3.Effects of sevoflurane preconditioning and postconditioning on activity of NF-κB during lung ischemia-reperfusion in rats

Objective To evaluate the effects of sevoflurane preconditioning and postconditioning on the activity of NF-κB during lung ischemia-reperfusion (I/R) in rats.Methods Fifty-four adult male Sprague-Dawley rats,weighing 300-350 g,were randomly divided into 3 groups (n =18 each) using a random number table:sham operation group (S group),I/R group and sevoflurane preconditioning and postconditioning group (SPP group).Lung I/R was induced by clamping the left pulmonary hilum for 45 min followed by 120 min reperfusion in I/R and SP groups.The left pulmonary hilum was only isolated but not ligated in group S.In group SP,2.1% sevoflurane was inhaled for 30 min followed by 10 min washout,the left pulmonary hilum was then ligated for 45 min,and 2.1% sevoflurane was inhaled for 30 min again starting from onset of reperfusion.Six rats were chosen at 30,60 and 120 min of reperfusion and sacrificed in each group,lungs were removed for determination of wet/dry lung weight (W/D) ratio and content of tumor necrosis factor-α (TNF-α) (by ELISA) and expression of nuclear transcription factor κB (NF-κB) p65 of nuclear protein (by Western blot) and for microscopic examination of the pathologic changes of lungs (with light microscope).Results Compared with S group,W/D ratio,TNF-α content and nuclear protein NF-κB p65 expression in lung tissues were significantly increased at each time point of reperfusion in I/R and SPP groups.Compared with I/R group,W/D ratio,TNF-α content and nuclear protein NF-κB p65 expression in lung tissues were significantly increased at each time point of reperfusion,and the pathological changes of lungs were attenuated in SPP group.Conclusion Sevoflurane preconditioning combined with postconditioning can reduce the lung I/R injury in rats through inhibiting NF-κB activity in lung tissues.
5.Effect of dietary fiber on carbohydrate metabolism in healthy volunteers with 13C breath test

Objective To investigate the effect of dietary fiber on carbohydrate metabolism of healthy volunteers after the intake of corn starch meal. Methods Totally 12 healthy volunteers aged (25. 8 ± 5. 3) years were enrolled in this study, and they were equally randomly divided into two groups (group A and group B). This was an open, randomized, cross-over, and two-period study, and each period lasted for one day. In period 1, the subjects in group A received fiber-free corn starch and group B received high-fiber corn starch (containing 16 g dietary fiber). In period 2, the two groups are crossed. There was a one-week wash-out time between the two study days. On the study day, breath samples of fasting and 0. 5, 1.0, 1.5, 2.0, 2.5, 3.0, 3.5, 4.0, 5.0, 6.0,7. 0, 8. 0 hours post meal were collected to measure13 CO2. Results The delta over baseline at 0. 5, 1.5, 2. 0,2. 5, 3. 0, 4. 0 hour after test meal in fiber free group and in high fiber group were 0. 79, 2. 03, 2. 57, 2. 86,3. 02, 3. 18 and 0. 16, 1. 33, 1.77, 2. 10, 2. 34, 2.42, respectively (the P value was 0. 014, 0. 014, 0. 011,0.018, 0. 036, and 0.020, respectively). Peak concentration of delta over baseline of fiber free group and high fiber group was 3.18 and 2. 56 respectively, there was no significant difference between two groups (P > 0. 05) ,peak time of the group at 4. 0 hour and 3. 5 hour respectively, showed significant difference (P = 0. 032). The cumulative percentage dose recovered 0. 5-6. 0 hours after test meal in fiber-free group and in high-fiber group were 0.41, 1.46, 3.15, 5.50, 8.28, 11.30, 14.42, 17. 62, 23. 65, 28. 78 and 0. 09, 0. 55, 1.61, 3.22,5.23,7.53, 10.09, 12.68, 17.60, 22.27 respectively (the P value was 0.014, 0.018, 0.018, 0.014, 0.013,0.014, 0.018, 0.020, 0.025, and 0.044, respectively). However, there was no significant difference 6.0 hours after meal (P > 0. 05 ). Conclusion The dietary fiber used in this study can delay the absorption of carbohydrate 6. 0 hours within intake without influencing its total absorption amount.
6.Influencing factors of iron deficiency in 3 to 6 months-old infants in Dongcheng District of Beijing

Objective To explore the prevalence of anemia,iron deficiency and iron deficient anemia (IDA),and to identify their risk factors in 3 to 6 months infants in Dongcheng District of Beijing.Methods All the 845 infants aged 3 to 6 months who received bacille calmette guerin (BCG) vaccine during December 2010 and October 2011 in Hepingli Hospital of Beijing were invited to complete the investigation.Data (months,gender,birth weight,birth height,etc) were collected through a questionnaire survey.Infants were tested for hemoglobin (Hb),mean corpuscular volume (MCV),mean corpuscular hemoglobin (MCH),mean corpuscular hemoglobin concentration (MCHC),serum ferritin (SF) and zinc protoporphyrin (ZPP) and mothers underwent measurement for Hb,SF and serum iron (SI).Descriptive epidemiology and non-conditional logistic regression model were used to analyze the prevalence of nutritional anemia.Results The prevalence of anemia,iron deficiency and IDA was 12.06%,7.38% and 2.11%,respectively.Non-conditional logistic regression analysis showed that the associated factor of anemia in infants was month age (vs.3-4 months age group,4-5 months age group:odds ratio (OR) =0.496,95% confidence interval (CI) 0.272-0.905 ; 5-6 months age group:OR =0.494,95% CI 0.234-1.043).The factors associated with iron deficiency were month age (vs.3-4 months age group,4-5 months age group:OR =0.539,95% CI 0.266-1.088 ; 5-6 months age group:OR =0.334,95% CI 0.125-0.891) and time of first breast-feeding (vs.the first day,the second day:OR =2.359,95% CI 1.191-4.675; the third day:OR =1.154,95% CI 0.450-2.963).Conclusions Our data show that iron nutrition in 3 to 6 months-old infants in Dongcheng District of Beijing was in good situation.The influencing factors of iron nutrition in infants may be month age and the time of first breast feeding.
7.ANALYSIS OF L-VALINE BIOSYNTHESIS MECHANISM BY METABOLIC FLUX BALANCE MODEL

Using metabolic flux balance model , the metabolic flux balance of L- v aline synthesis was established in this paper by material balances and linear p rogramming method. The analysis results indicate that 62.8% metabolic flux ent er ed EMP pathway and 38.2% metabolic flux entered the HMP pathway. And only 9.2 % c arbon entered the TCA cycle. But comparing to the optimal flux distributions of 92.31, the production of L-valine should be improved from the genetic manipula ti on and fermentation control through reducing byproduct of amino acid and decreas ing the metabolic flux.
8.The result of the antibiotics susceptibility testing of Neisseria gonorrhoeae

Objective To analyse the antibiotics susceptibility result of Neisseria gonorrhoeae(N.g) isolates.Method 95 strains of N.g were performed the in-vitro susceptibility test.MICs of penicillin, tetracycline, ciprofloxacin, levofloxacin, ceftriaxone, cefotaxime, cefepime, and spectinomycin were determined by agar dilution.Results Almost all of the N.g strains were susceptible to the first-line antibiotics used for treating the infection. The resistance rate to ciprofloxacin, tetracycline and penicillin were 100%, 78.9% and 68.4% respectively.Conclusion Although N.g is susceptible to first-line antibiotics used for treating the infections at the clinics, the situation of resistance to penicillin, tetracycline and ciprofloxacin is very serious.
9.Current status of the application of translational medicine in the early diagnosis of pancreatic cancer

Pancreatic cancer is a rapidly progressive and highly malignancy of the digestive system. In recent years, the diagnosis and treatment of pancreatic cancer has been in a slow stage of development, and the 5-year survival rate of patients remains very low. The main objective of translational medicine is to remove the barriers between basic medical research and clinical medical applications, to achieve practical integration between laboratory and clinic, and to accelerate the translation of the results obtained from basic research into clinical diagnosis, evaluation and treatment of diseases, thus promoting the development of life sciences. With the rapid development of the concept and technology of translational medicine, its application in the early diagnosis of pancreatic cancer can bring new hope for effectively improving the overall prognosis of patients. The authors comprehensively analyzed the latest research progress of translational medicine in the early diagnosis of pancreatic cancer in order to improve the early diagnosis and long-term survival of pancreatic cancer patient.
10.Bone regeneration on hard palate after palatal surgery on cleft palate and the relative factors.

OBJECTIVEAttending to observe the bony healing on hard palate after palatal repair, and to discuss the factors affecting on it.
METHODS52 patients with repaired cleft palate over 5 years postoperatively were examined, the CT scan of head was taken. The incidence of the bone regeneration among the patients examined was calculated, the position and quality of bone tissue were measured according to the CT images. After all, analysis was applied to evaluate the factors affecting on the bone tissue formation.
RESULTSFormation of bone bridge was found in the 37 cleft gaps out of 52 patients (71%), the ratio of the sex among the patients who had bone bridge was 1:1, there was no obvious difference between unilateral and bilateral cleft palate. Considering the operation age with the bone formation, the highest percentage of bone bridge formation fell into the group of 4-7 years old, and the most occurring region were in premolar and anterior part of molar area.
CONCLUSIONThere could be bony healing(regeneration bone tissue RBT) after palatal repair on cleft palate patients. The operation age could be an important affecting factor to RBT, but the sex and the clinical type of cleft palate make no difference on the bone tissue formation.
