1.Application of different biomaterials in the preservation of vital pulp in carious deciduous teeth: a prospective, single-center, randomized, controlled clinical trial

BACKGROUND:The success of the preservation of vital pulp in carious deciduous teeth depends not only on determining the health status of the pulp,but also on selecting the most suitable pulp capping agent.At present,the commonly used pulp capping agent materials contain calcium hydroxide preparation and mineral trioxide aggregate (MTA).Recently,new biomaterials,such as Bioaggregate,Biodentine,and TheraCal,with good biocompatibility and good edge sealing,have been developed and extensively used.OBJECTIVE:To compare the efficacies of MTA,calcium hydroxide preparation,Biodentine and TheraCal LC that are commonly used in pediatric dentistry in the preservation of vital pulp in carious deciduous teeth.METHODS:This prospective,single-center,randomized controlled clinical trial will be conducted in Chengdu Maternal and Child Center,China.Children with deep caries of deciduous teeth,who will be scheduled to undergo indirect pulp capping,direct pulp capping,and vital pulpotomy,will be selected and randomized into four groups according to pulp capping agents:MTA group,calcium hydroxide group,Biodentine group and TheraCal group.Patients will receive return visit at postoperative 3,6 and 12 months during follow-up.By using X-ray,we will observe and compare the speed and quality of the formation of the dentin bridge with different biomaterials,and assess the reason for the failure of treatment.This trial was registered in ClinicalTrials.gov identifier:NCT03220360,on July 14,2017.The study protocol has been approved by the Ethics Committee of Chengdu Maternal and Child Center,China (approval number:20170506).All protocols will be performed in accordance with the Ethical Principles for Medical Research Involving Human Subjects in the Declaration of Helsinki.Written informed consent will be provided by each patient and their family members after they indicated that they fully understood the treatment plan.RESULTS AND CONCLUSION:This study will compare the efficacies of various biomaterials in the preservation of vital pulp in carious deciduous teeth.New materials Biodentine and TheraCal will be selected in classical technique to verify their efficacies and explore their superiorities.Classical materials calcium hydroxide and MTA will be selected as controls,which will be separately used in indirect pulp capping,direct pulp capping,and vital pulpotomy to provide reliable choices for clinicians.
2.Research on the relationship between field cognition style and self-handicapping of medical students

Objective To explore the relationship between field cognitive style and self-handicapping of medical students.Methods 124 medical students were measured by embedded figure test (EFT) and self-handicapping questionnaire (SHS).Results ①The scores of self-handicapping questionnaire were significantly different in whether they like their major or not(33.61±5.21,35.94±5.16,t=-0.218,P=0.031).Differences were also found in whether they could obtain the scholarships (33.20±6.19,35.12±4.12,t=-0.247,P=0.043),and in whether they were in a relationship(35.35±5.48,33.39±5.01,t=2.063,P=0.041).②Compared with field-independent cognitive style students,Field-dependent cognitive style students ' self-handicapping score was higher (35.69±3.91,32.63±6.08,t=3.303,P=0.001).③Field-dependent cognitive style,dislike their major and be in a relationship were the significant predictors of self-handicapping.Conclusion Correlation exists between field cognitive style and self-handicapping of the medical students.Students who belong to field-dependent cognitive style are more prone to self-handicapping than field-independent cognitive style students.
3.The expression and function of retinoic acid-inducible gene Ⅰ in monocyte-derived dendritic cells in patients with hepatitis B virus infection

Objective To investigate the expression and function of retinoic acid-inducible gene Ⅰ(RIG-Ⅰ) in monocyte-derived dendritic cells (MoDC) at different stages of hepatitis B virus(HBV)infection and to explore the role of RIG-Ⅰ in the disease progression after HBV infection. Methods Peripheral blood samples were collected from 28 hepatitis B virus-infected persons, including 21 cases of chronic hepatitis B (CHB) and 7 of acute hepatitis B (AHB). Eighteen healthy subjects were recruited as controls. Purified CD14~+ monocytes were isolated by CD14 microbeads. MoDCs were induced from CD14~+ monocytes with granulocyte macrophage-colony stimulating factor (GM-CSF) and interleukin (IL)-4 for 7 days, and then were infected with vesicular stomatitis virus (VSV) to stimulate RIG-Ⅰ expression. The mRNA expression levels of RIG-Ⅰ, interferon (IFN )-promoter stimulating factor-1 (IPS-1) and IFN-β at 16 hours and 24 hours after infection with VSV were measured by real-time quantitative polymerase chain reaction (PCR). Data with normal distribution were tested by analysis of variance. Continuous variables between groups were compared using Mann-Whitney U test. Comparison among multiple groups was done by Kruskal-Wallis test. Results The expression levels of RIG-Ⅰ in MoDCs from CHB patients were significantly lower than those in AHB patients and healthy controls at 16 hours (2.44±2.03, 19. 54±3. 15, 21. 48±8. 39, respectively; F=7.451,P=0.002) and 24 hours (2. 68±2. 93, 10. 31 ±3. 88, 14. 01 ±5. 04, respectively, F = 7. 908, P = 0. 001)following VSV stimulation. The IPS-1 levels in both CHB patients and AHB patients were higher than those in healthy controls at 16 hours (2. 05±l. 08, 1. 99±1. 56, 0. 60±0. 31, respectively) F=7.246,P =0.003) and 24 hours (2. 27±2. 16, 3.24 ± 1.21, 1. 08±0. 73, respectively; F= 13. 598, P = 0. 001).Furthermore, the IFN-β expression levels were significantly lower in CHB patients compared to AHB patients and healthy controls at 16 hours and 24 hours after VSV stimulation. Conclusions The expressions of RIG-Ⅰ and IPS-1 in MoDC are abnormal in HBV infected persons, which indicates that RIG-Ⅰ signaling pathway might be blocked by HBV. The impaired function of MoDC may play a role in HBV infection and chronicity.

Objective To elucidate the roles of TANK-binding kinase-1(TBKl)in hepatitis B virus (HBV)infection induced interferon antiviral immunity.Methods Peripheral blood monocytes were separated by CD14 magnetic microbeads from healthy volunteers(HV)and chronic hepatitis B(CHB)patients.Purified mDCs were induced and proliferated in the culture medium with human granulocyte-macrophage concentration of 25 mg/L were stimulated.The mRNA expressions of TBK1,interferon regulatory factor (IRF)3 and interferon(IFN)-βwere quantified by real time polymerase chain reaction(PCR).The levels of IFN-β in supernatants were determined by enzyme-linked immunosorbent assay(ELISA).Reslllts The mRNA levels of TBK1,IRF3 and IFN-β did not change significantly at 0,12,24 and 48 h after the significantly at 0, 12, 24 and 48 h in CHB group, whereas, it was significantly up-regulated at 12 h in HV group. Conclusions Our results suggest that there may be some disorders in host antiviral signal transduction pathways downstream the binding between ligands and receptors on mDC surface. The insufficient IFN-β expression after HBV infection may result in persistent chronic infection.
5.Elongated needle for post-stroke indigestion.

OBJECTIVETo observe the clinical efficacy difference among elongated needle, filiform needle and medication for post-stroke indigestion.
METHODSSixty cases of post-stroke indigestion were randomly assigned into an elongated needle group, a filiform needle group and a medication group, 20 cases in each one. Acupuncture of restoring consciousness and inducing resuscitation and conventional medical treatment were applied in the three groups. 125 mm elongated needles were perpendicularly inserted at Zhongwan (CV 12), Liangmen (ST 21) and Tianshu (ST 25) in the elongated needle group; 20 to 30 mm perpendicular filiform at the same points in the filiform needle group. All the treatment was given for 4 weeks, 6 times a week, 1 time a day. Domperidone was prescribed orally in the medication group for 4 weeks, 3 times a day and once 10 mg. The indexes were dyspepsia TCM symptom score, Liz dyspepsia questionnaire (LDQ), Nepean Dyspepsia Index of quality of life (NDLQI), Safety Data Sheet (SDS) self rating scale and side effect scale (TESS). The effects were evaluated.
RESULTSThe scores of TCM symptom, LDQ, SDS scores in the three groups decreased and NDLQI increased after 7-day, 14-day, and 28-day treatment as compared with those before treatment (<0.05,<0.01). The TCM symptom score and LDQ score in the elongated needle group after 28-day treatment were lower than those in the filiform needle and medication groups (<0.05,<0.01). The SDS score in the medication group was lower than that in the elongated needle and filiform needle group (both<0.05). There were no significant statistical difference for NDLQI score in the three groups (>0.05). The total effective rate was 90% (18/20) in the elongated needle group; those in the filiform needle group and medication group were 70% (14/20) and 75% (15/20) respectively, indicating statistical significance (<0.01).
CONCLUSIONAcupuncture with elongated needle can obviously improve symptoms in the patients with post-stroke indigestion, which is better than filiform needle and medication.
6.Clinical profiles of circulating plasmacytoid dendritic cells in chronic hepatitis B patients in response to pegylated-interferon alfa-2a treatment.

OBJECTIVETo investigate the changes in circulating plasmacytoid dendritic cells (pDCs) in patients with chronic hepatitis B (CHB) during the course of treatment with pegylated-interferon alfa-2s (peg-IFNa-2a) and to determine the correlations with therapeutic response.
METHODSForty-one patients with CHB who were receiving peg-IFNa-2a antiviral treatment for 48 weeks were enrolled in the study.Expression of the Toll-like receptor 9 (TLR9) on and frequency and functionality of the pDCs were analyzed at treatment weeks 0, 2, 12, 24, 36 and 48.
RESULTSAll patients exhibited an initially rapid decrease in the numbers of circulating pDCs and showed CpG-induced endogenous IFNa production within the first 2 weeks of treatment.Subsequently, all responders displayed a continuous increase in pDC numbers as well as functionality, both of which peaked around week 12 of treatment; in addition, these treatment responses were accompanied by significantly increased levels of type 1 T helper cytokines (P less than 0.05), which did not occur in the non-responders.
CONCLUSIONpDCs are involved in the initial therapeutic immune response stimulated by peg-IFNa-2a treatment.Recovery of blood pDC number and functionality may represent a predictor of favorable response to peg-IFNa-2a antiviral treatment in patients with CHB.

7.Establishment of BGC-823/pBaBb-puro-MACC1 gastric cancer cell line stably expressing MACC1 and its tumor-related gene expression profiles.

OBJECTIVETo establish a gastric cancer cell line with stable expression of metastasis-associated in colon cancer 1 (MACC1) and detect the changes in tumor-related gene expression profiles for investigating the possible regulation mechanisms between MACC1 and the differentially expressed genes.
METHODSThe full-length MACC1 cDNA was amplified from human embryonic kidney 293FT cells and cloned into the pBaBb-puro vector. The recombinant pBaBb-puro-MACC1 expression vector, after identification with restriction enzyme digestion, was transfected into 293FT cells, and the expression of fluorescent reporter gene was observed. pBaBb-puro-MACC1 vector was transfected into human gastric cancer BGC-823 cell line to establish BGC-823/pBaBb-puro-MACC1 cell line stably expressing MACC1. Quantitative RT-PCR and Western blotting were used to detect MACC1 expression in both BGC-823/pBaBb-puro-MACC1 and control BGC-823 cells. High-throughout cDNA microarray was used to screen the effects of MACC1 on the gene expression profiles of gastric cancer cells.
RESULTSThe recombinant pBaBb-puro-MACC1 plasmid was successfully constructed and verified by PCR and sequencing. BGC-823/pBaBb-puro-MACC1 cells showed significantly increased MACC1 mRNA expression as compared with the control cells. The results of cDNA microarray identified 33 up-regulated and 24 down-regulated genes in the cells after MACC1 transfection involved were in various cellular functions.
CONCLUSIONThe established BGC-823/pBaBb-puro-MACC1 gastric cancer cell line show some important molecular changes caused by MACC1.

8.Relationship between abuse experience with suicidal ideation and behavior of junior middle school students in Chongqing

Objective:
To explore the relationship between abuse experience with suicidal ideation and suicide attempts of junior middle school students, and to provide a reference for suicide prevention of junior middle school students.
Methods:
Cluster sampling method were used to selct 10 289 junior middle school students from 25 districts and counties of Chongqing were included in the analysis of this study from July to September in 2020. And Questionnaire on Abuse in Childhood and Mental Health Scale for Middle School Students were applied to collect the data about demographic information, suicide ideation and behavior.
Results:
The prevalence of suicidal ideation and suicide attempts among junior middle school students were 20.93% and 10.83%, respectively. Multivariate Logistic regression model found that after controlling for demographic variables and mental health, emotional abuse ( OR =2.07) and emotional neglect ( OR =2.03) showed higher correlations with suicidal ideation than the other three types of childhood abuse( OR physical neglect =1.19, OR physical abuse =1.60, OR sexual abuse =1.37, P <0.05); and sexual abuse ( OR =2.29) and physical neglect ( OR =1.87) showed higher associations with suicide attempt than the other three types of abuse( OR emotional abuse =1.63, OR emotional neglect =1.59, OR physical abuse =1.50, P <0.01).
Conclusion
All five types of child abuse were independent risk factors for suicidal ideation and suicide attempts, and emotional neglect and emotional abuse had a greater effect on suicidal ideation, sexual abuse and physical neglect had a greater effect on suicide attempts.
9.Clinical and imaging features of idiopathic intracranial hypertension.

OBJECTIVES:
Idiopathic intracranial hypertension (IIH) is a syndrome that excludes secondary causes such as intracranial space-occupying lesion, hydrocephalus, cerebrovascular disease, and hypoxic ischemic encephalopathy. If not be treated promptly and effectively, IIH can cause severe, permanent vision disability and intractable, disabling headache. This study aims to explore the clinical and image features for IIH, to help clinicians to understand this disease, increase the diagnose rate, and improve the outcomes of patients.
METHODS:
We retrospectively analyzed 15 cases of IIH that were admitted to Xiangya Hospital, Central South University, during January 2015 to September 2020. The diagnosis of IIH was based on the updated modified Dandy criteria. We analyzed clinical data of patients and did statistical analysis, including age, gender, height, weight, medical history, physical examination, auxiliary examination, treatment and outcome.
RESULTS:
There were 10 females and 5 males. Female patients were 22 to 42 years old with median age of 39.5. Male patients were 27 to 52 years old with the median age of 44.0. The BMI was 24.14-34.17 (28.71±2.97) kg/m
CONCLUSIONS
IIH primarily affects women of childbearing age who are overweight. The major hazard of IIH is the severe and permanent visual loss. Typical image signs have high specificity in IIH diagnosis. Prompt diagnosis and effective treatment are significantly important to improve the outcomes of patients.

10.A preliminary study on age⁃related changes in the crown of the first maxillary molar in children

Objective :
To measure the anatomical parameters of the first maxillary molars in children of different age groups and evaluate the age⁃related changes in dental crowns.
Methods :
A retrospective analysis was conducted on cone beam computed tomography (CBCT) images of 4⁃8 ⁃year⁃old children. NNT software was used to analyze multiple important indicators of maxillary first molar.
Results :
A total of 308 first maxillary molars , including 154 pediatric patients , were evaluated in this study. The thickness of the pulp apex H1 (left , P = 0. 01 ; right , P = 0. 02) and the thickness of the pulp chamber floor H3 (left and right P < 0. 01) were positively correlated with age , while the height of the pulp cavity H2 (left and right P < 0. 01) and the height of the palate tip D1 (left P = 0. 003 , right P = 0. 002) showed a negative correlation with age. There was no significant correlation between the height of the buccal tip and age (P > 0. 05) . There were significant differences in H1 and H3 between the 4⁃year⁃old and 5 ⁃year⁃old age groups between the 8 ⁃year⁃old age group ( P < 0. 05) , as well as significant differences in H2 and D1 between the 4⁃year⁃old and 5 ⁃year⁃old between the 6⁃year⁃old , 7 ⁃year⁃old and 8 ⁃year⁃old age groups (P < 0. 05) .
Conclusion
The age⁃related changes in the crowns of the first maxillary molars are important references for the clinical treatment , and can be accurately measured through CBCT data.
