1.Development and verification of plaque method for detection of tick-borne encephalitis virus titer

Objective To develop and verify a plaque method for detection of infectious titer of tick-borne encephalitis virus(TBEV)strain(PHKT strain for short)adapted to primary hamster kidney(PHK)cells.Methods PHK cells were infected with TBEV,a primary mouse brain adaption strain,and passed consecutively for 12 passages.The titer of PHKT was detected by plaque method(Monolayer BHK-21 cells were infected with PHKT of various passages at different dilution ratios,and the plaque number was calculated by neutral red staining)and challenge titration in mouse brain(Mice were challenged with PHKT of various passages at different dilution ratios through brain cavity,0.03 mL for each,observed continuously for 14 days,and calculated for the median lethal dose(LD50)by Reed-Muench method)respectively,and the correlation between the results of two methods was analyzed.The developed plaque method for the detection of TBEV titer was verified for specificity,repeatability and intermediate precision.Results The plaque titer of PHKT virus was up to8.9 lgPFU/mL;The correlation between the results of plaque method and mouse brain challenge titration method was good(r = 0.92);The specificity of plaque method for detecting infectious titer of PHKT virus was good,and the coefficients of variation(CVs)of repeatability and intermediate precision were both less than 5%.Conclusion A plaque method for detecting infectious titer of PHKT virus was developed,which may be used as an alternative method for challenge titration in mouse brain.
2.Clinical significance of changes of postures in ECG examination in the judgment of pathological atrioventricular block

Objective To analyze the clinical significance of ECG changes in postures in the judgment of pathological atrioventricular block.Methods 62 patients with primary atrioventricular block were diagnosed by ECG examination.According to the complication,there were 31 cases with complication and 31 cases without complication.The two groups were taken sitting and squatting (1 min squat exercise,immediately lying position) ECG examination,the ECG abnormalities of the two groups were compared.Results In the comorbidity group,among the 31 patients,sitting electrocardiogram P-R interval returned to normal in 9 cases,abnormal in 22 cases,the normal rate was 29.0% (9/31).Among the 22 cases of abnormal patients,squatting electrocardiogram P-R interval returned to normal in 1 case,abnormal in 21 cases,the normal rate was 4.5% (1/22).Sitting,squatting electrocardiogram PR interval total returned to normal rate was 32.3% (10/31).In the no complications group,among the 31 patients,sitting electrocardiogram PR interval returned to normal in 21 cases,abnormal in 10 cases,the normal rate was 67.7% (21/31).Among the 10 cases of abnormal patients,squatting electrocardiogram P-R interval returned to normnal in 4 cases,abnormal in 6 cases,the normal rate was 40.0% (4/10).Sitting,squatting electrocardiogran P-R interval total returned to normal rate was 80.7% (25/31).Conclusion ECG changes in postures can early effectively determine the pathological and functional atrioventricular block.
3.The effect of non-medical factors on the quality of life in maintenance peritoneal dialysis patients

Objective To evaluate the quality of life (QOL) in continuous ambulatory peritoneal dialysis (CAPD) patients and analyze the non-medical factors influencing QOL in order to explore the possible way to improve QOL in peritoneal dialysis (PD) patients.Methods Seventy-nine chronic renal failure patients treated with PD included in the present study.Investigated their age,sex,working status,education,medical burden and family support.Patients' QOL was evaluated by international questionnaire-KDQOL/SF-36.Patients' anxiety/depmssion status was evaluated by Hamilton depression questionnaire.Results (1)The SF-36 with its eight scales of CAPD patients were markedly lower than ordinary people (P<0.05 or<0.01).(2) Comparing with those on work,the patients without work got lower SF-36 scores[(45.78±16.93) seores vs (32.65±12.26) scores,P<0.05].Comparing with those who had medical care,the patients without medical care got much lower SF-36 scores [(49.62±13.20) scores vs (33.85±6.24) scores,P<0.05].Patients with higher income had better QOL.(3) Patients cared by their children and companion had the higheat SF-36 [(68.33±14.73) scores],while those cared by nursemaid or themselves had the lowest [(27.98±15.77) scores].(4) Among the 79 patients,43 patients (54.4%) were anxiety and 12 patients (15.2%) were depression.QOL was negatively correlated with anxiety index and depression index.Conclusion The QOL of CAPD patients are remarkably low,which is deeply influenced by many non-medical factors such as working status,medical burden,family support and emotional status.
4.Subclinical hypothyroidism and ischemic stroke

Subclinicalhypothyroidism(SCH)maybeassociatedwithcarotidatherosclerosis, dyslipidemia, hypertension, hyperhomocysteinemia, and coronary heart disease, and these factors are often regarded as the risk factors for ischemic stroke. The correlation betw een SCH and the risk of stroke remains unclear now . The hypothesis of SCH increasing the risk of stroke has not been confirmed. Even studies have show n that SCH may have a beneficial effect for the prognosis of stroke. This article review s the relationships betw een SCH and risk factors for stroke, and risk of stroke and prognosis.
5.Research progress of the new tumor suppressor gene SLC5A8

Current research suggests that SLC5A8 is a new candidate tumor-suppressor gene whose expression is reduced or deficient in human thyroid cancer,esophageal cancer,gastric cancer,colon cancer and acute myeloid leukemia.The gene is in human chromosomes 12q13-23,and encoding proteins that belongs to the members of the family of sodium/glucose transporters.It is sodium-coupled carrier that transfer short chain fatty acid,lactic acid,nicotinic acid and iodide.The exon 1 of GPG island methylation and histone acetylation can make SLC5A8 gene express silenced and promote the occurrence of tumor.of all transporter substrates of SLC5A8 expresses,butyrate,propionate and pyruvate function as histone deacetylase inhibitors and can be mediated apoptosis of many kinds of tumor cells,which may be the mechanism of inhibiting tumor.

As the target of medicines, PPAR ? agonists may be powerful drugs for epidermal wound and metabolic syndrome X.
8.Protective effect of Bu Shen Yang Xue Ming Mu Formula on oxidative injury in ARPE-19 cells

AIM: To explore the effect of Bu Shen Yang Xue Ming Mu (BSYXMM) Formula on hydroquinone-induced oxidative stress injury in ARPE-19 cells.METHODS: The oxidative injury model of ARPE-19 cell was induced by exposure to various concentrations of hydroquinone (HQ) to determine the optimal concentration.Intestinal absorption solutions of BSYXMM Formula were prepared.Effect of intestinal absorption solutions of BSYXMM Formula on the cell viability was detected by CCK-8 assay,and the percentage of apoptotic cells was measured by TUNEL assay.The levels of superoxide dismutase (SOD) and glutathione peroxidase (GSH-Px) in ARPE-19 cells were detected by means of chemical colorimetry.RESULTS: It was found that ARPE-19 cell viability significantly decreased when the concentration of HQ was higher than 90μmol/L.Compared with the model group,1% and 2% intestinal absorption solutions in the pre-treatment group could significantly alleviate HQ-induced injury (P<0.01) and 0.5% and 5% intestinal absorption solutions in the pre-treatment group could alleviate the injury in certain degree(P<0.05).While in the treatment group 1% and 2% intestinal absorption solutions could alleviate the injury to some extent (P<0.05).TUNEL results showed that the apoptosis rate decreased significantly in the pre-treatment group (P<0.01)and to some extent in the treatment group (P<0.05)compared with the model group.It was shown that both levels of SOD and GSH-Px in pre-treatment group and treatment group were markedly higher than that of model group(P<0.05),and pre-treatment group had more significant effect (P<0.01,P<0.05).CONCLUSION: BSYXMM Formula could protect against HQ-induced oxidative stress injury in ARPE-19 cells,which may be related with the increasing of antioxidant enzyme in the cells.
9.Therapeutic Effect of Entecavir Combined with Vitamin E in the Treatment of Chronic Hepatitis B Complicated with Non-alcoholic Fatty Liver

Objective To explore the clinical efficacy of entecavir combined with Vitamin E in treating patients with Chronic hepatitis B complicated with non-alcoholic fatty liver.Methods Retrospective analysis was done by reviewing the clinical data of 103 patients, who suffered with chronic hepatitis B complicated with non-alcoholic fatty liver and treated in our hospital from 2015 to 2016.The patients were divided into the control group with 61 cases and the observation group with 42 cases based on different therapies.The control group was treated with only entecavir while the observation group was treated with entecavir and Vitamin E, and both the courses of treatment lasted for 6 months.The negative conversion ratio of HBV-DNA, levels of ALT, TBIL, ALP, the disease state and the adverse reactions of the two groups were compared before and after treatment.Results The negative conversion ratio of HBV-DNA of the observation group was obviously higher than that of the control group, and the patients of the observation group showed significant improvement.Meanwhile, the levels of ALT, TBIL and ALP in the two groups were both lower than before treatment.Compared with the control group, those indexes in the observation group were significantly lower (P<0.05).Conclusion Entecavir combined with Vitamin E for the treatment of chronic hepatitis B complicated with non-alcoholic fatty liver could significantly decrease the viral infections so that the state of patients can be improved.
