Objective:Toinvestigate the expression of RHAMMgene and TPX2 gene in esophageal squamous cell carcinoma(ESCC)and their relationships with clinicopathological factors.Methods:The expression of RHAMM gene and TPX2 gene were detected by reverse transcriptase polymerase chain reaction(RT-PCR)in ESCC tissues,para-cancer tissues,and matched esophageal normal mucosa tissues of 40 patients.Results:The positive expressions of RHAMM mRNA in ESCC tissues(65.0%)were significantly higher than those in para-cancer tissues(37.5%)and esophageal normal mucosa tissues(22.5%).Significant relationship was observed between positive expression of RHAMM mRNA and lymphnode metastasis(P
2.Inhibitory effects of human cloned lymphokine-activated killer (LAK) cells on growth of myeloid progenitor cells

Human LAK cells induced from peripheral blood mononuclear cells with recombinant interleukin2 (rIL-2) incubation, can be cloned in semisolid medium and proliferated well in liquid cultures containing rIL-2. Phynotypie analysis by flow cytometry showed that these cloned LAK cells were T_3(+), T_4(-), T_8(+), Tac(+), DR(+) and NKH1(-)/(+). Cultured with 1?10~5/ml normal bone marrow mononuelear cells (BMMNC)for 0.5-hr prior to semisolid culture system for CFU-GM, the cloned LAK ceils at 2?10~5/ml showed inhibitory effect on CFU-GM that the colonies were decreased by 43.8% of control though 0.5 or 1?10~5/ml LAK cells did not. Morever, a dose-dependent inhibition of CFU-GM was noted when LAK cells pre-inbated with BMMNC for 4-hr in doses of 0.5 -4?10~5/ml that the inhibitory rates of CFU-GM were 58.8, 67.5, 88.8 and 96.3% at0.5, 1, 2, 4?10~5/ml LAK cells addition respectively. Using double-layer agar cultures, inhibitory activity for CFU-GM was also observed only when LAK cells at 4?10~5/ml without contacted with BMMNC. No significant difference of inhibitory effects on CFUGM growth was found between autologous and allogenie clotted LAK cells under same condition. These data suggeste that LAK cells inhibited growth of myeloid progenitor cells without MHC-restriction and the mechanism might involve cell-to-cell interaction and/or some soluble factors secreted by LAK cells.

Peutz-Jeghers syndrome ( PJS ) is a rare autosomal dominant inherited disease characterized by mucocutaneous pigmentation and multiple gastrointestinal polyposis. Recent studies suggested that mutation in STK11 gene on chromosome 19p13. 3 was considered as the major cause of PJS. In this paper,we summarized the clinical features, pathogenesis,diagnosis and treatment of PJS through literature review.
4.Treatment of paclitaxel combined with oxaliplatinchemotherapy to the non-small cell lung cancer mouse model

Objective To investigate the therapeutic effect of paclitaxel plus oxaliplatin chemotherapy to the transplanted non-small cell lung cancer of nude mice and the effect to the apoptosis protein expression of PDCD5 and XIAP with mice model.Methods A tumor-bearing mice were randomly divided into blank group, normal saline group, oxaliplatin group, paclitaxel group, paclitaxel plus oxaliplatin group.The gene expression of PDCD5 and XIAP was assayed by real-time quantitative PCR(q-PCR).The apoptosis related PDCD5 and XIAP protein were detected by Western blot.Finally, the tumor weight of each group was measured for statistical analysis.ResultsThe mRNA expression of PDCD5 was highest and the gene expression of XIAP was lowest in paclitaxel plus oxaliplatin group(P<0.01).The expression of PDCD5 protein was highest and the expression of XIAP protein was lowest in paclitaxel plus oxaliplatin group (P<0.01).Finally, compare the tumor weight of each group, paclitaxel plus oxaliplatin group has the least mass(P<0.01).Conclusions Paclitaxel plus oxaliplatin group chemotherapy significantly increases PDCD5 expression and reduce XIAP expression.Meanwhile, paclitaxel plus oxaliplatin chemotherapy can significantly reduce the tumor weight of happened non-small cell lung cancer.
5.Induction of Specific CTLs and Their Tumor Suppressing Activity in BALB/c Mice by Peptide HER2/neu Immunization

Objective: To study the cellular immune responses and the anti-tumor effects induced by peptides derived from onco-protein HER2/neu. Methods: Two HER2/neu peptides compatible with H-2Kd binding motif as tumor rejection antigens were synthesized. The ability of inducing peptide-specific CTLs and suppressing the tumor growth in BALB/c mice immunized by HER2/neu peptide was analyzed. Results: HER2/neu peptides could promote the proliferation of BALB/c lymphocytes both in vitro and in vivo. HER2/neu peptide-specific CTLs could be induced from peptide-immu-nized BALB/c mice, which showed specifically killing effects on HER2/neu positive SP2/OHER2 cells but not on HER2/neu negative SP2/O cells. Moreover, the growth of SP2/OHER2 tumor were significantly suppressed in BALB/c mice immunized with HER2/neu peptides. Conclusion: These results suggested that it was feasible to use MHC-I epitopes derived from tumor-specific antigens as vaccines for cancer immunotherapy.
6.Comparative proteomics analysis of taxol resistance in human ovarian cancer

Objective To analyze the differential expression of protein in taxol-resistance from elderly females with ovarian cancer, by compared proteomics analysis and identified the candidated marker in the clinical tissues. Methods Soluble fraction proteins in two SKOV3 taxol-resistant cells, SK-TR30 and SK-TR2500 and one A2780 taxol-resistant cell, A2780-TR paired taxol-sensitive ovarian cancer cells were separated and prepared by two dimensional gel electrophoresis (2-DE). The differentially expressed proteins were selected and identified by matrix-assisted laser desorption/lonization time of flight mass spectrometry(MALDI-TOF-MS) and database search. 2-DE profiles with high resolution and reproducibility were obtained. Then highly expressed protein-cofilin1 which exist in two forms of de-phosphorylate was detected and compared between human ovarian cancer specimens including 22 elderly female with chemosensitive and 21 elderly female with chemoresistant by immunohistochemistry. Results Among the identified proteins, there were sixteen over/under-expressed proteins in taxol-resistant cell lines as compared with taxol-sensitive cell lines. In particular, overexpression of cofilin1 and destrin were displayed in all three taxol-resistant cell lines. The expression of phosphorylation of cofilin (P<0.05), but not cofilin1, displayed much higher in chemoresistant than in chemosensitive group. Conclusions Cofilin1 may play a role of taxol resistance by phosphorylation of ovarian cancer cell. However, the mechanisms need to be elucidated further.
7.Treatment of spinal fracture occurring in the wenchuan earthquake

[Objective]To investigate the application of thoracolumbar injury classification and severity thoracolumbar injuty chassification and severityscdre(TLICS)score in spinal fracture caused by earthquake.[Method]Night-five patients with spinal fractures were treated in Chendu Army General Hospital and Chendu Air Force Hosptial by Wenchuan earthquake medical team from Chinese PLA General Hospital.There were 49 males and 46 females with mean age of 48.6 years.The general patient data,injury mechanism,and injury time were recorded.The different treatments were decided by the TLICS score.[Result]There were 107 vertebral fractures in the 95 patients,out of whom 76 patients were injured by building collapsed,and the other 19 by falling from buildings.TLICS score was more than 4 in 36 patients,4 points in 15 patients,less than 4 points in 44 patients.40 patients were treated with open operation,15 patients with vertebraplasty,and the other patients with conservative treatment.[Conclusion]The earthquake spinal fractures can be treated with different treatments based on the different TLICS scores.
8.Investigation on current status of advanced schistosomiasis in Shanggao County

Objective To understand the current status of advanced schistosomiasis patients, so as to provide the reference for improving the medical care of them in Shanggao County. Methods The data of advanced patients in Shanggao County were collected from the Information Management System for the Advanced Schistosomiasis Chemotherapy and Assistance in Jiangxi Province and a field survey including the epidemiological investigation physical examinations and B ultrasound examinations was performed in 2014. All the data were analyzed statistically and compared with those in 2010. Results Among the 128 ad?vanced schistosomiasis patients the male female ratio was 2.46 1 the average age was 68.02±6.19 years and the patients old?er than 65 years contributed to 64.84%for the all. The proportions of clinically types of ascites splenomegaly multiple granulo?ma in the colon dwarf general and haemorrhage were 57.81% 12.50% 2.34% 0 26.57% and 0.78% respectively. The ultrasound examinations revealed the average diameter of the main trunk of portal vein was 12.70 ± 1.91mm in 2014 which was significantly broader than that of 12.05 ± 0.34mm in 2010 P < 0.01 . Conclusions The advanced schistosomiasis patents in Shanggao County decreases with years and shows an aging trend. Some advanced schistosomiasis patients are worse therefore the chemotherapy and salvation work still should be strengthened.
10.Effect of salidroside on brain edema and neurological function in global ischemia-reperfusion injuly in rats

Objective To investigate the effect of salidroside on brain edema and neurological function in global cerebral ischemia-reperfusion injury in rats.Methods A total of 100 Sprague Dawley rats were randomly divided into sham operation,ischemia-reperfusion and salidroside 12,24 and 48 mg/kg groups (n =20 in each group),and than redivided into 6 h,24 h,72 h and 7 dsubgroups (n =5 in each subgroup).A rat model of global cerebral ischemia was established using the four-vessel occlusion method.Immediately after modeling,all groups were administered intragastrically for 7 days.The brain water content was quantitated by the wet-dry weight method.The neurological evaluation was performed using a neurological deficit score (NDS).Results After modeling both the ischemia-reperfusion group and all the salidroside groups had significant neurological deficit,and as time went by,it was improved gradually.Compared to the ischemia-reperfusion group at the corresponding time points,neurological deficit in all the salidroside groups was improved significantly (all P < 0.05),and showing a dose-dependent trend.Compared to the salidroside 12 mg/kg and 24 mg/kg groups,neurological deficit in the salidroside 48 mg/kg group was improved significantly at 72 hours and 7 days (all P < 0.05).The brain water contents began to increase at 6 hours after modeling in the the ischemia-reperfusion group and all the salidroside group.They reached the peak at 72 hours,and significantly higher than that in the sham operation group (all P < 0.05).The brain water contents in all the salidroside group were significantly lower than those in the ischemiareperfusion group at 24 and 72 hours after modeling (all P < 0.05) and showing a dosedependent trend.The brain water content in the salidroside 48 mg/kg group was close to that in the sham operation group at 7 days after modeling.Conclusions Salidroside may significantlydecrease brain edema and improve neurological function in global cerebral ischemia-reperfusion injury in rats,and it has a neuroprotective effect.
