Objective To investigate the effects of siRNA silencing of inhibitor of apoptosis protein gene on the proliferation of airway smooth muscle in bronchial asthma mice .Methods Tweenty female BALB/c mice were randomly divided into experimen‐tal group and control group .Mice in the experimental group were constructed asthma models .The trachea and bronchi were collect‐ed for cell culture .Respectively ,the cultured airway smooth muscle cells in the experimental group and the control group were grouped:PBS control group (added to PBS solution) ,missense chain control group (transfected with missense chain) and siRNA transfection group (transfected with siRNA Survivin) .The expressions of Survivin mRNA ,Survivin protein and caspase‐9 protein in airway smooth muscle cells were detected .The levels of IL‐6 and CCL5 in cell supernatant were detected .Results RT‐PCR re‐sults showed that ,the relative expression levels of Survivin RNA of airway smooth muscle cells in the siRNA transfection group of the experimental group were lower than the PBS control group and missense chain group ,the differences were statistically signifi‐cant (P<0 .05) .Western blot results showed that ,the expressions of Survivin proteins in the siRNA transfection group of the ex‐perimental group were lower than the PBS control group and missense chain control group ,while the caspase‐9 protein were higher , In the experimental group ,the synthesis and secretion levels of IL‐6 and CCL5 of airway smooth muscle cells in the siRNA transfec‐tion group were lower than the PBS control group and missense chain control group ,the cell proliferations at 2-4 d in the siRNA transfection group were lower than the PBS control group and missense chain control group ,the proportion of G0/G1 phase in the siRNA transfection group were higher than the PBS control group and missense chain control group ,but the proportion of S/G2 phase were lower than the PBS control group and missense chain control group ,cells apoptosis rates of the siRNA transfection group were lower than the PBS control group and missense chain control group ,the differences were statistically significant (P<0 .05) .Conclusion siRNA specific silencing Survivin gene could accelerate the airway smooth muscle cell apoptosis ,and inhibit cell abnormal proliferation and secretion .

Objective To explore the clinical efficacy of entecavir combined with Vitamin E in treating patients with Chronic hepatitis B complicated with non-alcoholic fatty liver.Methods Retrospective analysis was done by reviewing the clinical data of 103 patients, who suffered with chronic hepatitis B complicated with non-alcoholic fatty liver and treated in our hospital from 2015 to 2016.The patients were divided into the control group with 61 cases and the observation group with 42 cases based on different therapies.The control group was treated with only entecavir while the observation group was treated with entecavir and Vitamin E, and both the courses of treatment lasted for 6 months.The negative conversion ratio of HBV-DNA, levels of ALT, TBIL, ALP, the disease state and the adverse reactions of the two groups were compared before and after treatment.Results The negative conversion ratio of HBV-DNA of the observation group was obviously higher than that of the control group, and the patients of the observation group showed significant improvement.Meanwhile, the levels of ALT, TBIL and ALP in the two groups were both lower than before treatment.Compared with the control group, those indexes in the observation group were significantly lower (P<0.05).Conclusion Entecavir combined with Vitamin E for the treatment of chronic hepatitis B complicated with non-alcoholic fatty liver could significantly decrease the viral infections so that the state of patients can be improved.

Objective To observe the intervention effect of acupuncture on mammalian target of rapamycin(mTOR)of hypoxic-ischemic encephalopathy(HIE)model in newborn rats.Methods Seven-day-old SD rats were randomly devided into six groups.Sham-surgery group:the bilateral common carotid arteries were separated,without ligation,and the rats were without hypoxia.HIE model group:the bilateral common carotid arteries were separated and ligated,and the rats were in low oxygen.In the sham-acpuncture group,four positions were acupunctured:from Baihui,Dazhui,Quchi and Yongquan by 1 cm after hypoxia ischemia.In acpuncture group,four positions were acupunctured:Baihui,Dazhui,Quchi and Yongquan after hypoxia ischemia,positioned according to the ordinary acupoints for acupuncture in rats.In the antagonists group,the HIE model rats were injected wortmannin 5 μl to the left ventricle.The acupuncture and antagonists group had the same operations of antagonists group and acupuncture group at the same time.All the groups were subdivided into four subgroups in terms of four time periods of 1 d,3 d,7 d and 21 d.The morphological changes of organizational structures of hypoxic ischemia side in brain tissue were observed by pathological methods.The levels of mTOR mRNA expression were detected by RT-PCR.Results In acupuncture group, acupuncture treatment for 21 d showed that acupuncture group had clearly organizational structure,regular arrangements of neurons,normal cell outline,relatively clear nucleolus,mitigated swell neuron and hyperplas-tic glial cells.Compared with that of the HIE model group,acupuncture treatment increased mTOR mRNA dramatically on day 1 ,day 3,day 7 and day 21 (Q =7.099 1 ,9.009 9,7.894 0,9.914 7,P ﹤0.05 respective-ly).The expression of mTOR mRNA in acupuncture and antagonists group was significantly higher than that in sham-surgery group on day 1 ,day 3 day 7 and day 21 (Q =9.868 9,15.843 4,21.007 2,P ﹤0.05 respec-tively).In day 21 ,compared with the sham operation group and the HIE model group,the expression of mTOR mRNA in acupuncture and antagonists group were significantly higher (Q =18.236 0,4.729 7, P ﹤0.05 respectively).Conclusion Electroacupuncture treatment can promote the expression of mTOR at the mRNA level and protect brain of newborn rats with HIE.

Objective To study the changes and significance of the hyaluronic acid (HA) and collagen in the larynx of old guinea pig.Methods Six young guinea pigs (4 months old) and 6 old guinea pigs (2 years old) were selected as control group and aged group for this study (n=6 for each)The expressions of HA and collagen in the larynx of guinea pigs were detected using Alcian blue and Masson trichrome methods,respectively.The images were managed using Image-Pro Plus 6.0 For Window.Results The optical density (expression level) of HA in the larynx was lower in aged group than in control group (0.69 vs.0.80,F=13.45,P＜0.05).The optical density(expression level) of collagen in the larynx was higher in aged group than in control group (0.71 vs.0.63,F=9.56,P＜0.05).Conclusions The content of HA in larynx is decreased and collagen in larynx is increased in old guinea pig which may result in presbyphonia change through higher viscoelastic properties of vocal cords.

Objective To investigate the expression and correlation of hypoxia-inducible factor-1?(HIF-1?),vascular endothelial growth factor(VEGF)and sFlt-1 in the preeclampsia placenta,and discuss their significance in the pathogenesis of preeclampsia.Methods Placentas were collected from 20 pregnant women with preeclampsia as study group and 15 normal pregnant women as control group.The expressions of HIF-1?,VEGF and sFlt-1 protein were semi-quantitatively analyzed with immunohistochemical assay and mRNA level was determined using reverse transcription polymerasc chain reaction(RT-PCR)technique. Results(1)the expression of HIF-1?and sFlt-1 protein in preeclampsia group obviously increased.Strong (+++)positive expression was observed in 9 and 11 cases respectively,significantly higher than in control group(2 and 3 cases)(P＜0.05),however,VEGF expression obviously reduced in preeclampsia group(P＜0.01).(2)the level of HIF-1?and sFlt-1 mRNA in preeclamptic placenta was 0.604?0.013, 0.898?0.041,significantly higher than 0.208?0.007 and 0.559?0.244 in normal placenta(P＜0.05). Although the level of VEGF mRNA increased in preeclampsia placenta,it was not significantly different from that in normal placenta(P＞0.05).The ratio of VEGF mRNA/sFlt-1 mRNA obviously reduced in preeclampsia group and was significantly lower than in control group(P＜0.05).(3)in preeclampsia group,HIF-1?mRNA expression was positively correlated with the expression of sFlt-1 mRNA(r=0.577, P＜0.05),and negatively correlated with the ratio of VEGF mRNA/sFlt-1 mRNA(r=-0.376,P＜0.05).Conclusion Abnormal high HIF-1?expression in preeclampsia placenta indicates that HIF-1?might play an important role in the pathogenesis of preeclampsia,possibly through affecting the cytotrophoblastic invasion and placental vascular reconstruction via the modulation of VEGF and sFlt-1 gene transcription.

Objective Cardiovascular diseases comorbid with mental disturbances are described as psycho-cardiology diseases, which have been paid attention increasingly. The aim of this study is to summarize and analyze the findings about the study of psycho-cardiology over a year , and to obtain the baseline data of psycho-ca rdiology diseases in inpatients of cardiology department. Methods Subjects in this study were inpatients of cardiology department in the First Affiliated Hospital of Kunming Medical University in the past 21 months. The demographic and clinical data of the inpatients were collected. Anxiety and depression were evaluated by the Hospital Anxiety and Depression Scale. The prevalence of anxiety and depression in patients with various cardiovascular diseases were calculated respectively. To detect the potential risk factors of anxiety or depression, two logistic regression models were established. Results The prevalence of anxiety, depression, anxiety and/or depression, and anxiety comorbid with depression was 38.6%, 12.4%, 51.0%, and 7.9%,respectively. The prevalence of depression in patients with chronic heart failure was 24.6%, which was higher than the general level and that of patients with other cardiovascular diseases. The prevalence of anxiety in patients with acute myocardial infarction, arrhythmia, and hypertension ranged from 39.6% to 41.7%. The long course of diseases, multiple diseases, and the history of cardiac interventional operation were the common risk factors for anxiety and depression. Conclusion Anxiety and depression are common in patients with cardiovascular diseases. Cardiologists should take mental disturbances seriously, and pay more attention to the patients with above risk factors.

Objective To investigate the influence factors of central venous catheter related infections (CVC-RI) in postoperative patients with gastric carcinoma treated with parenteral nutrition,in order to provide scientific basis for prevention and treatment.Methods Using the retrospective study method,150 patients who treated with parenteral nutrition after operation of gastric carcinoma from December 2013 to March 2014 were selected.Information including age,sex,anamnesis,catheter indwelling time,infusion rate of nutrient solution,and so on were collected.Patients with central venous catheter related infections were set as infection group,patients without infection as the control group,and the group logistic regression analysis was used to analyze the collected data.Results 21 patients in infection group had CVC-RI,and the incidence was 14.00％ (21/150),while 129 patients in control group had CVC-RI.Univariate analysis showed there was significant difference between infection group and control group in age,anamnesis,catheter indwelling time and infusion rate of nutrient solution (x2=17.126,5.447,12.795,7.973,P＜0.05),however there was no significant difference in sex,health insurance,the success rate of catheterization and whether glutamine was in nutrient solution (P＞0.05);the group logistic regression analysis showed that age (OR=15.927,95％CI 3.305-76.756,P＜0.05)and catheter indwelling time (OR=3.511,95％CI 1.028-11.992,P＜0.05) were independent risk factors for CVC-RI,however infusion rate of nutrient solution (OR=0.140,95％CI 0.033-0.597,P＜0.05) was a protective factor.Conclusions For the postoperative patients with gastric carcinoma treated with parenteral nutrition,CVC-RI is closely related to age,catheter indwelling time and infusion rate of nutrient solution.So reducing catheter indwelling time,restoring the enteral nutrition under condition permission as so on as possible can reduce infection of elderly patients.

Objective To investigate the prognosis correlation factors of sudden hearing loss.Methods A retrospective analysis was performed in 137 cases (149 ears) of sudden hearing loss.The relationships between the age,prehospital delay time,type of hearing loss,degree of hearing loss,and whether with dizziness,tinnitus,diabetes mellitus,high blood pressure and therapeutic effect were observed.Results The total effective rates in ＜ 45 years,45-59 years and ＞ 59 years patients were 86.36％(38/44),77.78％ (56/72) and 57.58％ (19/33) respectively.There were statistical differences in the total effective rate between ＜ 45 years,45-59 years patients and ＞ 59 years patients (x2 =8.128,P =0.004; x2 =4.525,P =0.033).The total effective rates in patients who were treated ≤7 d,8-14 d and ≥ 15 d were 83.52％ (76/91),76.67％(23/30) and 50.00％(14/28) respectively.There were statistical differences in the total effective rate between patients who treated ≤7 d,8-14 d and patients who were treated ≥ 15 d (x2 =13.050,P =0.000; x2 =4.459,P =0.035).The total effective rates in low-mid frequency type,all frequency type,total deafness type and mid-high frequency type were 89.66％ (52/58),84.44％ (38/45),7/12,47.06％ (16/34) respectively.There were statistical differences in the total effective rate between low-mid frequency,all frequency type and total deafness type,mid-high frequency type (P ＜ 0.05).The total effective rates in hearing loss mild degree,middle degree,severe degree,extremely severe degree and total deafness were 87.23％ (41/47),86.11％ (31/36),61.90％ (13/21),7/15 respectively.There were statistical differences in the total effective rate between mild degree,middle degree and severe degree,extremely severe degree and total deafness (P ＜ 0.05).The hearing loss prognosis with the dizziness,diabetes mellitus,high blood pressure was worse.Conclusions It is considered that the age,prehospital delay time,type of hearing loss,degree of hearing loss,and whether with dizziness,high blood pressure,diabetes mellitus are related to the total effective rate.The prognosis of hearing loss can be estimated according to these correlation factors.

[Objective]To investigate the significance of the high-risk human papilloma virus (HPV)detection in the screening and diagnosis of cervical lesions.[Methods] The high-risk HPV DNA test results of 797 patients with cervical lesions who all accepted cytology and histopathology test were collected and analyzed retrospectively.[Results]The high-risk HPV DNA positive rates in cervicitis,cervical intraepithelial neoplasia(CIN)Ⅰ,CIN Ⅱ,CIN Ⅲ and cervical cancer were 53.41％(188/352),70.91％(117/165),87.63％(85/97),97.90％(140/143),97.50％(39/40),respectively.The sensitivity and negative predictive value of the high-risk HPV DNA detection for CIN Ⅱ and more serious lesions were 96.66％(318/329),93.29％(153/164),respectively.The detection rate of CIN Ⅱ and more serious lesions in patients with atypical squamous cells of undetermined significance(ASCUS)and positive high-risk HPV DNA was 30.03％(94/313),while the rate in patients with negative high-risk HPV DNA was 1.55％(2/129).[Conclusions] The more serious the cervical lesion is,the higher high-risk HPV DNA positive rate is.It is most closely related with CIN 11 and cervical cancer.The high-risk HPV DNA detection has high sensitivity and negative predictive value for CIN Ⅱ and more serious lesions.The high-risk HPV DNA detection has high negative predictive value in CIN Ⅱ and more serious lesions in ASCUS.
