2.Expression of interferon regulatory factor 1 in patients with systemic lupus erythematosus and the influence of prolactin upon it

Objective To study the expression of interferon regulatory factor 1 (IRF-1) in patients with systemic lupus erythematosus (SLE) and the influence of prolactin (PRL) upon it. Methods The level of serum PRL in quiescent condition was examined by electrochemiluminescence-meter in 30 patients with SLE and 20 healthy controls. Peripheral blood mononuclear cells (PBMC) were separated from all the subjects by gradient centrifugation density, and cultured with or without the presence of recombinant human PRL (rhPRL) for 24 hours. The expression of IRF-1 gene in cultured PBMC was detected by reverse transcriptase-PCR (RT-PCR) with gel image scanning. Results The relative value of IRF-1 gene expression was significantly higher in SLE patients than in normal controls (0.89±0.21 vs 0.78±0.18, P=0.026), and in SLE patients with high PRL than in those with normal PRL (1.06±0.26 vs 0.82±0.21, P=0.005). However, there was no significant difference between SLE patients with normal PRL and healthy controls in regard to the expression of IRF-1 gene (P=0.514). The stimulation with rhRPL significantly elevated the relative expression of IRF-1 gene in SLE patients with normal PRL (0.99±0.22 vs 0.82±0.21, P=0.036), but had no obvious effect on that in the normal controls. Conclusion The study reveals a high expression of IRF-1 gene in SLE patients, which may be related to the high level of PRL.
3.Diagnostic significance of plasma total tau and phosphorylated tau protein in Alzheimer's disease

Objective To investigate the plasma total tau(T-tau)and phosphorylated protein tau(P-tau)protein in the diagnosis of Alzheimer's disease(AD).Methods 22 352 medical patients aged over 60 years in Beichen Area Community Service Center were involved in this survey.Random sampling principle was used for screening.Mini-mental state examination(MMSE)and activities of daily living assessment form(ADL)were conducted for the cognitive function and ability of daily score firstly.When the score below the standard,the Hamilton depression scale and Hachinski ischemic scale were re-used for diagnostic score.Related laboratory tests were conducted to exclude other central nervous system and other systems and material causes of dementia patients.Ultimately,105 cases of AD,diagnosed by the neurologist,and 42 cases of non central nervous system disease,non dementia non nervous system disease patients as the normal control group were involved.T-tau and P-tau levels in the two groups were determined by ELISA.Results the T-tau concentration in AD group(15.93+6.59)ng/L was higher than control group(14.10±6.32)ng/L,no significant difference was found (P > 0.05).However,compared with control group (0.69 ± 0.24) ng/L,P-tau protein in AD group (1.26 ± 0.75)ng/L increased significantly(P<0.05).Conclusion Plasma levels of phosphorylated tau protein might have diagnostic value for AD patients.
4.Application of systematic training methods for new nurses in central sterile supply department

Objective To discuss the systematic training method for new nurses in central sterile supply department(CSSD),in order to improve the training effects of new nurses and special practice ability,and make them to be qualified for jobs in CSSD.Methods 18 new nurses in CSSD were divided into the control group and the experimental group with 9 nurses in each group.The experimental group adopted systematic training method combining with unit tests and comprehensive evaluation.The control group chose the traditional training method.The testing results were compared between two groups.Results The experimental group all passed through the unit evaluation and comprehensive evaluation,could work independently after the training,got excellent achievement in comprehensive assessment of the department after 3 months on-the-job,and could finish all the work and solve various emergencies.4 passengers failed in unit assessment in the control group,and 3 failed in the comprehensive evaluation.The average results in the experimental group were obviously higher than that of the control group.Conclusions Scientific training content,the systematic training method,comprehensive evaluation method can avoid the blindness and arbitrariness in training and improve training effect.The implementation of unit access system and post access system in standardized training of new nurses can improve the study enthusiasm and strengthen the risk and responsibility consciousness of nurses,which is beneficial to the growth of new nurses and professional value guidance.
5.Investigation and analysis of the anxious condition of nurses and correlative factors

Objective To explore the anxious condition of nurses and correlative factors,and to explore the relationship between the nurses' anxiety and the correlative factors,so that can help the management echelon and nurses to correctly understand the relationship of anxiety and pressure,and control and handle the anxiety problem of nurses fundamentally,and improve the physical and mental health,work efficiency and nursing quality of nurses. Methods A questionnaire investigation was conducted in 85 nurses from a certain hospital in Harbin about the anxiety,life events,work stressor and the relationship between them. Results 30 nurses had anxiety(the anxiety group),another 55 nurses showed no anxiety(the nonanxiety group).The comparison of items in life events inventory showed that great difference existed in pregnancy,addition of new family members,the difficulty of training their children,property damage or being stolen,living apart with the spouse,magnificent changes of the life style,having got severe illness or wound.In the aspect of work sttessor scale,professional and career issues,workload and allocation of time,working environment,patient care and interaction showed significant difference.The work pressure was significantly associated with the level of anxiety. Conclusions Our finding suggest that effective stress management strategies should be implemented to reduce or eliminate work pressure of nurses,carry out training to improve the self-regulation ability of nurses,and if necessary biofeedback therapy can be introduced.
6.Expression of Caspase-3 in Renal Glomerulus Cells in Rats with Sub-chronic Arsenic Poisoning

Objective To investigate the influence of caspase-3 on the apoptosis of glomerulus cells in chronic arsenic poisoning. Methods Sprague-Dawley(SD) rats in cleanliness grade were randomly divided into high-dose group,low-dose group and control group,10 in each (5 males and 5 females). The rats in high and low groups were treated with As2O3 through drinking water,10 and 0.4 mg/(kg?d) respectively,for four months. The content of arsenic in the blood and urine was determined. The expression of caspase-3 in the glomerulus cells was detected by SABC immunohistochemistry and analyzed through image patterns. Results Compared with the control group,a higher arsenic content in the blood and urine,more positive cells of caspase-3 and lower OD value the glomerulus cells were found in both of high-dose and low-dose groups. Compared with low-dose group,a higher arsenic content in the blood and urine,more positive cells of caspase-3 and the lower value of grey degree in the renal glomerulus cells were all found in high-dose group. Conclusion The obvious increase of caspase-3 in the glomerulus cells may play a role in the apoptosis of the glomerulus cells induced by chronic arsenic poisoning.
7.Biological characteristics, osteogenic and adipogenic differentiation of mesenchymal stem cells from human umbilical cord blood

BACKGROUND: Up to now, there are few reports addressing the biological properties and differentiation potential of umbilical cord blood-derived mesenchymal stem cells (UCB-MSCs). OBJECTIVE: To observe the biological characteristics, osteogenic and adipogenic differentiation potential of UCB-MSCs. METHODS: MSCs were harvested and cultured from UCB at various gestational ages (GA). Harvested UCB-MSCs were cultured primarily and subcultured, and then induced to differentiate into osteoblasts and adipocytes. RESULTS AND CONCLUSION: Under the inverted phase contrast microscope, UCB-MSCs adhered to the wall, showing fibroblast like morphology and whirlpool like growth alignment. Observation of the ultramicrostructure under transmission electron microscope showed that UCB-MSCs had a big cellnucleus, fewer cellular organelles and big karyoplasmic ratio.Allofthe growth curves of primary and passaged UCB-MSCs presented S-shaped. The 3rd and 5th passages of MSCs showed the strongest proliferation activity. The count of colony forming unit fibroblasts varied with GA, significant difference was found among the three GA groups (P < 0.05), and the lower GA group had a higher count of colony forming unit fibroblasts than that in the older GA group. Flow cytometry showed that these cells expressed CD29, CD44 and CD90 positively, but they failed to express hematopoiesis related molecules such as CD34 and CD45. When the MSCs were induced to osteogenic and adipogenic differentiation for 3 weeks, strong expression of alkaline phosphatase was found and the formation of a mineral extracellular matrix was detected by alizarin red staining were detected; and neutral lipid vacuoles were detected by oil red O staining. UCB-MSCs have similarmorphologicaland biological characteristics and cell surface molecule markers with MSCs derived from bone marrow, both of which have great capability of proliferation and regeneration. UCB-MSCs can be induced to osteoblasts and adipocytes in a suitable condition in vitro.

Based on the plasma activation and the sensing ability of cataluminescence, a low temperature plasma-assisted cataluminescence sensor was developed for ethylene detection using the low-cost and abundant alkaline-earth oxides of MgO nanomaterials as the sensing materials.Taking advantage of the high activity of the plasma, the working temperature of this method was greatly decreased than that of traditional detection method (300-500℃), and the sensing of ethylene was realized at room temperature without any heating device.This ethylene cataluminescence sensor gave a linear range of 112-4997 ng/mL (90-3998 ppm, R=0.97669) with a detection limit of 37 ng/mL (30 ppm).Besides, the sensor showed good selectivity and stability in ethylene detection.Due to the absence of the heating element, the present sensor was simple, rapid, low-cost, low energy-consumption and stable for ethylene sensing.This study improved the applicability of cataluminescence sensors and might promote the development of cataluminescence sensors.

10.Bioinformatic Analysis of Differentially Expressed Genes in Peripheral Blood of Human Immunodeficiency Virus/Tubercle Bacilli Co-infected Patients.

Objective To analyze the differentially expressed genes in peripheral blood of human immunodeficiency(HIV)/tubercle bacilli co-infected patients and explore the biological regulatory mechanism and network of key proteins,so as to provide new evidence for early diagnosis and clinical treatment of HIV/TB co-infected patients. Methods Microarray gene chip data of HIV/TB co-infected patients were downloaded from public databases GEO and imported into the analysis software GEO,STRING,PANTHER,and GenClip. The gene expression profiles,protein interaction networks,processes of molecular biology,and gene functions were analyzed. Results The expression profiles of 15 529 genes between the two groups of patients were similar,and gene expression profiles from 44 subjects were highly correlated. The 251 differentially expressed genes had good diagnostic capabilities in the differential diagnosis of HIV/TB infection. RPLP1 might be a key gene in the diagnosis of HIV/TB infection. The differentially expressed genes and positive regulators showed certain functions such as external stimuli,signal transduction pathways in cells,migration of neutrophils,and immunological and other relevant functionalities. Meanwhile,they may also be involved in free radical-related apoptosis,inflammation,and activation pathways. Conclusions A total of 251 differentially expressed genes are found to be able to distinguish simple HIV infection from HIV/TB infection. Protein-protein interaction network of top 40 differential expression genes includes RPLP1 gene,which is possibly associated with HIV/TB co-infection and may be involved in and the positive regulation of external stimuli,signal transduction pathways in cells,migration of neutrophils,and immunological functions. These findings may provide certain evidence for the diagnosis and treatment of HIV/TB infection.
