1.Effect of Curcumin on Expression of Hepatocyte Nuclear Transcription Factor in Liver Cancer Cell Hepg-2 Induced by TNF-α

Objective To investigate the effect of curcumin on the expression of nuclear transcription factor (NF-kappa B) and its inhibitory protein (Iκ-Bα) in liver cancer cells (Hepg-2) induced by TNF-α, and explore its effect on non-alcoholic steatohepatitis. Methods The cells were divided into normal group, TNF-alpha group, and different concentrations of curcumin treatment group. MTT and Western blotting were used to assay curcumin’s effect on Hepg-2 proliferation activity and changes of NF-κB and Iκ-Bα’s in Hepg-2. Results Compared with the normal group, TNF-αgroup enhanced NF-κB and Iκ-Bαexpression, but the increase of Iκ-Bα had no statistical significance (P>0.05). Curcumin treatment group’s NF-κB expression was significantly weakened than TNF-α group, while Iκ-Bα was significantly enhanced than that of TNF-α group. Conclusion Curcumin can antagonize NF-κB inflammatory signaling pathway activation induced by TNF-α, thus reduce the inflammatory injury of the liver cells.
2.Effect of mongolian medicine Menggenwusenrile on correlation factor in imiquimod-induced psoriasiform skin inflammation in mice

Objective To investigate the efficacy and mechanism of mongolian medicine Menggenwusenrile in the treatment of imiquimod -induced psoriasiform skin inflammation in mice.Methods Fifty cases of BALB/c female mice were randomly divided into normal control group, model group, Menggenwusenrile low, middle and high dose groups, 10 cases in each group.Normal control group was given 0.3% sodium car-boxyl methyl cellulose(CMCNa) 2 mL/100 g, once a day intragastric administration for consecutive 21 days.Modal group was given 0.3%CMCNa 2 mL/100 g, once a day intragastric administration for consecu-tive 21 days.Menggenwusenrile low, middle and high dose groups were given 0.33, 0.66, 1.32 g? kg-1 , once a day intragastric administration for consecutive 21 days.The expression of proliferating cell nuclear anti-gen ( PCNA) , transforming growth factor-α( TGF-α) , tumor necrosis factor-α( TNF-α) , platelet endothelial cell adhesion molecule-1/CD31 (CD31), vascular endothelial growth factor (VEGF)and Toll -like receptors-2 ( TLR-2 ) were detected by immunohistochemical stai-ning.Results Six kinds of immunohistochemical analysis showed the each drug dose group was higher than the model group, high dose is bet-ter than middle dose group, the middle dose group is better than low dose.Conclusion Menggenwusenrile have a better therapeutic effect in imiquimod-induced psoriasiform skin inflam-mation in mice.
3.Preparation Technology of Zucapsaicin Cream

Objective To select the optimal preparation technology of zucapsaicin cream and compare it with zuacta cream.Methods Cold stability,thermal stability,centrifugal stability,and appearance were used as indicators to select the ratio of prescription.In this study preparation technology was optimize by using orthogonal experiment method,and transdermal absorption of the homemade zucapsaicin cream and zucata cream were investigated by employing modified Franz diffusion pool.Results The optimized preparation technology was as follows,emulsifying temperature was maintained at 70 ℃,the stirring speed was set at 2 000 r·min-1,the main medicine was added into the oil phase,emulsifying time was 30 min.The results showed that there was no significant difference between the homemade zucapsaicin cream and the reference Zuacta cream.Conclusion The homemade zucapsaicin were tested to have reasonable ratio of prescription,stability,and definite transdermal effect,which was basically in accordance with zuacta cream.
4. Bioinformatics analysis and fluorescence quantitative PCR results of Ricinus communis PIP5Ks

Objective To predict the expression of seven genes in the PIP5K gene family, and to determine the relative expression of PIP5Ks in different stages of type Lm gynoecious Ricinus communis aLmAB2. Method Bioinformatics analysis of online software protein physicochemical properties analysis, protein hydrophobicity analysis subcellular localization prediction, transmembrane region analysis, etc. Bioinformatics analysis of hemp PIP5K gene family and DNAMAN for sequence alignment and real-time PCR. Results The results showed that there were seven members in the R. communis PIP5Ks, which were PIP5K1, PIP5K2, PIP5K4, PIP5K6, PIP5K8, PIP5K9, and PIP5K11. The amino acid sequence homology of the corresponding protein of R. communis PIP5K reached 48.06%, and the protein of R. communis PIP5Ks all are hydrophilic proteins. Except that PIP5K9 and PIP5K11 were unstable proteins, the rest were stable proteins, and the corresponding proteins of PIP5Ks have no transmembrane domain, and all were non-transmembrane proteins. Subcellular localization results showed that the proteins PIP5K1, PIP5K4, PIP5K6, PIP5K9, and PIP5K11 were less likely to have a lead peptide, and there was no corresponding amino acid cleavage site, ie, localized in other organelles or possibly cytoplasmic proteins; Protein PIP5K2 was localized in chloroplasts, the chloroplast transit peptide has a higher value, and the protein PIP5K8 was located in the secretory pathway. From the perspective of the relative expression of PIP5Ks in the differential expression, except that no fluorescence was detected in PIP5K4, the other PIP5Ks had a certain differential expression. The changes of relative expression of PIP5Ks showed similar trends, the levels of PIP5K1, PIP5K2 relative expression were high, followed by the relative expression of PIP5K8, PIP5K9, and PIP5K6 and PIP5K11 only have fewer expressions. Conclusion It is concluded that PIP5Ks may have a certain effect on the inflorescence axis traits in ramie. The differential expression of PIP5K on the inflorescence axis has the certain regularity with the development of inflorescence.
6.Influences of Deduhonghua -7 powder on matrix metalloproteinases and transforming growth factor beta expression in liver fibrosis

Objective To observe the therapeutic effect of Deduhonghua-7 powder on liver fibrosis and its correlation with matrix metalloproteinases ( MMPs) and transforming growth factor β1 ( TGF-β1 ) .Methods The rats were divided into 4 groups, blank group, model group, control group, experiment group, 8 rats each group.The CCl4 was used to induce rat liver fibrosis.The control group was given colchicine pill, CMC sodium cellulosate was used to prepare 2% of colchicine suspen-sion, once time per day, 0.2 mL · kg-1 each time.The experiment group was given Deduhonghua-7 powder, CMC sodium cellulosate was used to prepare 3% of Deduhonghua-7 powder suspension, once time per day, 0.2 mL· kg -1 each time; blank group and model group were given same volume of pure water.All rats were sacrificed after 40 days of medication.Western blot was acted to detect the matrix metalloproteinase-1 (MMP-1), matrix metalloproteinase-13 (MMP-13),tissue inhibitor of metalloproteinases-1 (TIMP-1) protein expression.The PCR was used to detect the changesoftype Ⅰ, Ⅲ collagen, TGF -β1 mRNA gene expression.Results The MMP-1, MMP -13 protein expression in rats of model group was down -regulated compared with blank group ( P<0.05 ).The TIMP-1 protein expression was up-regulated compared with blank group ( P<0.05 ) . The mRNA expressions of typeⅠprocollagen, typeⅢprocollagen and TGF-β1 were increased compared with blank group(P<0.05).The MMP-1, MMP-13 protein expression of control group and experiment group were significant-ly up-regulated compared with model group.The TIMP-1 protein expression significantly down-regulated compared with model group.The mRNA gene expression of type I procollagen, typeⅢprocollagen and TGF-β1 were down-regulated compared with model group ( P<0.05 ).Conclusion Deduhonghua-7 powder regulate and maintain the stabilization of MMP/TIMP to block the expression of TGF-βand inhibit the transcription of type I,Ⅲ collagen and TGF-β1 mRNA to reach the purpose of detention of occurrence and development of liver fibrosis.
7.Expression of MCP-1 in the pancreas of piglet with chronic obstructive pancreatitis and its significance

Objective To determine the serum concentration of MCP-1 and the expression of MCP-1 protein in the pancreas in the piglet with chronic obstructive pancreatitis and to explore the role of MCP-1 protein in pancreatic fibrosisits.Methods The piglet model of chronic obstructive pancreatitis was established by incomplete ligation of the pancreatic duct.The piglets were sacrificed at 4, 6, 8 weeks after induction.Pathological changes of pancreas were examined.Pancreatic fibrosis was assessed by VG staining.Serum MCP-1 concentrations were detected by ELISA method.MCP-1 and α-SMA, PDGF, TGF-β1 and NF-κB protein expression were detected by immunohistochemistry.Results The induction was successful in 14 piglets ( 58.3% ).Mild atrophic changes, interstitial fibrosis, chronic inflammatory cell infiltration could be observed in the body and tail of pancreas from the 4th week in the experimental group.The most obvious changes occurred in the 8th week.Stage Ⅰ pancreatic fibrosis occurred in 5 piglets (35.7%), stage Ⅱ in 4 piglets (28.6%), stage Ⅲ in 5 rats ( 35.7% ).Seurm MCP-1 at 4, 6, 8 weeks was ( 102.44 ± 36.25 ) pg/ml,(97.84 ± 28.67) pg/ml, ( 94.32 ± 28.42 ) pg/ml, respectively, and was significantly higher than that in control group [ ( 10.42 ±5.86) pg/ml, (8.58 ±4.86) pg/ml, (8.22 ±4.58) pg/ml, P <0.01 ].There was no MCP-1 protein expression in the control group;MCP-1 protein was detected in the successful induction group, and MCP-1 expression was positively correlated with expressions of the PDGF, TGF-β1, α-SMA and NF-κB.Conclusions MCP-1 may play an important role in the course of pancreatic fibrosis in chronic obstructive pancreatitis.
8.Relationship between Wound Age and the Expression of Fzd2 in Rats Skeletal Muscle after Contusion

Objective To investigate the relationship between wound age and the expressions of frizzled-2 (Fzd2) mRNA and its protein in rats skeletal muscle after contusion,and to explore its possibility of being an index for wound age estimation.Methods The mRNA and protein expressions of Fzd2 in rats skeletal muscle of the control group and the experimental group within 4-48 h after contusion were detected per 4 h by RT-qPCR and Western blotting.Results The relative expression of Fzd2 mRNA increased at 24h,36h and 40h after contusion,and the expression at 24h was twice as the control group (P<0.05).The relative expression of Fzd2 protein changed inconspicuously after contusion (P>0.05).Conclusion The changes of Fzd2 mRNA expression after contusion in a certain time can be a basis to estimate wound age by combined multi-indicators.
9.Study of acute toxicity of Meng gen wu sen ri le in mice

Objective To investigate the study of acute toxicity of mon-golian medicine Meng gen wu sen ri le, to understand the dose of acute toxicity and distribution and death of acute toxicity after the administra-tion, to determine lethal dose 50 ( LD50 ) of Meng gen wu sen ri le. Methods According to the results of preliminary experiments, using Conn synthesis method ( modified Karber method ) , divided into six groups dosage are 14.30, 9.28, 6.04, 3.92, 2.55, 1.66 g? kg-1 , af-ter experiments observing 14 day, to record the change of the weight and adverse reaction.Results LD50 of Meng gen wu sen ri le is 5.1597 g? kg-1 ( 95%CI:3.6652-7.2637 g? kg -1 ).In 14 days, it does not appear obvious symptoms of adverse reaction and body weight has growth trends.Conclusion According to the results of experiments, LD50 of Meng gen wu sen ri le is 100 times of the clinical dosage, tip single oral is safer.
10.Expression of interleukin-6 in rat model of doxorubicin-induced nephropathy.

OBJECTIVEThe pathogenesis of minimal change nephrotic syndrome (MCNS) remains unclear. This study aimed to investigate the expression of interleukin-6 (IL-6) in rats with doxorubicin-induced nephropathy and its possible roles in the pathogenesis of MCNS.
METHODSEighty-three male Wistar rats were randomly assigned into a control group (n=32) and a nephropathy group (n=51). Nephropathy was induced by a single tail vein injection of doxorubicin (5 mg/kg). The control group was injected with normal saline. Twenty-four-hour urinary protein excretion was measured 7, 14, 28 and 42 days after doxorubicin injection. IL-6 expression in urine and renal tissues was determined using ELISA 7, 14, 28 and 42 days after doxorubicin injection.
RESULTSThe urinary protein excretion increased significantly in the nephropathy group 7, 14, 28 and 42 days after doxorubicin injection compared with that in the control group (P<0.01). IL-6 expression in urine and renal tissues increased significantly 7, 14, 28 and 42 days after doxorubicin injection compared with that in the control group (P<0.01). IL-6 expression in urine and renal tissues was positively correlated with 24-hour urinary protein excretion in the nephropathy group (r=0.794, P<0.01; r= 0.870, P<0.01). IL-6 expression in urine was positively correlated with that in renal tissues (r=0.739, P<0.01).
CONCLUSIONSIL-6 expression in the urine and renal tissues is increased in MCNS rats. IL-6 might play an important role in the pathogenesis of MCNS.
