1.MicroRNA and cerebral ischemia.

MicroRNAs (miRNAs) are a class of highly conserved single-stranded RNA molecules that modulate gene translation. By targeting the mRNA of protein- coding genes, miRNAs play a critical role in neuronal differentiation, cell proliferation, apoptosis and metabolism. There are a lot of miRNAs in central nervous system, which are not only closely linked to development, differentiation and function of nerve cells, also play an important role in nerve lesions and dysfunction after cerebral ischemia. Specific miRNA through their own changes affect their target gene expression levels after focal cerebral ischemia, involving in the protection against apoptosis in neurons and regeneration after cerebral ischemia. A full understanding of the specific microRNA function and its underlying mechanism in the brain can provide a new strategy for the prevention, diagnosis, and treatment of ischemic cerebral injury at gene level.

2.The historical responsibility of Chinese medical ethics in the process of promoting a community with a shared future for mankind

Reflecting on the historical responsibility of Chinese medical ethics at present, it is necessary to place oneself in the grand context of promoting the process of building a community with a shared future for mankind. Safety, development, and civilization constitute the three important ethical dimensions of a community with a shared future for mankind, which are not only ethical orientations, but also ethical appeals. These provide an important direction for Chinese medical ethics to deeply consider its role in the process of a community with a shared future for mankind. The historical responsibilities of Chinese medical ethics in the process of promoting a community with a shared future for mankind mainly include: confronting moral and interest issues, adhering to the patient-centered approach, and ensuring patients’ life safety; playing the role of ethical regulation, achieving the practical effects of medical ethics, and maintaining social security and stability; implementing the responsibility of medical ethics education, shaping medical professionalism, and promoting the development of healthy China; inheriting the excellent traditional culture, pursuing medical excellence and justice, and promoting the development of social morality; pursuing common health values, building a community with a shared future between doctors and patients, and promoting the progress of human. A global community of health for all provides the most direct practical platform for Chinese medical ethics to fulfill its historical responsibility in the process of building a community with a shared future for mankind. It directly promotes Chinese medical ethics to the world, and Chinese ethics facing the world will play an increasingly important influence in self-renewal.
3. Influence on the distribution of rehydrated particle sizes of freeze-dried docetaxel liposome

OBJECTIVE: To investigate factors that affected the distribution of rehydrated particle size of freeze-dried docetaxel liposome and develop a technique of producing docetaxel liposome that can meet clinical requirements. METHODS: The single factor method was utilized in this study. Docetaxel liposome were prepared using different types of negatively charged phospholipids, or lecithins with varying phosphatidylcholine (PC) content, or various cryoprotectants at different gradient concentrations, followed by freeze-drying with different protocols. The particle sizes of corresponding rehydrated products were compared and the effects of different factors were analyzed. RESULTS: The optimal liposomal membrane was composed of dipalmitoyl phosphatidic acid (DPPA), 20% trehalose and lecithin containing 92% PC. The best freeze-drying protocol included following steps; flash freezing, 19 h of sublimation-drying and 9 h of desorption-drying. CONCLUSION: The parameters, including negatively charged phospholipids, cryoprotectants, PC content of lecithin and lyophilization protocols, had various effects on the particle sizes of reconstituted docetaxel liposome.
4.Experimental study on the glucagon-likepeptide 1 receptor agonist modulation of insulin resistance and hepatic oxidative stress in rats with diabetes mellitus combined with nonalcoholic fatty liver disease

To study the effect of glucagon-likepeptide 1(GLP-1)receptor agonist on insulin resistance and hepatic oxidative stress in rats with diabetes mellitus combined with nonalcoholic fatty liver disease. 36 male SD rats were served as the experimental animal and randomly divided into control group, model group, and GLP-1 group. The rats of control group were given routine diet with intraperitoneal injection of normal saline, those in model group were given high fat diet and intraperitoneal injection of normal saline, while GLP-1 group rats were fed with high fat diet and intraperitoneal injection of liraglutide. After 4 weeks of treatment, insulin resistance, lipid metabolism, liver injury and oxidative stress were all assessed. Serum fasting blood glucose, fasting insulin, total cholesterol, triglyceride, alanine transaminase(ALT), aspartate transaminase(AST)levels and total cholesterol, triglyceride contents in liver tissue, and as well as homeostasis model assessment for insulin resistance(HOMA-IR)levels of model group were significantly higher than those of control group, complex insulin sensitivity index(ISIcomp)level was significantly lower than that of control group; serum fasting blood glucose, fasting insulin, total cholesterol, triglyceride, ALT, AST contents and HOMA-IR levels of GLP-1 group were significantly lower than those of model group, ISIcomp level was significantly higher than that of model group; superoxide dismutase(SOD), glutathione peroxidase(GSH-Px), catalase(CAT)contents in liver tissue of model group were significantly lower, while malondialdehyde content and SOD, GSH-Px, CAT, NF-E2 related factor-2(Nrf-2), antioxidant response element(ARE), heme oxygenase-1(HO-1), quinone oxidoreductase-1(NQO-1), glutathione thiol transferase(GST)mRNA expression were significantly higher than control group; SOD, GSH-Px, CAT contents and SOD, GSH-Px, CAT, Nrf-2, ARE, HO-1, NQO-1, GST mRNA expression in the liver tissue of GLP-1 group were significantly higher, while malondialdehyde content was significantly lower than that of model group. GLP-1 receptor agonist reduces insulin resistance and liver oxidative stress injury in diabetic rats with nonalcoholic liver disease.
5.Research progress on signaling molecules involved in articular cartilage repair.

After the articular cartilage injury, the metabolic level is increased during the progressive degeneration, the chondrocytes secrete a variety of inflammatory factors, and the original cell phenotype is gradually changed. For a long time, a large number of researchers have done a lot of researches to promote anabolism of chondrocytes and to maintain the stability of chondrocyte phenotype. There are many molecular signaling pathways involved in the process of promoting cartilage repair. This review focuses on the key signaling molecules in articular cartilage repair, such as transforming growth factor-beta and bone morphogenetic protein, and reveals their roles in the process of cartilage injury and repair, so that researchers in related fields can understand the molecular mechanism of cartilage injury and repair widely and deeply. Based on this, they may find promising targets and biological methods for the treatment of cartilage injury.

6.Definitive postponed surgery for the casualties with multiple injuries in the earthquake.

OBJECTIVETo explore the treatment of casualties who suffered from multiple injuries in earthquake and had been treated by damage control (DC) immediately,and the definite postponed surgery time by way of APACHE II score.
METHODSFrom May to June 2008, 21 casualties who had undergone DC accepted further treatment of medicine and surgery, and recovered physical potentiality in the intensive care unit (ICU). APACHE II score was adopted to evaluate the casualties' physical potentiality. Then, internal fixation of intra-articular fractures and centrum fractures, alternation from external fixation to internal fixation, reconstruction of soft tissue was applied by stages according to the plan.
RESULTSAll the patients were effectively cured after definite postponed surgery. There was less complication, low deformity ratio and no death happened.
CONCLUSIONTimely evaluation of the casualties' physical potentiality, appropriate surgery time and exact surgery can reduce the casualties' complication, deformity rate and death rate.

7.Advance in research on regulatory mechanism and functions of neutral sphingomyelinse 2.

Neutral sphingomyelinase 2 (nSMase2), which located mainly on the plasma membrane, hydrolyzes sphingomyelin into ceramide and plays an important role in the physiological and pathological regulation of cell apoptosis, cell growth arrest, and inflammation. nSMase2 is also involved in the development of Alzheimer's disease and the bone growth.Under neutral pH and the presence of Ca(2+), Mg(2+), and Mn(+), the activity of nSMase2 is induced by oxidative stress through phosphorylation. Furthermore, the induced interaction of anionic phospholipids and the signaling molecules like receptor for activated C-kinase 1/embryonic ectodermal development with nSMase2 are also crucial mechanisms of protein activation. In the review, recent research advances in the structure and function of nSMase2 and its underlying mechanisms are summarized.

8.Protective effect of H2S pretreatment on cerebral ischemia-reperfusion injury and its mechanisms in rats.

OBJECTIVETo investigate the protective effect of H2S pretreatment after cerebral schemia/reperfusion injury and its mechanisms in rats.
METHODSThe rat model of global cerebral ischemia/reperfusion injury was established by bilateral common carotid arteries occlusion combined with hemorrhagic hypotension.30 rats were randomly divided into four groups(1)sham group(n=5),in which rats received sham surgery only,with their bilateral vertebral artery and bilateral common carotid artery exposed but without ischemia treatment;(2)global cerebral ischemia/reperfusion model group(IR group,n=5),in which the global cerebral ischemia was induced by 10-min occlusion of bilateral common carotid arteries combined with hypotension;(3)H2S pretreatment group(n=15),in which H2S(12,24,48 Μmol/kg)was intraperitoneally injected before operation;(4)NaCl pretreatment group(n=5),in which the rats were intraperitoneally injected with saline 30 minutes before operation.The activities of superoxide dismutase(SOD)and the levels of malondialdeehyde(MDA)in brain were measured by spectrophotometry.Brain water content was detected.The expression of heat shock protein 70(HSP70) in the hippocampus was determined by Western blotting.
RESULTSThe SOD activities were significant increased in groups pretreated with 12Μmol/kg H2S(P=0.042),24Μmol/kg H2S(P=0.002),and 48Μmol/kg H2S(P=0.000),and the SOD activity was significantly lower in the ischemia group than in the Sham group(P=0.003).The MDA activities in the 24Μmol/kg group(P=0.026)and the 48Μmol/kg group(P=0.015)groups were significantly lower than in the IR group.The brain water content was decreased in H2S pretreatment group(24Μmol/kg and 48 Μmol/kg)compared with IR group(P=0.018,P=0.008),and it was also significantly higher in the IR group than in the sham group(P=0.009).The expression of HSP70 were decreased in H2S pretreatment group(24 Μmol/kg)compared with the IR group(P=0.000),and the expression of HSP70 were significantly higher in the IR group than in HSP70 group(P=0.000).The expression of HSP70 also significantly differed between NaCl group and HSP70 group(P=0.000).
CONCLUSIONH2S has protective effects on cerebral ischemia and reperfusion,which may be achieved by improving SOD activity,removing oxygen free radicals,inhibiting lipid peroxidation,and down-regulating the expression of HSP70 in the hippocampus.

9.Species diversity and host of Thesium chinense community.

OBJECTIVETo study species diversity and the host of Thesium chinense community.
METHODThe investigation on phytocoenology was carried out on the T. chinense community in 7 areas of Anhui Province and Jiangsu Province. The correlation between species diversity of community and T. chinense abundance was analyzed regarding to the abundance of species, the abundant index of species, species diversity index and community even index.
RESULT AND CONCLUSIONThe results showed that T. chinense liked light, warm climate, liked acidic to neuter soil. The majority of T. chinense distributed in the humid wasteland and herbaceous community. There was a positive correlation between the abundance of T. chinense and the unity numbers of community, and a negative correlation between the abundance of T. chinense and the abundance, coverage, diversity index, Pielou even index of community. T. chinense host species was various. This investigation found 28 host species belong to 11 Families, among them 5 Families and 18 species were found for the first time.

10.Quality evaluation of Schisandra chinensis at different producing areas in northeast China.

OBJECTIVETo investigate the quality of Schisandra chinensis, and thus provide the useful reference data for its quality standard establishment and standardized cultivation.
METHODBase on the Chinese Pharmacopoeia and literature relating to S. chinensis, the appearance, water-soluble extractive, total ash, acid-insoluble ash and volatile oil of 10 S. chinensis sampes collected from different producing sites in northeast China were studied or determined, and the contents of major active lingans were measured by HPLC.
RESULTThe content of total ash and acid-insoluble ash in S. chinensis was less than 7.0%, 1.0%, respectively. The contents of water-soluble extractive were higher than 45.0%. HPLC analyses showed that the content of schisandrol A was higher than 0.40%. All these results in accordance with the requirements of Chinese Pharmacopoeia were published in 2010.
CONCLUSIONThe differences of the appearance and the internal quality are different for the samples collected from different areas. And the establishment of a scientific and comprehensive quality standard system will be very important role in ensuring the quality of S. chinensis.
