1.The Immune Protective Effect of Astragalus Polysaccharide on Mice Genital Tract Infected with Chlamydia Trachomatis

[Objective]To probe into the immune protective effect of Astragalus polysaccharide on mice ,whose genital tract was infected with Chlamydia trachomatis. [Methods]After the murine model of genital tract infected with chlamydia trachomatis was established, we could obtain the minimum effective infective dose. Mice were immuned by different doses(10 mg·kg-1, 20 mg·kg-1, 40 mg·kg-1) of Astragalus polysaccharide(APS) and 0.9%NS , meanwhile the blank group was regarded as the control group. After mice were immuned, we detected serum IgG antibody levels by ELISA method,and took the twice effective dose of chlamydia trachomatis to attack the genital tract after a week. Then the mice genital shedding of epithelial cells were cultured at each interval 2d. After 10 days, we kil ed the mice to calculate spleen(thymus) index, and observed the pathological changes in the reproductive tract of the mice. [Results]Compared with the control group, spleen(thymus) index and serum IgG antibody levels of Astragalus polysaccharide group are significantly higher. The results of vaginal exfoliated cells from Ct training show that, Ct positive rate of Astragalus polysaccharide group is significantly reduced, the number of inclusion bodies is also significantly decreased. And with the increasing concentration of Astragalus polysaccharide, the protective effect is more obvious. The NS group has no significant difference from the control group in al indicators. [Conclusions]Astragalus polysaccharide can induce immune protection on Ct genital tract attacked mice, and in the concentration range, with the increase of the concentration, the protection effect is better.
2.c-Myc promoted migration and invasion of malignant melanoma B16-F10 cells through regulating PIK3 R3

Objective:To investigate the effect and the related mechanism of c-Myc on the proliferation,invasion and migration ability of malignant melanoma B16-F10 cells. Methods:We detected the expression of PIK3R3 in malignant melanoma and normal tis-sues. Efficiency of gene silencing of c-Myc and PIK3R3 was determined by qPCR and Western blot. We detected the proliferate ability of B16-F10 cells after silencing c-Myc and PIK3R3 using EdU assay. We detected the migration and invasion ability of B16-F10 cells after silencing c-Myc and PIK3R3 using wound healing assays and Transwell matrigel invasion assays. The expression of miR-199a after silencing c-Myc and PIK3R3 using qPCR. The expression of c-Myc and PIK3R3 was detected by qPCR after transfecting miR-199a mimics or miR-199a inhibitor. Dual-luciferase reporter assay system was used to detect miR-199a regulating c-Myc and PIK3R3. Results:Compared normal skin tissue,expression of PIK3R3 was significantly increased in malignant melanoma tissue;after silencing c-Myc and PIK3R3 gene,the proliferation,invasion and metastasis of melanoma cell line B16-F10 were significantly reduced;expression of miR-199a upregulated after silencing c-Myc and PIK3R3 genes, expression of c-Myc and PIK3R3 decreased after transfecting miR-199a mimics, expression of c-Myc and PIK3R3 upregulated after transfecting miR-199a inhibitor, dual luciferase reporter system test results revealed miR-199a can directly regulate c-Myc and PIK3R3 transcription activity. Conclusion: miR199a regulated the expression of c-Myc,then promote proliferation,migration and invasion in malignant melanoma cells.
3.Design and clinical application of modified fixed Maryland Bridge for restoration of geriatric anterior dentition defects

Objective The anterior dentition defects of geriatric patients were restored with modified fixed Maryland Bridge.The clinical design,tooth preparation,fabrication of prosthesis and clinical indication were summarized.Method 24 cases of geriatric patients with anterior dentition defects were restored with modified fixed Maryland Bridge.All selected cases were accompanied with different degree of loosening of neighboring teeth,gingival atrophy and exposure of root.The dentition defects were successfully restored and followed up for 3 to 5 years.Results The overall success rate was 87.5%.Three patients had the prosthesis loosened and re-bonding was necessary.No loosening was found in other patients.There was no swelling or inflammation of gingival underneath the prosthesis.All patients were quite satisfactory with the function and appearance of the prosthesis.Dental calculus was observed in 12 patients,and 2 patients complained of a feeling of foreign body on the side of tongue.Conclusion The modified fixed Maryland Bridge,when used in restoring the dentition defect of geriatric patients,showed the advantages of less tooth reduction,perfect esthetic effect,good fixation of loosening teeth as well as restoration of missing teeth with its splint wings.However,the long-term effect of this technique needs to be further observed.

5.Investigation on current status of advanced schistosomiasis in Shanggao County

Objective To understand the current status of advanced schistosomiasis patients, so as to provide the reference for improving the medical care of them in Shanggao County. Methods The data of advanced patients in Shanggao County were collected from the Information Management System for the Advanced Schistosomiasis Chemotherapy and Assistance in Jiangxi Province and a field survey including the epidemiological investigation physical examinations and B ultrasound examinations was performed in 2014. All the data were analyzed statistically and compared with those in 2010. Results Among the 128 ad?vanced schistosomiasis patients the male female ratio was 2.46 1 the average age was 68.02±6.19 years and the patients old?er than 65 years contributed to 64.84%for the all. The proportions of clinically types of ascites splenomegaly multiple granulo?ma in the colon dwarf general and haemorrhage were 57.81% 12.50% 2.34% 0 26.57% and 0.78% respectively. The ultrasound examinations revealed the average diameter of the main trunk of portal vein was 12.70 ± 1.91mm in 2014 which was significantly broader than that of 12.05 ± 0.34mm in 2010 P < 0.01 . Conclusions The advanced schistosomiasis patents in Shanggao County decreases with years and shows an aging trend. Some advanced schistosomiasis patients are worse therefore the chemotherapy and salvation work still should be strengthened.
6.Inhibitory effect of galangin on polyphenol oxidase and study of mechanism

Objective:To investigated the activity inhibition and inhibitory type of polyphenol oxidase (PPO) induced by galangin and the interaction mechanism of galangin with polyphenol oxidase was preliminarily indicated,and prove the related mechanism of galangin on proliferation of on human melanoma A375 cells.Methods: The activity inhibition and inhibitory type of PPO induced by galangin were investigated by spectrophotometric method,and interation mechanism of galangin with PPO was preliminarily indicated by fluorescence quenching and molecular docking,and chelating copper ions with the inhibitory mechanism of galangin on polyphenol oxidase was measured.Results: Galangin was a competitive inhibitor,the IC50 and Ki on PPO were obtained to be (47.86±3.33) and (24.83±1.45)μmol/L,respectively.Fluorescence spectrum indicated the fluorescence of PPO was quenched effectively by galangin and the binding constant Ka was obtained to be (4.67±0.43)×104 L/mol.Chelating copper ions and molecular simulation further showed that galangin was combined with active center of copper ions,and formed hydrogen bonds with catalytic site His259.Luteolin could induce the apoptosis of A375 cells significantly,and the tyrosinase activity and melanin synthesis were decreased.Conclusion: Galangin as a competitive polyphenol oxidase inhibitor and reduced the activity of polyphenol oxidase.which provides the theoretical basis for the clinical anti skin cancer.
7.Construction of two retroviral vectors for interleukin-1 receptor antagonist and interleukin-10 and in vitro transfection of rabbit synoviocytes

Objectives To construct two retroviral vectors, one containing human interleukin-1 recep-tor antagonist (hIL-1Ra) gene and the other containing human interleukin-10 (hIL-10) gene and to transfect rabbit synoviocytes in vitro and detect the expression level of target genes. Methods RNA from human peripheral blood mononuclear cells were extracted and target genes were amplified by RT-PCR. The target genes were cloned into retroviral vector pLXSN, which was then transducted into GP2-293 cells to produce recombinant retrovirus. Rabbit synoviocytes were transfected and the expression of target genes was detected by RT-PCR, immunohistochemistry and western-blot. Results The retroviral vector containing hIL-1Ra gene or hIL-10 gene was constructed successfully. The hIL-1Ra gene and hIL-10 gene were transduced respectively into rabbit synoviocytes in vitro. The mRNA level of both genes reached peak in 5 days. In positive cell clones, the protein level of hIL-1Ra reached peak within 30 days and maintained at least 60 days; the protein level of hIL-10 maintained at least 40 days. Conclusion The hIL-1Ra gene and hIL-10 gene can be transduced successfully into rabbit synoviocytes by recombinant retrovirus.
8.Syphilis in China from 2000 to 2013: epidemiological trends and characteristics

Objective To investigate the trends in and characteristics of syphilis epidemiology in China,and to provide a scientific basis for the development of control strategies.Methods An epidemiological study was performed on syphilis cases reported from 31 provinces,autonomous regions and municipalities between 2000 and 2013.Results The reported syphilis incidence increased yearly from 6.43 per 100 000 person-years in 2000 to 32.86 per 100 000 person-years in 2013,with an average annual growth rate of 13.37%.Syphilis was reported in most (99%) counties/districts in China mainland except for a few remote mountain areas,with a significant difference in the incidence of syphilis between different regions.The regions with a high incidence of syphilis included minority areas in Northwest China (Xinjiang,Qinghai and Ningxia),Minjiang River area,Yangtze River Delta (Zhejiang,Shanghai),Zhujiang River Delta (Guangxi,Guangdong),etc.There had been a rapid increase in the incidence of syphilis in some regions previously reported to have a low incidence (such as Guizhou,Shaanxi,Henan,etc).Among the reported cases,female patients predominated with an average male/female ratio of 0.92 ∶ 1.The incidence of syphilis was highest in the age group of 20-39 years,but increased yearly by more than 30% in the population aged > 60 years.Of 20 occupations,peasant workers accounted for the highest proportion (31.56%) of syphilis cases,and retired population showed the most rapid increase by about 27.35% per year.The constituent ratio of primary and secondary syphilis in all the syphilis cases had declined yearly,while that of latent syphilis increased by more than 50% after 2010.Conclusions Syphilis remains a major public health issue in China,and measures based on the epidemiological features are urgently needed to effectively control epidemic syphilis.
9.Valuation of MR Imaging for Diagnosis of Carotid-cavernous Sinus Fistulae

Objective To discuss the diagnostic value of MR in carotid-cavernous sinus fistulae(CCF). Methods Conventional MRI and 3D TOF MRA were performed in 23 cases of CCF confirmed by DSA. The MRI and MRA appearances of CCF were reviewed. Results The main MRI and MRA findings of CCF included enlargement of cavernous sinus, distortion and dilatation of the superior ophthalmic vein in the involved side in all cases.Dilatation of the inferior ophthalmic vein in 3 cases, superficial cerebral veins in 5 cases,dilatation of pterygoid venous plexus in 2 cases, superior petrosal sinus in 3 cases, and inferior pertrosal sinus in 2 cases. In the counterside, dilatation of cavernous sinus were found in 3 cases. Opening of posterior communicating artery in the circle of Willis happened in 17 cases. Conclusion MRI combined with MRA is able to make a reliable diagnosis of CCF, and is also able to demonstrate the condition of compensatory blood supply in brain, which is useful in the decision of the theraputic plan.
