Objective To investigate the immunomodulatory effects of heme oxygenase-1 ( HO-1) modified rat bone marrow mesenchymal stem cells ( BM-MSCs) on lymphocytes .Methods Rat BM-MSCs were cultured and identified in vitro.HO-1 gene was transduced into BM-MSCs via recombinant adenovirus (rAdV) to construct HO-1/BM MSCs, which were then co-cultured with rat spleen lymphocytes .The cellu-lar activity of lymphocytes was detected by using MTT method .The cell cycle distributions were analyzed by propidium iodide staining and flow cytometry .ELISA was used to detect the level of cytokines in the super-natant of cultured cells .The expression of lymphocyte activation markers , CD25 and CD71, were measured by using fluorescence labeled antibodies and flow cytometry .Results Compared with the control group , the lymphocytes co-cultured with HO-1/BM MSCs showed a suppressed proliferation and cell cycle progression . HO-1/BM MSCs inhibited the secretion of IL-2, TNF-αand IFN-γ, but enhanced the secretion of IL-10 in lymphocytes.Furthermore, the expression of CD25 and CD71 were also down-regulated by HO-1/BM MSCs. Conclusion Compared with BM MSCs , HO-1/BM MSCs show higher immunosuppressive effects on lym-phocytes .

Objective To establish a new flow-injection chemiluminescence(CL) method for the determination of hydrocortisone. Methods In H2SO4 solution, hydrocortisone could obviously enhance the chemiluminescence intensity of the reaction of KMnO4-Na2SO3 system.Based on this,a new flow-injection CL method for the determination of hydrocortisone was developed. Results There was a good linear relationship between CL intensity and the concentration of hydrocortisone in the range of 1.0?10-9-1.0?10-6g/mL.The detection limit was 4.0?10-10g/mL at a signal-to-noise ratio of 3∶1.The RSD of 11 assays was 2.2%.Conclusion This method can be successfully used to determine the quantity of hydrocortisone in injection. It is rapid, accurate, simple, and has high sensitivity and wide linear range.

Chinese Journal of Endocrinology and Metabolism 2010;26(9):758-761
A cross-sectional study was conducted in Heilongjiang in a representative sample of 2 875Chinese adults aged from 20 to 74 years. Questionnaire was conducted. Their height, weight, waist circumference, blood pressure, blood glucose, serum insulin, lipids, and other indexes were determined. The criterion of International Diabetes Federation was applied for metabolic syndrome (MS) diagnosis. The results showed that the preliminary prevalence rate of MS was 19.34% and the age-adjusted prevalence rate of MS was 21.92% ( male 20.41% , female 23.11% ). The prevalence rate of MS increased with the age. The prevalence rate in the urban area was higher than that in the town and rural area ( both P < 0.05 ). The result of multiple factor logistic regression analysis showed that body fat content, insulin resistance, age, residential difference, drinking habit, daily staple food quantity, urine albumin, education, family history of obesity, and gender were related with MS.

BACKGROUND:Early detection and accurate staging diagnosis of heart failure are the basis of good clinical therapy efficacy. Due to lack of simple and effective staging model for the diagnosis of heart failure, it is difficult to diagnose heart failure in clinics, leading to poor control of heart failure. OBJECTIVE:To establish the disease staging model based on Adaboost and SVM for heart failure, and improve the accuracy of diagnosis and staging of heart failure. METHODS:A total of 194 cases were roled into this study, including heart failure patients and healthy physical examination persons. According to the stage standards formulated by American Colege of Cardiology and American Heart Association, specific clinical feature parameters closely related to heart failure were colected and selected. Based on clinical diagnosis results and using Adaboost model and SVM model, we trained the models for heart failure diagnosis and staging, thus obtaining diagnosis model. RESULTS AND CONCLUSION: The parameters included stroke volume, cardiac output, left ventricular ejection fraction, left atrial diameter, left ventricular internal diameter at end-systole, N-terminal pro-brain natriuretic peptide and heart rate variability. As for the Adaboost model, its sensitivity and specificity was 100% and 94.4%, respectively. At the same time the SVM model had good sensitivity and specificity, 86.5% and 89.4% respectively. Adaboost classification model can be accurate in the diagnosis of heart failure symptoms, the accuracy reached 89.36%. On the basis of the diagnosis of heart failure, the SVM classification model is effective in staging the severity of heart failure, staging accuracy for staging B and C was 86.49% and 81.48%, respectively. The findings indicate that, combining Adaboost and SVM machine learning models could provide an accurate diagnosis and staging model for heart failure.

Objective:To study the surface structure and antibacterial property of Ag-containing coating(AgCC)on casting cobalt chromium alloy(Co-Cr).Methods:Plasma spraying technique was adopted to prepare AgCC on Co-Cr.Scanning electron microsco-py,energy dispersive analysis and X-ray diffraction analysis were used to evaluate the surface properties. The antibacterial effects of AgCC against staphylococcus aureus and candida albicans were examined by film attachment method.Fluorescence microscope was a-dopted to evaluate the bacteria adhesion property. Results:AgCC surface was uniform and compact, combined perfectly with sub-strate material. The content of the surface was mainly Ag,Cr and a small amount of Ag2 O and Cr2 O3 .The results of antibacterial as-say showed that on AgCC the number of bacterial colony was fewer(P<0.05)and bacterial adhesion was less than those on Co-Cr(P<0.05).Conclusion:AgCC surface was stable and has antibacterial property.
