitro studies mainly focused on morphological and nerve marker aspects in the study of bone marrow mesenchymal stem cells (BMSCs)-derived neuron-like cells, but less focused on the olectrophysiological properties of neuron-like cells following differentiation.OBJECTIVE:To study the electrophysiological changes of differentiation from BMSCs into neuron-like cells after induction by brain-derived neurotrophic factor (BDNF)/basic fibroblast growth factor (bFGF)/alltrans-retinolc acid (AT-RA).DESIGN,TIME AND SETTING:The cytological comparative in vitro study was performed at the Department of Neural Cell Laboratory,Tianjin Huanhu Hospital and College of life Science of Nankai University from June 2005 to October 2007.MATERIALS:Totally 3 male Wistar rats (6-week old, weighing about 160 g) were used in this study.METHODS:BMSCs were cultured and purified by their characteristic of plastic adhesion in vitro, and then induced by BDNF,bFGF and AT-RA,and differentiate into neuron-like cells.Whole-cell patch damp technique was used to detect cell membrane current prior to and 3 days following induction.MAIN OUTCOME MEASURES:MSC phenotype was determined by flow cytometry.Cell morphology was observed under the inverted microscope before and after differentiation.Neuron specific enolase expression was assessed by immunocytochemistry.Whole-cell current results were measured.RESULTS:Flow cytometry results demonstrated that CD90 positive rate (99±3)%,CD31 (3.4±0.8)%,and CD34 (0.3±0.1)%.This indicated that these cells were undifferentiated stem cells, with purity of 95%.Undifferentiated MSCs under the optical microscope were mostly fiat cells with processes, similar to fibroblast-like cells.Neuron-like cells appeared 3 days following induction.Immunocytochemistry results showed that MSCs before induction were weakly positive for neuron specific enolase, but strongly positive for neuron specific enolase. At 72 hours, the differentiated rate was (24.01±3.76)%.The peak currents of outward currents in neuron-like cells were significantly higher in the induction group compared with the control group (P < 0.05),but no inward Na~+ current was detected.CONCLUSION:(bFGF & BDNF)/AT-RA could induce the differentiation of MSCs into neuron-like cells. These cells showed the tendency to differentiate into mature neurons, though having no electrophysiological properties of mature neurons.

Objective To detect the effect of osthole on proliferation and apoptosis of HL-60 cells and its molecular mechanism. Methods HL-60 cells proliferation was measured through the CCK8 assay method. The cell morphological changes were observed by Hoechst33342 staining after 8 h of drug effect.Induction of apoptosis was determined by flow cytometry and fluorescent microscopy.Expressions of Bcl-2 and Bax mRNA were evaluated by RT-PCR,and the expressions of cleaved caspase-3,caspase-8,caspase-9,Fas and FasL were evaluated by using western bolt assay. Results Osthole could inhibit the proliferation of HL-60 cells, the maximum inhibiting rate was (90.7 ±4.5)%,F =138.46,P =0.000; the apoptosis rate was 33.6%,F =27.75,P =0.006.The changes of apoptosis of cells and nucleus were shown in cell morphological observation.Osthole affected the decrease of the mRNA levels of Bcl-2 and the increase of the Bax mRNA levels via a dosedependent manner(F =210.12,P =0.000).Western blotting demonstrated that osthole could lead to the increase of the expression levels of cleaved caspase-3,caspase-8,caspase-9,Fas and FasL in the HL-60 cell line via a time-dependent manner. Conclusion Data suggests that osthole inhibits proliferation and induces apoptosis of HL-60 cells through mitochondria-dependent pathway and death-receptor pathway.
