1.The effects of Leptin in proliferation of MCF-7 breast cancer cells

Objective To investigate the effects of leptin on proliferation of the breast carcinoma MCF-7 cells in terms of protein and nRNA of Rab-25 and C-myc.Methods MCF-7 cells were treated with leptin and normal saline (NS) as control.The 490 nm OD values of cells were read by MTT and the protein and mRNA expressions of Rab-25 and C-myc were measured by western blot and RT-PCR methods in two groups.Results Compared with control group,leptin can promote the proliferation of MCF-7 cells in a dose and time dependent manner within certain limit and up-regulate the protein expressions of Rab-25 and C-myc in the breast carcinoma MCF-7 cells (P < 0.05).However,leptin also can up-regulate the mRNA expressions of Rab-25 but without significance (P =0.05).On the contrary,leptin can also up-regulate the mRNA expressions of C-myc (P < 0.05).Conclusion Leptin promotes breast cancer cells proliferation,which may be related to up-regulate the protein and mRNA expressions of Rab-25 and C-myc.It could provide theoretical foundation and new target of gene therapy in the clinical treatment of breast cancer.
2.AN IMMUNOELECTRON MICROSCOPIC STUDY ON THE SYNAPTIC RELATIONSHIP BETWEEN DOPAMINERGIC AND GABAergic NEURONS IN THE BASOLATERALNUCLEUS OF THE RAT AMYGDALA

Objective To explore the relationship between dopaminergic axon terminal and GABAergic neurons and explore the neuroanatomic mechanism of their effects in schizophrenia. Methods The co-location and the association of dopaminergic axon terminal and GABAergic neurons in the basolateral nucleus (BL) of rat amygdala were examined by using double labeling immunoelectron microscopic techniques. Dopaminergic axon terminal and GABAergic neurons were labeled with the antidopamine (anti-DA) and the anti-glutamic acid decarboxylase (anti-GAD) antibodies respectively. Results 43% of the DA-input synapses was observed to relate directly or indirectly to GAD-immunoreactive(IR) dendritic structures(DA/GAD), which includes single (38%), convergent (30%), serial (20%), and axoaxonic (12%) contact types.And 57% of the DA-input synapses was found to associate with unlabeled elements (DA/UE), which includes unlabeled perikarya (10%), dendrites (82%) and axons (8%). All of the synapses that show DA-IR terminals profiles were found to be symmetric (inhibitory) synapses.Conclusion These results suggest that the dopaminergic system in the BL of rat amygdala controls the mediations of the GABAergic interneurons via symmetric synapses. In addition, the dopaminergic axon terminal associates with the glutamatergic projection neurons and exerts influence on its activity.
3.Correlation between the abnormal expression of ICAM-1 and CD44s and lymph node metastasis in human non small cell lung cancer

Objective: To study the mRNA and protein expressions of ICAM 1 and CD44s in non small cell lung cancer as well as nearby normal lung tissue. Methods: RT PCR and SP immunohistochemistry were used to detect the mRNA and protein expressions of ICAM 1 and CD44s in 42 non small cell lung cancer and nearby normal lung tissue. Results: The expression level of ICAM 1 and CD44s increased in NSCLC tissue than in normal tissue. No correlations were noted in expression of ICAM 1 and CD44s and pathological type, histological differentiation and TNM stage. But there was significant correlation between abnormal expressions of ICAM 1 and CD44s and lymph node metastasis. Conclusion: These results suggest that the level of ICAM 1 and CD44s genes expression in lung cells increases as they progress from normal to the transformed stage, that indicate these two genes are involved in the process of lung cancer development.
