In university research institutions,when in charge of several projects at the same time,the project leader will inevitably pay not enough attention to every project.In order to ensure the progress and quality of each research project,the study designs and builds project managercentered management mechanisms,redesigns the research and development structure from the system perspective,establishes the crossbar management model,and further analyzes the internal communication mechanism,the power-and-responsibility division mechanism,and oversight mechanism.
2.Retrospective Evaluation of Clinical Efficacy and Safety of Ticagrelor in the Patients with Acute Coronary Syndrome

Objective: To evaluate the clinical efficacy and safety of ticagrelor in the patients with acute coronary syndrome ( ACS) . Methods:A retrospective study was applied to investigate the ACS patients treated with ticagrelor in our hospital from July to December in 2015. The basic information of patients, drug administration, platelet aggregation induced by ADP, major adverse cardio-vascular events ( cardiac death,nonfatal myocardial infarction,target vessel revascularization and stent thrombosis) , and adverse drug reactions ( ADR) were recorded. The incidence of end point events was calculated and the change of platelet aggregation induced by ADP before and after the drug administration was analyzed by SPSS statistical software. Results:A total of 161 patients were collected. The incidence of cardiovascular adverse events was 1. 2%, while the incidence of adverse drug reactions was 30. 4% including bleeding (15. 5%) without severe bleeding events and dyspnea (10. 6%) with 3 severe ones. The platelet aggregation rate before and after the ticagrelor treatment respectively was (54. 96 ± 14. 654)%and(24. 37 ± 13. 183)% in 122 patients with low reaction to clopidogrel( P<0. 01). Conclusion:Ticagrelor at the recommended dose can further reduce the platelet aggregation induced by ADP. In spite of high incidence of ADR, ticagrelor has slight ADR with good tolerance.
3.The Effect of Histone Deacetylases-1 siRNA on the Growth and Apoptosis of HeLa Cells

Objective To investigate the effect of histone deacetylases-1(HDAC-1)siRNA on the HDAC-1 protein expression,cell growth and apoptosis in HeLa cells.Methods The HDAC-1 protein was knocked down by HDAC-1 siRNA.HDAC-1 protein was detected by Western blot.The cell growth inhibition was assesses by MTT and clone forming assay.Apoptosis was measured by flow cytometry.Results Forty-eight h after transfection of HDAC-1 siRNA,HDAC-1 protein expression in HeLa cells was down-regulated obviously.The down-regulation of HDAC-1 significantly reduced the colony formation rate,inhibited cell proliferation and induced apoptosis in HeLa cells.Conclusion HDAC-1 siRNA may play an anti-proliferation role in HeLa cells by inducing apoptosis.

5.The clinical features and treatment of autoimmune hemolytic anemia in patients after orthotopic liver transplantation

Objective To analyze the clinical feature,pathogenesis and treatment of autoimmune hemolytic anemia (AIHA) in patients after orthotopic liver transplantation.Method A retrospective analysis was conducted on 7 patients who developed AHA after orthotopic liver transplantation.The clinical feature,pathogenesis and treatment of AIHA were reviewed.Result The incidence of AIHA was 0.98% (7/713).AIHA occurred (7.9-± 4.0) (3-14) days after transplantation.All patients were characterized by progressive anemia and jaundice.Laboratory examination showed lower hemoglobin and platelet count,higher lactate dehydrogenase,higher indirect bilirubin and higher reticuloeyte count.The causes of AIHA included ABO-incompatible liver transplantation,infection and repeated transfusion.Conclusion AIHA is a rare and serious complication after liver transplantation.The most cause is ABO-incompatible liver transplantation.The patients can be successfully cured if the diagnosis is clarified and the treatment is definitive.
8.Quantitative determination of bosentan by proton nuclear magnetic resonance with internal standard method.

The study aims to establish a quantitative nuclear magnetic resonance (QNMR) method for the determination of the absolute content of bosentan. Proton nuclear magnetic resonance spectroscopy [1H NMR] spectra were obtained in CDCl3 with the internal standard dimethyl terephthalate and zg30 pulse sequence by using a Bruker AVANCE II 400 spectrometer. The content of bosentan is determined with QNMR in comparison with the result obtained by mass balance method. The result is 96.25% by QNMR and 96.54% by mass balance method. A rapid and accurate QNMR method has been established for the quantitative determination of the absolute content of bosentan. The study provides a new way for the quality control and calibration of a new reference standard material, it could be the complementary with the mass balance method for the assay of standard reference.

9.Breeding of Arachidonic Acid Producting Strain with Mortierella Sabellina by Ultraviolet Mutation

In order to gain the strain which has high growth vigor and whose biomass concentration satisfy further fermentation to produce arachidonic acid, we took the yield of microbes olein and arachidonic acid as evaluative index, adopted twice ultraviolet mutation, determined the time of ultraviolet irradiation by single factor expriment, and then the content of the arachidonic acid was measured by GC. The results of this experiment showed that the power of viltalight lamp was 20 W, exposure distance was 30 cm and exposure time was 80 s, lethality of spores of Mortierella isabellina was 76.4%. After twice ultraviolet mutation and repeatedly screen, a mutation of Mortierella isabellina Z80s2-109 whose arachidonic acid concentration in biomass were 3.77 times of the control strains was obtained and genetic stability.
10. Effects of total flavonoids of Clerodendrum bungei on epithelial-mesenchymal transition (EMT) of A549 cells by regulating Wnt/β-catenin signal pathway

Objective To observe the effect of the total flavonoids of Clerodendrum Bungei (TFCB) on proliferation, invasion and migration of A549 cell lines by Wnt/β-catenin signal pathway. Methods Transfecting A549 cell lines with empty vector pcDNA3.1 and β-catenin plasmid which has the mutation at S45 (S45A-β-catenin), which were intervened with TFCB. MTT assay detected cell proliferation, scratch test observed cell migration, Transwell experiment detected the cell invasion. The expression of β-catenin, E-Cadherin, vimentin, Slug, GSK-3β, and p-GSK-3β protein in each group was detected by Western blotting. Results The proliferation, invasion and migration of A549 cells were enhanced significantly after transfected with S45A-β-catenin plasmid (P < 0.05 or 0.01), along with the increasing expression of β-catenin, vimentin, and the reducing expression of E-cadherin, GSK-3β, P-GSK-3β (P < 0.01). TFCB can inhibit the proliferation, migration and invasion of A549 cells (P < 0.05), expecially in S45A-β-catenin group (P < 0.001). A549 cells transfected with empty vector had the ability of up-regulating the expression of E-cadherin, GSK-3β and P-GSK-3β, down-regulating the expression of β-catenin and vimentin with TFCB. A549 cells transfected with S45A-β-catenin plasmid had the ability of down-regulating the expression of β-catenin and vimentin with TFCB. Conclusion The mechanism of inhibiting lung cancer of TFCB maybe associate with the inhibitory expression of β-catenin and regulate the downstream factors, with view to inducing EMT by activating Wnt/β-catenin pathway.
