1.Comparative study of negative emotion and behavioral problems between left-behind and non-left-behind children in rural areas during hospitalization

ObjectiveTo discuss the incidence of negative emotion and behavioral problems between left-behind and non-left-behind children in rural areas during hospitalization. Methods175 participants included 90 cases of non-left-behind children patients and 85 left-behind children patients.The negative emotion and behavioral problems of all children were recorded and compared. ResultsCompared with the non-left-behind children patients,left-behind children manifested negative emotions such as anger,anxiety,depression and fear,more probability and incidence of behavioral problems such as dysphonia,special learning disabilities,sleep disorders,twitch disease,multiple tourtte's syndrome,attention deficit disorder,enuresis sickness and so on.There was significant differences between the two groups. ConclusionsCompared with the non-left-behind children,left-behind children patients are more likely to have negative emotion and behavioral prob-lems,it may have lasting influence on children and eventually influence the development of the physical and mental health,cause learning and adaptation difficulties.So more care and attention should be given to left-behind children,and given timely discovery and timely treatment of negative emotions and behavioral problems.

3.Naringin reduced polymethylmethacrylate-induced osteolysis in the mouse air sacs model.

OBJECTIVETo evaluate the influence of naringin on PMMA-induced osteoclastic bone resorption using the mouse air sacs model.
METHODSTotal 48 female Balb/c mices with the age of 8 to 10 weeks were chosen in the study. Air were injected into the back in 32 mices and formed the air sacs, 6 d later, the skulls (originated from other 16 mices) were implanted to the air sacs. Thirty-two animals were divided into naringin treatment group (with 2 concentrations of 150 mg/kg and 30 mg/ kg) , DMSO group and PBS blank group, 8 animals in each group. Polymethylmethacrylate (PMMA) particles were injected into the air sacs in naringin treatment groups and DMSO group so as to irritate inflammatory reaction. Naringin with 2 concentrations of 150 mg/kg and 30 mg/kg were dissolved in DMSO of 0.2 ml, and were injected into air sacs, respectively. In PBS black group, no stimulation with PMMA particles, only injected PBS, and in DMSO group, injected DMSO without naringin. Tartrate resistant acid phosphatase (TRAP), Ca2+ release, modified Masson stain and histological analysis were performed on the 7th day after stimulation.
RESULTSCompared with DMSO group, naringin treatment group's cellular infiltration decreased (P < 0.01); concentration of 150 mg/kg was better than that of concentrations of 30 mg/kg (8.90 ± 1.75 vs 15.23 ± 1.86). Naringin can decrease calcium release in the lavage of the air sacs bone resorption model, especially obvious in naringin with concentration of 150 mg/kg. Naringin can ameliorate the inflammatory reaction and the subsequent bone resorption (including bone collagen loss, TRAP positive cells amount and so on) in air sacs with bone implant and PMMA particles. Naringin with concentration of 150 mg/kg appeared to be an optimal dosage to deliver the therapeutic effects.
CONCLUSIONNaringin inhibits PMMA-induced osteoclastogenesis and ameliorates the PMMA-associated inflammatory reaction and the subsequent bone resorption.

5.Content Determination of Asperosaponin Ⅵ in Yulin Mixture by HPLC

OBJECTIVE:To establish HPLC method for the content determination of asperosaponin Ⅵ in Yulin mixture.ME-THODS:The separation was performed on Zorbax EcLipse XDB-C18(150 mm?4.6 mm,5 ?m) column with mobile phase consisted of methanol-0.01 mol?L-1 hydrochloric acid (69:31) and flow rate of 1 mL?min-1.The detection wavelength was set at 212 nm and column temperature was 20 ℃.RESULTS:The linear range of asperosaponin Ⅵ was 0.022~0.23 mg?mL-1(r=0.998 9) with an average recovery of 101.33% (RSD=2.35%,n=9).CONCLUSION:The method is convenient,simple,accurate and reproducibility for the quality control of Yulin mixture.
6.The pleural membrane biopsy accumulates the fluid patient's diagnosis value to the chest cavity

Objective To appraise the diagnosis value of pleural membrane biopsy on pleural effussion.Meth- ods 218 patients with pleural effusion who were had pleual menbrane biopsy were retrospectively analyzed.Among them 165 cases were done only once,42 cases were done twice,11 cases were done three times.They were done 271 times of biopsies.Results The pleural membrane biopsy always masculine gender rate was 72.0%,1 pleural mem- brane biopsy succeeds the masculine gender rate was 59.6%,after the masculine gender rate which 2 pleural mem- branes biopsies succeeds was 69.7%.The illness complication formation rate was 8.7%.Conclusion Pleural mem- brane biopsy is a simply,safe and higher diagnosing-rate method.The different spot pleural membrane biopsy may cnhance diagnosis rate.
7.Clinical application of E-tracking technique in T2DM patients common femoral artery elastidties

Objective To study the elastic features of common femoral artery by echo-tracking(ET)technique in type 2 diabetic(T2DM)patients.Method 60 eases of T2DM and 60 cases in control group,were separated into groups with age,ET technique was used to evaluate the common femoral artery modulus,including pressure strain elastic modulus(Ep),stiffness parameter(β),arterial compliance (AC),augment index(AI).Results Compared with the normal group ,in the T2DM group,β and Ep of common femoral artery were significantly higher(P<0.01,P<0.05),and AC was significantly lower(P<0.05),and the difference in AI was not obvious (P>0.05).The indexes were most obviouse in 40 to 59-year-old.The correlation parameter of age with β,Ep,AC,AI was 0.74,0.76 (P<0.001)and 0.62,0.51 (P>0.05).Conclusion The elasticity declination of common femoral artery in T2DM patients occur in the early stages,the change in 40-59 years old is obvious.β,Ep are apparently age-related indexes,and AC,AI are not.
8.Clone,expression and identification of human recombinant IL-4

Objective Construction of human IL‐4 recombinant expression vector and then conduct the expression ,purification and identification of human recombinant IL‐4 .Methods the open reading frame of IL‐4 was amplified by nest PCR with total RNA from PBMC of healthy volunteer .And then the amplified IL‐4 was inserted into pET101/D‐TOPO ,transformed into BL21 ,ex‐pressed ,purified and indentified .Results The size of amplified open reading frame of IL‐4 was about 460 bp and the sequence was correct .After transformed into BL21 ,the IL‐4 clone with higher expression level was selected by selection of different clones insert‐ed with IL‐4 and the size of expressed ,purified IL‐4 was about 28 × 103 .Western blot results showed that the size of single band was identical with the expected protein .Conclusion Human IL‐4 recombinant protein was got successfully .
10.Eosin Y-water test for sperm function examination.

Based on the principles of the in vitro staining technique, hypotonic swelling test, and water test, the Eosin Y-water test method was developed to simultaneously detect the integrity of the sperm head and tail and sperm membrane structure and function. As a widely used method in clinical laboratories in China, the Eosin Y-water test is methodologically characterized by three advantages. Firstly, both the sperm head and tail can be detected at the same time, which allows easy and comprehensive assessment of membrane damage in different parts of sperm. Secondly, distilled water is used instead of the usual formula solution to simplify and standardize the test by eliminating any potential effects on the water molecules through the sperm membrane due to different osmotic pressure or different sugar proportions and electrolyte solutions. Thirdly, the test takes less time and thus can be repeated before and after treatment. This article focuses on the fundamental principles and modification of the Eosin Y-water test and its application in sperm function examination and routine semen analysis for male infertility, assessment of the quality of sperm retrieved by testicular fine needle aspiration, semen cryopreservation program development, and evaluation of sperm membrane integrity after microwave radiation.
