1.AN ANTl-TSH ANTI-IDIOTYPIC ANTISERUM WITH ACTIVITY OF THYROID-STIMULATING INHIBITOR IMMUNOGLOBULIN (TSH)

Rabbit anti-TSH anti-idiotypic antiscra were raised by immunization with auti-TSH antibody. The antiserum from rabbit No. 551 was found to have the activity of inhibiting the binding of TSH receptor on thyroid cell membrane and 125I-TSH, that is, TBII (TSH-binding inhibitor immunoglobulin) positive. This antiserum was also shown to inhibit cAMP release stimulated by 100 mU/L bTSH. The characteristics of No. 551 antiserum were similar to of patients with myxedema.
2.Measurement of soluble IL-2 receptor by sandwich ELISA with monoclonal and polyclonal antibodies

In this work, a simple and sensitive assay for soluble IL-2 receptor (sIL-2R) was established. The main procedures are as follows: the microplate was coated with anti-Tac monoclonal antibody (McAb); after sIL-2R in samples bound with the McAb, the rabbit anti-IL-2R serum was added;and HRP-goat anti-rabbit IgG was used to display and amplify this binding. The sensitivity of this method is about 100u/ml, which is near that in "Cellfree IL-2 Receptor Test Kit".
3.Recent advances in breast cancer stem cell phenotypes and clinical application

The cancer stem cell theory suggests that cancer develops from a subset of tumor cells that possess characteristics of stem cells. Breast cancer stem cells comprise a sub-population, which possesses the capacity of self-renewal and the potential for differentiation and high tumorigenicity. Evidence from bothin vitro andin vivo studies demonstrates breast cancer stem cells are responsible for tumor relapse, invasion and metastasis, chemo- and radio-resistance and epithelial-mesenchymal transition (EMT). Herein, this review highlighted the recent advances in breast cancer stem cells.
4.Observation the effect of double path endoscopic surgery in the treatment of fungal sinusitis

Objective To analyze the curative effect of double path endoscopic surgery in the treatment of fungal sinusitis,and to compare with the effect of simple in endoscopic nasal passages windowing.Methods The clinical data of fungal sinusitis patients were analyzed.According to the different surgery way,the patients were divid-ed into two groups.The observation group:nasal endoscopic nasal passages open a window in the joint of fangs nest small windows double path;Control group:simple in nasal endoscopic fenestration.Results The observation group had 35 cases and the control group had 37 cases.After treatment,the symptoms score of the observation group was sig-nificantly lower than the control group [(1 .09 ±0.83)points vs (1 .47 ±0.77)points;t =2.01 5,P =0.048].The effective rate of the observation group was significantly higher than the control group (94.29% vs 75.68%;χ2 =4.81 3,P =0.028).7 days after treatment,the C -reactive protein(t =4.971 ,P <0.01 ),TNF alpha(t =9.1 9,P <0.001 ),beta IL -1 (t =5.635,P <0.01 ),white blood cells (t =3.25,P =0.002)and IL -6(t =3.86,P =0.002) in the observation group were significantly lower than the control group (P <0.05).The preoperative plasma level of 1 -3 -β-glucan D between the two groups had no statistical difference (P >0.05).2(t =5.978,P <0.01 ),4(t =6.335,P <0.01 ),6(t =9.348,P <0.01 ),and 8 days(t =8.226,P <0.01 )postoperatively,1 -3 -β-glucan D levels were significantly lower than the control group,the differences were statistically significant (P <0.05).The postoperative recurrence rate of the observation group was significantly lower than the control group (8.57% vs 27.03%,the Log -rank χ2 =4.061 ,P =0.044).Conclusion In fungal sinusitis treatment,the use of endoscopic nasal passages open window joint fangs nest small windows double path can more effectively remove lesions,its forward curative effect is superior to pure in nasal endoscopic fenestration.
5.Serum soluble IL-2 receptor level in patients with leukemia

Serum soluble IL- 2 receptor (sIL-2R) level of 34 patients with leukemia were measured by ELISA. The sIL- 2 R levels are 296?99u /ml in normal, 660 ?449u /ml in AML, 1323 ?755 u/m in ALL, 1577 ? 759 u /ml in AMML and 1815 ? 858 u /ml in NHL. 5 of 6 patients with 2000u /ml sIL- 2 R died in one month after detection of sIL-2R. There is no apparent relationship between sII -2 R level and mononuclear cells count in peripheral blood. The results suggest that it is possible to be a new parameter for the observation and prognosis estimation to patients with leukemia by detection of sIL-2R.

Nanotechnology has been extensively applied in the diagnosis and treatment of cancer, and is currently a major focus of research worldwide.Among the various nanomaterials, gold nanoparticles (GNPs) are of particular interest to many researchers due to their superior properties of low cytotoxicity, good biocompatibility, and preferential accumulation in tumors (the enhanced permeability and retention effect), which in turn gives GNPs a potential application in cancer diagnostics, imaging, photothermal therapy, and radiotherapy.Many published studies have shown that GNPs can be used as a new ideal radiosensitizer, and therefore understanding the radiosensitizing effects of GNPs both in vitro and in vivo will be of great significance in the clinical translation of nanomedicine.

Objective To analyze the factors related to glare occurred after the phakic intraocular lens (P-IOL) implantation,and to raise the standard for the P-IOL selection.Design Retrospective case series.Participants 104 patients with high myopia underwent P-IOL implantation in Beijing Tongren Hospital.Methods In 104 cases,22 cases (22 eyes) were implanted angle supported Phakic6 designed accord with corneal symmetry,54 cases (54 eyes) were implanted iris fixed Verisyse,and 28 cases (28 eyes) were implanted posterior chamber PRL.Selection of the diameter of P-IOL optics depended on P-IOL diopters.The pupil diameter,the P-IOL optic center position,the pupil and corneal center position were measured with anterior segment optical coherence tomography (OCT).The contrast sensitivity (CS) under different condition (day,day and glare,night,night and glare) and in different space frequency (1.5,3,6, 12,18c/d)were examined with Optec6500 system.Main Outcome Measures Glare incidence,CS,and the influence of pupil diameter and P-IOL location to glare.Results None of the 104 P-IOL eyes occurred glare in day time.59 eyes (56.73%) complained of glare in dime condition.The glare incidence was 63.18% (14/22) in Phakic6 eyes,55.56% (30/54) in Verisyse eyes,and 53.27% (15/28) in PRL eyes (P=0.691).CS of all surgical eyes were improved in day time with or without glare luminous environment (P=0.000).And the CS of halo-complaining eyes was lower than of no halo eyes in night time with or without glare luminous environment (P=0.000).Glare oc- curred more frequently in eyes with the pupil diameter larger than P-IOL diameter in clime condition after surgery (10 in Phakic6 eyes, 20 in Verisyse eyes,12 in PRL eyes,all P
8.The establishment of minipigs model of no-reflow after percutaneous coronary intervention of acute myocardial infarction

Objective To investigate the extablishment of minipigs no-reflow models after percutaneous coronary intervention of AMI. Methods A total of 20 wuzhishan minipigs (9?1 months old with body weight of 25?5 kg) were chosen. Left ventriculography and coronary angiography (CAG) were performed sequentially by femoral approach,AMI and no-reflow was set up by diliating balloon to occlude the coronary coronary and reperfusion after the injection of microthrombus into the left anterior descending artery. At the same time, left ventricular hemodynamics parameters were recorded. Changes in ECG and IC-ECG were monitored. Results (1) Sixteen animals survived after the procedures and fourteen of them reached the criterion of AMI with no-reflow (TIMI≤2, CTFC≥36.2). The model extablishment success rate was 70%. (2)The hemodynamics parameters including heart rate, PRI, LVEDP, and PCWP increased, but blood pressure decreased at the time no-reflow was formed compared with those before LAD was occluded. (3) During the experiment, the changes in ECG and IC-ECG were similar to those observed in human in AMI. Conclusion Minipig is an ideal species for the establishment of animal no reflow model reperfusion after microthombus injection and balloon occlusion of LAD.

Objective To construct recombinent retroviral vectors containsins the expression sequence of hIL-4,and detect the expression of target genes.Methods Design the primer with restrictive enzyme spot and amplify target gene by PCR from plasmid including hIL-4 gene.Clone the target gene into retroviral vector pLXSN,and identify the aquired plasmid by sequencing.Transfect human synoviocytes with acquired virus in vitro.Detect the protein expresssion by western blotting.Results We successfully constructed the recombinant retrovirus-rRV-hIL-4.The hIL-4 gene was transduced into human synoviocytes by recombinant retrovirus in vitro.The protein expression of genes was detected by Western blotting.Conclusion Recombinant retrovirus rRV-hIL-4 is constructed successfully.The hIL-4 gene is transduced successfully into the human synoviocytes in vitro and the transduced synoviocytes can express hIL-4 protein.
