2.Genotypes of human papilloma virus in male condyloma acuminata patients in Lishui area.

OBJECTIVETo explore the genotypes of human papilloma virus (HPV) in male patients with condyloma acuminate in Lishui area of Zhejiang Province, China.
METHODSUsing HybriMax, we identified the genotypes of HPV of the verruca samples from 110 male condyloma acuminate patients aged 16 - 65 years in Lishui area.
RESULTSHPV infection was detected in 107 (97.27%) of the condyloma acuminate patients, including 76 cases (71.02%) of simple infection and 31 cases (28.97%) of multiple infection. The peak age of infection was 21 - 40 years old. Totally, 14 genotypes of HPV were identified, including the high-risk genotypes as HPV16, 53, 52, 33, CP8304, 58, 51, 45, 66, 68, and 59, which accounted for 44.85% of the cases, and the low-risk ones as HPV6, 11, and 19 (43), with an infection rate of 93.45%.
CONCLUSIONThe low-risk genotype is a dominant type of HPV infection and the rates of high-risk genotype and multiple infection are high among the male condyloma acuminate patients in Lishui area.

Background There is no effective preventing way for the posterior capsular opacification(PCO) after cataract surgery.Objective The aim of this study was to investigate the preventing effect of adenovirus-mediated herpes simplex virus thymidine kinase/ganciclovir(HSV-tk/GCV) system on PCO.Methods Twelve New Zealand white rabbits were divided into BSS+BSS group,BSS+GCV group,HSV-tk+BSS group and HSV-tk+GCV group at random.Phacoemulsification was performed in bilateral eyes of rabbits,and 0.1ml of BSS or adenovirus vector with HSV-tk gene were injected into lens capsular bag during the operation based on the grouping.In 12 hours after operation,0.1ml BSS was injected into the anterior chamber daily for 3 times in BSS+BSS group and HSV-tk+BSS group,and 0.1ml GCV was used in the same way in BSS+GCV group and HSV-tk+GCV group.The situations of the anterior segment and the posterior capsule were examined under the slit lamp postoperatively.All the wet weight of capsular bags was assessed using electronic balance.The capsular membrane was histopathologically examined in the 4th week after injection.PCO was graded according to the method of Couderc.Results Mild corneal edema and aqueous humor opacity were observed in all the eyes within 3 days and returned to normal level gradually within 2 weeks postoperatively.PCO scores were higher in BSS+BSS group,BSS+GCV group,HSV-tk+BSS group than the HSV-tk+GCV group,showing the significant differences between them(H=2.647,P=0.008;H=2.939,P=0.003;H=2.884,P=0.004).No obvious difference was seen in PCO score among BSS+BSS group,BSS+GCV group,HSV-tk+BSS group(P>0.05).The wet weight of the capsular bags in HSV-tk+GCV group was reduced in comparison with BSS+BSS group,BSS+GCV group,HSV-tk+BSS group with the statistically significant difference between them(q=9.93,q=10.15,q=10.07,P<0.05).Histopathological examination revealed that mild proliferation of lens epithelial cells(LECs) at the capsular equator and migration toward the posterior capsule to form a thin soemmering ring in the periphery of the capsular bags and thin fibromembrane at the center of the capsular bags without cells or cortices in HSV-tk+GCV group.However,in BSS+BSS group,BSS+GCV group and HSV-tk+BSS group,the proliferation and migration of LECs were more obvious to form a dense soemmering ring in the periphery of the capsular bags and thick fibromembrane in the center of capsular bags.Conclusion Adenovirus-mediated HSV-tk/GCV system can effectively prevent PCO.
5. Effect of cereblon on inhibition of human lung cancer A549 cell migration by thalidomide

Objective:To investigate the influence of cereblon(CRBN)on the inhibitory effect of thalidomide on the migration of human lung adenocarcinoma A549 cells and related mechanism. Methods: The effect of thalidomide on the A549 cell proliferation was assayed by the MTS method. CRBN in A549 cells was knocked down(A549CRBN cells) with RNA interference and lentivirus infection. Transwell assay was performed to detect the effect of thalidomide on the cell migration of A549 and A549CRBN cells. RT-qPCR assay was performed to detect the gene expression of N-cadherin and vimentin. Results: The thalidomide at the 1, 10, 50 and 100 μmol/L concentration did not affect the proliferation of A549 cells, while the 100 μmol/L thalidomide could significantly inhibit the migration of A549 cells. After the CRBN gene knockdown the migration ability of A549 cells was significantly decreased(P<0.01), and the gene expression of N-cadherin and vimentin was also significantly reduced in the A549CRBN cells. The inhibitory effect of thalidomide on the cell migration in the A549 cells disappeared after CRBN gene knockdown. Thalidomide could inhibit the gene expression of N-cadherin and vimentin in a CRBN-dependent manner. Conclusion: Thalidomide could inhibit the migration of A549 cells and the expression of the migration-related genes via CRBN.
6. Optimization of extraction technology for Asteris Radix et Rhizoma in Zibai Zhisou Capsules with orthogonal test

Objective: To optimize the extraction technology of Asteris Radix et Rhizoma in Zibai Zhisou Capsules. Methods: Orthogonal test was carried out. The influences of concentration of solvent, the dosage of solvent, duration of extraction, and frequency of extraction on extraction results were investigated by using the content of shionons as index. Results: The optimal extraction technique was to extract pueraria in 80% alcohol with 8 times the weight of herbal medicine for 3 times, with 60 min once. Conclusion: High yield of extractum and high content of shionon are obtained with the present technology. The results with better repeatability are stable, which can provide the reference for the production of Zibai Zhisou Capsules.
7. Determination of costunolide and dehydrocostuslactone in Liuwei Muxiang Capsules by HPLC

Objective: To establish an HPLC method for the determination of costunolide and dehydrocostuslactone in Liuwei Muxiang Capsules. Methods: The determination was performed on VP-ODS (250 mm × 4.6 mm, 5 μm) with column temperature set at 25℃. The mobile phase consisted of methanol-0.4% phosphoric acid water (72∶28) at a flow rate of 1.0 mL/min. The detection wavelength was set at 225 nm. Results: The linear range of costunolide and dehydrocostuslactone was 203.12-2 031.20 ng and 207.84-2 078.40 ng (r = 0.999 9), with average recovery at 101.1% and 101.2%, RSD = 0.48% and 0.73% (n = 6). Conclusion: The method proved is simple, accurate, reproducible, and specific, and it can be used for the quality control and evaluation of Liuwei Muxiang Capsules.
8.Simplified urodynamic test for stress urinary incontinence

This study was to investigate the diagnosis value of simplified uredynsmic test in stress urinary incontinence. Fifty patients with stress test-confirmed urine incontinence underwent simplified urodynamic test before routine urodynamic test. Detrusor instability, max bladder volume, average flow rate, and residual urine of simple urodynamic test were compared. Simplified urodynamic test showed a sensitivity of 87%, specificity of 9/12, positive predictive value of 92%, and negative predictive value of 9/14. There was no significant difference in max bladder volume, average flow rate, and residual urine between the two tests. Simplified urodynamic test might be a reliable method for diagnosis of stress urinary incontinence, and could be used for detrnsor instability screening.
9.Clinical study on tension-free vaginal tape and tension-free vaginal tape obturator for surgical treatment of severe stress urinary incontinence

Objective To investigate the clinical efficacy and improvement of patients' quality of life in tension-free vaginal tape(TVT)and tension-free vaginal tape obturator(TVT-O)for surgical treatment of severe female stress urinary incontinence.Methods This study was a randomized,singleblinded,controlled trial.Patients were randomized by a computer-generated randomization schedule with allocation to either TVT or TVT-O procedure.TVT procedure was performed in 35 cases and TVT-O in 34 cases.None had received surgery for urinary incontinence or was in pregnancy.Transvaginal hysterectomy and prolapse reparation were done simultaneously in some of the patients.All patients were requested to complete the Urinary Distress Inventory(UDI-6)and Incontinence Impact Questionnaire(ⅡQ-7)as part of their pre-and postoperative assessment.Results All patients were evaluable and the mean follow-up was 14.5 months.The mean operative time was(18±5)min in the TVT-O group,Significantly shorter than in the TVT group(27±5)min(P<0.01).The two groups did not differ significantly in perioperative blood loss,postoperative complications(including tape erosion,pain in thigh or behind pubis),postvoid residual volume,hospital stays or expenses(all P> 0.05).Sixty patients were successfully treated for stress urinary incontinence(88.6%and 85.3%for TVT and TVT-O groups,respectively).There were significant improvements in postoperative scores for both the ⅡQ-7 and the UDI-6(P<0.01),except in subscale measuring symptoms of voiding dysfunction(P>0.05).Conclusions Both techniques appear to be equally effective in the surgical treatment of severe stress urinary incontinence in a short term review.Significant improvements could also be seen in patients' quality of life.However.TVT-O has a shorter operative time.No evidence of increasing risk of urethral obstruction after the operation could be found.Long term followups axe necessary to evaluate outcomes of different types of surgery for stress urinary incontinence.
