Induced pluripotent stem cells(iPS)are derived from the differentiated somatic cells that regain the differentiation capabilities by the direct nuclear reprogramming with the exogenous factors.The methods for iPS induction evolved from transcription factors to RNA binding proteins,small molecules,and extracellular signals with the efforts in improving biosafty.The cellular and epigenetic characterization for iPS generation is a progressive and time-dependent process,which is tightly associated with the cellular differentiation status.However,epigenetics of iPS is not same as the embryonic stem cells.In combination with gene therapy and cellular transplantation therapy,the achievements of iPS had been applied in animal disease model treatment.

Objective To explore the diagnosis and treatment of liver trauma.Methods A retrospectively analysis of clinical data of 160 cases of liver trauma, including 96 cases of blunt trauma and 64 cases of open trauma, was made.Results Among the 160 cases, one case with stage IV trauma and severe thoracic injury died 30min after admission. Among the other 159 cases, 45cases (9 cases of stage Ⅰ, 31 cases of stage Ⅱ and 5cases of cases Ⅲ) received nonoperative treatment, and 114cases (7 cases of stage Ⅱ, 67 cases of stage Ⅲ and 50 cases of stage Ⅳ) received operative therapy. In non-operative treatment group, the cure rate was 100 %(45/45). In operation treatment group the cure rate was 98.2 %(112/114), the mortality rate was 1.8 %(2/114), 1 case died of hemorrhage after operation and 1 case died of ARDS. Five patients with postoperative complications were cured, including hepatic abscess in 2 cases, biliary fistula in 2 cases, and hemorrhage after operation in 1 case. Conclusions B-ultrasound is the method of choice for diagnosis of liver trauma, and CT can be used if conditions permit. Cases of lives truma of stages Ⅰ and Ⅱ and some cases of stage Ⅲ that have stable hemodynamics, can receive nonoperative treatment under close observation. Some cases of stage Ⅱ, most cases of stage Ⅲ and all cases of stages Ⅳ to Ⅵ injury should preferably undergo surgical treatment.

Objective To study the effect and mechanism of Shenshaoruangan Decoction on liver fibrosis in rats.Methods Seventy-two male Wistar rats were selected and normally fed for 1 week.Then they divided into the control group(n=12) and model constructing group(n=60).The model constructing group was subcutaneously and intraperitoneally injected by 40％ CCl4 olive oil for constructing fibrosis rat model.After successfully constructing model,which was divided into the model group(equivalent normal saline gavage),positive control group(colchicines 0.154 mg · kg-1 · d-1),low-dose group(2 mL per 100 g body mass,containing Shenshaoruangan Decoction crude drug 0.23 g/mL),medium-dose group(2 mL per 100 g body mass,containing Shenshaoruangan Decoction crude drug 0.46 g/mL) and high-dose group(100 g/2 mL,containing Shenshaoruangan Decoction crude drug 0.69 g/mL),12 cases in each group.The differences of related indicators rat hepatic fibrosis after 8-weeks treatment were observed.Results The control group was the stage 0 standard,the liver fibrosis degree in the positive control group was significantly better than that in the model group(P＜0.05);the liver fibrosis degree in the low-dose group,medium-dose group and high-dose group was gradually alleviated;the liver fibrosis degree in the high-dose group was significantly slighter than that in the model group and-dose group(P＜0.05).The detection values of PC-Ⅲ,LN,Ⅳ-C and HA in the model group were significantly higher than those in the control group(P＜0.05),and the detection values of PC-Ⅲ,LN,Ⅳ-C and HA in the positive control group,lowdose group,medium-dose group and high-dose group were significantly lower than those in the model group(P＜0.05).Serum SOD detection value of the model group was significantly lower than that in the control group(P＜0.05),the serum MDA level in the model group was significantly higher than that in the control group(P＜0.05);serum SOD level in the positive control group,low dose group,medium-dose group and high-dose group was significantly higher than that in the model group(P＜0.05),while serum MDA level in those group was significantly lower than that in the model group(P＜0.05).Conclusion Shenshaoruangan Decoction can effectively alleviate the liver fibrosis degree,and its mechanism is to reduce the levels of liver fibrosis indicators,inhibit the prodmotion of peroxides and alleviate the damage of free radicals on liver cells.

Objective:To establish and validate a method for the determination of particle size and size distribution of gefitinib. Methods:A Malvern Mastersizer 3000 laser size analyzer and a Hydro LV wet autosampler were used,a light scattering method for the determination of particle size and size distribution was adopted to analyze the particle size of gefitinib,and then the methodological study was carried out. The pump speed was 2 000 r·min -1 ,the obscuration was 8% -20% ,the measurement time of background and sample was 10 s,the sample RI was 1. 500,and the sample absorbance was 0. 00. Results:The D10 of 3 batches of gefitinib was less than 7 μm,the D50 was less than 15 μm and the D90 was less than 25 μm,which all met the preparation requirements of final products. Conclusion:The method is accurate,simple and repeatable,and suitable for the particle size analysis of gefitinib.

Objective To investigate the therapeutic efficacy of combining Kangfuxin solution with compound Bingpengsan on patients with diabetes mellitus complicated with pressure sores. Methods Fifty-two diabetes mellitus patients complicated with pressure sores admitted in Department of Emergency in the PLA 155th Central Hospital were divided into observation group and control group by lot method,26 cases in each group. Observation group was treated by combining Kangfuxin solution with compound Bingpengsan,the control group was given Mepliex application therapy,they all had change of dressing once a day. After treatment for 20 days,the therapeutic efficacy of pressure sore,healing time and frequency of changing dressing were observed. Results The total effective rate in observation group was significantly higher than that of control group〔96.1%(25/26)vs. 80.6%(21/26),P<0.05〕, in the observation group,the pressure sore healing time was significantly shorter than that of the control group(day:Ⅱstage:9.5±1.7 vs. 13.0±2.1,Ⅲstage:13.1±3.1 vs. 18.1±5.1,Ⅳstage:15.3±3.7 vs. 19.6±5.9,all P<0.05)and the number of times of changing dressing was significantly reduced compared with that of control group (times:Ⅱ stage:16.39±1.89 vs. 19.32±2.26,Ⅲ stage:19.56±2.52 vs. 22.36±2.69,Ⅳ stage:23.54±2.86 vs. 26.47±3.96,all P<0.05). Conclusion The Kangfuxin solution combined with compound Bingpengsan for treatment of deep pressure ulcers in patients with diabetes mellitus has significant effect,its cure rate is relatively high,the pressure sore healing time is reduced and the patients' suffering is alleviated.

Objective To investigate the relationship between waist-to-height ratio and metabolic syndrome,in order to identify the optimal cut-off point of waist-to-height ratio for predicting metabolic syndrome.Methods In this cross-sectional study,we recruited 343 people who received physical examination in First Hospital of Quanzhou between January 2012 and June 2014,and collected the information of their waist circumference,height,weight,blood pressure,laboratory test results (including fasting blood glucose,2-hour glucose after oral glucose tolerance test,triglyceride,high-density lipoprotein cholesterol) and visceral fat area assessed by computed tomography.Then a receiver operating characteristic (ROC) curve analysis was used to estimate the optimal cut-off points of waist-to-height ratio for the prediction of metabolic syndrome.Results Among the 343 people,there were 195 metabolic syndrome patients,the prevalence rate was 56.8％,which was 70.2％ in men (127/181) and 42.0％ in women (68/162).In ROC curve analysis,the area under the curve of waist-to-height ratio for the prediction of metabolic syndrome was 0.664 for men,and 0.673 for women.The optimal cut-off point of waist-to-height ratio for predicting metabolic syndrome was 0.543 0 (sensitivity 88.2％,specificity 44.4％) for men,and 0.568 3 (sensitivity 86.8％,specificity 46.8％).Conclusion The optimal cut-off point of waist-to-height ratio for predicting metabolic syndrome in Quanzhou population is 0.543 0 for men and 0.568 3 for women.

Objective To provide a scientific basis for nursing managers to promote the professional quality of life of nurses.Methods Totally 373 nurses from three general hospitals in Jinan by convenience sampling technique were recruited and assessed with the Professional Quality of Life Scale Version 5 (ProQOL-V) and the Five Facet Mindfulness Questionnaire.The relationship between professional quality of life and mindfulness was analyzed.Results The average score of compassion satisfaction was (29.71±6.61) points,the average score of compassion fatigue was (52.82±8.04) points,which were in the middle level of them.Compassion satisfaction was correlated with mindfulness,observation,description,non-reactiveness and non-judgment (P ＜ 0.05).Compassion fatigue was correlated with mindfulness,observation,awareness of the action,non-reactiveness and non-judgment (P ＜ 0.05).Multiple regression analysis showed that the three dimensions (observation,non-reactiveness and non-judgment) of mindfulness could predict 15％ variance of compassion satisfaction and the three dimensions (observation,awareness of the action and non-judgment) of mindfulness could predict 34.1％ variance of compassion fatigue after controlling demographic variables.Conclusions Mindfulness can effectively predict nurses' professional quality of life.Nursing managers should take some mindfulness training in order to improve their professional quality of life.

Objective To explore the therapeutic strategy and clinical value of intraoperative chole-dochoscopy and electrohydraulic lithotripsy for refractory intrahepatic bile duct stones.Methods Liver pa-renchyma,intrahepatic bile duct and bile duct stones were explored under direct vision and intraoperative choledochoscope in 1 1 cases of refractory intrahepatic bile duct stones.Electrohydraulic lithotripsy and lithot-omy were performed to remove the stones and protect the liver parenchyma.If the stones could not be re-moved once,a secondary lithotripsy and lithotomy was performed through the fistula tract.Results All re-fractory calculi were crushed after one or two procedures and the clearance rate were 100%.No complica-tions occurred.Ten patients were followed up from 1 to 3 years except one.Three cases revealed recurrent stone during follow-up due to withdrawal of ursodeoxycholic acid capsules in 1 to 2 years.Seven others showed no stone recurrence within follow-up time.Conclusion Intraoperative choledochoscopy and electro-hydraulic lithotripsy is an easy technique and can effectively protect the liver parenchyma.The life quality of patients can be improved with low surgical risk and postoperative complications.

Objective To investigate toe-brachial index ( TBI) in the diagnosis of peripheral artery disease ( PAD) and its risk factors in the patients with type 2 diabetes.Methods TBI was examined in the 238 patients with type 2 diabetes.The patients were divided into the group with low TBI ( TBI≤0.7 ) and the group with normal ABI ( TBI>0.7 ) .The two groups were compared for clinical parameters.Results Thirty two patients (13.4%)with abnormal ABI (TBI≤0.7) showed older age [(63.8 ±9.9) yrs vs (54.9 ±10.8) yrs, P =0.000] , lower diastolic blood pressure [(70.5 ±6.9) mmHg vs (74.9 ±9.1) mmHg, P =0.003], more frequency of hypertension (56.3%vs 38.3%, P =0.043), coronary artery heart disease (28.7%vs 10.7%, P =0.020) and cere-bral vascular disease (15.6%vs 4.4%, P =0.025).Step-wise analysis screened that age and diastolic blood pressure were the inde-pendent risk factors for TBI .Conclusions Aging and hypertension were the risk factors in the patients with abnormal TBI in type 2 di-abetes.TBI was an easy and economical method for diagnosing PAD in patients with type 2 diabetes.

Objective To evaluate the value of three-dimensional color power angiography(3D-CPA) in the differential diagnosis of benign and malignant breast tumors.Methods 3D-CPA vascular parameters were collected and analyzed in 121 breast solid lesions (63 malignant,58 benign) in the 114 female patients.The diagnostic performances of each parameter were compared by receiver operating characteristic (ROC)cure.Results The parameters of intra-tumor flow index (FI1),flow index of shells with outside thickness of 3mm surrounding the breast tumors(FI2 ) and the ratio of FI2 and FI1 (G2) in malignant group were significantly higher than those in benign group( P ＜0.05,respectively).Analysis of the areas under ROC curve showed that the areas under FI2 curve was the largest.No significant difference( P ＞0.05) was found between the area under FI2 curve and the area under G2 curve.Comparing the area under FI2 curve to that under FI1,there was significant differences ( P ＜ 0.05).Taking FI2 ≥ 27.385 as the critical value,the sensitivity and specificity for the diagnosis of malignant breast tumors were 88.9％ and 81.0％,and the positive and negative predictive values were 83.6％ and 87.0％ respectively.Conclusions The sensitivity and specificity of 3D-CPA parameter FI2 in the diagnosis of malignant breast tumors were higher than others,FI2 with the optimal critical value of ≥27.385 could help to identify malignant from benign breast tumors.
