1.Expression and clinical significance of telomerase and hTERT in ovarian neoplasm tissues

Objective To discuss the expression and clinical significance of telomerase and hTERT in ovarian neoplasm tissues . Methods The activities of telomerase and the expression levels of hTERT mRNA were detected in the tissues of malignant ovarian neoplasm ,benign ovarian neoplasm and normal ovarian respectively ,and the results were analyzed .Results Comparing with the tis‐sues of benign ovarian neoplasm and normal ovarian ,the activity of telomerase and the expression level of hTERT mRNA increased in malignant ovarian neoplasm tissues ,and the differences were statistically significant(P< 0 .05) .Conclusion The telomerase plays an important role in the occurrence and development of malignant ovarian neoplasm .The detection of telomerase activity can help in early diagnosis of malignant ovarian neoplasm .
2.Monitoring of Drug-resistance in Mycoplasma Isolated from Female Genitourinary Tract

OBJECTIVE To investigate the infection status and drug resistance of Ureaplasma urealyticum (Uu) and Mycoplasma hominis (Mh) isolated from female genitourinary tract. METHODS The specimens of 1890 cases were collected from Jan 2003 to Dec 2006. The results of mycoplasma cultivation and susceptibility test were analyzed retrospectively. RESULTS Among 1890 cases, there were 756 cases with positive mycoplasma cultivation. Positive rate was 40.0%. Positive rate was the highest at the age of 21 to 40. Among 11 commonly used antibiotics, sensitivity of tetracyclines was the highest,followed by macrolides. Four quinolones all showed the higher resistance. CONCLUSIONS The resistance of commonly used antibiotics in mycoplasma isolated from female genitourinary tract is greatly severe.We should think highly of it.
3.Pathogen Distribution and Drug Resistance in Infection of Operative Incision

OBJECTIVE To investigate pathogen distribution and drug resistance in infection of operative incision and provide the reference for clinical prevention and cure.METHODS Referring to National Rules of Operation in Clinical Laboratory,strains were isolated and identified,and drug-sensitivity test was performed by DADE BEHRING panel.RESULTS Among total 140 strains of pathogenic bacteria,the most were Gram-negative bacilli that accounted for 58.6%(82 strains),followed by Gram-positive cocci that accounted for 41.4%(58 strains).Staphylococcus aureus and Pseudomonas aeruginosa had the higher detection rate.The former was 29.3% and the latter was 27.1%.Among the total S.aureus,meticillin-resistant S.aureus(MRSA) accounted for 31.7%.Detection rate of extended-spectrum beta-lactamases(ESBLs) producing Escherichia coli and Klebsiella pneumoniae was 40.9%.CONCLUSIONS Effective measurement should be taken to prevent and control infection of operative incision.
4.Application value of combined detection of AFP-L3,GP73 and GPC3 in diagnosing primary hepatic cancer

Objective To study the clinical value of the combination detection of serum tumor markers alpha fetal protein hetero-geneity-L3(AFP-L3) ,Golgi protein 73(GP73) and phosphatidylinositol proteoglycan-3(GPC-3) in the diagnosis of primary hepatic cancer .Methods The serum AFP-L3 ,GP73 and GPC-3 levels were measured in 34 patients with primary hepatic cancer (PHC) ,20 patients with liver cirrhosis ,20 patients with chronic hepatitis B ,37 patients with other tumors by the enzyme linked immunosor-bent assay(ELISA) .Meanwhile ,20 individuals with healthy physical examination were selected as the control group .The expression situation of various indexes were compared and analyzed .Results The expression levels of AFP-L3 ,GP73 and GPC-3 in the PHC group were significantly higher than those in the control group and the other tumors groups ,difference was statistically significant (P<0 .05) .The areas under the receiver operating characteristic (ROC) curve of PHC in the single item detection of 3 markers were 0 .909 ,0 .832 and 0 .817 respectively .The areas of the ROC curve in the combination examinations of two items were 0 .935 , 0 .945 and 0 .912 respectively .The area of the ROC curve in the 3-item combined detection of PHC could be up to 0 .960 .Conclusion The combined detection of AFP-L3 ,GP73 and GPC-3 can increase the detective rate of PHC and has important clinical significance to early diagnosis of PHC .
5.Clinical value of alpha-fetoprotein heterogeneity 3, golgin 73 and glypican 3 in diagnosis of primary hepatic cancer

Objective To investigate the clinical value of serum tumor marker alpha-fetoprotein heterogeneity 3 (AFP-L3),golgin 73 (GP73) and glypican 3 (GPC-3) in primary hepatic cancer by ROC curve.Methods The AFP-L3,GP73,GPC-3 levels in serum were detected in 34 patients of primary hepatic cancer by enzyme immunoassay.Meanwhile,same terms from 20 healthy volunteers were detected as normal group.The area under curve was made by SPSS 17.0.Each laboratory indicator levels were compared and analyzed.Results The AFP-L3,GP73,GPC-3 levels in primary hepatic cancer group were significantly higher than those in normal control [(1890.13±506.47) ng/L vs (623.40±317.89) ng/L,(219.53±136.33) ng/ml vs (56.40± 25.63) ng/ml,(14.28±7.15) μg/L vs (7.33±3.71) μg/L,P < 0.01].The areas under the concentration-time curve of receiver operating characteristic of single tumor marker were 0.909,0.832,0.817,the areas of AFP-L3+ GP73,AFP-L3+GPC-3,GP73+GPC-3 were 0.935,0.945,0.912,the area of combined detection of primaryhepatic cancer was 0.96.Conclusion AFP-L3 combines with GP73 and GPC-3 can increase the positive rate in patients with primary hepatic cancer and has important clinical significance.
6.In vivo confocal microscopic observations of 49 patients with posterior polymorphous corneal dystrophy

AIM: To explore the imaging features of 49 patients with posterior polymorphous corneal dystrophy(PPCD)by in vivo confocal microscopy(IVCM).METHODS: Retrospective case series study. A total of 49 patients(86 eyes), including 32 males and 17 females diagnosed with PPCD between January 2013 and January 2021 were collected. The mean age was 42.5±22.9 years. All patients were scanned by IVCM to analyze the density of corneal endothelial cells and described IVCM characteristics of different types of PPCD.RESULTS: The number of endothelial cells in the lesion area of all patients was lower than that in the peripheral area. Under IVCM, 44 eyes(51%)were categorized into type 1 PPCD(vesicular lesions), characterized by single or multiple, central round or irregular crater-like lesion on paracentral corneal endothelial layer; 16 eyes(19%)were categorized into type 2 PPCD(band lesions), which displayed curved and raised edge with scattered or banded-distributed gutta-like lesion between edges. Type 3 PPCD(diffuse lesion)were in 26 eyes(30%), which showed that endothelial cells were missing in many areas. The blurred images of endothelium in most areas featured with spikes lined in a streak, and the clear images in some areas featured with a band lesions. Two patients were followed up for 4-5a. The IVCM images showed different lesions, including the decrease of central corneal endothelial cell density and the iron deposit in the corneal epithelium, etc.CONCLUSION: IVCM is able to scan the characteristic microstructural alterations at the level of endothelium and Descemet membrane in patients with PPCD, and provide an effective image diagnosis for PPCD.
