1.Preparation and In Vitro Permeability Study of Dihydroartemisinin Ointment

Objective To optimize the formulation of Dihydroartemisinin ointment and study its characteristics of penetration enhancers.Method A method of transdermal experiment in vitro was established for DHA.The penetration rate in vitro was examined by HPLC,and coincided the DHA cumulative permeation curve with the Higuchi equation.Result The penetration rate of DHA from DHA ointment made up of water-soluble carbopol and Azone was the highest in all formulations,up to 27.262 ?g/(h?cm2).The cumulative permeation curve which coincided with the Higuchi equation was better.Conclusion The formulation which made up of water-soluble carbopol and Azone was optimized.

Azone. PG combined with azone showed a higher synergic effect of enhancing effectiveness. Conclusion National Starch polyacrylate pressure-sensitive adhesive was the most effective pressure-sensitive adhesive matrix, PVP-K30 was the most effective crystallization inhibitor, PG combined with azone were the most effective penetration enhancers.
3.Effect of Permeation Enhancers on the Percutaneous Absorption of Artesunate in Vitro

IPA.Skin permeation was significantly enhanced by increasing the loading concentration of OA or NMP,larger IPA amount(8%) or PG amount(20%) could inhibit the permeation of artesunate.Conclusion Oleic acid was found to be the most efficient enhancer for artesunate,10% is the most effective concentration.
4.Study on protective effects of pharmacological preconditioning on rabbits with myocardial ischemia/reperfusion

Objective: To observe the effects of ginaton and nitroglycerin injection on expression of heat shock protein 70 (HSP70) and cardioprotective mechanism in rabbits with ischemia/reperfusion (I/R). Methods : Twenty New Zealand white rabbits were randomly divided into four groups, 5 rabbits being in each group. The model was not replicated in the sham-operated group. Myocardial I/R models were replicated in animals in normal saline, ginatone and nitroglycerin injection groups and were administered with normal saline, ginatone and nitroglycerin injection respectively 0.5 hour before ischemia. Western blot was used to measure HSP70 of ischemia and non-ischemia myocardium and the expression of HSP70 was analyzed semiquantitatively . Serum nitric oxide (NO), malondialdehyde (MDA) contents and total superoxide dismutase (TCD*2SOD) , creatine kinase (CK) activity were measured. Results: HSP70 was less expressed in sham-operated group and more expressed in normal saline group, ginaton group and nitroglycerin injection group. Expression of HSP70 of ischemia and non-ischemia myocardium in normal saline group was 2.5CD*2 and 2.1CD*2fold, in ginaton group 17.6CD*2 and 20.7CD*2fold and in nitroglycerin group 28.1CD*2 and 29.1CD*2fold to that in the shamCD*2operated group, respectively. 5.An Automated Dual-gradient Liquid Chromatography-MS/MS Method for Simultaneous Determination of Four Kinds of Plants Hormones in Different Parts of Soya Beans and Its Application to a Real Analysis

There are some common analysis challenges in the hormone detection in agriculture science, including matrix interference, complicated sample preparation, poor reproducibility, trace analyte content. An automated on-line SPE and innovative fast polarity switch analysis method employing dual-gradient liquid chromatography ( DGLC ) coupled with tandem mass spectrometry ( DGLC-MS/MS ) was established and validated for the simultaneous determination of gibberellic acid ( GA3 ) , indole acetic acid ( IAA ) , zeatin ( ZT) and abscisic acid ( ABA) in the soybean plant ( leaf, grain and pod) . The method was applied in the actual sample detection successfully. In order to acquire higher sensitivity, recovery, stability and precision, some conditions including SPE column, analytical column, mobile phase, additive etc were optimized according to the selection and retain of hormone. Beans were cryogenically grinded by liquid nitrogen, extracted by 80% methanol, certrifugatel and dilluted with water, and then injected directly. Samples were transported and gradient eluted on the analytical column Acclaim PA2 by 0 . 1% formic acid in water and methanol, after retaining and separation on the SPE column Hypersep Retain AX. All analytes were detected in selection reaction monitoring ( SRM) mode in both positive and negative channels. The quantification was based on linear regression. The linear ranges of GA3, IAA and ZT were 0. 1-50 μg/L with the LOQ of 0. 0002 μg/g, and the linear of ABA was 0. 5-50 μg/L with the LOQ of 0. 0010μg/g. The recoveries of four kinds of plants hormones were 76 . 1%-93 . 5%, and RSDs were 0 . 82%-6 . 02% at low ( 0 . 8 μg/L ) , medium (4. 0μg/L) and high (40μg/L). The results noted that the content of ABA in seeds was apparently higher than others. This method could be used for the rapid and accurate detection of hormone in different parts of soya beans.
6.Anaphylactoid reactions induced by polysorbate 80 on Beagle dogs.

OBJECTIVETo evaluate the sensitization effect of polysorbate 80 from different factories on Beagle dogs.
METHODBeagles dogs were randomly divided into 5 groups, 3 in each group, received respectively the intravenous infusion of polysorbate 80 made by four factories in the concentration of 0.5%, with the constant infusing speed of 5 mL x min(-1) and volume of 10 mL x kg(-1). Changes were observed before infusion and in the 24 h after infusion, time of symptom appearance and disappearance was recorded, and the grade of response was determined. Moreover, blood pressure and heart rates were tested by the machine of Bp-98E, blood samples of animals were collected before infusion and at 10 min after ending infusion for measuring histamine content in plasma using ELISA. Then the sensitization effect was comprehensively estimated by combined consideration of the responding grade and histamine level.
RESULTTypical symptoms of anaphylactoid reactions were found in sample 3 group, atypical symptoms were found in other polysorbate 80 groups. Different degrees of heart rate speeding and blood pressure downing were found in polysorbate 80 groups. No over 1-fold increase of plasma histamine level was found in all groups. The atypical anaphylactoid reactions and blood pressure of sample 2 group was lighter than other polysorbate 80 groups. Estimation showed that the sample 3 induced the suspicious anaphylactoid reactions, other test solutions did not induce the typical anaphylactoid reactions on Beagle dogs.
CONCLUSIONFor allergies and other special populations, there is still a certain risk to applicate polysorbate 80 in the concentration of 0.5%. Production process of polysorbate 80 have a certain influence on allergenicity, refined polysorbate 80 increase the security, but further reasearchs are needed to confirmed.
