1.Protective effects of polysaccharides from Hyriopsis cumingii on hepatocytes injured by CCl_4

Objective To study the protective effect on CCl4-injured L-02 hepatocytes of polysaccharides from Hyriopsis cumingii(HCP) and try to explain the mechanism.Methods The L-02 hepatocytes were incubated and then injured by CCl4.The levels of AST,ALT,MDA and SOD in cultural supernatant were detected by general methods.Cell viability was assayed by MTT method.Results The polysaccharides(25,250 and 1000?g?L-1)could reduce the levels of AST,ALT,MDA in cultural supernatant which increased by CCl4.It also could boost the hepatocytes viability and elevate the level of SOD which reduced by CCl4.Conclusion The results suggest that polysaccharides from Hyriopsis cumingii have direct protective activity on hepatocytes injured in vitro.It might be associated with the anti-oxidative activity of polysaccharides from Hyriopsis cumingii.
2.Determination of major nucleosides in Hyriopsis cumingii L. by HPLC

Objective A HPLC method was established to measure the contents of major nucleosides in Hyriopsis cumingii.Method Separation was performed on a Lichrosper-C_(18) column, pure water was used as mobile phase,the flow rate was 1.0mL min~(-1),the column temperature at 30℃and the detective walvelength at 254 nm.Results The contents of uracil,hypoxanthine, xanthine,uridine and thymine were 153.2~173.3?g?g~(-1),106.6~360.8?g?g~(-1),117.6~172.8?g?g~(-1),70.3~153.1?g?g~(-1) and 343.9~511.4?g?g~(-1),respectively. Conclusion This method showed good repeatability and flexibility.It could be used as a conventional method for determination of major nucleosides in H.cumingii.
3.Quantification of $\alpha$-Methylene-$\gamma$-butyrolactone Extracted from Different Parts of \it{Alstroemeria wilhelmina} and Evaluation of it's Antigenicity Using the Guinea-Pig Maximization Test

To detect the type of contact dermatitis caused due to the handling of Alstroemeria wilhelmina, 1% α-methylene-γ-butyrolactone (α-MBL) dissolved in physiological saline and a five-fold diluted saline solution of original extracts of flowers, leaves and stems of the flower were applied to guinea-pigs for induction treatment, and 0.1% α-MBL physiological saline and a ten-fold diluted solution of the original extracts were applied to the animals as the challenge treatment in compliance with the guinea-pig maximization test (GMT). As a consequence, not only primary irritant dermatitis was observed, but also delayed type allergic contact dermatitis due to Alstroemeria wilhelmina was observed. α-MBL determined in the extracts using high-performance liquid chromatography (HPLC) was found to be the biochemical material cause of the contact dermatitis. The flower region contained α-MBL in the highest concentrations compared with those of the leaves and stems. Therefore, the quantification of α-MBL in the extracts was concluded as being a useful evaluating method for contact dermatitis due to the handling of Alstroemeria.

4.Determinants of Life satisfaction among Japanese Agricultural Workers

The purpose of this study was to investigate the factorial structure of life satisfaction in agricultural workers. One hundred and sixty−five (87 males and 78 females) agricultural workers mainly engaged in plastic greenhouse cultivation in southern Japan, ranging in age from 19 to 72 years (mean = 44.3 years), completed a questionnaire. Scores of life satisfaction showed a higher tendency in males than in females, and those in elders were higher than in younger people. Conversely, females showed higher stress scores than males, and younger people’s score were higher than the elders. Multiple regression analysis was performed to analyze the factorial structure of satisfaction. It is suggested that the level of life satisfaction is positively related to social support, and is negatively related to the impact of stressful life events, globally perceived stress and subjective symptoms of ill health. Stress levels depended on factors concerning the psychological demands of work, work posture, work hours and work environment. Different factorial structures were observed between female and male workers. Findings from this study suggest that a strong social support network may assist in enhancing life satisfaction. Furthermore, the ill health and stress experienced in agricultural work were associated with a reduced life satisfaction.

5.Community-based Analysis of the Factorial Structures of the Recent Increase in Low Birthweight Infants

This study was conducted to determine whether the regional factors were related to the increase in the percentage of low birthweight(LBW: <2, 500g) infants in Kumamoto Pref., and to establish a tentative structure model for predicting low birthweight infants. Analyses for frequency of LBW infants between 1974 and 1997, and a multiple regression model and covariance structure model were conducted using data from the vital statistics between 1992 and 1997 and regional indicators concerned with LBW infants from official registered statistical data between 1992 and 1997. The 72 regional factors were clustered into four groups linked with agricultural areas such as Urban, Flat, Hilly and Mountainous areas. The recent increase in the incidence of LBW infants resulted from the increase in moderate−LBW(MLBW: 2, 000−2, 500g) infants of full term−LBW infants. There was a steady annual increase in the Urban agricultural area LBW infants since 1992. The two structure analyses revealed that the Urban area had a marked effect on the increase in LBW infants, whereas, farm villages in Hilly or Moutainous areas had less effect on the increase in LBW infants. These findings suggest that the regional factors relating to the mothers’ life−style or regional environments play a key role in the etiology and prevention of LBW, and will be a useful in the analyses using official registered material.

6.Dynamic Changes of Phenotypes and Secretory Functions during the Differentiation of Pre-DCs to Mature DCs

The dynamic expression of cytokines and phenotypes during the differentiation process of dendritic cell precursors (pre-DCs) to mature dendritic cells (DCs) was investigated,and the effects of inflammatory stimulation with lipopolysaccharide (LPS) on DCs differentiation were understood.The differentiation of bone marrow cells isolated from Balb/c mice was induced to DCs in an 8-day cell culture system with RPMI-1640 complete culture medium containing 10% FBS,20 ng/mL recombinant mouse granulocyte-macrophage colony-stimulating factor (rmGM-CSF) and 10 ng/mL recombinant mouse interleukin-4 (rmIL-4).On the day 3,6 and 7 after culture,DCs were divided into non-LPS group and LPS group,given 500 ng/mL LPS for 24 h stimulation and no stimulation respectively.The expression levels of CD 1 l c+,MHC-Ⅱ +,CD40+,CD80+ and CD86+ were detected by flow cytometry,and those of IL-2,IL-4,IL-10,IL-12 p70 and IFN-γ in the supernatant by ELISA.On the day 3 and 6 after culture,the expression of IL-2,IL-4,IL-10 and IFN-γ in DCs showed no significant differences between non-LPS group and LPS group,whereas the differences were significant at day 7.The expression levels of cytokines (for IL-2,IL-4,IL-10,IFN-γ and IL-12 p70:152.86±6.91,778.33±8.35,44.55±2.54,5.8.26±1.09 and 2423.00±57.21 pg/mL respectively) in LPS group were higher than those in non-LPS group,especially IL-12 p70 increased obviously at day 7.It was concluded that during the differentiation process of pre-DCs to mature DCs,LPS stimulates DCs producing large amounts of IL-12 p70 and Thl-type cytokines as compared with Th2-type cytokines,and day 7 may be a key time-point for DCs polarization.
7.Studies on the chemical constituents in root of Rheum rhizastachyum.

OBJECTIVETo investigate the chemical constituents from the root and rhizome of Rheum rhizastachyum.
METHODThe compounds were isolated by column chromatography on silica gel, Sephadex LH-20, MCI-gel CHP-20P separately and their structures were elucidated by chemical and spectral technology.
RESULTSix compounds were isolated and identified as chrysophanol, emodin, gallic acid, sucrose, 1-O-beta-D-glucopyranosyl-chrysophanols, 1-O-beta-D-glucopyranosyl-emodin.
CONCLUSIONAll the compounds were obtained from this plant for the first time.

8.Effects of different modes of intermittent hypobaric hypoxia on ischemia/reperfusion injury in developing rat hearts.

The aim of the present study was to explore the effects of two different modes of intermittent hypobaric hypoxia (IHH) on myocardial ischemia/reperfusion injury in developing rat hearts. Postnatal male sprague-Dawley rats (n=72) were divided randomly into 3 groups: intermittent hypoxia at 3 000 m (IHH3000) group, intermittent hypoxia at 5 000 m (IHH5000) group and control group. The isolated hearts were perfused in the Langendorff apparatus, undergoing 30 min of global ischemia and 60 min of reperfusion. Cardiac function, coronary flow and lactate dehydrogenase (LDH) activity were recorded at 5 min before ischemia and 1, 5, 10, 20, 30, 60 min during reperfusion, respectively. The heart weight was measured at the end of the experiment. The results showed that: (1) There was no difference in body weight gaining between IHH3000 and control groups. The gain of body weight in IHH5000 group was much lower than that in IHH3000 and control groups (P<0.01). (2) Compared with that in the control group, the recovery of cardiac function in IHH3000 group was enhanced at 60 min after ischemia/reperfusion, coronary flow was increased, and LDH activity was decreased (P<0.05), meaning a cardioprotective effect occurred. There was no significant difference in heart weight between IHH3000 and control groups. In addition, cardiac function restored better in IHH3000 group after 42 d of hypoxic exposure than that after 28 d of hypoxic exposure (P<0.05). (3) Compared with that in the control group, the recovery of cardiac function in IHH5000 group was lower, coronary flow was decreased, and LDH activity was increased (P<0.05). There was a hypertrophy in the right ventricle in IHH5000 group. All changes indicated definitely that a detrimental effect developed in IHH5000 group. The results suggest that proper IHH can protect developing rat hearts against ischemia/reperfusion injury while this effect could be affected by the modes of intermittent hypoxic exposure.
