1.Prevalence of Resistance to Macrolide,Lincosamide and Streptogramin B Antibiotics in Staphylococcus epidermidis Caused Nosocomial Infection

OBJECTIVE To determine the prevalence of antibiotic resistance to macrolide,lincosamide and streptograminB(MLS_B) in Staphylococcus epidermidis caused nosocomial infections and give guide for clinical antibiotics usage. METHODS Totally 126 S.epidermidis isolates were collected to test the minimal inhibitory concentrations(MIC) of erythromycin,clindamycin,clarithromycin,and azithromycin,quinupristin/dalfopristin and cefoxitin.Inducible type of MLS_B resistance(iMLS_B) and MS phenotypes were distinguished by double disk diffusion method(D test) as detailed by the NCCLS. RESULTS Among 126 S.epidermidis isolates,90(71.4%) isolates were meticillin resistant(MRSE),36(28.6%) were meticillin susceptive(MSSE).The rates of antibiotic-resistance to erythromycin,clindamycin,clarithromycin,axithromycin were 92.8%,73.8%,89.7% and 91.3%,respectively.Percentages of strains resistant to erythromycin and clindamycin were 96.7% and 85.6% in MRSE,and 83.3% and 80.6% in MSSE.Constitutive phenotype,inducible phenotype and MS phenotype were detected in 93(73.8%),13(10.3%),and 11(8.7%) isolates,respectively.In constitutive phenotype, the MRSE proportion appeared high(78.5%),in contrast,in inducible phenotype,MSSE proportion appeared high(75%).No resistance to quinupristin/dalfopristin was detected in all isolates(MIC≤1mg/L). CONCLUSIONS The prevalence of antibiotic resistance to MLS_B is high in the collected isolates.The main antibiotic phenotype is constitutive phenotype.MLS_B antibiotics should be used judiciously.

6.Clinical Experience of Professor Zhang Juan in Treating Elderly Diabetic Constipation

Objective] To explore the clinical experience of Professor Zhang Juan on Elderly Diabetic Constipation and hope to provide a new perspective for future study about Chinese medicine treatment of Diabetes Mellitus(DM).[Method] Reviewed 103 cases from Sep. 2014 till now who were older than 65,diagnosed with DM and had constipation as primary symptom. Analyzed and summarized the syndrome differentiation and treatment of the cases. [Results] According to the etiology and pathogenesis of Elderly Diabetic Constipation ,Professor Zhang deems that its pathogenesis takes place in two stages:one is spleen deficiency,the other is kidney deficiency and proposes the therapy of“treat the middle-jiao if others are not working”,“nourish the natal qi with post-natal qi”,“combine disease,syndrome and symptom”and“treat in stages”.[Conclusion] The treatment on Elderly Diabetic Constipation raised by Professor Zhang is effective and her clinical experience has important clinical value .
7.The mechanism study of EGCG on improvement of cognitive function and alleviation of hippocampal insulin resistance in APP/PS 1 transgenetic mice

Objective To explore mechanism of epigallocatechin-3-gallate (EGCG) on improvement of cognitive function and alleviation of hippocampal insulin resistance in APP/PS 1 transgenetic mice. Method 12 months old female APP/PS 1 mice were randomly divided into 3 groups:model group(Tg), EGCG low dose group (Tg/EGCG-L), high dose group(Tg/EGCG-H). C 57 BL/6 J mice were utilized as control. learning and memory ability in 4 group mice were detected by morris water maze test(MWM). The hippocampal TNF-α/JNK signal and IRS-1 pSer 312 expression were detected by Western blot and immunohistochemical staining. Results Compared with NT mice, Tg mice showed a marked prolongation of the escape latency and swimming distance in the MWM test(P<0.05);Abnormal activation of TNF-α/JNK signaling and increased IRS-1 pSer 312 expression in the hippocampus of Tg mice(P<0.05). EGCG-treated Tg mice showed significantly improvement of all these abnormal changes(P<0.05). Conclusion EGCG treatment is able to alleviate hippocampal insulin resistance and improve cognitive function in the APP/PS 1 mice. which may be partly attributed to the reduction of TNF-α/JNK signaling activity in this AD mouse model.

Hepatitis C and its complications are one of the highlights in liver disease prevention and control in China,and from the clinical practice 20 years ago to a cure rate of nearly 100% at present,standardized diagnosis and treatment of hepatitis C,a large number of scientific studies,and drug research and development are the basis for the clearance of HCV.Direct-acting antiviral agents play an important role in the clearance of HCV,and the clearance of HCV does not equal to the cure of chronic hepatitis C.For patients with hepatitis C who have achieved a high sustained virologic response rate after using direct-acting antiviral agents,their long-term prognosis should be taken seriously.Correct and reasonable application of hepatitis C treatment is still a hot issue that needs to be discussed at present.

10.Induction of Specific CTLs and Their Tumor Suppressing Activity in BALB/c Mice by Peptide HER2/neu Immunization

Objective: To study the cellular immune responses and the anti-tumor effects induced by peptides derived from onco-protein HER2/neu. Methods: Two HER2/neu peptides compatible with H-2Kd binding motif as tumor rejection antigens were synthesized. The ability of inducing peptide-specific CTLs and suppressing the tumor growth in BALB/c mice immunized by HER2/neu peptide was analyzed. Results: HER2/neu peptides could promote the proliferation of BALB/c lymphocytes both in vitro and in vivo. HER2/neu peptide-specific CTLs could be induced from peptide-immu-nized BALB/c mice, which showed specifically killing effects on HER2/neu positive SP2/OHER2 cells but not on HER2/neu negative SP2/O cells. Moreover, the growth of SP2/OHER2 tumor were significantly suppressed in BALB/c mice immunized with HER2/neu peptides. Conclusion: These results suggested that it was feasible to use MHC-I epitopes derived from tumor-specific antigens as vaccines for cancer immunotherapy.
