4.Regulation of Jagged1 expression by its receptor Notch1 in prostate cancer PC3 cells

Objective To investigate the effect of Notch1 and Jagged1 on the growth of prostate cancer PC3 cells and the regulation of Jagged1 expression by its receptor Notch1 in PC3. Methods Notch1 and Jagged1 expression were inhibited by siRNA interference. The growth rate of PC3 cells was measured by MTT. Notch1 expression was inhibited by siRNA interference and promoted by transfection of Notch1 cDNA. The changes of Notch1 and Jagged1 protein and mRNA levels were measured by Western blot and Real-time RCR. Results The growth of PC3 cells decreased after inhibiting Notch1 and Jagged1 expression. Jagged1 protein level decreased after inhibiting Notch1 expression and increased after over-expressing Notch1. Moreover, the change of Jagged1 protein level was discordant with the situation of mRNA. Conclusion In PC3 cells, Notch1 and Jagged1 are important to cell growth. Notch1 may regulate Jagged1 expression.
6. Active components in six kinds of ground bamboo leaves and their anti-oxidant activities

Objective: To study the effective components of the leaves in six kinds of ground bamboo (Shibataea chinensis, Pleioblastus kongosanensis f. aureostriatus, Indocalamus latifolius, Sasa pygmaea, S. argenteastriatu, and S. veitchii) and their anti-oxidant activities. Methods: The contents of total phenolics, total flavonoids, and triterpenoids in bamboo leaves and their anti-oxidant activities were detected and investigated by using Forint reduction assay reagent, aluminum nitrate-sodium nitrite colorimetry, and vanillin-acetic acid assay, respectively. Eight characteristic compounds (orientin, isoorientin, vitexin, homovitexin, p-coumaric acid, chlorogenic acid, caffeic acid, and ferulic acid) were quantitatively analyzed by UPLC synchronously. The anti-oxidative and reducing activities of extracts from bamboo leaves were investigated by DPPH and FRAP. Results: Six kinds of ground bamboo leaves contained abundant botanic secondary metabolites. The content of total phenolics, on dry basis of leaves, varied in a range of 4.15%-9.12% in the six kinds of ground bamboo, total flavonoids and triterpenoids located in 1.62%-4.00% and 0.44%-0.57%, respectively. The sequence of the eight characteristic compounds in total phenolics was S. veitchii 6.81% > S. argenteastriatus 4.80% > S. pygmaea 4.35%, P. kongosanensis 4.35% > S. chinensis 1.54% > I. latifolius 1.08%. The content of eight characteristic compounds and their anti-oxidant activities in each ground bamboo leaves were significantly changed (P < 0.05). Conclusion: Among the six kinds of ground bamboo, there are much more effective components and more outstanding anti-oxidant activities of radical scavenging and iron reduction in two species (S. chinensis and P. kongosanensis) than those in any other banboos.
7.Study of the extract of averrhoacarambola L.root on renal function in diabetic mice and its anti-oxidative action

Aim To investigate the effect of the extract of Averrhoacarambola L.root (EACR)on renal func-tion in diabetic mice and its anti-oxidative action. Methods Diabetic mice were established by tail vein injection with 120 mg·kg-1 streptozotocin (STZ)and were divided into 5 groups:model control group,val-sartan control group,and low-,middle-,high-dose of EACR groups (300,600,1 200 mg·kg-1 ).And 10 normal mice consisted of normal control group.The fasting blood glucose (FBG)of mice was detected be-fore and after administration of drugs.After last admin-istration,the blood and urine samples were collected for creatinine (Cr),urea nitrogen (BUN),urine and 24 h urinary protein determination.The activities of superoxide dismutase (SOD ),glutathione peroxidase (GSH-Px)and malonaldehyde (MDA)content were determined using kits.HE staining was conducted to observe the pathological changes of kidney tissues. ELISA method was utilized to detect the contents of catalase (CAT)and reactive oxygen species (ROS ). The expressions of Cyto-C,AIF and caspase-3 proteins in kidney tissues were analyzed by Western blot.Re-sults Compared with model group,the serum bio-chemical indexes and 24 h urinary protein of valsartan and moderate-,high-dose of EACR groups were de-creased with statistical significance (P<0.05 ).After the treatment, the MDA content was decreased by EACR treatment,and SOD,GSH-Px and CAT activities were enhanced.Meanwhile the expressions of ROS, Cyto-C,AIF and caspase-3 were down-regulated.The pathological changes of kidney tissues were ameliorated by EACR through HE results.Conclusions The ex-tract of Averrhoacarambola L.root can decrease the se-rum levels of Cr and BUN,reduce the MDA and ROS contents in kidney tissue and enhance the activities of SOD,GSH-Px and CAT,down-regulate the expres-sions of Cyto-C,AIF and caspase-3 proteins in kidney tissues,elevate the anti-oxidative effect of kidney. Therefore,the renal function of diabetic mice is melio-rated.
8.Clinical study of monitoring of transcranial Doppler ultrasonography to seriously illed children with cerebral dysfunction

Objective To investigate the application value of transcranial Doppler ultrasonography(TCD) in critically ill children with cerebral dysfunction.Methods We detected the cerebral blood flow parameters[systolic velocity(Vs),mean velocity(Vm),end of diastolic velocity(Vd),pulsatility index(PI),resistance index(RI)] of middle cerebral artery in cerebral dysfunction group(n=20) and control group(no cerebral dysfunction,n=20),and observed the differences of those parameters between two groups.Children in cerebral dysfunction group were also divided into groups according to Glasgow Coma Score(GCS) and prognosis.The differences of each TCD parameter in diverse groups divided by GCS and by prognosis were analysed.Children in cerebral dysfunction group were checked TCD everyday until TCD parameters were normal.The relation between the days of TCD parameters′ reaching normal and GCS,duration of disorder of consciousness with correlation analysis were analysed.Results Vs,PI,RI in cerebral dysfunction group were higher than those in control group,while Vd was lower (P<0.05).(2)PI in the group of GCS less than 6 points(0.91±0.21) was higher than that in group of GCS 7-13 points(0.83±0.14)(P<0.05).The other parameters between two groups of GCS less than 6 points and GCS 7-13 points were not significantly indistinctive.Between the good prognosis group and the bad prognosis group,there were no significant differences in all parameters.(3) The days of TCD parameters′ reaching normal were negatively correlated with GCS(r=-0.653,P<0.01),but positively correlated with the duration of disorder of consciousness(r=0.923,P<0.01).Conclusion The cerebral hemodynamics of children with cerebral dysfunction is abnormal,the more serious cerebral dysfunction,the higher PI and the longer time for regaining normal TCD parameters.Constantly monitoring TCD could reflect the changes of cerebral hemodynamics and is valuable to assess the state of illness and prognosis.
9.Evaluation of genome degradation of plasma hepatitis C virus during methylene blue photochemistry treatment

Objective To study the degradation of hepatitis C virus(HCV) genome before and after methylene blue photochemistry(MB-P) treatment of the plasma.Methods MB was added to HCV positive plasma to a final concentration of 1.0?mol/L.The plasma was then exposed to 30000 Lux fluorescence.Plasma samples were then collected at different times of exposure.Real-time PCR was used to quantitatively study the course of the HCV-RNA degradation.The whole genome was screened for integrity by RT-PCR with 8 pairs of specific primers which targeted sequential overlapping sections of HCV genome.Results Results of real-time PCR showed that the copy number of HCV-RNA was continuously decreasing during MB-P treatment.RT-PCR results showed that the reactivity of different sections of the HCV genome to MB-P was significantly different and indicated that the 5' end and 3' end were more stable. Conclusion MB-P treatment could degrade HCV RNA with various sensitivities to different sections of the genome.RNA degradation may play an important role in plasma virus inactivation.Detection of HCV genomic RNA might be clinically useful to monitor the process and efficiency of virus inactivation.
10.Exploration of the teaching model of presentation-assimilation-discussion class in the course of medical physics

There are some problems in the teaching of medical physics course,such as low learning initiative,deficient grasp of concept,and obstacles to realize the internalization and practical application of knowledge.The PAD (presentation-assimilation-discussion) class is applied in the course,in order to improve the students' learning state.PAD class is a teaching mode proposed from a psychological point,which advocates that teaching is the first before students start to learn and has an equal treatment on students' discussions and teachers' instruction,supporting independent learning.The teaching measures include:selecting course content to reach the balance between simplification and detailed interpretation;assigning open homework based on the course;dividing the class discussion into four steps;assessing students based on the performances of homework and final examinations.The research has shown that the PAD class facilitates improving the students' initiative of study and promoting the absorption and application of knowledge.
